Chemical reduction of 3-oxo and unsaturated groups in fatty acids of diphosphoryl lipid A from the lipopolysaccharide of Rhodopseudomonas sphaeroides. Comparison of biological properties before and after reduction.

Unlike the diphosphoryl lipid A (DPLA) derived from toxic lipopolysaccharide of Escherichia coli and Salmonella strains, the DPLA from nontoxic lipopolysaccharide of Rhodopseudomonas sphaeroides ATCC 17023 is biologically inactive. This could be due to the presence of 3-oxotetradecanoic and delta 7-tetradecenoic acids. These two fatty acids in R. sphaeroides DPLA were catalytically reduced in platinum oxide/H2 to the 3-hydroxy and saturated fatty acids, respectively. The biologically active E. coli DPLA was also treated with platinum oxide/H2, but as expected, the reduction step did not change the structure. These two preparations were then compared with the untreated samples for biological activity in three select in vitro assays. Over a range of 0.01-100 ng/ml, both normal and reduced DPLA from R. sphaeroides were inactive in priming phorbol myristate acetate-stimulated superoxide anion release in human alveolar macrophages. Over a range of 10-10(3) ng/ml, both samples failed to induce tumor necrosis factor in the RAW 264.7 murine macrophage cell line. The reduced DPLA marginally activated 70Z/3 pre-B cells at concentrations of 0.1-30 micrograms/ml. In every case, both normal and platinum oxide/H2-treated E. coli DPLA were biologically active. These results indicate that the lack of biological activity of R. sphaeroides DPLA is not due to the presence of 3-oxo and unsaturated fatty acids, but rather to one or more of the following: (i) presence of only five fatty acyl groups (compared to six in active lipid A); (ii) presence of 3-hydroxydecanoic acids (rather than 3-hydroxytetradecanoic, in active lipid A); (iii) greater variation in size of the fatty acids.

Identification of the ancestral haplotype for apolipoprotein B suggests an African origin of Homo sapiens sapiens and traces their subsequent migration to Europe and the Pacific.

The probable ancestral haplotype for human apolipoprotein B (apoB) has been identified through immunological analysis of chimpanzee and gorilla serum and sequence analysis of their DNA. Moreover, the frequency of this ancestral apoB haplotype among different human populations provides strong support for the African origin of Homo sapiens sapiens and their subsequent migration from Africa to Europe and to the Pacific. The approach used here for the identification of the ancestral human apoB haplotype is likely to be applicable to many other genes.

Schistosome female reproductive development.

Schistosome parasites have evolved to produce a number of unique features in their life history; one of these is separate sexes. This has, in turn, led to a novel interplay between the male and female parasite that has been recognized for over 50 years: the growth and reproductive development of the female parasite is in some way regulated by the male schistosome. Early classical and later experimental studies established that the presence of the male schistosome is necessary not only for the initiation of female development but also for the maintenance of her mature state. The male parasite regulates the reproductive development of the female, partly by providing a stimulus that is necessary for the development of the vitelline gland. The cells of the vitelline gland provide nutrients and shell precursors for the egg. Also in this review by Philip LoVerde and Li-ly Chen, it is interesting to note that recent molecular studies have confirmed early work by showing that gene expression in the female parasite is developmentally regulated in a tissue-specific manner and that this gene expression is controlled by the presence of a male parasite.

A comparative electrophysiological study on neurotisation in rats.

A comparative experimental study has been carried out in rats with denervated gastrocnemius muscles. Three groups of five rats were treated by three different types of implantation of nerve directly into muscle (neurotisation). In the fourth group, the common peroneal nerve was sutured to the transected tibial nerve. The fifth group was left denervated as a control. The muscles were studied by serial electrodiagnostic studies and later histologically. The results showed little difference between nerve suture and implantation of nerve directly into muscle. Extension of the common peroneal nerve with a pair of sural nerve grafts did not produce a detrimental effect.

