Label-free protein biosensor based on aptamer-modified carbon nanotube field-effect transistors.

We have fabricated label-free protein biosensors based on aptamer-modified carbon nanotube field-effect transistors (CNT-FETs) for the detection of immunoglobulin E (IgE). After the covalent immobilization of 5'-amino-modified 45-mer aptamers on the CNT channels, the electrical properties of the CNT-FETs were monitored in real time. The introduction of target IgE at various concentrations caused a sharp decrease in the source-drain current, and a gradual saturation was observed at lower concentrations. The amount of the net source-drain current before and after IgE introduction on the aptamer-modified CNT-FETs increased as a function of IgE concentration. The detection limit for IgE was determined as 250 pM. We have also prepared CNT-FET biosensors using a monoclonal antibody against IgE (IgE-mAb). The electrical properties of the aptamer- and antibody-modified CNT-FETs were compared. The performance of aptamer-modified CNT-FETs provided better results than the ones obtained using IgE-mAb-modified CNT-FETs under similar conditions. Thus, we suggest that the aptamer-modified CNT-FETs are promising candidates for the development of label-free protein biosensors.

Label-free immunosensor for prostate-specific antigen based on single-walled carbon nanotube array-modified microelectrodes.

We have fabricated a label-free electrochemical immunosensor using microelectrode arrays modified with single-walled carbon nanotubes (SWNTs). Label-free detection of a cancer marker, total prostate-specific antigen (T-PSA), was carried out using differential pulse voltammetry (DPV). The current signals, derived from the oxidation of tyrosine (Tyr), and tryptophan (Trp) residues, increased with the interaction between T-PSA on T-PSA-mAb covalently immobilized on SWNTs. The selectivity of our biosensor was challenged using bovine serum albumin (BSA) as the target protein. The detection limit for T-PSA was determined as 0.25 ng/mL. Since the cut-off limit of T-PSA between prostate hyperplasia and cancer is 4 ng/mL, the performance of our label-free electrochemical immunosensor seems promising for further clinical applications.

Anomalously large formula unit volume and its effect on the thermal behavior of LiBF4.

The crystal structure of LiBF4 has been determined by single-crystal X-ray diffraction measurements. LiBF4 crystallizes as a merohedral twin in the trigonal space group P3(1)21 with a=4.892(5) A, c=11.002(12) A, V=228.0(4) A3, and Z=3 at 200 K. The twin is generated by a 2-fold rotation about the [10] direction. The lithium cation is coordinated by four fluorine atoms in a distorted tetrahedral manner, wherein two Li-F distances of 1.862(5) and 1.846(5) A are observed. The formula unit volume (FUV=V/Z) of 77.9 A3 for LiBF4 at 298 K is considerably larger than 72.7 A3 for NaBF4 and 72.5 A3 for AgBF4, despite the smaller size of Li+, indicating loose ionic packing of LiBF4. The thermodynamic evaluation of the decomposition temperature for LiBF4 was performed using the empirical relationship between the standard entropy and the FUV obtained. The results indicate that the large FUV of LiBF4 contributes to its higher decomposition temperature compared to that of LiPF6.

[Malignant melanoma].

There have been many prospective randomized clinical trials of IFNalpha as post-surgical adjuvant therapy for high-risk melanoma patients. High dose IFNalpha therapy(HDI) has been reported to be effective in disease-free survival (DFS). However, the usefulness of HDI therapy is still controversial due to its grave toxicity. Low dose IFNalpha therapy (LDI) has no consistent effects on DFS. Most trials showed little activity in patients with metastatic melanoma. In Japan, IFNbeta, in combination with chemotherapy, has been used as post-surgical adjuvant therapy, and has been considered to improve the prognosis of stage III melanoma patients. In addition, we have recently performed a phase I/IIa trial of IFNbeta gene therapy for advanced melanoma patients.

Intratumoral injection of immature dendritic cells enhances antitumor effect of hyperthermia using magnetic nanoparticles.

Dendritic cells (DCs) are potent antigen-presenting cells that play a pivotal role in regulating immune responses in cancer and have recently been shown to be activated by heat shock proteins (HSPs). We previously reported that HSP70 expression after hyperthermia induces antitumor immunity. Our hyperthermia system using magnetite cationic liposomes (MCLs) induced necrotic cell death that was correlated with HSP70 release. In the present study, we investigated the therapeutic effects of DC therapy combined with MCL-induced hyperthermia on mouse melanoma. In an in vitro study, when immature DCs were pulsed with mouse B16 melanoma cells heated at 43 degrees C, major histocompatibility complex (MHC) class I/II, costimulatory molecules CD80/CD86 and CCR7 in the DCs were upregulated, thus resulting in DC maturation. C57BL/6 mice bearing a melanoma nodule were subjected to combination therapy using hyperthermia and DC immunotherapy in vivo by means of tumor-specific hyperthermia using MCLs and directly injected immature DCs. Mice were divided into 4 groups: group I (control), group II (hyperthermia), group III (DC therapy) and group IV (hyperthermia + DC therapy). Complete regression of tumors was observed in 60% of mice in group IV, while no tumor regression was seen among mice in the other groups. Increased cytotoxic T lymphocyte (CTL) and natural killer (NK) activity was observed on in vitro cytotoxicity assay using splenocytes in the cured mice treated with combination therapy, and the cured mice rejected a second challenge of B16 melanoma cells. This study has important implications for the application of MCL-induced hyperthermia plus DC therapy in patients with advanced malignancies as a novel cancer therapy.

