Platelet aggregation inhibitors in a Bhutanese medicinal plant, shug chher.

The 90% methanol-soluble fraction of a Bhutanese medicinal plant, Shug Chher, exhibited inhibition of platelet aggregation induced by platelet activating factor. Bioassay-directed fractionation led to the isolation of four new labdane diterpenoids, 3 alpha, 15-dihydroxy-labda-8(17), 13E-diene (5), 3 alpha-hydroxy-labda-8(17), 13E-dien-15-oic acid (6), 3 alpha-hydroxy-labda-8(17), 12E, 14-trien-19-oic acid (7), and 3 alpha-acetoxyisocupressic acid (8) and four known diterpenoids, manool (1), 3 alpha-hydroxymanool (2), 3 alpha-hydroxy-12, 13E-biformene (3), and isocupressic acid (4). The structures of the new compounds were determined spectroscopically. Compounds 2, 3, and 5 inhibited platelet aggregation.

Formation of beta-fructosyl compounds of pyridoxine in growing culture of Aspergillus niger.

Two pyridoxine compounds were found to be formed in a culture filtrate of Aspergillus niger and A. sydowi, when grown in a medium containing sucrose and pyridoxine. Each of the two compounds I and II was obtained as a white powdered preparation by preparative paper chromatography, gel filtration on Toyopearl HW-40S and Sephadex G-10 columns, DEAE-cellulose column chromatography, and lyophilization. Compounds I and II were identified as 5'-O-(beta-D-fructofuranosyl)-pyridoxine and 5'-O-[beta-D-fructofuranosyl-(2-->1)-beta-D-fructofuranosyl]-pyridoxine, on the basis of the various experimental results, viz., elementary analyses, UV, 1H-, and 13C-NMR spectra, products by hydrolysis with acid and yeast beta-D-fructofuranosidase, migration on paper electrophoresis, and Gibbs reaction in the presence and absence of boric acid. Levansucrase from Microbacterium laevaniformans and yeast beta-D-fructofuranosidase did not catalyze the beta-D-fructofuranosyl transfer from sucrose to pyridoxine to give rise to beta-D-fructofuranosyl-pyridoxine.

Eosinophilic meningitis in two cows.

A neurological disorder was noted in 5 cows on a farm. Histopathological examination in 2 of these 5 animals revealed an infiltration of eosinophils with some macrophages in the meninges and around arterioles of the cerebral cortex. The endothelial cells of the reactive blood vessels were swollen. From these observations, the brain lesions were diagnosed as "eosinophilic meningitis". In one case, pulmonary arterioles were also infiltrated with eosinophils and macrophages in a granulomatous tissue. No bacteria, fungi, protozoa, or parasitic worms were detected in the present cases.

Characteristics of Staphylococcus aureus isolated from peracute, acute and chronic bovine mastitis.

Fifty-eight Staphylococcus aureus strains isolated from bovine mastitic milk at 27 dairy farms in Japan during the period from November 1988 to May 1989 were examined for their productivity of virulence associated factors. The positive rates of the total isolates for various virulence factors were as follows: toxic shock syndrome toxin-1 (TSST-1, 27.6%), staphylococcal enterotoxins (SEs, 34.5%), alpha-haemolysin (74.1%), beta-haemolysin (65.5%), delta-haemolysin (12.1%), DNase (100%), egg-yolk factor (25.9%), clumping factor (70.7%) and protein A (58.6%). All of S. aureus isolates from peracute mastitis produced TSST-1, SEC, alpha-haemolysin and beta-haemolysin. While none of clumping factor and protein A were detected among peracute isolates, these factors were produced at a relatively high frequency by isolates from chronic mastitis. In coagulase typing, the most predominant type was VI (36.2%), and types IV, V and VIII were not observed. TSST-1 positive isolates showed interesting characteristics which all of the isolates produce both SEC and coagulase type VI but lack egg-yolk factor, clumping factor and protein A except for one isolate. We could infer that TSST-1 and SEC contribute to bovine mastitis, especially to peracute mastitis from the present study.

Proto-oncogene of genomic DNA, related to the human epidermal growth factor receptor (EGFR) gene, from clinically normal domestic animals.

Genomic DNAs of cattle, horses, pigs, dogs, cats and chickens were surveyed using Southern blot hybridization analysis, with a human EGFR cDNA fragment. Several bands with different numbers and molecular weights were observed under the condition of low stringency in the individual animal species. The bands showing DNA polymorphism were observed among bovine genomic PstI-digested DNAs from 4 individuals and EcoRI-digested genomic DNAs from 4 chickens. These results may provide basic data which are useful for analysis of tumorigenetic mechanisms in domestic animals.

