Interaction of rat liver glucocorticoid receptor with histones.

The activated glucocorticoid-receptor complexes (GRC) from rat liver bind tightly to histone (from calf thymus)-agarose and cannot be eluted with 3 M KCl or 50% ethylene glycol, but can be eluted with 20 mM pyridoxal 5'-phosphate (PALP). Purified activated GRC were found to have much higher affinity for histones H3 and H4 (arginine-rich histones) than for histones H2A and H2B (slightly lysine-rich histones) and to have negligible affinity for histone H1 (lysine-rich histone). The GRC bound to immobilized histones could be eluted with PALP, but not with its related compounds, such as pyridoxamine, pyridoxamine 5'-phosphate, pyridoxal, and pyridoxine, suggesting a specific effect of PALP. Not only activated GRC but also unactivated GRC were found to bind to immobilized histones. However, from their profiles of elution from a histone-agarose column by 20 mM PALP, the activated GRC seemed to have higher affinity than unactivated GRC for histone. Our results suggest that the binding of GRC to histones could be associated with a mechanism for alteration of the chromosome structure by the hormonal signals, before the binding of the GRC to a specific sequence of DNA in regulatory elements of glucocorticoid-responsible genes.

Properties of an adenosine triphosphate-stimulated factor that enhances the nuclear binding of activated glucocorticoid-receptor complex: binding to histone-agarose.

Recently we have purified and characterized a factor in rat liver cytosol that enhances nuclear binding of the activated glucocorticoid-receptor complex in the presence of ATP [an ATP-stimulated translocation promoter (ASTP)]. It has a mol wt of 93,000, S value of 6.5, and pI of 4.5. Here we report studies on the binding of ASTP to histones from calf thymus. Higher salt concentrations were required to disrupt the binding of ASTP to histone-agarose than to disrupt its binding to the anion exchanger diethylaminoethyl-cellulose (half-maximal concentration for inhibition: 200 mM vs. 60 mM KCl). ATP decreased the binding of ASTP to histone-agarose in a dose-dependent fashion, but ADP and AMP had no appreciable effect on the binding. Pyridoxal 5'-phosphate (10 mM) inhibited the binding of ASTP to histone-agarose, whereas 10 mM pyridoxal, pyridoxamine 5'-phosphate, pyridoxamine, and pyridoxine were not inhibitory. ASTP did not bind to histone-agarose that had been pretreated with pyridoxal 5'-phosphate and then reduced with sodium borohydride. In contrast, ASTP pretreated in the same way could bind to histone-agarose. Therefore, modification of lysine residues of histones by pyridoxal 5'-phosphate (Schiff base) presumably inhibited the binding of ASTP to the immobilized histone. Competition experiments with various histones (H1, H2A, H2B, H3, and H4) indicated that ASTP bound to the arginine-rich histones H3 and H4, with preference for H4. These observations together suggest that lysine residues in H4 and H3 are essential for the binding of ASTP to histone. The implications of this interaction are discussed.

Silent myocardial ischemia in the aged: a retrospective study from the evaluation of necropsy hearts.

The diagnosis of silent myocardial ischemia in the aged remains controversial. In this study to evaluate the basic state of silent ischemia in the aged, a total of 972 consecutive autopsy cases (mean 79.0 yrs of age) were analyzed in terms of coronary atherosclerotic diseases. The following results were obtained: (1) Severe coronary occlusive lesions simultaneously detected in the three major coronary arterial branches were found in 85 cases (8.7%), of which 10 (12%) had neither history of angina nor myocardial infarction (MI), which may correspond to one representation of silent ischemia. (2) There were 218 cases of MI (22.4%), of which 106 (49%) were never documented to have MI during life. The latter may correspond to unrecognized MI. It was considered to be caused by 1) small or middle-sized MI in 66 cases and 2) lateral or nontransmural MI in 29 out of 40 large MI cases (more than 4.9 cm in diameter). (3) 30 cases (54%) out of 56 fresh and large MI with a correct diagnosis revealed no chest pain. This is an incidence of painless MI in the aged. This painless group showed a significant difference in the incidence of a correct diagnosis of MI (51% vs 89%), combined arrhythmia (47% vs 79%) and the association of CVD (47% vs 11%) compared with chest pain group.

Mycobacterium avium infection through the alimentary tract in mice.

