Absorption and metabolism of lipids in rats depend on fatty acid isomeric position.

This study determined the effect of dietary (dipalmitoyl) triacylglycerol (TAG) structure on the absorption and metabolism of fat in the presence or absence of cholesterol. Sprague-Dawley male rats weighing approximately 180 g were fed diets containing 10 g/100 g diet 1,3-dipalmitoyl-2-oleoyl-glycerol-rich oil (POP) or 1(3),2-dipalmitoyl-3(1)-oleoyl-glycerol-rich oil (PPO), either with or without cholesterol supplementation (0.5 g/100 g diet), for 2 wk. There were no differences in the growth of rats fed the various diets. The apparent absorption efficiency of dietary fat especially the apparent absorption efficiency of palmitic acid, was significantly lower in the POP groups than in the groups fed the PPO diets. The excretion of cholesterol in feces of rats fed the POP diet with cholesterol was significantly greater than those fed the PPO diet containing cholesterol. The excretion of total bile acids in the rats fed cholesterol was significantly greater than in those fed the diets without cholesterol. Liver cholesterol was significantly greater in rats fed cholesterol and was not affected by the TAG structure. Plasma cholesterol was significantly lower in the POP dietary groups than in the PPO groups. These results suggest that the absorption and metabolism of fat depend on the positional isomer of the fatty acid.

Shortcut link between the fast and slow pathways and the mechanism of cure in atrioventricular nodal reentrant tachycardia by catheter ablation.

UNLABELLED: The mechanism of cure in AV nodal reentrant tachycardia (AVNRT) by catheter ablation has not been fully clarified. We hypothesized that disruption of a shortcut link between the fast and slow pathways is responsible for the elimination of tachycardia. RESULTS: AVNRT was eliminated in 20 patients by catheter ablation. In five patients (25%; group I) slow pathway conduction disappeared 1 week after ablation. In six patients (30%; group II), the effective refractory period of the slow pathway was prolonged by more than 50 ms (212 +/- 81 ms vs 340 +/- 81 ms; P < 0.05). In the remaining nine patients (45%; group III), there was no change in the refractory period (270 +/- 65 ms vs 273 +/- 74 ms), although tachycardia was not inducible. A shortcut link between the fast and slow pathways was examined by comparing the A-H intervals over the slow pathway during the tachycardia and during atrial pacing at the tachycardia cycle length. Prior to ablation, a shortcut link was assumed in 1 of group I patients, 2 of group II patients, and 8 of group III patients. Of the 9 patients in whom the slow pathway was not impaired after ablation (group III), 8 patients were found to have a shortcut link, while 8 of 11 patients with impairment of the slow pathway after ablation (groups I and II) had no shortcut link between the fast and slow pathways (P < 0.05). CONCLUSION: In patients with a shortcut link between the fast and slow pathways, slow pathway conduction itself does not need to be impaired to eliminate the AVNRT, whereas in patients without this shortcut link, slow pathway conduction must be impaired.

Isolation of group B porcine rotavirus in cell culture.

While group A and C rotaviruses have been grown in cell culture, group B rotavirus has never been cultured. In this study we successfully isolated porcine group B rotavirus in swine kidney cells. Pancreatin treatment is essential for the propagation of group B rotavirus.

Serological and genomic characterization of porcine rotaviruses in Thailand: detection of a G10 porcine rotavirus.

A total of 557 fecal specimens collected from piglets with diarrhea in Thailand were examined for rotavirus RNA by polyacrylamide gel electrophoresis. Twenty-three, one, and two samples were positive for group A, group B, and group C rotaviruses, respectively. Two samples exhibited two segments found in picobirnavirus RNA. RNA electropherotyping of 23 group A rotaviruses showed that they were classified into five patterns. By serotyping by enzyme-linked immunosorbent assay and PCR, viruses in 3 and 14 specimens were found to be serotype G3 and serotype G10, respectively. For one specimen, containing a serotype G10 virus (strain P343), virus was isolated in MA-104 cells, and the nucleotide sequences of the VP7 and VP4 genes were determined. Comparative sequence analysis and cross-neutralization tests showed that strain P343 has B223-like G10 and UK-like P7 serotype (or VP4 genotype 5) specificities. Rotaviruses having such antigenic specificities have not been detected in piglets. Thus, the interspecies transmission of rotaviruses between cows and pigs was suggested.

Nondefective rotavirus mutants with an NSP1 gene which has a deletion of 500 nucleotides, including a cysteine-rich zinc finger motif-encoding region (nucleotides 156 to 248), or which has a nonsense codon at nucleotides 153-155.

