Quantification and localization of galactose-specific binding sites in rat liver during postnatal development.

We investigated the number and distribution of galactose-specific binding sites in developing livers from suckling rats of various ages using Lac-BSA-Au5 (lactosylated bovine serum albumin adsorbed onto colloidal gold particles 5 nm in diameter) as electron-dense ligand, and performing transmission electron microscopy of the specimen. It has been reported that the number of galactose-specific binding sites increases rapidly during organ development post partum (p.p.) and this was ascribed to hepatocyte receptor increase only. We now have investigated in in situ and in vitro experiments whether the binding sites of identical sugar specificity but located on sinusoidal cells show the same increase in expression or are independently regulated. We therefore quantified the number of particles bound by isolated hepatocytes and liver macrophages and found a gradual increase of both binding activities with age, the binding levels of adult liver cells being reached at day 15 p.p. This was confirmed with experiments using in situ prefixed organs thus proving the validity of this finding also for the intact organ. In both sets of experiments--in vitro as well as in vivo--ligand was found binding statistically distributed as single particles on hepatocytes of all ages, whereas on liver macrophages the binding pattern changed during development. On liver macrophages from rats 15 days of age ligand binding occurs in the preclustered pattern described for macrophages from adult rat livers whereas liver macrophages of newborn rats express a different binding pattern: they bind the ligands mostly as single particles with only few and small microaggregates.(ABSTRACT TRUNCATED AT 250 WORDS)

Hormonal control of cholesterogenesis and related enzymes in isolated rat hepatocytes during pre- and postnatal development.

The effect of glucagon and insulin on the incorporation of 1-14C-acetate into cholesterol and fatty acids and on the enzymes involved in the first steps of cholesterol synthesis (3-hydroxy-3-methyl-glutaryl-coenzyme A reductase, 3-hydroxy-3-methyl-glutaryl-coenzyme A synthase, and acetoacetyl-coenzyme A thiolase) was investigated. Isolated rat hepatocytes at different stages of fetal and postnatal development were employed. Data obtained show the appearance of hormonal control on the 18th day of fetal life, indicating the same pattern, as regards acetate incorporation and HMGCoA reductase prepared and assayed in the presence of NaF. On the contrary, HMGCoA reductase, prepared without NaF, HMGCoA synthase, and acetoacetyl CoA thiolase, does not appear to respond to hormonal stimulation. In the perinatal period, the hormonal effect is no longer detectable, probably because of a hormone resistance of this metabolic pathway.

Cholesterogenesis and related enzymes in isolated rat hepatocytes during pre- and postnatal life.

Cholesterogenesis pathway during pre- and postnatal development was studied in isolated rat hepatocytes. No modified activity of cytosol acetoacetyl coenzyme A (CoA), thiolase, or 3-hydroxy-3-methylglutaryl CoA (HMGCoA) synthase was detectable at the different stages examined. Minimal levels of 1(14)C-acetate incorporation into cholesterol and HMGCoA reductase activity were present at 16 days of fetal development in newborn and suckling rats, whereas both parameters increased rapidly before birth. The pattern of NaF nonsuppressible reductase activity showed a different activation state of the enzyme, suggesting the appearance of a modulation state, probably related to the development of some short-term regulatory mechanisms.

Electron microscopic evidence for the presence of an asialoglycoprotein receptor on isolated foetal rat hepatocyte surface.

The ontogeny of asialoglycoprotein receptor was investigated by electron microscopic cytochemistry in hepatocytes isolated from foetal and adult rat. The binding capacity for asialofetuin coupled to horseradish peroxidase was lacking before the 18th day of intrauterine life; it arises at this time and increases with developmental age. The ligand-receptor complexes form small patches. The distribution pattern of positivity is very similar in pre and postnatal age, covering the entire cell surface. These results indicate a rather late appearance of the galactose-binding capacity related to the asialoglycoprotein clearance function, which is typical of adult mammalian liver.

Reflux esophagitis: radiologic-endoscopic correlation in 39 symptomatic cases.

Thirty-nine consecutive patients with symptoms suggestive of reflux esophagitis underwent a double contrast upper gastrointestinal series and subsequently had endoscopy with biopsy. In a control group of 164 consecutive patients without symptoms of esophagitis a double contrast examination was done with the same method. We have found a significant increase of the diameter of the esophagus in its distal or cardiac segment (IDCE) in patients with esophagitis of Grades 1 and 2 when compared with the control group (p less than 0.001). Radiology was found to have correctly diagnosed 35 of the 39 cases (89.7%) and the majority of the patients had endoscopic signs of mild esophagitis.

Small benign tumors of the esophagus: radiological diagnosis with double-contrast examination.

Benign tumors of the esophagus are rare. So far radiological examination has been useful in the diagnosis of fairly large benign tumors. In 4100 consecutive double-contrast studies we have found 22 incidental cases of protruded lesions of the esophagus with the appearance of submucosal tumor (8 cases) or polyp of the mucosa (14 cases). The lesions were small: 14 were less than 1 cm and 8 were between 1 and 3 cm. The histological data showed leiomyoma in the submucosal tumors and squamous papillomas in the polyps of the mucosa. A noteworthy feature is the unusual frequency of small squamous papillomas detected. The radiological diagnosis of benign tumors of the esophagus cannot be confined to large tumors in symptomatic patients but may include the accidental detection of small tumors. These call for histological verification and possibly endoscopic removal.

Isolated rat hepatocytes as a tool for the determination of antibodies against human liver cell membrane.

Antibodies against liver cell membrane are measured by counting the percentages of fluorescent rat hepatocytes ( FRH ), obtained by an indirect immunofluorescence method after incubation of isolated rat hepatocytes with sera of patients with chronic liver diseases. A close relationship exists between the percentages of FRH and the serological and histological parameters of diseases activity, there was no difference between HBsAg-positive or -negative sera.

Insulin binding and internalization in rat hepatocytes during prenatal and postnatal life.

Insulin binding to isolated rat hepatocytes was studied during prenatal and postnatal life. Results show that in hepatocytes isolated from prenatal, postnatal and adult rat there is a constant increase in the number of insulin binding sites per cell, whereas the affinity of plasma membrane receptors for the hormonal ligand remains unaltered from prenatal to adult hepatocytes. Autoradiographic studies indicate a greater internalization of hormone during prenatal life and, taking into account the increase of cell size, suggest an unchanged surface density of receptor sites before and after birth.

[Problems of gastrointestinal angiography].

The subject of abdominal angiography is discussed, for informational rather than technical reasons, on the basis of seven years personal experience. The method used to prepare the patient for angiography and emergency treatment of the various episodes of reaction to the contrast medium or of complications of arterial catheterism are illustrated and some of the most significant aspects of abdominal angiographic symptomatology described.