Application of carbon nanotubes for detecting anti-hemagglutinins based on antigen-antibody interaction.

Carbon nanotube sensors detected anti-hemagglutinin binding to immobilized hemagglutinins. An ultra-sensitive detection method for antibodies or antigens in serum is required. Hemagglutinins were immobilized on the reverse side of a carbon nanotube, thereby producing a source and a drain. Electrode pads covered each edge of the nanotube. The I-V curves between the source and the drain were measured after incubation of anti-hemagglutinins with immobilized hemagglutinins in a buffered solution on the reverse side of the nanotube. The sensitivity of the CNT sensor was higher than that of an ELISA system. This method constitutes a new tool to analyze interaction among biomolecules on a substrate.

The effect of the anion fraction on the physicochemical properties of EMIm(HF)nF (n = 1.0-2.6).

A series of molten salts EMIm(HF)nF's with different n values has been synthesized by the reaction of EMImHF(2) and anhydrous hydrogen fluoride. The salts contain EMIm cation and some oligomeric fluorohydrogenate anions, (HF)nF-, of which the fraction changes with the change of n. A phase diagram of EMIm(HF)nF's (n = 1.0-2.6) has been constructed which suggests the presence of the stoichiometric compounds, EMIm(HF)1.5F and EMIm(HF)2F, in this range. Compared to the EMIm(HF)2.3F previously reported, EMIm(HF)nF's (n = 1.8-2.0) possess wider liquid temperature ranges because of their similar melting points and superior thermal stabilities at elevated temperatures. The electrochemical windows of EMIm(HF)nF's (n = 1.0-2.6) falls in the range of 2.9-3.4 V. The conductivity of EMIm(HF)nF's (n = 1.0-2.6) increases with the increase of n.

Ionization state and ion migration mechanism of room temperature molten dialkylimidazolium fluorohydrogenates.

The ionization state of room temperature molten salts, alkylimidazolium fluorohydrogenates (RMIm(HF)(2.3)F: R = alkyl group, M =methyl group), was evaluated from the observed diffusion coefficient and viscosity, using the Stokes-Einstein relation. Assuming that the dissociation degree of the salt is acceptable for representation of the ionic state of the molten salts, the larger the cation size with elongation of the alkyl chain was, the higher the dissociation degree of the salt. Further, we proposed that an idea of the "degree of ordering of cations and anions" was more suitable to represent the ionization state without solvent species. On the basis of this idea, the smaller the cation size of RMIm(HF)(2.3)F salt was, the higher the ordering of the ion, indicating formation of domain particles of aggregated ions as a unit of mobile species such as A(AX)m+ and X(AX)n- for A+ X- salt. It was found that highly ordered particles, with large numbers for m and n, showed a high diffusion coefficient.

Mutation analysis of the ADAR1 gene in dyschromatosis symmetrica hereditaria and genetic differentiation from both dyschromatosis universalis hereditaria and acropigmentatio reticularis.

Dyschromatosis symmetrica hereditaria (DSH) (also called "reticulate acropigmentation of Dohi") is a pigmentary genodermatosis of autosomal dominant inheritance. We have clarified for the first time four pathological mutations of the double-stranded RNA-specific adenosine deaminase gene (ADAR1 or DSRAD) in four DSH pedigrees. In this paper, we report 16 novel mutations containing six missense substitutions (p.V906F, p.K1003R, p.G1007R, p.C1036S, p.S1064F, p.R1078C), two splice site mutations (IVS2+2T>G, IVS8+2T>A), six frameshift mutations (p.H216fs, p.K433fs, p.G507fs, p.P727fs, p.V955fs, p.K1201fs), and two nonsense mutations (p.R426X, p.Q600X) found in Japanese patients with DSH. We did not establish any clear correlation between the clinical phenotypes and the genotypes of ADAR1 gene mutations in our examination of 16 cases plus four pedigrees. None of the different mutations identified in our studies of 20 cases suggested any founder effect. Furthermore, we did not identify any mutations in the ADAR1 gene of three patients with dyschromatosis universalis hereditaria or three patients with acropigmentatio reticularis, indicating that the two diseases are completely different from DSH, although they have sometimes been suggested to be phenotypical variations of DSH.