We isolated two nondefective bovine rotavirus mutants (A5-10 and A5-16 clones) which have nonsense mutations in the early portion of the open reading frame of the NSP1 gene. In the NSP1 gene (1,587 bases long) of A5-10, a nonsense codon is present at nucleotides 153 to 155 just upstream of the coding region (nucleotides 156 to 230) of a cysteine-rich Zn finger motif. A5-16 gene 5 (1,087 bases long) was found to have a large deletion of 500 bases corresponding to nucleotides 142 to 641 of a parent A5-10 NSP1 gene and to have a nonsense codon at nucleotides 183 to 185, which resulted from the deletion. Expression of gene 5-specific NSP1 could not be detected in MA-104 cells infected with the A5-10 or A5-16 clone or in an in vitro translation system using the plasmids with gene 5 cDNA from A5-10 or A5-16. Nevertheless, both A5-10 and A5-16 replicated well in cultured cells, although the plaque size of A5-16 was extremely small.

Agents for the treatment of overactive detrusor. IX. Synthesis and pharmacological properties of metabolites of N-tert-Butyl-4,4-diphenyl-2-cyclopentenylamine (FK584) in human urine.

We synthesized the racemates of the five presumed metabolites (1b-f) of (S)-(--)-N-tert-butyl-4,4-diphenyl-2-cyclopentenylamine hydrochloride (FK584, S(--)-1a), a novel agent for the treatment of overactive detrusor syndrome, in order to confirm the structures of the metabolites and also to evaluate their inhibitory activity against detrusor contraction. (+/-)-N-tert-Butyl-4-(4-hydroxyphenyl)- and 4-(4-hydroxyphenyl)- and 4-phenyl-2-cyclopentenylamines (1b--e) were synthesized via 5-(4-methoxyphenyl)- and 5-(4-benzyloxy-3-methoxyphenyl)-5-phenyl-2-cyclopenten-1-one (9g, h), respectively. Compounds 1b-f prepared in this study were identical with the metabolites in human urine in gas chromatography-mass spectrometry and analytical HPLC. The inhibitory activity of compounds 1b-f against detrusor contraction in vitro induced by electrical field stimulation in guinea-pigs was less potent than that of FK584.

Agents for the treatment of overactive detrusor. V. Synthesis and inhibitory activity on detrusor contraction of N-tert-butyl-4,4-diphenyl-2-cyclopentenylamine.

N-tert-Butyl-4,4-diphenyl-2-cyclopentenylamine ((+/-)-3) was designed to restrict the conformation of terodiline 1 and was synthesized in a 6-step approach starting with diphenylacetaldehyde (10) or in a 4-step approach starting with 2,2-diphenyl-4-pentenoic acid (17). Using di-p-toluoyltartaric acid as a resolving agent, the synthetic (+/-)-3 was resolved into its optically pure forms, (-)- and (+)-3. The (-)-enantiomer (-)-3.HCl (FK584) showed about ten times more potent inhibitory activity on urinary bladder rhythmic contraction in rats (ED30 = 0.18 mg/kg, i.v.) than terodiline (ED30 = 1.9 mg/kg, i.v.), while the (+)-enantiomer (+)-3.HCl showed no inhibitory activity at 1.0 mg/kg i.v. Compound (-)-3.HCl (FK584) has pharmacological properties similar to those of terodiline, as evaluated by in vitro assay and is currently in clinical development for the treatment of overactive detrusor.

Development of the Bowman's and Jacobson's glands in the Japanese reddish frog, Rana japonica.

The olfactory organs are equipped with their own associated glands, Bowman's glands (BG) of the olfactory epithelium (OE), and Jacobson's glands (JG) of the vomeronasal organ (VNO). Histology and ultrastructure of these glands in the adult were investigated in the Japanese reddish frog, Rana japonica, along with their development from hatching to the end of metamorphosis. In the adult, BG cells contained large, electron-opaque secretory granules, intensely PAS-positive by light microscopy, while JG cells contained middle-sized secretory granules with moderate electron density, only faintly PAS-positive. Embryologically, BG appeared within the OE at 44 days after hatch, increased in number and were situated in the lamina propria under the OE at 52 days after hatch. BG cells contained large, electron-opaque granules, and well-developed rER at this time. While, JG appeared much earlier than BG. The VNO appeared as a concave of the ventral part of the OE at 4 days after hatch, and JG appeared under the VNO at 10 days after hatch. JG cells contained well-developed rER at 12 days after hatch. Secretory granules appeared in a small number in JG at 24 days after hatch, and increased thereafter. These findings suggest that JG may take part in secretion earlier than BG in ontogeny.

Development of the olfactory epithelium and vomeronasal organ in the Japanese reddish frog, Rana japonica.

