Binding of environmental carcinogens to asbestos and mineral fibres.

A rapid method has been developed for measuring the binding capacity of asbestos and other mineral fibres for environmental carcinogens. Benzo(alpha)pyrene (B(alpha)P), nitrosonornicotine (NNN), and N-acetyl-2-aminofluorene (NAAF) were assayed in the presence of Canadian grade 4T30 chrysotile, chrysotile A, amosite, crocidolite, glass microfibres, glasswool, attapulgite, and titanium dioxide. Chrysotile binds significantly more carcinogens than the other mineral fibres. This binding assay is reproducible with coefficients of variation of less than 8% and 6% respectively for inter and intra assay. The influence of pH was also studied, and there is good correlation between the carcinogen binding and the charge of the tested mineral fibres. The in vitro cytotoxicity on macrophage like cell line P388D1 and the haemolytic activity of various mineral fibres were also measured; a good correlation was found between the binding capacity and the cytotoxicity of tested mineral fibres on P388D1 cells. These results give some explanations for the reported synergism between exposure to asbestos and the smoking habits of workers.

A baby has died: the impact of perinatal loss on family social networks.

OBJECTIVES: To describe the impact of a baby's death on the family's social network and to design nursing interventions to support families and their networks. DESIGN: Descriptive, with a qualitative approach. SETTING: An urban area of western Quebec. PARTICIPANTS: Twenty parents (mothers and fathers) who had experienced a perinatal loss (abortion, miscarriage, in-utero death, stillbirth, or death of a newborn within the 1st week of life) within the last 6 years. MAIN OUTCOME MEASURES: Self-administered questionnaires developed by the authors were completed by each parent. RESULTS: Family members' quality and quantity of ties with their network were profoundly affected by the perinatal loss. Some families experienced reinforcement of their bonds with their social network, but most suffered permanent losses of relationships with friends, colleagues, or extended family members. CONCLUSIONS: The quality and quantity of ties with one's network are associated with improved health status and life satisfaction. Considering the changes participants noted in their relationships within their network, further studies of the impact of these changes on family members' grieving process would be useful to guide nursing interventions.

Coupling a preactivated daunorubicin derivative to antibody. A new approach.

Many procedures have been reported for coupling anthracycline drugs to antibody for drug targeting. But they either yield conjugates with lower pharmacological activity or antibodies which have lost much of their immunological specificity. The use of glutaraldehyde for coupling saves both pharmacological and immunological activities but it causes a considerable polymerization of the antibody which is undesirable for in vivo use. We report a new coupling procedure which uses an activated daunorubicin derivative which is later added to the antibody. Using this procedure for daunorubicin coupling to monoclonal anti-CEA we found no significant polymerization of the conjugate and a full recovery of the pharmacological activity as tested in vitro on CEA producing human colon adenocarcinoma cells. The activated drug was found stable for one week at 25 degrees C and the whole coupling procedure is highly reproducible.