Intestinal infection by Mycobacterium avium was investigated in C57BL/6 and BALB/c mouse strains. Single intragastric administration of a massive dose (10(8] or multiple administration of a lower dose (10(7), 10 times) established infection principally in the mesenteric lymph-node (MLN); a continuous or intermittent fecal excretion of the bacilli was detected by 6-8 weeks after the administration. Based on three criteria--isolation of the organisms from the MLN and from feces, and detection of acid-fast bacilli in sections of the MLN--germ-free (GF) BALB/c mice exhibited clearer dose-effect relations than the flora-bearing (FB) counterparts. After intragastric administration, the organisms were probably trapped in the Peyer's patch and then transferred to the MLN at an early period (by 4-7 days), persistent infection thus being established in the MLN. Systemic involvement evolved both in athymic and euthymic mice after a prolonged period of time (more than 40 weeks) showing far more severe involvement in the former regardless of the presence of floral organisms.

Influence of diet on the development of antral gastrin-like immunoreactivity in the stomach of rats.

The effects of individual food constituents on antral gastrin-like immunoreactivity concentrations were studied in young rats. Rats aged 7 to 20 days were given only rat breast milk and then weaned by various nutrients (regular laboratory chow, protein (ovalbumin)-, fat- or carbohydrate (starch)-rich food). Rats receiving rat breast milk only until 27 days of age were also studied. In rats on regular laboratory chow, antral gastrin-like immunoreactivity increased and reached adult levels on day 25. In rats on ovalbumin, fat-rich food or starch, it increased on day 23 but dropped thereafter. The increment by laboratory chow was higher than that by the individual nutrients. No increase was observed during milk feeding alone. Gel filtration of antral gastrin-like immunoreactivity from 25-day-old rats on laboratory chow or three essential nutrients showed the same results.

Sheehan's syndrome with hypomagnesemia and polymorphous ventricular tachycardia.

A 40 year old woman with Sheehan's syndrome demonstrated polymorphous ventricular tachycardia and hypomagnesemia. The diagnosis of Sheehan's syndrome was based on her history, physical examination and endocrinological data. Serum magnesium level was 1.5 mg/dl. After the treatment of polymorphous ventricular tachycardia by lidocaine, an electrocardiogram showed low voltage, T-wave inversion or flattening and QT-prolongation, and these electrocardiographic abnormalities gradually improved after the initiation of cortisone therapy. A chest roentgenogram, an echocardiogram, thallium-201 scan of the myocardium and hemodynamic parameters indicated no abnormal findings. It is suspected that hypomagnesemia caused polymorphous ventricular tachycardia. However, the cause of hypomagnesemia and its relation to Sheehan's syndrome in the present case were not determined, although, hypomagnesemia was improved after the administration of cortisone.

[A questionnaire about the dental treatment for visiting patients at welfare center for the aged people].

A questionnaire about the dental treatment for visiting patients at Welfare center for the aged people. Nowadays, 11 percent of the population is over 65 in Japan. According to the Minister of Health and Welfare's Population Projection for Japan, it will grow to 20% at the beginning of the 21st century. Problems concerning old people will become some of Japan's biggest challenges. In that situations, the dental treatments for visiting aged patients will become more important. Then, authors sent out a questionnaire about it on aged people who have no need of anyone's assistance in their home lives. And we examined concrete countermeasures and problems about the dental treatments for visiting patients. The findings were as follows: 1. Even the aged people who have no need of anyone's assistance in their home lives, they hope much carrying out the dental treatments for visiting them. 2. The dental treatments for visiting aged patients have some problems. But it will become one of the most important way to supply their demand for dental treatments, in spite of their latent requirements, a lot of old people give up to undergo dental treatments.

[Sialidase in rat salivary glands. Characteristics of soluble sialidase in salivary glands and other tissues].

In this study, we elucidated some characteristics of soluble sialidases in the rat salivary glands, brain, liver and kidney. 1) Soluble sialidases in the submandibular and parotid glands were inactivated by about 75 and 60%, respectively, and the enzymes in the brain, liver and kidney were also inactivated by 16-48%, when preincubated at 40 degrees C for 1h. When preincubated at 60 degrees C, the enzymes in all tissues were completely inactivated within 10 min. 2) Ca2+ ion in low concentrations tended to activate soluble sialidases in both the submandibular and parotid glands, but had no apparent effect on the enzymes in the brain, liver and kidney. The enzyme in the submandibular gland tended to be activated also by low concentrations of Mg2+. Both Cu2+ and Hg2+ ions caused a marked inhibition on the soluble sialidases in all tissues. 3) The molecular weight of soluble sialidases in the submandibular and parotid glands was determined to be about 68,000 and 46,000, respectively, by means of Sephadex G-200 gel filtration. The molecular weight of the enzymes in the brain and liver was almost similar to that of the enzyme in the parotid gland. In the kidney, soluble sialidase was separated into two enzymes, one having high (more than 500,000) and the other having low (about 46,000) molecular weight. 4) By isoelectric focusing analysis, three isozymes were detected in the submandibular gland and brain sialidases and two isozymes detected in the enzymes of other tissues. The isoelectric point of the major isozymes in the submandibular and parotid glands was 6.4 and 6.9 respectively. In the brain, liver and kidney sialidases, the isozymes with isoelectric point in the range of 4.4 to 6.7 were detected.

