Amylin-insulin relationships in insulin resistance with and without diabetic hyperglycemia.

To determine if increased secretion of amylin can be implicated in the pathogenesis of non-insulin-dependent diabetes mellitus (NIDDM) in vitro and in vivo, we studied its relationships to insulin in insulin-resistant rats with and without NIDDM. In obesity-associated and dexamethasone-induced insulin resistance without diabetes, basal and stimulated secretion of amylin and insulin by isolated pancreata were proportionately elevated, leaving the amylin-to-insulin ratio (A/I) unchanged. By contrast, whenever diabetes occurred in dexamethasone-treated rats or in spontaneously diabetic obese insulin-resistant ZDF-drt male rats, a doubling of A/I was invariably observed due to an increase in amylin without a proportional increase in insulin secretion. Correction of dexamethasone-induced hyperglycemia with the glucocorticord receptor antagonist RU-486 was accompanied by a decline in A/I. Longitudinal in vivo studies demonstrated in both spontaneous and dexamethasone-induced models of NIDDM an increase in plasma A/I at the onset of hyperglycemia. In dexamethasone-induced diabetes, the increased A/I was associated with a high proamylin mRNA relative to proinsulin mRNA. We conclude that amylin and insulin expression and secretion rise in concert in compensated insulin-resistant states, but when hyperglycemia is present the increase in amylin exceeds that of insulin. Although a role of an increased A/I in the pathogenesis of NIDDM has not been established directly, these studies indicate that such a role could be possible.

Intracellular Ca2+ and PKC activation do not inhibit Na+ and water transport in rat CCD.

Experiments examined the effects of elevation of intracellular calcium concentration ([Ca2+]i) or activation of protein kinase C (PKC) on Na+ and water transport in the rat cortical collecting duct (CCD). We measured the lumen-to-bath 22Na+ flux (J1-->b), transepithelial voltage (VT), and water permeability (Pf) in CCD from deoxycorticosterone (DOC)-treated rats. Ionomycin (0.5 and 1 microM) and thapsigargin (1 and 2 microM) were used to increase [Ca2+]i. Phorbol 12-myristate 13-acetate (PMA; 0.3 and 1 microM) and oleoyl-acetyl-glycerol (OAG; 100 microM) were used as activators of PKC. [Ca2+]i was measured in isolated perfused tubules using the fluorescent dye fura 2. When added to the bathing solution, 220 pM arginine vasopressin (AVP) failed to affect [Ca2+]i, whereas 1 microM ionomycin increased [Ca2+]i by 103 +/- 15% and 2 microM thapsigargin increased [Ca2+]i by 24 +/- 4%. In flux studies, neither ionomycin nor thapsigargin affected J1-->b or Pf, although ionomycin caused marked morphological changes. Ionomycin also failed to alter either parameter in tubules from non-DOC-treated rats. Neither 100 microM OAG nor 1 microM PMA affected J1-->b or Pf. OAG at 50 microM had no effect on VT or transepithelial resistance, indicating no inhibition of conductive Na+ transport. We conclude that increased [Ca2+]i and PKC activation do not affect J1--b or Pf in the rat CCD. These findings may account for the sustained increase in J1--b produced in the rat CCD by AVP.

Soft-palate myogenesis: a developmental field paradigm.

Surgical correction of clefts of the soft palate leads to varying degrees of normal function although the repair itself is successful. Explanations for this include structural abnormalities of the muscles. Previous studies have focused primarily on gross anatomical features of late fetal and postnatal cleft palate musculature; however, infrequent reference has been made to early prenatal morphologic patterns of soft-palate development, beginning with the embryo. Thus we evaluated the chronology of prenatal myogenesis of the soft palate from its early mesenchymal phase through the appearance of definitive palatal muscles and associated structures in a sample of 22 human fetuses that represented postfertilization weeks 6.5 to 20.5 (18- to 192-mm crown-rump length). Specimens were histologically prepared for descriptive and morphometric light microscopy. Data were collected on the earliest appearance times of identifiable soft palate and associated structures within the mesenchymal field and on their individual stages of myogenesis (e.g., for muscles, from mesenchyme to myoblasts to fascicles). Analyses showed that (1) palatal muscles and related bony structures emerge sequentially as densely staining mesenchymal subfields within the larger mesenchymal soft-palate field during the 6- to 9-week period, with the tensor veli palatini muscle appearing earliest, and the musculus uvulae latest; (2) further morphogenesis of the soft palate and associated structures follows a definite timeline; and (3) by 16 to 17 weeks the postnatal palatal morphology is in place.

