Stimulation by thyroid hormone of Na+-H+ exchange activity in cultured opossum kidney cells.

Cultured opossum kidney (OK) cells were used to determine whether thyroid hormone has a direct stimulatory effect on Na(+)-H+ exchange activity in an intact cellular preparation. Na+ uptake or intracellular pH recovery was measured in confluent monolayer cells following acid loading with NH4Cl. Triiodo-L-thyronine (T3) had no effect on cell number or protein and DNA contents but stimulated amiloride-sensitive Na+ uptake in a dose- and time-dependent manner. Maximal stimulation of Na+ uptake was observed at 10(-7) M T3 and 10(-6) M L-thyroxine (T4) with half-maximal effects at 10(-9) M T3 and 3 x 10(-8) M T4. The T3 specific binding capacity of OK cells was 96 +/- 15 fmol/mg DNA with a KD of 1.1 +/- 0.2 x 10(-9) M T3. Neither T3 nor T4 had any effect on amiloride-insensitive Na+ uptake. In kinetic studies of Na+ uptake, T3 increased the Vmax from 123 +/- 22 to 157 +/- 24 nmol.mg-1.min-1 without changing the Michaelis-Menten kinetics (Km) for Na+ (21 +/- 1 in control and 22 +/- 4 mM in T3-treated cells). Studies of intracellular pH (pHi) showed that the resting pHi and the buffering capacity were unaffected by T3. However, after an acid load, OK cells treated with T3 exhibited a greater rate of Na(+)-dependent pH recovery than untreated control cells. These results indicate that thyroid hormone can stimulate Na(+)-H+ exchange activity directly in renal cell line without apparent changes in pHi, cellular hypertrophy, or hyperplasia.

Dopamine stimulation of cAMP production in cultured opossum kidney cells.

Dopamine receptors have been identified in many tissues including the kidney. To establish an in vitro system as a model for dopamine action, we studied the effect of dopamine (DA) receptor agonists and antagonists on adenosine 3',5'-cyclic monophosphate (cAMP) formation in opossum kidney (OK) cells. The stimulation of cAMP production in these cells by dopamine was dose dependent, and markedly higher levels were observed in the presence of dopamine plus a phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine. Half-maximal stimulation was found with 1.15 +/- 0.22 microM dopamine. A DA1-receptor agonist, SKF 82526J, stimulated cAMP production, whereas a DA2-receptor agonist, Ly 171555, did not. The stimulatory effects of dopamine and SKF 82526J were abolished by a specific DA1-receptor antagonist, Sch 23390 with half-maximal inhibition concentrations of 1.24 +/- 0.18 and 4.0 +/- 0.5 nM, respectively. In contrast, the DA2-receptor antagonist, spiperone, had no inhibitory effect on dopamine- and SKF 82526J-stimulated cAMP production. Beta-Adrenergic antagonists failed to attenuate the stimulatory effects of dopamine and SKF 82526J on cAMP production. In addition, the beta-adrenergic receptor agonist, isoproterenol, did not stimulate cAMP production. These results suggest that the action of dopamine was not mediated through beta-adrenergic receptors. Furthermore, our results clearly demonstrated the existence of DA1-receptors linked to adenylate cyclase in OK cells.

Downregulation of parathyroid hormone receptors in renal membranes from aged rats.

The mechanism of the inhibition or blunting of parathyroid hormone (PTH)-stimulated Na(+)-Ca2+ exchange activity in renal cortical cells from aged rats was examined. The number of PTH binding sites in basolateral membranes prepared from adult (6 mo) and old (24 mo) rats was quantitated by the binding of the synthetic analogue 125I-labeled [Nle8,18, Tyr34]bPTH-(1-34) amide to the membrane. The maximum number of specific PTH binding sites, Bmax, was 92.7 +/- 9.3 and 36.7 +/- 6.1 fmol/mg protein, respectively, in membranes prepared from adult and old rats. The affinity of the receptor to PTH was unaffected with age. The level of PTH binding components (68 and 70 kDa) estimated by a ligand affinity blot technique using biotinylated bPTH-(1-34) as the ligand was similarly reduced in membranes isolated from senescent rats. To test the hypothesis that change in the number of PTH binding sites and level of PTH binding components represented an adaptive response to a high serum PTH level, rats were parathyroidectomized (PTX) and the changes were reexamined. Decreases in the number of PTH binding sites and PTH binding components were either partially or completely negated by the surgery. These findings suggest that the blunting of both the PTH-stimulated Na(+)-Ca2+ exchange and adenylate cyclase activities in the kidneys of aged rats was due, in part, to be loss of PTH receptors in basolateral membranes and that this defect could be partially reversed by removal of the parathyroid gland.