Malignant histiocytosis involving pancreas at initial presentation.

A 10-year-old boy with malignant histiocytosis presented with fever, hepatosplenomegaly, and diffuse pancreatic enlargement, mimicking acute pancreatitis. Although malignant histiocytosis involving pancreas at initial presentation is exceptional, this entity should be included in the differential diagnosis of acute pancreatitis, especially when hepatosplenomegaly and pancytopenia are present.

Neuronal organization of the supracommissural ventral telencephalon and the nucleus preopticus periventricularis in the himé salmon (landlocked red salmon, Oncorhynchus nerka): a Golgi study.

The neuronal organization of the supracommissural ventral telencephalon (Vs) and the nucleus preopticus periventricularis (NPP) of the medial preoptic area (MPOA) in the himé salmon (landlocked red salmon, Oncorhynchus nerka) was studied by means of the Nissl staining and the rapid Golgi methods. The Vs was composed of many laterally-orienting spiny neurons and a few sparsely-spined polymorphous neurons. Some of the laterally-orienting neurons extended their dendrites into the lateral forebrain bundle (LFB), suggesting that they may receive inputs from the telencephalofugal and telencephalopetal fibers in the LFB. The NPP consisted of five types of sparsely-spined neurons: laterally-orienting, bipolar, cerebrospinal fluid (CSF)-contacting, commissural, and multipolar neurons. Some of the laterally-orienting neurons extended their dendrites into the medial forebrain bundle (MFB) and regions near the LFB, suggesting that they may also receive inputs from the telencephalofugal and telencephalopetal fibers running there. The presence of the CSF-contacting neurons in the NPP suggests that the NPP may receive humoral informations from the CSF. Involvement of these CSF-contacting neurons in the control of the reproductive functions was discussed. The neuronal organization of the NPP in the himé salmon was compared with that of the MPOA in other vertebrates.

Infection of rabbits with Sendai virus.

Rabbits were either inoculated with Sendai virus (SV), strain MN, or caged with virus-inoculated rabbits on the same day of the viral inoculation, and examined for viral shedding and detection of viral antigens in the respiratory tract, histopathologic changes, and serum antibodies. Infectious virus was recovered from nasal swabs at postinoculation day (PID) 3 and disappeared by PID 10. Viral antigens were detected by immunofluorescence in epithelial cells of the nasal cavities, but not of the trachea and lungs from PID 3 to PID 10, and antibodies were detected after PID 7. Rabbits had no clinical manifestations and only exhibited a moderate increase in goblet cells of the nasal epithelium. In the transmission study, virus was recovered from one of three uninoculated rabbits at postexposure day (PED) 10 and antibodies were detected at PED 15 in the same rabbit. These data suggest that, although viral multiplication was limited to the nasal epithelium, laboratory rabbits are susceptible to Sendai virus infection.

Pulmonary lymphangiomyomatosis in childhood? Marked smooth muscle cell proliferation of the lung in a preadolescent girl with repeated pneumothorax and progressive dyspnea.

A 13-year-old girl with repeated spontaneous pneumothorax and progressive dyspnea is described. The biopsy specimen of the lung showed marked proliferation of smooth muscle cells in the thickened bullous wall and alveolar septa, which was similar to the findings of the pulmonary lymphangiomyomatosis. Pulmonary lymphangiomyomatosis is one of the diseases which should be considered when children have progressive dyspnea, chylous effusions and repeated spontaneous pneumothorax.

[Spontaneous remission of four months' duration in hypoplastic leukemia with tetraploid chromosome after blood transfusions and infection].

Spontaneous complete remission of four months' duration was observed in a 51-year-old male with hypoplastic leukemia. Cytogenetical analysis revealed that leukemia cells of this patient were tetraploid. The diameter of leukemia cells involving myeloid cells ranged from 30 to 50 mu. The remission was apparently associated with repeated blood transfusions and severe infection. Complete remission was confirmed by normal morphology and karyotype of the bone marrow cells, although in vitro marrow stem cell growth did not return to normal. Thus, normal hematopoiesis may not have recovered when the diagnosis of spontaneous remission was made.