GLUT2 expression and function in beta-cells of GK rats with NIDDM. Dissociation between reductions in glucose transport and glucose-stimulated insulin secretion.

GLUT2 underexpression has been reported in the beta-cells of Zucker diabetic fatty rats and db/db mice, models of spontaneously occurring NIDDM with antecedent obesity. To determine whether the beta-cells of a nonobese rodent model of NIDDM exhibit the same abnormalities in GLUT2, we studied Goto-Kakizaki rats. In these mildly diabetic animals glucose-stimulated insulin secretion was reduced at all ages examined from 8 to 48 wk. In normal control Wistar rats, immunostainable GLUT2 was present on all insulin-positive cells in the pancreatic islets. Only 85% of beta-cells were GLUT2-positive in GK rats at 12 wk of age, and only 34% were positive at 48 wk of age. GLUT2 mRNA was 50% of normal in 12-wk-old GK rats. In the latter age-group, glucose-stimulated insulin secretion was only 28% of normal at a time when 85% of beta-cells were GLUT2-positive and initial 3-O-methyl-D-glucose transport rate was 77% of the control value. We conclude that although GLUT2 is underexpressed, neither the magnitude of the underexpression of GLUT2 nor of the reduction in GLUT2 transport function in islets of GK rats is sufficient by itself to explain the profound reduction in glucose-stimulated insulin secretion.

Comparison of the electrophysiological effects of S49, CSH087 and CSH068 in rat ventricular cells.

S49, CSH087 and CSH068 are aporphine alkaloids and are synthesized from vanillin. In the present study, the effects of these compounds on the action potential and membrane currents of rat ventricular cells were examined by using the whole cell recording technique with single suction pipettes. At a stimulation frequency of 0.1 Hz, S49 (1.5 microM), CSH087 (3 microM) and CSH068 (3 microM) could prolong the action potential durations (APD50 and APD90). Voltage clamp studies revealed inhibition of the transient outward currents by S49, CSH087 and CSH068 with KD of 2.0, 1.8 and 5.2 microM, respectively, but a lack of enhancement of calcium influx. Therefore, the inhibition of the potassium conductance may explain the prolongation of APD induced by these compounds. In contrast, the late potassium outward current was more resistant to CSH087 and S49, but was blocked by CSH068 with a KD of 6.4 microM. The sodium inward current (INa) also was inhibited by S49, CSH087 and CSH068 in a dose-dependent manner with KD of 1.2, 6.8 and 7.5 microM, respectively (at a holding potential of -80 mV). The inhibition of INa) was enhanced at a less negative holding potential. The voltage-dependent inhibition of INa was associated with the shift of the INa availability curve to more negative potentials. These results indicate that these compounds have high affinity for inactivated Na+ channels. A twin-pulse experiment revealed that these compounds increased the recovery time constant of INa. The potency for the shift of the INa availability curve and the retardation of INa recovery from inactivation were S49 > CSH087 and CSH)68.(ABSTRACT TRUNCATED AT 250 WORDS)

[The role of hemorheologic disturbance in experimental acute pancreatitis].

The role of hemorheologic disturbance in experimental acute pancreatitis was studied in Wistar male rats. It was found that pancreatic duct obstruction with secretin stimulation could induce acute interstitial pancreatitis (AIP), but when high molecular dextran intravenous injection being added, acute necrotizing pancreatitis (ANP) was induced. The authors suggest that the high molecular dextran can increase blood viscosity, aggregation of erythrocytes and decrease erythrocyte deformability, which results in pancreatic microcirculatory disturbances and promotes progressive pancreatic necrosis. Hemorheologic disturbance is one of the key factors in the pathogenesis of ANP.

[Lamellar keratoplasty for treatment of corneal dermoid].

The results of lamellar keratoplasty for treatment of 53 cases with corneal dermoid were analysed. The visual acuity of postoperation remained or was improved in 79.5% of the cases. Keratometry measured in 12 eyes after surgery showed the corneal curvature remained unchanged in 7 eyes, was reduced in 2 eyes, and increased in 3 eyes. The astigmatism of cornea was not changed or decreased in 6 eyes. Three eyes got increased astigmatism less than 0.5D, the other 3 eyes respectively 1D, 1.5D and 4.4D. The refractive states estimated by photorefraction in 7 eyes showed there was no changed astigmatism in 3 eyes. One eye decreased 2.5D and three eyes respectively increased 0.5 D, 1.0D and 2.0D. Indication and time of surgery for corneal dermoid were also discussed.