Double-labeling immunohistochemical studies on canine senile plaques and cerebral amyloid angiopathy.

The relationship of senile plaques to neuronal cells, neurites, glial cells, or capillaries was examined using double labeling-immunostaining methods on the Bouin's solution-fixed serial brain sections from dogs. Compact deposits of beta protein (amyloid plaques) in the cerebral cortex always contained microvessels labeled by anti-collagen type IV antibody and some of them might be formed as the result of fusion of several perivascular beta amyloid deposits. In the periphery of those plaques swollen neurites recognized with anti-neurofilament antibody were sometimes present, but the relation between such plaques and neuronal cells or glial cells were unclear. Diffuse deposition of beta protein (diffuse plaques) was frequently developed beside neuronal cells, while most plaques did not contain glial cells. Some of those plaques were closely contact with microvessels, but some had no relation. Intact or irregularly arranged neurites were present in diffuse plaques. Such irregularity of the neurites were obvious in the plaques in the hippocampus as compared with those in the cerebral cortex. These results indicate the possibility that canine amyloid plaques would be formed as the result of amyloid degeneration of cortical capillaries, and diffuse parenchymal deposition of beta protein would originate from neuronal or neuritic processes.

Systemic ceroid-lipofuscinosis in a Japanese domestic cat.

An 11-month-old castrated male Japanese domestic cat was euthanized because of neurological symptoms such as shivering and difficulty of walking. Histopathological examination showed glial proliferation and marked deposition of ceroid-lipofuscin in the neuronal and glial cells of the brain. Ceroid-lipofuscin was deposited also in reticuloendothelial cells of the liver, spleen and some lymph nodes.

[Early lesions of pancreatic islets in small-ob mice (C57BL/6J-ob/ob)].

Pancreatic islets of C57BL/6J-ob mice were microscopically examined at the ages of 5, 13, and 21 days, to clarify the time of the increase in number of A-cells, which appears to lead to diabetic symptoms in Small-ob mice (C57BL/6J-ob/ob). The findings of islet in 5-day-old C57BL/6J-ob mice were the same as those of C57BL/6J-+/+ mice. At 13 days of age, changes in islet of C57BL/6J-ob mice were classified into 3 groups. They were no significant changes (putative normal), hypertrophic B-cells with clear cytoplasm (Ob mice), and an increase of A-cells (Small-ob mice), respectively. Electron microscopically, a tendency toward dilatation of endoplasmic reticulum was found in the hypertrophic B-cells. In the islets that had an increase in number of A-cells, only a few vacuoles, which were suggestive of degranulation, were found in the cytoplasm of B-cells, and premature secretory granules were seen in the A-cells. In part of the A-cells, development of rough-surfaced endoplasmic reticulum was also seen. Mild hypertrophy of the islets and the B-cells were observed in more non-diabetic Ob mice at the age of 21 days than non-obese mice at the same age, and only 2 of 6 mice had the increase in number of A-cells. In Small-ob mice, the increase in number of A-cells was more obvious than in the diabetic Ob mice, while other islet findings were similar to those of non-obese mice in extent.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of cardioplegic reperfusion on ATP recovery of rod-shaped myocytes isolated from rat ventricles.

ATP content [ATP] and fraction of rod-shaped myocytes (Rrod) were measured in ventricular myocytes isolated from the rat heart. [ATP] (nmol (10(6) cells)-1) and Rrod (%) were 72.0 and 60.5, respectively, and significantly reduced to 18.8 and 41.8 by no-flow ischemia for 30 min. Reperfusion for 15 min with cardioplegic solutions restored [ATP] of rod-shaped myocytes to the control level.

Chronic effects of camostate on growth and endocrine function of the pancreas in streptozotocin-induced diabetic rats.