Regulation of renal sodium calcium exchange by PTH: alteration with age.

Parathyroid hormone, when incubated with renal cells acting in vivo and in vitro, increased Na+/Ca2+ exchange activity. The effect of parathyroid hormone was specific for biologically active analogs and could be mimicked by cAMP and forskolin. Parathyroid hormone-sensitive Na+/Ca2+ exchange activity was markedly blunted in cells from senescent rats. Parathyroid hormone-stimulated adenylate cyclase was also decreased in aging. In contrast, forskolin-stimulated Na(+)-dependent Ca2+ efflux and adenylate cyclase did not change with senescence. Decrease of PTH binding sites was observed in cells from old rats. Further, cells from 24-month-old rats had decreased Gs and Gi proteins, as detected by ADP-ribosylation. Since serum iPTH level was elevated in the old rat and could contribute to the desensitization to PTH, we tested this hypothesis by comparing sham-operated and PTX animals. The decreases in PTH-sensitive Na+/Ca2+ exchange activity and adenylate cyclase activity in cells from 24-month-old rats could be completely negated by parathyroidectomy. Decrease in PTH binding sites and contents of Gs and Gi in cells from aged-rats was partially negated by the surgery. In conclusion, our results suggested that the age related blunting in responses of renal cells to PTH was due, at least in part, to the elevated serum iPTH level in old rats.

Early effects of subtotal nephrectomy and food restriction on urinary calcium in the rat.

Urinary calcium excretion (UCa) is normal several weeks following subtotal nephrectomy (Nx) in the rat, despite factors such as magnified solute load per nephron which should promote hypercalciuria. To define the levels of UCa during the first few weeks after Nx and to determine if the development of compensatory mechanisms governing renal conservation of calcium are detectable, we measured UCa in Nx and sham Nx rats with matched food intake (10 g/day) from 10 days before through 20 days following Nx. In the sham Nx animals, UCa rose during the first 5 days following surgery from 27 +/- 8 to 66 +/- 5 microM/day and then plateaued for the remaining time. In Nx rats, UCa also rose during the first 5 postoperative days from 23 +/- 5 to 110 +/- 10 microM/day (Nx vs. sham Nx, p less than 0.05), but then fell over the next 3 days to levels observed simultaneously in sham Nx animals. The rise in UCa after Nx, but not after sham Nx, was associated with a doubling of urine flow rate and increased urinary titratable acid excretion. In parathyroidectomized rats, UCa also rose following Nx; however, maximum UCa was then sustained for at least 4 days. In an additional sham Nx group fed 20 g/day, no increase in UCa occurred following surgery. Thus, hypercalciuria is present following Nx in the rat, in part possibly attributable to increased acid excretion. The transient nature of calciuria reflects an adaptive phenomenon, most likely hyperparathyroidism. Diminished food intake following surgery independently contributes to hypercalciuria, regardless of the status of renal mass.

[Role of "qi" in reaching affected area using acupuncture in "promoting blood circulation to remove blood stasis"].

In the present work, an experimental observation was carried out with the determination method of bioelectric impedance in 78 cases of chronic hepatitis, 58 cases of chronic obstructive pulmonary disease, 45 of obstructive thromboendarteritis and 65 of coronary heart disease, who were diagnosed definitely and possessed the symptoms and signs of blood stasis and produced change in hemodynamics. Yanglingquan (G. B. 34) and Zhangmen (Liv.13) acupoints on right side were needled and four indices of liver blood circulation were determined for chronic hepatitis. Bilateral Kongzui (Lu.6) acupoints were needled and seven indices of lung blood circulation were detected for chronic obstructive pulmonary disease. Acupoint along the pathway of channel were needled and six indices of blood circulation in the extremities were examined for obstructive thromboendarteritis. Bilateral Neiguan (P.6) acupoints were needled and four indices of the cardiovascular function were detected for coronary heart disease. Results showed 95 cases of the above four diseases having Qi reaching to affected area. 84 cases had propagated sensation along the channel. 67 cases had local sensation. Experimental results indicated that Qi reaching affected area produced obvious effects of dredging the meridian passage, and promoting blood circulation to remove blood stasis.

Transcriptional activator nuclear factor I stimulates the replication of SV40 minichromosomes in vivo and in vitro.