[Influence of 11 ginsenoside monomers on action potentials of myocardiocytes].

Eleven ginsenoside monomers were added to cultured myocardiocytes of neonatal Wistar rats in a concentration of 20 micrograms.ml-1. And the action potentials (AP) were led with microelectrodes. Rb1, Rb2, Rb3, Rc, Rg1, Rg2, Re, and Rh1 inhibited AP, decreasing APA by 20%, 13%, 5%, 18%, 30%, 37%, 28%, and 23%, respectively. The potency of the most effective monomer Rg2 20 micrograms.ml-1 was similar to that of the calcium channel blocker nimodipine 1.25 micrograms.ml-1. Rd, Rf, and Ro had no significant influence on AP. The average potency of the panaxatriol saponin (PTS) monomers were greater than that of the panaxadiol saponin (PDS) monomers. The above demonstrate the reason why the calcium channel blockade action of PTS is stronger than that of PDS results from the summation of monomer drug effects.

[The influence of pressing the meridian course on electroretinogram during acupuncture].

Observation was performed on 23 subjects with marked propagated sensation along meridians (PSM) and 30 without PSM. The electroretinogram (ERG) was recorded with surface electrode and added by an averager. The b wave of ERG evoked by flash stimulation was positive with a latent period about 60 ms and amplitude of 22 +/- 1.66 muv (n = 53). There were 2 types of ERG response during puncturing Hegu (LI 4). The amplitude of ERG b wave increased in 33 subjects and decreased in 20 subjects. However, the acupuncture effect was kept relatively stable in each subject. The spontaneous variation of ERG amplitude was about 10%. In contrast, it changed by 26.5 +/- 2.43% (n = 20) when Hegu (LI 4) was acupunctured alone. It is particularly to be noted that the amplitude of b wave almost remained unchanged, with in the range of 10.08 +/- 2.42% (n = 20), as the pressure was applied on Shousanli (LI 10) during acupuncture, no matter whether the subjects with or without PSM. However, the variation rate of b wave increased by 24.83 +/- 3.74% (n = 15) once again, if the pressure was applied on both side of Shousanli (LI 10), it was nearly equal to the value caused by acupuncture Hegu (LI 4) alone. The contrast between the two is striking. The result revealed again that there may be a certain substantial process advancing along the channel during acupuncture. It is the blockade of this process by mechanical pressure which leads to the decrease of the acupuncture effect.

[Observation on the relation between propagated sensation along meridians and the therapeutic effect of acupuncture on myopia of youngsters].

Nine hundred and ninety two eyes suffering from various degrees of myopia in 536 youngsters were treated with acupuncture. A method of exciting propagated sensation along meridians (PSM) and making it travel toward the affected region was employed in treatment. Bilateral Hegu (LI4) and Taichong (LR3); and bilateral Waiguan (ST5) and Guangming (GB37) were punctured on alternate days. The results showed that after 1-3 periods of treatment, the vision of 868 eyes (87.5%) were improved in various extent. Among them, the vision completely recovered in 131 eyes (13.31%), and the diopter decreased-0.75 to -1.00 D.S. in 13 eyes after acupuncture treatment. The therapeutic effect of acupuncture seemed to be considerably satisfactory and stable in two years following up. The extent of distinctness of PSM raised markedly with the increase of times of acupuncture. There was a close relation between the extent of distinctness of PSM and effectiveness of acupuncture. The more striking the centripetal PSM, the better was therapeutic effect of acupuncture. The excellent effect was always achieved when PSM arrived at the affected eye. Since PSM arrived at a large number of eyes in the subjects in younger year and so the better therapeutic effect was also achieved.

[Observation on the phenomenon of propagated sensation along meridians in youngsters].

That the opinion on the relation between the propagated sensation along meridians (PSM) and the age of the subjects was divergent in the results reported by some authors. In the last 10 years, we have performed a systematic investigation on PSM on a group of 536 youngsters. The main characteristics of PSM in youngsters were just the same as that of adults. However, the incidence of PSM in youngsters, particularly the degree of distinctness of PSM, was by far higher than that of adults. The results were stable and could be repeated in the experiments performed in different years. It showed that there was surely a close relation between the degree of distinctness of PSM and the age of subjects. This fact can not be appropriately explained at present. However, it provides an important clue to clarify the factors which influence the appearance of PSM, and in turn to promote the improvement of the therapeutic effect of acupuncture and cilacriation of the mechanism underlying the formation of PSM.

