Comparative activity of the topical quinolone OPC-7251 against bacteria associated with acne vulgaris.

The antibacterial activity of the topical quinolone OPC-7251 against bacteria commonly found in acne vulgaris was tested in vitro by an agar dilution method. The MIC50 was 0.25 mg/l for Propionibacterium acnes, 0.125 mg/l for Propionibacterium granulosum, 0.03 mg/l for Staphylococcus aureus and 0.06 mg/l for coagulase-negative staphylococci. Compared with seven other antibiotics tested (ciprofloxacin, penicillin, erythromycin, tetracycline, clindamycin, fusidic acid and gentamicin), OPC-7251 had potent activity against both propionibacteria and staphylococci and the lowest incidence of resistant strains.

Urease production by Helicobacter (Campylobacter) pylori.

Urease activity of 50 Helicobacter pylori (H. pylori) strains was assessed employing a photometric assay. Urea hydrolysis reached a maximum in the late log-phase and during the plateau phase of bacterial growth. The reaction time of H. pylori urease was significantly shorter than that of other urease producing bacteria (P. mirabilis, P. vulgaris, K. pneumoniae, K. oxytoca). Increasing the reaction temperature hardly led to an acceleration of the quick urea hydrolysis of H. pylori, in contrast to the situation with P. mirabilis. Acetohydroxamic acid showed a dose-dependent non-competitive suppression of urease production, whereas 9 antibiotics in subinhibitory concentrations did not influence urease production of H. pylori.

Quantum yield of CHAPSO-solubilized rhodopsin and 3-hydroxy retinal containing bovine opsin.

The quantum yields of bleaching for two artificial pigments, bovine opsin combined with (3R)-3-hydroxy retinal or (3R,S)-3-methoxy retinal, were determined in comparison to the value for regenerated bovine rhodopsin. Regeneration of the visual pigments was performed by incubation of 3-[(3-Cholamidopropyl)-dimethylammonio]-2-hydroxy-1- propanesulfonate (CHAPSO)-solubilized opsin with the 11-cis isomers of retinal and the respective retinal derivatives. The extinction coefficients of the pigments in CHAPSO were determined to 35,000 M-1 cm-1 (native rhodopsin), 35,300 M-1 cm-1 (regenerated rhodopsin) and 34,500 M-1 cm-1 (3-OH retinal opsin). With respect to rhodopsin (lambda max: 500 nm), the pigments carrying the substituted chromophores exhibit blue shifted absorbance maxima (3-hydroxy and 3-methoxy retinal opsin: 488 nm). In parallel experiments under absolutely identical conditions we find related to the value of CHAPSO solubilized rhodopsin (identical to 1) a quantum efficiency of bleaching for the 3-hydroxy pigment of 1.2.

Effect of bismuth subcarbonate against Campylobacter pylori: do citrate ions improve antibacterial activity?

The minimum inhibitory concentrations of three bismuth subcarbonate preparations: 1. bismuth subcarbonate, 2. a mixture of 50% bismuth subcarbonate and 50% citric acid, 3. a mixture of 50% bismuth subcarbonate and 50% potassium citrate were assayed against 30 strains of Campylobacter pylori employing the agar dilution method. Bismuth subcarbonate alone exhibited an MIC50 of 2 mg/l and was more effective than the two combinations (MIC50 4 mg/l). It is concluded that the addition of citrate ions does not improve antibacterial activity.

[A computer-assisted system for diagnosing rhinologic function].

We describe the new computerized rhinologic diagnostic system "CARIMA", a software-controlled system based on a measuring head and a PC. The currently available software allows active anterior rhinomanometry to be carried out with a special program for the nasal allergen test, as well as active posterior rhinomanometry and the determination of the laryngotracheal resistance in patients with an open tracheostoma. The measurement procedure is controlled by the patient by watching the screen during data collecting. The breaths are averaged after curve correction by splining, introducing a time-referred system into the computerized rhinomanometry. The advantage of the method is a detailed analysis of the increasing and decreasing part of the inspiratory and exspiratory half-wave, allowing clinical conclusions to be made about the nasal valve.

Successful pregnancy in advanced renal failure without dialysis.

We describe a successful pregnancy in a 22-year-old patient with advanced renal failure, who gave birth to a living boy in the 35th week of pregnancy. At the time of spontaneous delivery the mother had a serum creatinine of 851 mumol/l. No dialysis treatment had been instituted during this successful pregnancy.

Synergism between ciprofloxacin and fosfomycin against gram-negative bacteria in vitro.

A combination of ciprofloxacin and fosfomycin was evaluated for synergism employing time-kill experiments. A total of 59 clinical isolates of gram-negative bacteria was examined. All organisms were moderately sensitive or resistant to ciprofloxacin; 72.9% also showed moderate sensitivity or resistance to fosfomycin. After 24 h, synergism was seen in 22% of the strains, i.e. less than reported by other authors. There was no correlation between synergistic action and sensitivity to ciprofloxacin and fosfomycin, respectively.

The minimum inhibitory concentrations of various bismuth salts against Campylobacter pylori.

The minimum inhibitory concentrations of five bismuth salts (bismuth subcitrate, bismuth subgallate, bismuth subnitrate, bismuth subsalicylate and tripotassium dicitrato bismuthate, a water soluble compound of bismuth subcitrate) were assayed against 48 strains of Campylobacter pylori employing the agar dilution method. Tripotassium dicitrato bismuthate was most effective (MIC50 8 mg/l), the other bismuth salts exhibited somewhat lower inhibitory activities. It is concluded that bismuth salts are suitable agents for inhibiting growth of Campylobacter pylori.