SV40 DNA replication in vivo is greatly stimulated by cis-acting transcriptional elements. We studied a model viral chromosome containing a single binding site for the cellular transcriptional activator, nuclear factor I (NF-I/CTF), located adjacent to the replication origin. The presence of the NF-I recognition site increased replication efficiency over 20-fold in vivo. Purified NF-I had little effect on the replication efficiency in the standard SV40 cell-free system when the template was introduced as naked DNA. However, NF-I specifically prevented the repression of DNA replication that occurred when the template was preassembled into chromatin. Our data support a model in which the binding of a transcriptional activator perturbs the local distribution of nucleosomes, thereby increasing the accessibility of the origin region.

Glial fibrillary acid protein immunoreactivity in fine-needle aspiration of salivary gland lesions: a useful adjunct for the differential diagnosis of salivary gland neoplasms.

The value of immunocytochemical staining for glial fibrillary acid protein (GFAP) in salivary gland lesions was investigated in 33 fine-needle aspiration smears. The study utilized cytologic material from ten pleomorphic adenomas, six normal salivary glands, three cases of chronic sialadenitis, three Warthin's tumors, two adenoid cystic carcinomas, three adenocarcinomas, two malignant mixed tumors, one acinic cell carcinoma, and three mucoepidermoid carcinomas. All tested pleomorphic adenomas stained positively. The adenoid cystic carcinomas and the cases of chronic sialadenitis, along with the low-grade mucoepidermoid carcinoma, were negative for GFAP immunoreactivity. These results indicate that immunostaining for GFAP may be a valuable aid in the diagnosis of pleomorphic adenoma; GFAP may be especially helpful in distinguishing those cases for which the differential diagnosis includes the aforementioned salivary gland neoplasms.

Desensitization to parathyroid hormone in renal cells from aged rats is associated with alterations in G-protein activity.

Parathyroid hormone (PTH)-stimulated Na+/Ca2+ exchange activity, but not forskolin-sensitive Na+-dependent Ca2+ efflux, was blunted in renal cortical cells from aged rats. PTH-sensitive adenylate cyclase activity in renal membranes from senescent rats also declined, but forskolin-stimulated activity did not change. In addition, cholera toxin- and pertussis toxin-stimulated Na+-dependent Ca2+ efflux and cAMP formation were blunted in cells from aged animals. Further, cells from aged rats had decreased Gs-alpha and Gi-alpha proteins, as detected by ADP-ribosylation. These findings would be consistent with the proposal of an age-associated heterologous desensitization that involved the G-proteins. Serum concentrations of iPTH were increased in the old rat, suggesting that the desensitization to PTH in the aging rat represented an adaptive response to prolonged stimulation by the hormone. This hypothesis was supported by the findings that the attenuated PTH-sensitive Na+/Ca2+ exchange activity, cAMP formation, and adenylate cyclase activity in cells from old rats could be reversed by parathyroidectomy. The decreased label in cholera toxin-catalyzed ADP-ribosylated Gs-alpha and pertussis toxin catalyzed ADP-ribosylated Gi-alpha found in cells from aged rats was also largely negated by the surgery. In conclusion, the results suggest that the age-related blunting in the responses of renal cells to PTH was associated with a deficit in G-protein function and that this alteration could be reversed by removal of the parathyroid gland.

An investigation of the polymerization of orthodontic adhesives by the transillumination of tooth tissue.

Prepared samples of light-activated composite were cured by transillumination of tooth tissue in healthy volunteers. Positioning of the samples and the direction of illumination was designed to simulate curing during direct bonding. The effectiveness of curing was investigated by determination of surface microhardness. The degree of polymerization of the specimens cured by transillumination was found to be reduced compared to the control samples cured by direct exposure to the light source.

DNA ploidy analysis of cervical condyloma and intraepithelial neoplasia in specimens obtained by punch biopsy.

This study analyzed the feasibility of, and the strategy for, DNA ploidy analysis of cervical condyloma and intraepithelial neoplasia by a computerized digital imaging system. Paraffin-embedded tissue provided satisfactory single-cell preparations for DNA ploidy analysis after enzyme digestion and additional procedures. Negative endocervical curettings and normal squamous mucosa were used as internal diploid controls. With suitable controls, 21 (81%) of the 26 aneuploid lesions were identified as such in the single-cell preparations. The remaining five lesions (not recognized as aneuploid in the single-cell preparations) had ploidy levels between 2.08n and 2.30n and required DNA measurements on 12-microns sections. Criteria for these DNA measurements were defined: specimens intended for DNA ploidy analysis should contain abnormal epithelium of at least 3 mm to 4 mm in greatest dimension and should be accompanied by diploid controls, such as endocervical curettings or normal ectocervical squamous mucosa. With a combination of single-cell preparations and 12-microns tissue sections, it was possible to accurately determine the DNA ploidy patterns of the cervical lesion specimens obtained by punch biopsies. Available evidence suggests that ploidy analysis can provide useful diagnostic and prognostic information.

