Rat alpha1-acid glycoprotein. Purification and immunological estimation of its serum concentration.

A rapid method is described for the preparation of serum alpha1-acid glycoprotein from rats with inflammation induced with turpentine oil injection. The protein obtained by two purification steps, batchwise adsorption with DEAE-cellulose followed by chromatography on CM-cellulose, was proved to be native alpha1-acid glycoprotein in a high degree of purity by electrophoretical, immunological, ultracentrifugal and carbohydrate analysis. The monospecific and potent antiserum to this protein was prepared by immunizing rabbits with the desialyzed material emulsified with Freund's incomplete adjuvant. Using purified alpha1-acid glycoprotein and its specific antiserum, the concentration of alpha1-acid glycoprotein in rat serum was determined by single radial immunodiffusion. Abnormally high levels of its concentration (5-6 times higher than the control) were observed in inflammatory and tumor bearing rats.

Relation of food consumption to cancer mortality in Japan, with special reference to international figures.

The present study was designed to examine the epidemiological association of food consumption with cancer mortality in Japan, if any, in comparison with similar examinations in developed countries including Japan, in a search for dietary factors in cancer etiology. Whereas food consumption patterns in Japan was believed to be specific among developed countries, this study revealed that the correlation matrix among major foods both for international and Japanese data made it possible to categorize these foods into two groups with some similar patterns, since there should be observed a positive or negative correlation for each pair of foods. The patterns for correlation of food consumption to cancer mortality in Japan were found to be considerably different from those in developed countries, and this suggested that the epidemiological association between the two in Japan, if any, could be specifically evaluated from that in develop countries. The urban-type region for food consumption in Japan, however, indicated a possible westernization for the relation of food consumption to cancer mortality as well as for food consumption patterns, whereas the traditional Japanese region for food consumption did not indicate any similarity with westernization for the relationship between food consumption and cancer mortality, as well as for food consumption patterns. The above findings also suggested that there might be a possible significance for the regionalization of Japan in such studies, and that the urbanization of food consumption patterns in Japan would show that this country has a similar pattern of association between food consumption and cancer mortality as observed among developed countries in Europe and/or North America.

Role of the kidney in gastrin metabolism.

Fasting blood gastrin level among various clinical entities was measured by means of radioimmunoassay. Hypergastrinemia of various degrees was found in some cases of chronic renal failure. During angiography arteriovenous difference of gastrin level was investigated in the kidney in 22 patients without renal diseases. The high rate of gastrin removal from the blood circulation by the kidney was also confirmed in the synthetic human gastrin 1-17 (SHG 1-17) infusion experiment in dogs. In this study, hypergastrinemia seen in renal diseases was thought to be caused partly by impaired gastrin metabolism in the kidney.

Intrafamilial incidence of autism, cerebral palsy, and mongolism.

A comparative study of developmentally disturbed children as well as normal children is presented with a focus on their position in the family. Demographic data were compared for three groups of Japanese children, with a mean age of 4 years 7 months, who were diagnosed as autistic, cerebral palsied, and trisomic (trisomy 21 mongoloid). The areas compared were maternal age at birth, ratio of males to females, number of siblings, birth order, and age interval between the subject and the closest younger sibling. The normal kindergarten children with a mean age of 4 years 6 months, as well as the Japanese general population, were also compared for these demographic items. The findings on maternal age and sex ratio for these three nosological groups are consistent with recent studies. Although the reproduction of mothers of either cerebral palsied or mongoloid children ceases after the birth of the propositi, the reproduction of mothers of autistic children does not show significant difference from that of normal children's mothers. The age intervals between the subjects and the closest younger sibling do not differ among the three groups or in the normal group.

Diagnostic value of secretin provocation test.

Plasma gastrin response to the intravenously administered secretin was investigated in various clinical entities. The marked increase of plasma gastrin was found in response to secretin in a case of suspected Zollinger-Ellison syndrome in contrast to various degrees of plasma gastrin decrease seen in patients with ordinary or postoperative recurrent peptic ulcer. The diagnostic value of secretin provocation test was stressed especially in relation to differentiation between Zollinger-Ellison syndrome and recurrent ulcer due to retained pyloric antrum kept away from the food-passing route, both of which are characterized by hypergastrinemia and acid hypersecretion.

Significance of gastric analysis with particular reference to the diagnosis of Zollinger-Ellison syndrome.

Various criteria in gastric analysis currently used for the diagnosis of the Zollinger-Ellison syndrome were assessed in cases of endoscopically normal gastric mucosa, gastric ulcer and duodenal ulcer. New criteria consisting of BSV greater than 350 ml/hr, BAO greater than 25 mEq/hr, MAO greater than 60 mEq/hr, and BAO/MAO greater than 0.7 were proposed to differentiate the Zollinger-Ellison syndrome and peptic ulcer.