Heat immunotherapy using magnetic nanoparticles and dendritic cells for T-lymphoma.

Dendritic cells (DCs) are potent antigen-presenting cells that play important roles in regulating immune responses in cancer. Immunotherapy using these immunocytes has become an accepted therapeutic modality. We previously reported that hyperthermia using magnetic nanoparticles induces antitumor immunity, which could be activated by adjuvant including cytokines. In the present study, we investigated the therapeutic effects of hyperthermia combined with DC immunotherapy on mouse EL4 T-lymphoma. Magnetite cationic liposomes (MCLs) have a positive surface charge and generate heat in an alternating magnetic field (AMF) due to hysteresis loss. MCLs were injected into an EL4 nodule in C57BL/6 mice, which were subjected to AMF for 30 min. The temperature at the surface of the tumor reached 45 degrees C and was maintained by controlling the magnetic field intensity. Hyperthermia treatment was repeated twice with 24 h intervals. After hyperthermia, immature DCs were directly injected into the EL4 nodule. As a result, complete regression of tumors in 75% (6/8) of the mice was observed, while the percentage of complete regression of tumors was 12.5% (1/8) in the case of mice treated by hyperthermia alone. This novel cancer therapy, which we have termed "heat immunotherapy", may be applicable to patients with advanced malignancies.

Apoptosis-like cell death of human breast cancer cell line MCF-7 induced by buprenorphine hydrochloride.

The analgesic buprenorphine hydrochloride (Bph) induced apoptosis-like cell death in the caspase-3-deficient human breast cancer cell line, MCF-7. This apoptosis-like cell death activated key molecules in the mitochondrial apoptotic pathway: cytochrome c, caspase-9, caspase-7, and caspase-6. Bph caused the release of fluorescent protein from the mitochondria of MCF-7 cells transfected with the pDsRed2-Mito-vector in a time-dependent manner, suggesting disruption of the mitochondrial membrane. Zn(2+) as high as 2 mM did not inhibit the DNase that took part in this apoptosis. Thus, this unidentified DNase might resemble other DNases involved in apoptosis-like cell death whose activity is not inhibited by zinc ion.

Number of acquired melanocytic nevi in patients with melanoma and control subjects in Japan: Nevus count is a significant risk factor for nonacral melanoma but not for acral melanoma.

BACKGROUND: Epidemiologic studies have suggested that number of acquired melanocytic nevi is a risk factor for melanoma development in Japanese as it is in white populations. However, there are only a few population-based studies on acquired nevi in Asian populations, and no epidemiologic study on relationship between number of acquired nevi and melanoma in Japanese populations has been reported. OBJECTIVE: The purpose of this study was to assess number, size, and distribution of acquired melanocytic nevi in a Japanese population. Particular attention was paid to evaluation of relationship between number of acquired nevi and development of nonacral or acral malignant melanoma. METHODS: In all, 82 patients with malignant melanoma and 600 control subjects were included in this study. All participants were Japanese. The number of acquired melanocytic nevi, 2 mm or larger in diameter, on the whole body except the scalp and genital areas was counted by experienced dermatologists. The participants were divided into 5 age categories (0-19, 20-39, 40-59, 60-79, and >80 years old) for the statistical analyses. This categorization adjusted the age and sex distribution between patients with melanomas and control subjects in 40- to 59-, 60- to 79-, and over 80-year-old groups. RESULTS: In the control Japanese population, the number of acquired melanocytic nevi on the whole body increased with age in 0- to 19-year-old age group and reached the highest number, 6.7 +/- 8.1/person, in 20- to 39-year-old group. In patients with nonacral melanoma, the number of acquired nevi on the whole body in 40- to 59- and 60- to 79-year-old groups was significantly higher than that of the corresponding control group. In contrast, the rate of individuals who had acquired nevi on soles, palms, and nail apparatus was not significantly different between acral melanoma group and the control group in 40- to 59- and 60- to 79-year-old groups. CONCLUSION: This study has revealed that a large number of acquired melanocytic nevi is a risk factor for the development of nonacral melanoma in Japanese and white populations. However, acquired nevi on soles, palms, and nail apparatus do not seem to be a risk factor for acral melanoma in Japanese populations.

Skin infiltration of juvenile myelomonocytic leukemia.