Sustained potentiation of NMDA receptor-mediated glutamate responses through activation of protein kinase C by a mu opioid.

mu opioids, such as morphine and certain enkephalin analogs, are known to modulate glutamate-evoked activity in dorsal horn neurons in the spinal cord and caudal brain stem. Yet the molecular mechanism by which this modulation occurs is not understood. We examined the interactions between glutamate and a selective mu opioid receptor agonist, D-Ala2-MePhe4-Gly-ol5-enkephalin (DAGO), in spinal trigeminal neurons in thin medullary slices of rats. DAGO caused a sustained increase in glutamate-activated currents that are mediated by N-methyl-D-aspartate receptors. Intracellularly applied protein kinase C (PKC) mimics the effect of DAGO, and a specific PKC inhibitor interrupts the sustained potentiation produced by DAGO. Thus, PKC plays a key role in mediating the action of mu opioid peptides.

Interleukin-6 induces the (2'-5') oligoadenylate synthetase gene in M1 cells through an effect on the interferon-responsive enhancer.

Interleukin-6 (IL-6) activates (2'-5') A synthetase (2'-5' AS) gene expression in differentiating myeloleukemic M1 cells. Antibodies to type I interferon (IFN) inhibit 2'-5' AS induction but not differentiation. Analysis of the mechanism of 2'-5' AS induction shows that it does not result from increased IFN formation, but from a synergism between IL-6 and endogenously secreted IFN. IL-6 can activate expression of a CAT construct fused to the interferon response sequence (IRS) of the 2'-5' AS gene. In extracts of IL-6-treated M1 cells, changes in protein binding to IRS DNA can be demonstrated. One of the effects of IL-6 on M1 cells is, therefore, to induce DNA binding factors, some of which act on the same enhancer sequence as IFNs, resulting in a synergistic gene activation. M1 variants resistant to differentiation by IL-6 have lost the ability to induce the 2'-5' AS gene.

High intensity focused ultrasound for the treatment of rat tumours.

Discrete implanted liver tumours in the rat have been exposed to arrays of 1.7 MHz ultrasound lesions. Focal peak intensities in the range 1.4-3.5 k Wcm-2 were used for an exposure time of 10 s. It has been demonstrated that where the whole tumour volume was exposed to the focused ultrasound beam, no evidence of tumour growth could be detected histologically. Where the ultrasonic lesion array was not contiguous, regrowth occurred. Preliminary histological studies confirmed this finding.

Growth factors and cardiovascular structure. Implications for calcium antagonist therapy.

Abnormalities of cellular growth regulation are integral to the development of cardiovascular disorders such as atherogenesis, ventricular hypertrophy, and diabetic glomerulopathy. Moreover, cellular growth is in large measure controlled by peptide and nonpeptide growth factors that mediate their actions, in part, through the transcriptional regulation of normal cellular genes called protooncogenes. Because angiotensin II is one such growth regulatory factor and because changes in intracellular calcium are intimately involved in the action of angiotensin and other growth factors, it is likely that inhibitors of angiotensin action and calcium-channel-blocking agents will be found to have useful growth regulatory properties.

Na+ transport in isolated rat CCD: effects of bradykinin, ANP, clonidine, and hydrochlorothiazide.