Histological and ultrastructural development of the olfactory epithelium (OE) and vomeronasal organ (VNO) was investigated in the Japanese reddish frog, Rana japonica. Tadpoles, from hatching to the end of metamorphosis, and adult frogs were examined. In the adult, olfactory cells of the OE were equipped with olfactory vesicles with long cilia, but supporting cells with microvilli. The supporting cells of the OE contained secretory granules, PAS-positive by light microscopy, in their apical cytoplasm. On the contrary, sensory cells of the VNO were equipped with microvilli, and supporting cells of the VNO were equipped with cilia, but without secretory granules. Embryologically, the olfactory cells were indistinguishable from the supporting cells in the olfactory placode (primitive OE) lining the nasal pit, at hatch. The VNO appeared as a concave of the ventral part of the OE at 4 days after hatch. At the time, the olfactory and supporting cells of the OE became distinguishable from each other. Secretory granules were formed in the supporting cells of the OE at 36 days after hatch, and the OE was similar in fine structure to that in the adult. While, the VNO remained immature at 24-36 days after hatch, and did not complete its ultrastructural development at 60 days after hatch, the end of metamorphosis. These findings suggest that the OE may take part in the olfaction earlier than the VNO in ontogeny.

Effects of vitamin D3 injection on activity of thyroid parafollicular cells in pregnant rats.

Effects of vitamin D3 (VD3) injection on the activity of thyroid parafollicular cells (C cells) and calcium (Ca) metabolism were examined in rats of non-(NP), middle (MP) and late pregnancy (LP). At 3 days after injection, the average area of a C cell was significantly wider (P<0.01) in the VD3 groups than that in their control groups in NP, MP, and LP. On the otherhand, the plasma ca concentration in the VD3 groups decreased significantly (P<0.05) in comparison with that in their control groups in NP and LP, and tended to decrease in MP. These results suggest that injection of VD3 may accelerate the activity of C cells, which may result in the decrease of plasma Ca concentration in both non-pregnant and pregnant rats.

Immunohistochemical and enzyme histochemical characteristics of short axon cells in the olfactory bulb of the golden hamster.

Short axon (SA) cells in the olfactory bulb are subdivided into six types after Golgi impregnation, although their functional significance is not fully elucidated. In the present study, we examined the golden hamster olfactory bulb by immunohistochemistry to localize neurotransmitters, neuron-specific marker, and nitric oxide synthase (NOS) in the SA cells. Enzyme histochemical staining was also performed to detect the activity of nicotinamide adenine dinucleotide phosphate (NADPH)-diaphorase, which is identified with NOS. In the main olfactory bulb (MOB), neuropeptide Y (NPY)-, NOS-, and NADPH-diaphorase-positive SA cells were detected in the glomerular layer (GL), vasoactive intestinal polypeptide (VIP)-positive SA cells in the external plexiform layer (EPL), and NPY-, somatostatin (SOM)-, protein gene product 9.5 (PGP 9.5)-, NOS-, and NADPH-diaphorase-positive SA cells in the granule cell layer (GCL). In the accessory olfactory bulb (AOB), VIP- and PGP 9.5-positive SA cells were detected in the mitral/tufted cell layer (MTL), and NPY-, SOM-, NOS-, and NADPH-diaphorase-positive SA cells in the GCL. The common presence of NPY- SOM-, VIP-, PGP 9.5-, NOS-, and NADPH-diaphorase-positive SA cells in both the MOB and the AOB may suggest that respective types of cells with the same immunoreactivity play the same role no matter where these cells are located in the MOB or the AOB.

Evaluation of cardiac sympathetic nervous function by 123I-metaiodobenzylguanidine scintigraphy in insulin-treated non-insulin dependent diabetics with hypoglycemia unawareness.

The association between the lack of adrenergic symptoms during hypoglycemia and myocardial 123I-metaiodobenzylguanidine (MIBG) accumulation was investigated in 12 insulin-treated non-insulin-dependent diabetes mellitus (NIDDM) patients who had no evidence of heart disease. These patients were divided into 2 groups according to the presence (group A) or absence (group B) of adrenergic symptoms during hypoglycemia. Autonomic function tests revealed significantly severe autonomic dysfunction in group B compared to that in group A. Insulin infusion test indicated no significant difference in the catecholamine response between the two groups. 123I-MIBG scintigraphy showed that the heart/mediastinum ratio of MIBG uptake was significantly lower, and scintigraphic defect was greater in group B than in group A. There were no significant differences in the washout rate between the two groups. These results suggested that the lack of adrenergic symptoms during hypoglycemia may be associated with cardiac sympathetic nervous dysfunction in insulin-treated NIDDM patients, and this dysfunction is mainly due to cardiac sympathetic denervation.