Chronic feeding of normal rats with camostate, a synthetic trypsin inhibitor, stimulates the growth of the pancreas chiefly by increasing cholecystokinin release. We examined the effects of camostate on the growth and the endocrine function of the pancreas in streptozotocin-induced diabetic rats. The pancreatic weight of the diabetic rats given camostate (200 mg/kg/day) by oral gavage for 14 days was significantly elevated by 120% over that of diabetic rats not given camostate, and was comparable to that in the nondiabetic rats given camostate. The total pancreatic contents of DNA, RNA, and protein in diabetic rats given camostate were also significantly higher than those in diabetic rats not given camostate, and did not differ from those observed in nondiabetic rats given camostate. The pancreatic growth seen in diabetic rats treated with camostate was associated with moderate hyperplasia and pronounced hypertrophy. By contrast, treatment with camostate did not improve hyperglycemia, hypoinsulinemia, or low pancreatic content of insulin seen in diabetic rats. These findings demonstrate a marked growth of the pancreas in diabetic rats stimulated by camostate, and suggest that camostate-induced pancreatic growth is not affected by the reduced level of the endogenous insulin. The present study also indicates that camostate has no beneficial effects on the function of residual B cells, failing to improve diabetes.

[Detection of numerical chromosome aberrations in urologic malignancies using in situ hybridization from formarin-fixed paraffin sections].

It has been shown that molecular cytogenetics in interphase nuclei is applicable to paraffin embedded sections from formarin-fixed materials utilizing in situ hybridization (ISH). This allows precise identification of numerical abnormalities of chromosome in tumor cells without disruption of tumor morphology. Thirteen cases of bladder cancer and twenty six cases of prostate adenocarcinoma were evaluated for numerical aberrations of chromosome 1, 7, 10, 11, 17, 18, X and Y utilizing biotinylated probes specific for the alpha satellite region. In two cases from total prostatectomy, one case from total cystectomy and two cases from TUR-P specimen, hybridized chromosomes could not be detected in individual tumor cells. With respect to the appearance of the ISH signal, optical concentration and time for the digestion enzyme has to be established essentially. This technique can now be applied on the detection of biological activity in various types of urological cancer.

[Histopathological comparison of prostatic adenocarcinoma in Japan and the United States].

Histopathological comparison of clinical prostatic adenocarcinoma between Japan and the United States (US) has been made with retrospective analyses on 1,037 and 987 cases, respectively. All cases were histologically evaluated by the same pathologist (M.H.) without previous knowledge of clinical information according to application of the Japanese General Rules for Prostatic Cancer (JGRPC) based on the predominant degree of histological differentiation, the nuclear grading by WHO classification and the Gleason grading systems. The age distribution of the patients was slightly different among both groups. However, this difference appeared not to be essential, because there existed difference of the date on diagnosis between two groups. Even though considering this time difference on registration, much more cases from Japan had stage D disease. The observed incidence of the histological differentiation classified by JGRPC among over-all cases did not show significant difference, however, the incidence among the cases with same stage between two groups differed with some statistical significance. Much more cases from the US fell in well differentiated adenocarcinoma and frequency of moderately differentiated adenocarcinoma was higher in the Japanese even in the cases diagnosed at advanced stages. The observed incidence of nuclear grade 3 was also higher in the Japanese. Gleason primary and secondary grade 5 and score 9-10 appeared more frequently in the cases from Japan.(ABSTRACT TRUNCATED AT 250 WORDS)

[Histopathological comparison of stage A prostatic adenocarcinoma in Japan and the United States].

Histopathological characteristics of stage A prostatic adenocarcinoma from Japan (137 cases) and the United States (51 cases) were comparatively evaluated by retrospective analysis. All cases were examined by one referee pathologist (MH) using classification based on Japanese General Rules for Prostatic Cancer (JGRPC), nuclear grading by Mostofi-WHO system and Gleason grading. The incidences of the cases with well differentiated, low nuclear anaplasia and Gleason grade 1 or 2 on primary and secondary grades as well as histologic score 2-4 were much more higher in the cases from Japan as compared with those from the US. In the cases from the US proportion of the cases with moderately differentiated adenocarcinoma and grade 2 nuclear anaplasia exceeded those with well differentiated and grade 1 anaplasia. Adenocarcinomas with Gleason grade 3 in primary and secondary grade and 6-8 of histologic score were also more frequently observed in the cases from the US. No different distribution of each histologic grade could be obtained among the Japanese cases classified stage A2 and entire cases from the US. These results suggest that much more stage A adenocarcinoma in the US might fall into A2 if strict criteria was applied for subcalssification of stage A, which may reflected in the remarkable difference in the incidence of clinical prostatic carcinoma.

[Phenotypic difference of lymphocytes circulating in tumorous renal artery and renal vein in patients with renal cell carcinoma].

