Pseudouridine and uridine in normal kidney and kidney cancer tissues.

The tissue concentrations of a modified nucleoside, pseudouridine, and a normal nucleoside, uridine, were measured with high-performance liquid chromatography. Human kidneys were obtained from five patients with renal cell carcinoma and divided into a noncancerous part and a cancerous part. The pseudouridine concentration in the cancerous part of the kidneys ranged between less than 2-2.8 nmoles/g and in the noncancerous part 4.3-19.4 nmoles/g (mean 10,9 nmoles/g). The uridine concentration in the cancerous and noncancerous parts of the kidney ranged between 19.6-179.1 nmoles/g (mean 110.7 nmoles/g) and 117.5-235.6 nmoles/g (mean 191.5 nmoles/g), respectively. The pseudouridine concentration appeared to be approximately seven times higher in the noncancerous part as compared to the cancerous part of the kidney. In the case of uridine, the difference was less pronounced.

Loneliness, birth order and social loss among a group of elderly women.

This paper is concerned with birth order as a childhood experience which may effect how and when older adults experience loneliness. In this study 207 elderly women (mean age 77) were studied in terms of birth order, social loss, depressive disorder and loneliness. As hypothesized, firstborns will experience less loneliness than laterborns under socially stable circumstances. But, after a social loss, firstborns are likely to experience more loneliness than laterborns. The effect of social loss and loneliness was also examined in terms of two health outcome variables--depressive disorder and physical complaints. No direct or indirect (via loneliness) effect of social loss on the health variables was found. Loneliness was found to have an effect on depressive disorder but not on physical complaints. Finally, for firstborns an interaction between social loss and loneliness was observed on physical complaint outcomes.

Alcohol occlusion of segmental renal arteries as alternative to kidney resection.

Treatment of postoperative urine leakage after a lower pole resection by embolic occlusions of segmental arteries using pure ethanol in a thirty-four-year-old man is described. There were no postoperative complications, and follow-up examination at two years showed a functioning but small kidney in a patient without symptoms.

Hemorrhagic infarctions of testis due to intimal fibroplasia of spermatic artery.

Two cases of hemorrhagic infarctions of the testis are presented. The infarctions were due to intimal fibroplasia of the branches of the spermatic artery; to our knowledge, a condition described only in the kidney. The differences between intimal fibroplasia and the lesions of polyarteritis nodosa and thromboangiitis obliterans are discussed.

Intervention against loneliness in a group of elderly women: an impact evaluation.

An intervention program, based on an interpretation of everyday loneliness as consisting of two parts--emotional and social estrangement--is discussed. The main variables were availability of a confidant, social comparison and personal control (the CCC-design). The intervention took the form of small group meetings. The sample consisted of elderly women living in Stockholm and interviews were held before, and 6 months after participation in the program. Results showed that subjects had less feelings of loneliness, and also less feelings of meaninglessness, more social contacts, higher self-esteem, greater ability to trust and lower blood pressure (both systolic and diastolic) after the intervention. Analyses showed that women with several years of adult employment on the same job experienced the greatest decrease in feelings of loneliness. Also, women who had had much contact with their grandparents, and women who had experienced a serious or protracted illness in the family during childhood, had the greatest decline in blood pressure. It was concluded that change in blood pressure operates through some other mechanism than the one which effects feelings of loneliness. Finally, a model is presented for distinguishing between different levels of intimacy.

Interferon therapy in disseminated renal cell carcinoma.

Twenty patients with pulmonary metastases of renal cell carcinomas have been treated in a randomized study with either leucocyte interferon in daily doses of 3 x 10(6) I.U.i.m or irradiation of both lungs in a calculated mean central dose of 10 Gy in 4 weeks in combination with bleomycin and vincristine. Most patients had a short time to progression of their disease. Three patients in the interferon group had complete response (CR) and partial response (PR) and one patient in the other group had PR. The numbers of patients in both arms of the study are too small to allow any conclusions whether beneficial effects are more frequent in one group than in the other, but there is a more favourable tendency in the interferon group.

Irradiation therapy with multiple small fractions per day in urinary bladder cancer.

In a clinical trial 168 patients with carcinoma of the bladder, T2-T4, were randomized to one of two treatments; 1 Gy 3 times a day to a total of 84 Gy or 2 Gy once a day to a total of 64 Gy. Local eradication of the tumour in the bladder cystoscopically and cytologically at 6 months after completion of treatment and patient survival were analyzed. The results favoured significantly the patients treated with 84 Gy. All patients were followed 5-9 years. The survival was significantly improved in patients with T3 lesions treated with 84 Gy (p less than 0.01). Complications in the bowel requiring surgical treatment were not significantly different between the two groups of patients. The results indicate a therapeutic gain by hyperfractionated radiotherapy in comparison to conventional fractionated radiotherapy.