We examined the effects of bradykinin (BK), atrial natriuretic peptide (ANP), hydrochlorothiazide (HCTZ), and clonidine on Na+ transport in isolated perfused cortical collecting ducts from rats treated with deoxycorticosterone. Arginine vasopressin was present in the bathing solution at 220 pM. Clonidine (1 microM, bathing solution) depolarized transepithelial potential difference (PDT) from -11.9 +/- 2.0 (SE) to -7.4 +/- 1.7 mV (P less than 0.001), hyperpolarized basolateral membrane potential difference (PDbl) from -85 +/- 1 to -87 +/- 1 mV (P less than 0.01), and increased the fractional resistance of the apical membrane (FRa) from 0.81 +/- 0.02 to 0.86 +/- 0.02 (P less than 0.03), indicating that it inhibited the Na+ conductance of the luminal membrane. BK (1 or 10 nM) or ANP (10 nM) in the bathing solution had no effect on PDT, PDbl, or FRa. BK, ANP, or 0.1 mM luminal HCTZ also had no effect on lumen-to-bath 22Na+ flux (J1----b), whereas we showed previously that clonidine inhibits J1----b by 30% (L. Chen, M. Paris, S. K. Williams, M. C. Reif, and J. A. Schafer. Kidney Int. 37: 366, 1990). Luminal addition of Na+ channel blockers amiloride (10 microM) or benzamil (1 microM) reduced J1----b to a level not significantly different from bath-to-lumen 22Na+ flux measured previously (M. Reif, S. L. Troutman, and J. A. Schafer. J. Clin. Invest. 77: 1291-1298, 1986), and neither BK nor HCTZ had any further effect. These results show that transcellular Na+ transport occurs exclusively through the apical membrane amiloride-sensitive channel, and this conductance is inhibited by clonidine but not by BK, ANP, or HCTZ.

Clonidine and PGE2 have different effects on Na+ and water transport in rat and rabbit CCD.

We examined the effects of prostaglandin E2 (PGE2) and the adrenergic agonist clonidine on osmotic water permeability (Pf), lumen-to-bath 22Na+ flux (Jl----b), and transepithelial voltage (VT) in isolated perfused cortical collecting ducts (CCD) from rats and rabbits. Although PGE2 inhibited arginine vasopressin (AVP)-dependent Jl----b and Pf in CCDs from deoxycorticosterone (DOC)-treated and untreated rabbits, 0.1-10 microM PGE2 had no inhibitory effect on any of these transport parameters in CCDs from DOC-treated rats in presence of AVP. On the other hand, clonidine (1 microM in bathing solution) reversibly inhibited AVP-dependent Pf, Jl----b, and VT in the rat CCD by 30-40%, and 0.3 microM yohimbine, a specific alpha 2-adrenoceptor antagonist, reversed these effects. However, we were unable to demonstrate any inhibitory effect of 1-10 microM clonidine on Pf, Jl----b, or VT in the rabbit CCD using a variety of protocols. These results are consistent with the pattern of inhibition of AVP-dependent adenosine 3',5'-cyclic monophosphate (cAMP) production in the rat and rabbit CCD in that PGE2 inhibits both transport and cAMP production in the rabbit but not the rat CCD, and clonidine inhibits both transport and cAMP production in the rat but not the rabbit CCD [D. Epiglottic laryngoplasty after hemilaryngectomy for glottic cancer.

Epiglottic laryngoplasty is technically feasible as a one-stage procedure with excellent functional results. Although the Kambic-Sedlacek-Tucker (K-S-T) technique of glottic reconstruction offers early extubation with an adequate airway, a subsequent wide neoglottis may increase the chance of aspiration and a poor voice. To better restore the laryngeal functions of closure and phonation, we made some modifications on the original K-S-T technique as follows: (1) One of the lateral margins of the epiglottis with the aryepiglottic fold is sutured to the arytenoid region of the cricoid rather than a thyroid cartilage remnant. A neo-arytenoid is formed. (2) The other lateral margin of the epiglottis with the aryepiglottic fold is sutured to the cut edge of the false and true cord instead of a thyroid ala remnant. Therefore both margins of the epiglottis with the aryepiglottic folds are lowered as much as possible to the level of the glottis. A new pseudocord is formed. (3) A cartilage cut is made at the anterior aspect of the epiglottis, leaving its laryngeal surface of mucoperichondrial intact. A new anterior commissure with a sharp angle is shaped by this maneuver. Nineteen hemilaryngectomies with modified epiglottic laryngoplasty have been performed by members of the Department of Otolaryngology of Guangxi Medical College since 1984. Results in this series are fairly good and indicate that the modified epiglottic laryngoplasty is effective in enhancing functional results in terms of respiration, deglutition, and phonation.

Expression of reg/PSP, a pancreatic exocrine gene: relationship to changes in islet beta-cell mass.