Phenotypic difference of lymphocytes circulating in tumorous renal artery and renal vein in patients with renal cell carcinoma was studied. Fifteen patients with no prior treatment were studied, from whom blood samples were taken at the time of radical nephrectomy. Mononuclear cells separated from tumorous renal arterial and venous blood were analyzed for cell surface markers. CD3, CD4, CD8, CD11b, CD16, CD25, CD57, Leu8 HLA-DR were compared. CD25 positive cells were significantly increased in venous blood than in arterial blood in the kidneys with tumor. No significant difference was found regarding the other surface markers.

[A case report of an ectopic ureter in a patient treated for chronic prostatitis].

A male aged 20 had been treated for anal and lower abdominal pain after ejaculation for about 2 years under the diagnosis of chronic prostatis. Because of worsening of the pain, he was admitted to our hospital in September 24, 1991. On the rectal examination, a cystic tumor was palbable on the left side of the prostate. The left kidney was not detected by IVP and ultrasonography. CT revealed two retrovesical cystic lesions in the left side. Cystourethroscopy showed an absence of ureteral orifice and ureteral ridge on the left side. Left uretero-seminal vesiculectomy was performed on October 3rd, under the diagnosis of an ectopic ureter entering into the seminal vesicle or ejaculatory duct. Histopathologically, the left ectopic ureter entering into the lower portion of the seminal vesicle or ejaculatory duct and the left renal agenesis were diagnosed. It was remarkable that ureteral epithelium was not transitional but columnar epithelium.

[Retroperitoneal liposarcoma with intrarenal metastases which was difficult to distinguish from angiomyolipoma. A case report].

A 40-year-old man was admitted to our hospital with a fat-density extrarenal tumor and intrarenal nodules which were detected by ultrasonography (US) and CT scan obtained the general health check. After his admission, US, CT, MRI and angiography were performed. As the final diagnostic procedure, we performed a US-guided percutaneous needle biopsy of the extrarenal tumor, because this tumor was difficult to distinguish from liposarcoma and angiomyolipoma (AML). The pathological study revealed a well-differentiated liposarcoma, and the extrarenal tumor and the left kidney were removed en bloc. Pathologically, the tumor not only invaded into the renal parenchyma directly but also have metastasized independently to the intrarenal region. Postoperatively, he received a tumor dose of irradiation of 50 Gy. Liposarcoma arising in the perirenal space should be considered in the differential diagnosis of exophytic AML. If radiological studies can not confirm the appropriate diagnosis, we consider that biopsy of the tumor should be selected as an alternative diagnostic approach. And, we think that this is the first case report dealing with intrarenal metastases of liposarcoma.

Biological properties of aureobasidin A, a cyclic depsipeptide antifungal antibiotic.

Aureobasidin A (AbA) is a novel cyclic depsipeptide antifungal antibiotic. The antifungal activity of AbA was studied in vitro and in vivo in comparison with clinically effective antifungal agents, amphotericin B and fluconazole. AbA was highly active in vitro against many pathogenic fungi, including Candida albicans, Cryptococcus neoformans, Blastomyces dermatitidis and Histoplasma capsulatum. The activity was superior to amphotericin B in most cases. AbA exhibited fungicidal action toward growing cultures of C. albicans. It was highly tolerated by mice and showed good efficacy in the treatment of murine systemic candidiasis when given orally or subcutaneously. AbA's fungicidal action in mice with candidiasis was more effective than fluconazole and amphotericin B.

Ten-year experience in the use of double balloon catheter for kidney procurement from non-heart beating donors in cadaveric kidney transplantation.

One-hundred-and-twenty patients underwent first cadaveric kidney transplantation from the non-heart beating donors. All of the organs were procured with the use of double balloon catheter for in situ cooling. The mean warm ischemic time and cold ischemic time were 10.7 +/- 17.0 minutes and 18.9 +/- 11.4 hours, respectively. One- and 5-year graft survival rates were 85.0 and 72.7%, respectively. Among 120 recipients, 30 (25%) grafts functioned immediately (immediate function), 82 (68.3%) grafts functioned after varying length of oliguric periods (delayed function) and 8 (6.7%) grafts never functioned (non-function). The mean age of the donors in the group of immediate function (31.5 +/- 16.1 yr) was significantly lower than those of other two groups. The mean warm ischemic time in the group of immediate function (6.0 +/- 11.2 min) was significantly shorter than that of delayed function. However, there was no significant difference in donor hypotensive episode, types of preservation fluid and cold ischemic time between the groups. The conclusion is that the ultimate result of cadaveric kidney transplant from the non-heart beating donors with the use of double lumen catheter is acceptable despite a relatively high incidence of delayed graft function.