Catheter-related complications during continuous ambulatory peritoneal dialysis (CAPD): a retrospective study on sixty-two double-cuff Tenckhoff catheters.

From May 1980 to April 1985, a total of 62 double-cuff Tenckhoff catheters were surgically implanted in 54 patients through a low medial laparotomy. The follow-up on continuous ambulatory peritoneal dialysis accounted for 1,029 patient-monthly. The patients who used a detachable double-bag system developed a total of four exit-site infections, one tunnel infection, three outer cuff erosions, four catheter dislocations, and two dialysate leaks. In five patients, the catheter had to be removed (one exit-site infection, two catheter breaks, and two catheter dislocations). Cumulative catheter survival using the method of life-time analysis was 81% and 70% after 1 and 2 years, respectively, considering all catheters implanted, and 92% and 92%, respectively, when peritonitis-related removal was excluded.

Does retinol serve a sensitizing function in insect photoreceptors?

Spectral sensitivity of the dorsal compound eye of Simuliid males (Nematocera) shows a maximum in the u.v. at 340 nm, and a shoulder or second, smaller maximum around 430 nm. The visual pigment--based on retinal and therefore a rhodopsin--has its absorption maximum at 430 nm. The 340 maximum is due to a sensitizing pigment that transfers energy to the visual pigment. The properties of the Simuliid-photoreceptor hence are similar to most of the photoreceptors in higher flies (Musca, Calliphora, Drosophila), that also have a u.v.-absorbing sensitizing pigment. The difference is that in Simuliids the sensitizing pigment is not 3-hydroxyretinol as in the higher flies but a different substance, most likely retinol.

A complete library of point substitution mutations in the glucocorticoid response element of mouse mammary tumor virus.

The glucocorticoid response element (GRE) of mouse mammary tumor virus (MMTV) was chemically synthesized as two complementary DNA strands bearing cohesive termini. During automated synthesis, random mutations were introduced into the DNA by "doping" each of the four nucleoside phosphoramidites (A, G, C, and T) with a low level of the other three. These preparations were annealed and cloned into an M13 phage vector to produce a library of GRE mutants. Mutations within the synthesized region were identified by sequencing phage isolates at random. All of the chemically distinct classes of transition and transversion mutations have been observed. Statistical considerations indicate that the library contains all of the possible 90 point substitution mutations within a 30-nucleotide mutagenic target. So far 88 of these substitutions have been isolated, 74 as single mutants. At least two of the three possible single mutants at each of the 30 positions have been identified.

[Metastatic pulmonary calcification in patients with chronic renal insufficiency].

Case reports on two patients with metastatic pulmonary calcification are presented. Both suffered from long standing chronic renal failure and received immunosuppressive therapy for a (non-functioning) renal transplant. Laboratory tests disclosed hyperphosphatemia and secondary hyperparathyroidism. In the first patient, who presented with "pulmonary edema", the course was rapidly fatal. Diffuse pulmonary calcification was diagnosed only post mortem. Transbronchial biopsy was diagnostic for calcification in the second patient, who had exertional dyspnea and bilateral, asymmetric, interstitial infiltrations on chest X-ray. In patients with chronic renal failure, metastatic calcifications are due not only to disturbances of calcium-phosphate homeostasis but also to other, mostly unknown factors. Diagnostic procedures include biopsy and 99m-technetium-diphosphonate scintigraphy. Prophylaxis of pulmonary calcifications through normalization of serum phosphate and, if indicated, subtotal parathyroidectomy is of the utmost importance as regression of established calcifications rarely occurs.

The contribution of different colour receptors to a motor output in the fly.

A light flash given to the eye of Calliphora leads to a movement of the legs (light induced leg reflex) which most likely normally initiates flight of the animal. This reflex has a short latency (12 to 30 ms, depending upon light intensity) and is quite reproducible without habituation. The spectral sensitivity of the reflex shows that receptors R1-6 most likely govern the input to the reflex in dark adaptation, a contribution of receptors R7 can be demonstrated with selective chromatic adaptation.

[Dose-dependent effect of amrinone on hemodynamics, myocardial circulation and myocardial energy requirement. An experimental study].

This study was designed to assess the dose-dependent effects of amrinone (1, 2 and 4 mg/kg i.v.) on hemodynamics, myocardial blood flow and myocardial oxygen consumption in anesthetised closed chest dogs (n = 8). Heart rate (HR), cardiac output, mean aortic pressure (MAP), left ventricular end-diastolic pressure (LVEDP), maximum dp/dt (dp/dtmax), myocardial blood flow (MBF) and aorto-coronary sinus oxygen difference (AVDO2 cor) were measured. Cardiac index (CI), stroke volume index (SVI), ejection fraction (EF), total peripheral resistance (TPR), coronary vascular resistance (CVR) and myocardial oxygen consumption (MVO2) were calculated from standard formulas. Amrinone improved myocardial pump function by a direct positive inotropic effect on the myocardium. EF and SVI increased to a maximal degree with 1 mg/kg amrinone (28% resp. 30%). dp/dtmax increased dose dependent (46, 64, 71%). Following a systemic vasodilation due to amrinone, left ventricular filling pressure and TPR decreased significantly. With 1 and 2 mg/kg amrinone MAP remained unchanged. 4 mg/kg produced a distinct fall in MAP accompanied by an increase in HR. The improvement in myocardial contractility did not cause a comparable increase of myocardial oxygen consumption. Due to an unloading of the heart 1 and 2 mg/kg amrinone induced no significant and prolonged augmentation of the myocardial oxygen demand. In the coronary circulation a non energy dependent vasodilation occurred followed by a marked decrease of AVDO2 cor (10, 18, 28%).