A cDNA termed reg was recently isolated by differential screening of a library prepared from regenerating islets isolated from pancreatic remnants of rats subjected to 90% pancreatectomy and nicotinamide treatment. This led to speculation that this gene may be involved in expansion of beta-cell mass. In the current study we have measured reg expression after implantation and resection of a solid insulinoma tumor into rats, maneuvers known, respectively, to reduce and reexpand the volume of beta-cells in the islet. Animals with an implanted insulinoma tumor became profoundly hypoglycemic. Islet beta-cells declined from the normal 75% of total islet volume to less than 30%, in concert with a marked reduction in the reg mRNA level. Removal of the tumor resulted in a sharp increase in beta-cell replication, as measured by [3H]thymidine incorporation and a return to normal beta-cell volume within 4 days of tumor resection. This was associated with a transient induction in reg expression compared to that in tumor-bearing animals, effectively returning the amount of reg mRNA to the levels found in normal animals within 48 h; at later time points after tumor removal (3-7 days) reg expression declined, but then rose toward normal. In situ hybridization analysis localized the initial induction in reg mRNA expression to the exocrine pancreas. Continuous infusion of insulin into normal rats for 4 days, a maneuver that does not significantly reduce beta-cell mass, resulted in dramatically reduced insulin mRNA in islets, but no change in the levels of reg mRNA. We conclude that the diminution in pancreatic beta-cell mass caused by subcutaneous implantation of an insulinoma is associated with reduced reg gene expression and that the increase in beta-cell replication after resection of the tumor is preceded by return of reg gene expression toward normal.

Angiotensin-converting enzyme inhibition reduces neuroblastoma cell growth rate.

Because of the known capacity of angiotensin II to serve as a growth factor in multiple tissues, we elected to study the effects of renin-angiotensin system inhibition on the growth of human SH-SY5Y neuroblastoma cells. Cells were treated with captopril (0.05-5 mg/ml), enalapril, or enalaprilat (0.02-5 mg/ml) or saralasin (0.1-0.25 mg/ml). In all cases, statistically significant reductions in cell growth were seen over 5 days of culture. In additional experiments, captopril and enalaprilat significantly decreased thymidine incorporation into DNA in these cells. The administration of angiotensin II in the presence of captopril partially offset these suppressive effects.

[Early effects of explosion on hearing threshold and activities of Na+-K+-ATPase and succinic dehydrogenase in the stria vascularis of guinea pigs].

Slow cortex responses to several frequencies were recorded in guinea pigs before and after intense impulse noise exposure. Guinea pigs were decapitated 90 minutes after the exposure. Besides scanning electron microscopic analysis of cochleas, activities of Na(+)-K(+)-ATPase and SDH in the stria vascularis (SV) corresponding to 4 kHz were studied with histochemical methods under light microscope. Densities of histochemical reaction products and cross sections of the SV were measured with image analysis system. Activities of Na(+)-K(+)-ATPase and SDH in the SV decreased significantly in PTS group (P less than 0.01, P less than 0.05). There was no significant change in TTS group. It revealed that decrease of Na(+)-K(+)-ATPase and SDH in the SV may contribute to remarkable hearing threshold shift besides mechanical destruction by noise. Metabolic disturbance may aggravate hair cell damage. Direct damage of the SV caused by intense impulse noise may be one of the causes of PTS.

Terminal differentiation of myeloleukemic M1 cells induced by IL-6: role of endogenous interferon.

During terminal differentiation of myeloleukemic M1 cells triggered by IL-6, an induction of IFN-activated genes, such as IRF-1, class I MHC, and (2'-5')-A synthetase, is observed. Antibodies to murine type I IFN, inhibit most (2'-5')-A synthetase induction but do not inhibit IL-6-induced growth-arrest and differentiation. IL-6 induction of (2'-5')-A synthetase subforms, however, differs from that of IFN. IL-6 in fact induces a cell surface form of (2'-5')-A synthetase that is not induced by IFN.
