Decreased superoxide dismutase-2 activity in a patient with ring chromosome 6.

Manganese superoxide dismutase (SOD-2) activity was measured in erythrocytes and leukocytes of a patient with ring chromosome 6. Compared to normal control individuals, SOD-2 activity in leukocytes of the patient was lower, whereas cuprozinc SOD (SOD-1) activity was normal. These findings suggest the cause of decreased enzyme activity is the effect of the ring form on production of the enzyme or inherent instability of ring chromosomes.

[Study on relationships between sonographic endometrial images and ovarian hormone levels].

Sixty-seven and 109 sonographic endometrial images obtained from 67 infertile patients and 5 healthy volunteers with normal ovulatory cycles, respectively, were investigated with regard to serum estradiol (E2) and progesterone (P) levels. The ultrasound devices used were sector electronic scanners (3.5 MHz). 1) Endometrial images were able to be classified into four pattern types, according to the criteria reported by Sakamoto (1985), using a gray-scale contact compound scan. Pattern 1: linear cavity echo, pattern 2: thick hypoechoic endometrium surrounded by thin border echo, pattern 3: thickened border echo, pattern 4: entire echogenic endometrium. 2) Both E2 and P levels were low in pattern 1. The E2 level was significantly higher in pattern 2 than in pattern 1 (p less than 0.01), indicating pattern 2 reflects E2 surges prior to ovulation. The P level was high in pattern 3; and in pattern 4, it was significantly higher than in patterns 1 and 2 (p less than 0.01), indicating that patterns 3 and 4 are in the early and mid-luteal phase, respectively. 3) Sonograms obtained from 12 cases with suspicious luteal phase defects showed an abnormal pattern in the mid-luteal phase. These results indicate that the sonographic endometrial images reflect respective ovarian hormone levels.

Ring chromosome 21 and SOD activity of blood cells.

A female infant was found to have an abnormal karyotype with mosaicism: 45,XX,-21/46,XX,r(21) (p11.2 q22.3). She showed a slightly decreased level of superoxide dismutase (SOD) activity in erythrocytes, polymorphonuclear and mononuclear leukocytes. The cuprozinc SOD (SOD-1) activity of leukocytes was 70-80% of normal level, while their manganese SOD (SOD-2) activity was within the normal range. The relationship between the chromosomal aberration and the SOD activity is discussed in relation to the gene dosage effect.

Significance of erythrocyte lipid peroxidation and superoxide dismutase activity in a patient with idiopathic pulmonary haemosiderosis.

In a patient with idiopathic pulmonary haemosiderosis, lipid peroxidation of erythrocytes and their superoxide dismutase activity were determined. The latter was found to be decreased, and the erythrocytes showed easy peroxidisability, which was shown by vulnerability to hydrogen peroxide and a marked elevation of iron-catalysed lipid peroxidation in addition to a higher basal malondialdehyde level, an end product of lipid peroxidation. These findings were correlated with haemolysis, iron-deficiency anaemia and a shortened survival of erythrocytes. Although the aetiological significance of our observations still remains to be clarified, it appears that antioxidant enzymes and the peroxidative reactivity of erythrocytes should be carefully evaluated in other patients with idiopathic pulmonary haemosiderosis.

Zinc, copper, manganese, and selenium metabolism in thyroid disease.

This study was designed to evaluate trace metal metabolism in adults with thyroid diseases. Erythrocyte zinc values were significantly lower than normal in hyperthyroidism and higher in hypothyroidism. A significantly higher than normal urinary excretion of zinc was observed in hyperthyroidism. The mean concentrations of plasma and erythrocyte copper were significantly above normal in hyperthyroidism. Plasma selenium levels were significantly lower than normal in hyperthyroidism. No statistically significant difference was found in plasma zinc, erythrocyte manganese, or urine copper values between patients with thyroid diseases and healthy controls. The erythrocyte manganese content correlated well with thyroxine and triiodothyronine levels. Plasma prealbumin and retinol-binding protein correlated well with the erythrocyte zinc content but not with plasma zinc levels. There was no correlation between erythrocyte superoxide dismutase activity and erythrocyte copper or zinc concentrations. The results of this study suggest that the metabolism of zinc, copper, manganese, and selenium is abnormal in thyroid diseases.

Superoxide dismutase activity in leukemic blasts of children with acute leukemia.

Superoxide dismutase activity was determined in leukemic blasts of 24 patients with acute leukemia and was correlated to its type. The activity was significantly lower in five patients with T cell or B cell leukemia, and there was a wide range of distribution in 15 patients with non-T, non-B cell leukemia. Manganese superoxide dismutase activity was significantly lower in all types of acute lymphoblastic leukemia than in normal mononuclear leukocytes. Both total and manganese superoxide dismutase activities in acute myeloblastic leukemia, however, were significantly higher than those in normal polymorphonuclear leukocytes. The prognosis in patients with a low superoxide dismutase activity in leukemic blasts seemed to be poorer. Determination of superoxide dismutase activity in leukemic blasts may have a prognostic value in the management of children with acute leukemia.