Alpha-2-adrenergic modulation of the parathyroid hormone-inhibition of phosphate uptake in cultured renal (OK) cells.

Parathyroid hormone enhances the formation of cAMP and decreases the Na+-dependent uptake of phosphate in cultured renal cells derived from the American opossum (OK cells). Epinephrine, acting as an alpha 2-adrenergic agonist, inhibits the PTH-induced synthesis of cAMP by a pertussis toxin-sensitive mechanism and blunts the inhibition of phosphate transport by PTH. Na+-dependent alpha-methylglucoside and Na+ uptakes by the cells are unaffected by PTH and epinephrine. These findings suggest that alpha 2-adrenergic agonists may selectively modulate PTH-sensitive phosphate transport in the renal proximal tubule.

Malondialdehyde-altered protein occurs in atheroma of Watanabe heritable hyperlipidemic rabbits.

It has been proposed that chemically reactive lipids released during lipid peroxidation convert low density lipoprotein (LDL), the major carrier of plasma cholesterol, to an abnormal form and that receptor-mediated clearance of this altered LDL produces cholesteryl ester deposition in macrophage-derived foam cells of atheroma. Immuno-cytochemical analyses now reveal the presence of protein modified by malondialdehyde, a peroxidative end product, which colocalizes with the extracellular deposition of apolipoprotein B-100 protein of LDL in atheroma from Watanabe heritable hyperlipidemic rabbits. These findings provide direct evidence for the existence in vivo of protein modified by a physiological product of lipid peroxidation within arterial lesions.

Asymmetric distribution of gangliosides in rat renal brush-border and basolateral membranes.

Highly enriched brush-border and basolateral membranes isolated from rat renal cortex were used to study the distribution of endogenous gangliosides in the two distinct plasma membrane domains of epithelial cells. These two membrane domains differed in their glycolipid composition. The basolateral membranes contained more of both neutral and acidic glycolipids, expressed on a protein basis. In both membranes, the neutral glycolipids corresponding to mono-, di-, tri- and tetraglycosylceramides were present. The basolateral membranes contained more diglycosylceramide than the brush-border membranes. The major gangliosides found were GM4, GM3, and GD3 with minor amounts of GM1 and GD1a. The latter were identified and quantified by sensitive iodinated cholera toxin binding assays. When the distribution of individual gangliosides was calculated as a percent of total gangliosides, the brush-border membranes were enriched with GM3, GM1 and GD1a compared to the basolateral membranes, which were enriched with GD3 and GM4. The observation of a distinct distribution of glycolipids between brush-border and basolateral membranes of the same epithelial cell suggests that there may be a specific sorting and insertion process for epithelial plasma membrane glycolipids. In turn, asymmetric glycolipid biogenesis may reflect differences in glycolipid function between the two domains of the epithelial plasma membrane.

Keratin immunoreactivity in fine-needle aspiration of the prostate: an aid in the differentiation of benign epithelium from well-differentiated adenocarcinoma.

The efficacy of utilizing immunocytochemical staining of prostatic basal cells in separating benign from malignant prostatic epithelium was tested by staining fine-needle aspiration smears of prostatic lesions with the monoclonal antibody EAB-903. This antibody has been shown to stain keratin subtypes present in the prostate only in basal cells. The study utilized 12 benign, nine malignant, and four suspicious-for-carcinoma cases. Ten of 12 benign cases showed an intermingled pattern of staining, which was not found in the malignant cases. Our findings indicate that this distinctive pattern of staining may assist in distinguishing benign epithelium from well-differentiated prostatic adenocarcinoma.

Cranial base and cleft lip and palate.

Previous studies do not agree on relationships between cleft palate and the cranial base. This study comparing normal and cleft lip-and-palate samples finds no significant difference in cranial base angle, but significantly shorter clivus length in the cleft sample.

Detection of Mycoplasma pneumoniae-DNA within diseased gingiva by in situ hybridization using a biotin-labeled probe.

Isolation of Mycoplasma pneumoniae was accomplished in gingival biopsies of four patients exhibiting chronic inflammatory periodontal disease using in situ hybridization with biotin-labeled DNA probes. The distribution of the M. pneumoniae organisms was mainly cytoplasmic and evenly spread around the nuclei of connective tissue cells. Controls of nondisease sites in the same patients showed no infection with M. pneumoniae. Since M. pneumoniae is the only Mycoplasma known to be pathogenic, their role in the pathogenesis of periodontal disease needs further study.