[The effect of stimulating the auricular liver-gall point on the size of gallbladder of the rabbit].

After stimulating the auricular liver-gall point of rabbit with electrode. We observed marked contraction of gallbladder. Only one minute after stimulation, the contraction began. About ten minutes after stimulation the contraction was marked and the contraction lasted for 30-40 minutes. The average product of the length and width of gallbladder (15-25 minutes after stimulation) is markedly smaller than that of control (pre-stimulation). This result proves the liver-gall point of rabbit corresponds with its internal organ-gallbladder. The result also present a theoretic basis for clinical therapy of press over the auricular liver-gall point induced the excretion of gallstones.

[Studies on residual antimalarial activity of tripynadine in mice and monkeys].

This paper reports the experiments in which tripynadine free base at a dose 4.5 times that of ED50 was given to mice by intragastric administration. On the 20th day following the administration the mice were inoculated with 1 x 10(7) RBC infected with Plasmodium berghei ANKA strain. The infection rate was zero, implying that all mice had acquired protection. Although the residual activity time of tripynadine phosphate was longer than that of tripynadine free base or piperaquine phosphate, but tripynadine phosphate caused vomiting in monkeys during the medication. The residual antimalarial activity of tripynadine hydroxynaphthoate was less than that of tripynadine phosphate or tripynadine free base. A total dose of 200 mg/kg of tripynadine free base ensured residual antimalarial activity against P. cynomolgi bastianellii for 20 days. However, the residual activity decreased evidently when the total dose was reduced to 100 mg/kg. In short, it seems that the residual antimalarial activity of tripynadine free base is slightly less than that of piperaquine in monkeys.

A domestic cell bioreactor and its application in virus culture.

A cell culture bioreactor (CellCul-20) and its application in cell and virus culture are described in this paper. It has been evaluated with strict aseptic tests and one-year's operation shown that CellCul-20 bioreactor can keep its aseptic condition after being autoclaved. It can meet the requirement for the control of the main parameters for cell and virus culture and the finely adjustment of the main parameters to meet the changing conditions of the cultivation. A high cell density and a high level of virus titre were reached respectively for Vero cells and Japanese encephalitis virus (JEV) while they were cultured in this bioreactor. It is the first report on large-scale culture of JEV-infected Vero cells to prepare primary JEV vaccine. Some suggestions are made for the improvement of CellCul-20.

[Sodium channel blocking effect of scorpion venom on cultured mouse myocardiocytes].

Myocardiocytes of mice were cultured. Action potentials were recorded with microelectrodes inside the cells. Scorpion venom from Buthus martensii Karsch 3.75 or 7.5 micrograms.ml-1 decreased the duration of action potential and all of the depolarization concerned parameters of myocardiocytes. The Vmax, TP, APA behaved apparently in a dosage-dependent way. Restoration happened after washing out. Tetrodotoxin 2.5 micrograms.ml-1 acted in a similar way. Nimodipine 3.0 micrograms.ml-1 led to a decrease in action potential duration. BaCl2 0.1 mmol.L-1 elongated the action potential duration, while decreased the parameters concerned with depolarization. These results indicate that the scorpion venom has Na+ channel blocking action.

[Analysis on postoperative prognosis in relation to blood ketone body ratios in surgical patients with liver disease].

Eighty-one patients with liver disease were studied for the postoperative changes in the arterial blood ketone body ratio (KBR) and routine liver function tests. The postoperative patients were classified into three groups-according to the postoperative changes in KBR. In the 54 patients of group A, KBR did not decrease to below 0.7, only 2 cases with complications (3.7%); in the 25 patients of group B, KBR decreased transiently to 0.4-0.7 and then increased to over 0.7, 9 cases with complications (36.0%), but they were ameliorated concomitantly with the restoration of KBR; in the 2 cases of group C, KBR decreased irreversibly to below 0.4, and a variety of complications occurred, so the patients finally died of multiple organ failure (MOF). It was found that routine liver function tests were less reliable direct indicators for the onset of postoperative hepatic insufficiency than KBR, which reflected the oxidoreduction state of liver mitochondria.
