Functional changes in the lungs of guinea pigs exposed to sodium sulfite aerosols.

Guinea pigs were exposed head only for 1 hr to submicrometer sodium sulfite aerosols (mass median aerodynamic diameter = 0.36 micron, sigma g = 2.96) at 474, 669, and 972 micrograms SO3(2-)/m3. Respiratory mechanics were measured in unanaesthetized animals before, during, and after exposure. Dose-related increases in resistance and decreases in compliance were observed. At 972 micrograms SO3(2-)/m3, the sodium sulfite aerosol caused a 50% increase in resistance and a 19% decrease in compliance. These changes were still present 1 hr after the end of exposure. The results were used to assess the irritant potency of sodium sulfite aerosol. Another group of guinea pigs was exposed whole body for 1 hr to the same aerosol at 0, 204, 395, and 1152 micrograms SO3(2-)/m3. Immediately after the exposures, lung volume, diffusion capacity for carbon monoxide (DLCO), and wet lung weight were evaluated in anesthetized, tracheotomized animals. As compared to controls, total lung capacity, vital capacity, functional residual capacity, residual volume, and DLCO were all decreased with increasing concentrations of sodium sulfite. Dose-related increases in wet lung weights were also observed. These results were compared with the irritant responses of animals exposed to zinc oxide and sulfur dioxide mixed under different conditions of temperature and humidity.

Alpha-1-antitrypsin types in five Chinese national minorities.

The frequencies of alpha-1-antitrypsin alleles were determined for five Chinese national minorities: Uigur, Korean, Mongolian, Chuang, and Li. PI S and PI Z alleles are not found in the five populations studied. PI ETOK allele is present in Korean, Mongolian, and Chuang populations, and Etokyo is a very common alpha-1-antitrypsin variant in Chinese. Other alleles which occur in more than one of the minorities are PI X (Mongolian and Chuang) and PI M4 (Korean and Uigur). The high frequency of PI M4 in the Uigur population suggests a probably Occidental origin of this nationality.

Geographical variability of alpha-1-antitrypsin alleles in China: a study on six Chinese populations.

Alpha-1-antitrypsin phenotypes of six Chinese Han populations (20 degrees-45 degrees N. latitude) were determined. The frequency of allele M2 increases from North to South China. The north-south change of M2 appears to be mainly at the expense of alleles M1 and M3. Geographical variability of other variants was also observed. In the northern populations, the most common variants are M4 and Etokyo; in the southern populations, Pweishi, a variant which can not be distinguished electrophoretically from Pyasugi. These results form a distribution pattern of alpha-1-antitrypsin alleles in China.

Exposure-response relationship of bronchial mucociliary clearance in rabbits following acute inhalations of sulfuric acid mist.

Eight rabbits underwent 1 h oral inhalations of submicrometer sulfuric acid mist at concentrations ranging from approx. 100-1084 micrograms/m3, followed by measurement of the mucociliary clearance of a tracer aerosol from the bronchial tree. These data, plus those from a previous study, were used to construct an exposure concentration-response relationship for alterations in clearance produced by H2SO4. The response pattern is characterized by transient acceleration of clearance at low concentration exposures, and retardation at higher concentrations. In addition, comparison of these results with those from a similar study using human volunteers supports use of the rabbit as an appropriate model for studying mucociliary clearance alterations produced by inhaled irritants.

Study and models of total lead exposures of battery workers.

In an attempt to establish a more realistic and reliable model for relating environmental exposure measurements to the biological indices of exposure, a study was undertaken to quantify the total sources of lead exposure among lead storage battery workers. In addition to the usual personal and area lead air sampling, quantitative and repeatable measurements of removable lead from work surfaces and the workers' hands and faces were obtained daily for ten consecutive work days in the pasting and battery assembly departments. Mathematical correlations of blood lead and zinc protoporphyrin (ZPP) levels as the dependent variable with the lead exposure sources were derived and demonstrated most strongly as log-log relationships. Statistical analyses by computer programming indicated that the airborne, hand, facial and work surface levels have a high degree of inter-correlation with a very significant positive individual correlation with blood lead levels and a somewhat lower correlation with ZPP. The results suggest that contaminated personal and work surfaces may play a more significant role in toxic occupational and environmental exposures, generally, than had heretofore been demonstrated or suspected.

Response of the bronchial mucociliary clearance system in rabbits to inhaled sulfite and sulfuric acid aerosols.

A group of eight rabbits was exposed (orally) for 1 hr to a distilled water aerosol (sham control) or to submicrometer aerosols of either a transition metal sulfite complex (Fe(III)-S(IV], sodium sulfite (Na2SO3), or sulfuric acid (H2SO4). Mucociliary clearance was evaluated by external retention measurements of radioactively tagged tracer particles from the bronchial tree. Fe(III)-S(IV) in the range of 238 to 1227 micrograms/m3 (as SO2-3) produced no significant change from sham control in the mean residence time (MRT) of the tracer, indicating no effect upon mucociliary clearance rate. On the other hand, Na2SO3 at levels greater than or equal to 1200 micrograms/m3 (as SO2-3) resulted in clearance acceleration; the lack of effect of Fe(III)-S(IV) is possibly due to its stability. H2SO4 at 260 to 2155 micrograms/m3 produced a significant dose-related response, indicating clearance acceleration at low concentrations and a depression at higher levels.

Immunological phenotype of lymphomas induced by avian leukosis viruses.

The production of immunoglobulin by six cell lines derived from bursal tumors induced by avian leukosis virus follows two general patterns: (i) three cell lines that have been extensively passaged in culture synthesize and secrete light chains only; (ii) three cell lines that are recently isolated produce and secrete monomeric immunoglobulin M in addition to free light chains. All six cell lines synthesize and secrete both glycosylated and unglycosylated forms of light chain. We conclude that the cell lines established from lymphomas induced by avian leukosis virus represent relatively mature, but possibly abnormal, stages in the development of chicken B-lymphocytes. The immunoglobulin M produced by the cell lines failed to form detectable immune complexes with avian leukosis virus. It therefore appears that the immunoglobulin M is not directed against viral antigens and that autogenous antigenic stimulus cannot account for the sustained growth of the neoplastic B-lymphocytes.