Decreased superoxide dismutase activity of erythrocytes and leukocytes in Fanconi's anemia.

Superoxide dismutase (SOD) activity was determined in the erythrocytes and leukocytes of 5 patients with Fanconi's anemia (FA) and 1 with atypical Fanconi's anemia without any hematological disorder. SOD activity was decreased in the blood cells of the patients with Fanconi's anemia, but was normal in the atypical patient. The influence of SOD on the chromosome aberrations and hematological disorder in FA is discussed.

Favorable effect of dimethyl sulfoxide on secondary amyloidosis in juvenile rheumatoid arthritis.

A girl with secondary amyloidosis as a complication of juvenile rheumatoid arthritis was administered dimethyl sulfoxide by topical application to the skin. Her gastrointestinal symptoms and massive proteinuria improved. Decreased left ventricular function and creatinine clearance also improved remarkably. The favorable effect of dimethyl sulfoxide in this single patient deserves further study in a controlled trial.

Vitamin D metabolism in osteogenesis imperfecta during calcitonin therapy.

The effect of calcitonin on plasma concentrations of 25-hydroxyvitamin D (25-OHD) and 1,25-dihydroxyvitamin D [1,25-(OH)2D] was studied in six patients with osteogenesis imperfecta. The mean pretreatment value of plasma 1,25-(OH)2D was significantly higher than the mean value of age-matched control subjects (P less than .05). High or normal plasma levels of 1,25-(OH)2D before calcitonin therapy were decreased after 1 month of therapy and remained normal thereafter in all six patients. Plasma 25-hydroxyvitamin D concentrations, which were normal before calcitonin injection, remained normal during calcitonin administration. These results indicate that there may be acute and chronic effects of calcitonin on vitamin D metabolism.

A case of 21q-syndrome with half normal SOD-1 activity.

A male Japanese infant was found to have a chromosomal aberration of del(21)(qter leads to q22.1-2) and decreased superoxide dismutase (SOD) activity in erythrocytes and polymorphonuclear and mononuclear leukocytes. The cuprozinc enzyme (SOD-1) level was 40-50% of normal, while the cyanide-insensitive manganese enzyme (SOD-2) activity was within the normal range. Determination of SOD activity in blood cells is a valuable method of classification of the syndrome.

A case of hypophosphataemic vitamin D-resistant rickets treated with initial massive doses of 1 alpha-hydroxy-vitamin D3 alone.

A 1 5/12-year-old girl with hypophosphataemic vitamin D-resistant rickets was treated with initial massive doses of 1 alpha-hydroxy-vitamin D3 (1 alpha-OH-D3) alone. A dramatic improvement of bone lesions and growth rate and normalization of the characteristic biochemical defects were noted within four months after the start of massive 1 alpha-OH-D3 therapy. The results of this study suggest that early therapy with massive doses of 1 alpha-OH-D3 alone improves the radiological findings and restores to normal the biochemical defects and the growth rate as early as possible. This regimen may improve motor strength and endurance and obviate the need for corrective orthopaedic procedures.

[Studies on relationship between shape of uterine cavity on hysterosalpingography and menstrual disorder].

Eight hundred and twenty five infertility patients were investigated to reassess the association between uterine mild anomaly and menstrual disorder proposed as a new syndrome by Sørensen. A hysterosalpingogram we classified the shape of the uterine cavity into 4 types: Normal uterus, T-shaped uterus, arcuate uterus and others (included septate uterus, bicornuate uterus,--), and also classified the size of the uterine cavity into 3 groups; large uterus, medium-sized uterus and small uterus. After classification of uterine shape and size, menstrual disorder in each patient was reviewed and the correlation between uterine anomaly and menstrual disorder was examined. Five hundred and twenty eight out of 770 patients (68.6%) were assigned to normal uterus, 75(7.9%) to T-shape uterus, 148(19.2%) to arcuate uterus and 19(2.5%) to others. Their incidences of menstrual disorder were 31.4%, 37.2%, 39.2% and 42.1%, respectively, and no significant difference was found among them. There were 55 patients (7.3%) with a large uterus, 655(87.2%) with a medium-sized uterus and 41(5.5%) with a small uterus. The incidences of menstrual disorder were 16.4%, 35% and 34.1%, respectively, and there was significant difference in incidence between large uterus and medium-sized uterus. The uterine fundal depression index (FDI) was used as a parameter to evaluate uterine mild anomaly. The incidences of menstrual disorder were 29.8% in the FDI-0 group, 32.9% in the FDI-1 approximately 10 group, 42.7% in the FDI-11 approximately 20 group, 45.5% in the FDI-21 approximately 30 group and 22.2% in the FDI-over 31 group. However, it was difficult to find a difference in the pregnancy rate or delivery rate among different classifications of uterine shape, size or FDI. From these results, it can be said that mild uterine anomaly is hardly associated at all with menstrual disorder clinically.