Plasma flow and net nutrient flux across gut and liver of cattle fed twice daily.

The objectives of this study were 1) to quantify daily patterns of plasma flow and metabolite flux through portal-drained viscera (PDV) and liver in cattle fed twice daily and 2) to identify an interval for blood sampling that would approximate the average daily plasma flow and nutrient flux values. Data are from three experiments in which multicatheterized cattle were fed at or near ad libitum intake twice daily. Five lactating primiparous Holstein cows (506 kg, fed at 0730 and 1930) ate 17.3 kg DM/d as chopped alfalfa hay:corn grain plus supplement (urea and minerals) 50:50 (Exp 1). Five beef steers (474 kg, fed at 0900 and 2100) ate 8.3 kg DM/d as chopped switchgrass hay:corn grain plus supplement 37:63 (Exp 2). Six beef steers (306 kg fed at 0900 and 2100) ate 6.9 kg DM/d as chopped alfalfa hay (Exp 3). Plasma flow (by dilution of para-aminohippurate) was measured hourly for 24 h. Plasma flows (mean +/- SE) through PDV were 1,264 +/- 147, 538 +/- 56, and 499 +/- 26 L/h for Exp. 1, 2, and 3, respectively. Corresponding liver flows were 1,662 +/- 216, 642 +/- 41, and 591 +/- 30 L/h. The within-animal differences from their respective daily means were estimated as a function of time of day using nonparametric smoothing. Across experiments, PDV and liver flows were above the daily mean from 1200 to 1400, were not different from the daily mean from 1600 to 1700, and were below the daily mean from 1930 to 2130. Metabolites measured were ammonia, urea, alpha-amino N, and glucose. In general, metabolite flux was not different from the average daily mean values between 1200 and 1600. Blood sampling over 12 h or one 12-h feeding cycle is sufficient for daily plasma flow and metabolite flux estimation in cattle fed twice daily.

Ultrastructural study of neurosecretory cells in the nervous system in the cestode (Taenia hydatigena).

The neurosecretory cells in nervous system of the Taenia hydatigena was studied light- and electron-microscopically. On the basis of the cytological structure they were divided into two types: type I neurosecretory cells, which containing large dense-cored vesicles, small elongated mitochondria and a large amount of free ribosomes, were located mainly in the central and peripheral nervous system; type II neurosecretory cells were characterized by their moderate amount of free ribosomes, endoplasmic reticulum which contacted with the membranes of perikarya, large ovoid mitochondria, large dense-cored vesicles and their localization in the musculature near serve profiles. The synaptic and nonsynaptic contacts i.e. omega figure and exocytosis release sites were seen in the neuropile, musculature and excretory ducts. The present findings suggest that neurosecretory cells in nervous system may play an important integrative roles of both neuronal and endocrine in the flatworm.

[Stress hormone response during midazolam/fentanyl anesthesia combined with epidural anesthesia for abdominal total hysterectomy].

Stress hormone response was investigated during midazolam/fentanyl/oxygen/air anesthesia combined with epidural anesthesia in six patients undergoing abdominal total hysterectomy (group MF). A control group of six patients received nitrous oxide/oxygen/sevoflurane anesthesia combined with epidural anesthesia (group C). We evaluated plasma concentrations of ACTH, cortisol, aldosterone and beta-endorphin during the control period, 10 min, 60 min after incision, and in the recovery room. In group C, plasma concentrations of ACTH, cortisol, aldosterone and beta-endorphin increased significantly 60 min after incision compared with control concentrations. But in group MF, they did not increase significantly during anesthesia. In the recovery room there were significant differences compared with preinduction concentrations in both groups. It is concluded that midazolam/fentanyl anesthesia combined with epidural anesthesia is more effective than nitrous oxide/oxygen/sevoflurane anesthesia combined with epidural anesthesia to control the stress response to surgery.

[Anesthetic and postoperative care of a patient with Crigler-Najjar syndrome type II].

Crigler-Najjar syndrome is a rare inherited deficiency of bilirubin uridine diphosphate glucuronyl transferase, characterised by lifelong unconjugated hyperbilirubinemia. We report anesthetic and postoperative management of a patient with this syndrome. A 64-yr-old man with Crigler-Najjar syndrome type II underwent cholecystectomy. Preoperative laboratory values included total bilirubin 8.4 mg.dl-1 and direct-bilirubin 1.8 mg.dl-1. He received epidural anesthesia combined with general anesthesia with nitorous oxide and isoflurane. He had no remarkable perioperative complications.
