Control of initiation of pMB1 replication: purified Rop protein and RNA I affect primer formation in vitro.

We show that a protein of 63 amino acids is the product of the rop gene, a gene which negatively regulates the copy number of plasmids of the ColE1 family. Rop protein purified to homogeneity inhibits ColE1 plasmid replication in a bacterial extract. Furthermore, we show that Rop inhibition requires RNA I. In a purified in vitro system that can support primer transcription and processing, Rop affects primer formation in two ways: first, it elicits transcription termination oat nucleotide 220, and second, it increases the ability of RNA I to inhibit RNAase H processing of the primer. The analysis of these data and the comparison with the results obtained in vivo with transcription fusion experiments allow us to propose a tentative model of the molecular mechanism underlying Rop-RNA I inhibition.

[Biliary perforations caused by acute pancreatitis: considerations apropos of a case of necrosis of the intrapancreatic choledochus caused by acute cephalopancreatitis secondary to a transduodenal sphincterotomy].

The AA. refear a case of massive necrosis of intrapancreatic biliary tract secondary to an acute necrotizing postoperative cephalopancreatitis. The high mortality lesion risk was successfully treated by many factors: surgicals and medicals. The AA. think that the wise use of Somatostatine was the most important factor.

[Original operative technic in the surgical treatment of recto-sigmoid perforations (traumatic lesions, diverticular perforations, anastomotic dehiscences)].

In the modern rectal surgery the AA. think it be useful to preserve the sphinteric integrity to the greatest part of patients with non neoplastic rectosigmoid perforations (traumatic, diverticular, anastomotic leakage). They obtain to remove, by a personal modification, the difficulties and the disadvantages of intestinal restitution following the Hartmann's procedure.

[Our surgical choice in the treatment of tumors of the rectum].

The AA. suggest to rationalise the criterion of choice in the operation of rectum tumour. According to the AA., the choice is between abdomino-perineal and anterior resection operation. In accordance with that acknowledgement and the respect of the precise oncological and anatomical presuppositions, they define the heart of the tumour and regulated their surgical conduct. In practice they perform the anterior resection for the tumours situated between the superior and inferior haemorrhoidal arteries which, after dissection, can be found at least 8-9 cm. from the anus, while they always deal with the abdomino-perineal the neoplasms which develop in the area of inferior and middle haemorrhoidal arteries.

Cloning of several cDNA segments coding for human liver proteins.

A human cDNA library was constructed using M13 derivative vectors. The simple and rapid procedures for sequencing single-stranded DNA by the dideoxy chain termination method allowed a screening of individual clones directly by DNA sequence analysis. Some of these clones were identified as coding for: serum albumin, alpha1-antitrypsin, retinol-binding protein, prothrombin, haptoglobin, and metallothionein. Furthermore, a clone coding for aldolase B was tentatively identified on the basis of high sequence homology with rabbit muscle aldolase.

Processing of eukaryotic tRNA precursors: secondary structure of the precursor specific sequences affects the rate but not the accuracy of processing reactions.

The primary transcriptional product of eukaryotic tRNA genes is a precursor molecule with extranucleotides at the 5' and at the 3' end. We show that the 5' and 3' sequences, uniquely present in the RNA precursor molecule do not play any role in the efficiency and accuracy of processing reactions. If, however, as a consequence of in vitro manipulation, these extranucleotides form a base-paired extension of the aminoacid acceptor stem, the rate of processing is slowed down. The rate of processing is brought back to normal in a single base-pair deletion mutant probably as a consequence of a destabilization of the base-paired extension of the aminoacid acceptor stem.

Promoter of a eukaryotic tRNAPro gene is composed of three noncontiguous regions.

The 71-base-pair coding sequences of the tRNAPro gene from Caenorhabditis elegans contains all of the information required for transcription and processing in the injected oocytes. Several subclones of the DNA coding for the tRNAPro were constructed, carrying deletions or insertions, or both. Their transcriptional properties lead to the hypothesis that the tRNAPro gene promoter is composed of three discontinuous regions within the coding sequence.

[Reconversion after Hartmann's procedure. Our experience].

An increasing number of intestinal reconversion after Hartmann have been performed in recent years, especially due to improved surgical techniques and progressively lengthened lifespan. The authors report 33 cases of intestinal recanalization of 100 interventions according to Hartmann from 1984 to 1996 (21 not neoplastic pathologies, 12 neoplasias). The variables considered included: patient age, type of disease requiring intervention according to Hartmann, oncologic characteristic of patients with neoplasia, interval between the two interventions, preoperative examinations performed, morbidity and mortality after reconversion. Furthermore, the fundamental indications for reconversion are described, in particular in patients with neoplasias (CEA, transanal echo, total body Ct, anal manometry). The low frequency of preoperative complications, zero mortality, satisfactory long-term follow-up (only one patient with neoplastic relapse) indicate that colon-rectal reconversion can also be performed in the elderly and patients with neoplasias with favorable prognosis.

[Benign neoplasms of the small intestine].

The authors report four cases of benign tumors of the small bowel (2 leiomyomas and 2 lipomas). After a short review of the literature, they put in evidence the relatively infrequent occurrence and the difficulties involved in early preoperative diagnosis.

Pulmonary angiodysplasia: a case report.

A case of lung angiodysplasia is reported here in. The diagnosis of this disease before surgery is difficult. The intraoperatory diagnosis of vascular disease was possible since the pulsating mass disappeared after the inflow and outflow blood vessels were selectively clamped. The abnormal placement of a venous vessel made the resection problematical. A careful anatomic and clinical study of the lesion, and the correct instrumental diagnostic approach may lead to a suitable therapeutic stategy, which respects both functional integrity and surgical radicality. The usefulness of a systematical selective clamping of the inflow and outflow blood vessels for an exact delimitation of the lesion emphasised.
