A new allele of ocular retardation: early development and morphogenetic cell death.

The inheritance and some developmental effects of a new allele of ocular retardation (orJ) are described. Affected animals or 12 days of gestation, show reduced cell death in the eye cup and thickening of the inner wall of the optic fissure. At 11 to 13 dyas of gestation orJ/orJ eyes grafted to the testis do not produce retina as their orJ+ littermates do. Adult animals have small eyes with closed lids, abnormal retinal layers, and no optic nerve.

Development of teratomas from the ectoderm of mouse egg cylinders.

We studied the developmental capacities of the primary ectoderm and endoderm of 6-day embryos of hybrids between strains 129/Sv-SIJ C P and A/He mice by grafting these germ layers into the testes of adult mice for 30 days. Grafts of embryonic ectoderm gave rise to teratocarcinomas composed of undifferentiated embryonal cells and derivatives of all three germ layers including respiratory and alimentary epithelium. Grafts of extraembryonic ectoderm gave rise to invasive trophoblastic giant cells. Grafts of endoderm did not develop.

Ultrastructure of ovarian teratomas in LT mice.

Stem cells of these tumors formed nests consisting of undifferentiated embryonic cells in the center with more differentiated cells towards the periphery. A type viral particles were seen in the cisternae of rough endoplasmic reticulum. Ultrastructurally the stem cells of ovarian teratomas did not differ from stem cells of testicular or embryo-derived teratomas. They were however distinct from the cleavage stage embryonic cells and/or the unfertilized ovum, they stemmed from. They correspond both cytologically and developmentally to ectodermal embryonic cells from the egg-cylinder, the most advanced developmental stage of parthenotes observed previously in the ovary of LT mice.