We described a two-year-old boy who developed a skin infiltration from JMML. Several indurated erythematous lesions were seen on his back on his first visit to our department. Edematous erythemas had repeatedly appeared on his auricles and feet for the previous six months. He had had a high fever for a month. Hepatosplenomegaly and superficial lymphadenopathy were recognized. Laboratory investigation showed leukocytosis and anemia. The diagnosis of JMML was confirmed by the findings of myeloid hyperplasia in his bone marrow and the spontaneous colony formation and GM-CSF hypersensitivity in a culture of bone marrow cells. Histopathologically, large atypical mononuclear cells were infiltrated throughout the dermis in a perivascular and interstitial distribution in a skin biopsy specimen. These cells were CD3 (-), CD20 (-), CD45 (+), CD68 (+) and myeloperoxidase (+). Bone marrow transplantation and then cord blood stem cell transplantation were performed but soon rejected. The indurated erythematous lesions appeared again soon after the relapse of JMML. There are other reported cases of JMML with skin infiltration that preceded any other manifestations of the disease. JMML cells in some patients, including our case, seem to have a great affinity for the skin, and skin biopsy aids in early detection of this disease.

Syntheses, structures and properties of 1-ethyl-3-methylimidazolium salts of fluorocomplex anions.

Fluoroacid-base reactions of a room-temperature ionic liquid, 1-ethyl-3-methylimidazolium fluorohydrogenate (EMIm(HF)2.3F, EMIm = 1-ethyl-3-methylimidazolium cation), and Lewis fluoroacids (BF3, PF5, AsF5, NbF5, TaF5 and WF6) give EMIm salts of the corresponding fluorocomplex anions, EMImBF4, EMImPF6, EMImAsF6, EMImNbF6, EMImTaF6 and EMImWF7, respectively. Attempts to prepare EMImVF6 by both the acid-base reaction of EMIm(HF)2.3F with VF5 and the metathesis of EMImCl with KVF6 failed due to the strong oxidizing power of the pentavalent vanadium, whereas EMImSbF6 was successfully prepared only by the metathesis of EMImCl and KSbF6. EMImBF4, EMImSbF6, EMImNbF6, EMImTaF6 and EMImWF7 are liquids at room temperature whereas EMImPF6 and EMImAsF6 melts at around 330 K. Raman spectra of the obtained salts showed the existence of the EMIm cation and corresponding fluorocomplex anions. IR spectroscopy revealed that strong hydrogen bonds are not observed in these salts. EMImAsF6(mp 326 K) and EMImSbF6(mp 283 K) are isostructural with the previously reported EMImPF6. The melting point of the hexafluorocomplex EMIm salt decreases with the increase of the size of the anion (PF6- < AsF6- < SbF6- <NbF6- approximately TaF6-).

Effects of selected food factors with chemopreventive properties on combined lipopolysaccharide- and interferon-gamma-induced IkappaB degradation in RAW264.7 macrophages.

Degradation of IkappaB (IkappaB) is a key step for nuclear factor-kappaB (NF-kappaB)-induced transcription of certain proinflammatory genes, including inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2. We selected seven chemopreventive agents and examined their effects on combined lipopolysaccharide- and interferon-gamma-induced IkappaB degradation in RAW264.7 murine macrophages. IkappaB degradation was notably suppressed by 1'-acetoxychavicol acetate (ACA), zerumbone (ZER), and benzylisothiocyanate (BITC), however, not by auraptene (AUR), while the suppressive potencies of nobiletin (NOB), genistein (GEN), and resveratrol (RES) were low, but significant. These results suggest that ACA, ZER, and BITC suppress iNOS/COX-2 gene expression mainly by attenuating IkappaB degradation, while other chemopreventive agents use alternative pathway(s) to suppress the expression of proinflammatory genes.

Feasibility of a curvature-based enhanced display system for detecting cerebral aneurysms in MR angiography.

PURPOSE: To assess the feasibility of a curvature-based enhanced display system for detecting cerebral aneurysms in MR angiography. METHODS: MR angiography studies of 18 patients (eight male and 10 female, average age 65.7, age range 50 to 75 years old) with 23 known aneurysms were evaluated with a curvature-based display system. The two curvature features-the volumetric shape index and curvedness values-were calculated at each voxel. These were displayed independently on a workstation, overlaid on volume-rendered images. Two neuroradiologists evaluated the images for visibility and diagnosis of the cerebral aneurysms. The diagnostic results were compared with the original reports. RESULTS: The calculation time for each curvature index was 30 to 40 s for 120 to 140 slices of original MR angiography data. Shape index images emphasized smooth and round aneurysms more than aneurysms with irregular surfaces. Curvedness images revealed aneurysms well when the aneurysms had diameters that differed from those of the surrounding vessels. The computer-assisted-detection method detected 24 aneurysms, three of which were not pointed out in the initial report. CONCLUSION: Our results show that the curvature-based display system we have developed is feasible and that it may help to detect small aneurysms that are prone to be overlooked in routine readings.