The estimation of blood group gene frequencies: a note on the allocation method.

The principles for estimating gene frequencies in complex blood group systems by the allocation method are discussed. This numerical method is simpler than the maximum likelihood method, but gives the same estimate. It is shown that there are some cases in which the application of a previous description of the method gives incorrect results. The principles are discussed for how family information may be included in the estimation process in order to obtain more precise estimates.

Effect of a homologous series of halogenated methanes on pulmonary uptake of 5-hydroxytryptamine in isolated perfused rat lung.

The potency of halogenated methanes to inhibit uptake of 5-hydroxytryptamine (5-HT) from the pulmonary circulation was studied using an isolated, perfused and ventilated rat lung preparation. The agents were vaporized and mixed with the inlet air. The results indicate that the degree of chlorination is the most important factor for potency of the methanes to inhibit lung uptake of 5-HT. When hydrogen was substituted with fluorine the potency was decreased dramatically. Bromine seemed to have the opposite effect. The data also suggested that the degree of chlorination was more important rather than hydrophobic/hydrophilic balance of the solvent molecule. These effects seem to be correlated with the narcotic effects of the substances studied.

Genetic variation of haemoglobin alpha- and beta-chains in rabbits detected by isoelectric focusing and reversed-phase chromatography.

Previous studies on the amino acid sequences and on the amino acid composition of peptides revealed genetic polymorphism both of the haemoglobin alpha-chain (Hb alpha) and beta-chain (Hb beta) in rabbits. In this study, rabbit haemolysates were analysed by isoelectric focusing in a narrow pH range (6.7-7.7) and by reversed-phase chromatography. Two variants were found for both Hb alpha and Hb beta. The two methods detected the same variants in this material. Inheritance data were consistent with the hypothesis that the observed Hb alpha and Hb beta variants were each controlled by two codominant, autosomal alleles. Haemoglobin polymorphism appears to be frequent in domestic rabbits since both variants of each chain were observed in all the three breeds studied.

Fractionation of human serum proteins by immobilized metal affinity chromatography.

Human serum was fractionated on zinc and cadmium iminodiacetate and tris(carboxymethyl)ethylenediamine chelates. Zinc iminodiacetate was found to be useful in the isolation of hemopexin and alpha 2-macroglobulin. The chromatographic behaviour of serum proteins on zinc iminodiacetate could be modulated by the nature of the eluent; a sulphate-containing solvent increased the binding of serum proteins to immobilized metal ions.

International overview on the treatment of prostate cancer--Scandinavian experience.

The Scandinavian countries, especially Sweden and Norway, have a high incidence of prostate cancer. The tumor classification has been mainly uniform in recent years, according to the TNM system. Transrectal fine-needle biopsy and cytologic malignancy grading were introduced in Sweden and were gradually accepted in other countries. The treatment of prostate cancer has been mainly conservative as it is in most parts of Europe. For a long period endocrine therapy was given in the vast majority of cases. In recent years radiotherapy has also been used in cancer of low differentiation with no evidence of dissemination. In poorly differentiated and disseminated disease, Estracyt is the standard therapy. Estracyt is also given in disseminated carcinoma nonresponsive to ordinary hormone therapy. In this category of patients other chemotherapy is the second therapeutic alternative. Various techniques of endocrine therapy, chemotherapy, and radical surgery are subject to controlled trials.

The healing process of anastomoses of the colon. A comparative study using single, double-layer or stapled anastomosis.

In spite of modern suture materials and different techniques in colonic anastomoses after resection, leakage is still the most feared complication in colonic surgery. In female pigs of Swedish land breed, standardized 5-cm long colonic resection was performed 10 cm above the peritoneal deflection, using either a single layer of Gambee-stitches (n = 6, Vicryl 4-0), two-layer interrupted stitches (n = 6, Vicryl 4-0) or the intraluminal stapling device (n = 6, ILS Ethicon). After one week, the animals were sacrificed and an anastomotic index was calculated using in vitro x-ray. Also, anastomotic circulation (calculated by the microsphere technique), breaking strength, and histologic evaluation were performed. All animals survived and no leakage was observed. The time to perform the anastomosis was significantly shorter (P less than 0.05) for the stapling device compared with the manual techniques used. The anastomotic index was lower (P less than 0.05) for two rows (0.24) compared with Gambee-stitches (0.38) or stapler anastomoses (0.37). There were no differences in blood flow among the three groups and no differences in breaking strength. Macroscopic investigation revealed mucosal defects in two of the stapled anastomoses and histologic investigation showed small areas of necrosis. The stapling device did not induce any inflammatory reaction. On the other hand, a severe inflammatory reaction was seen when using conventional suture materials. In conclusion, this study shows that a single row of Gambee-stitches is equal to the ILS stapling device when performing colonic anastomoses and these two methods seem to be superior to the two-layer technique.
