[Influence of carrageenin inflammation on prostaglandin release from rat synovial membrane and action of anti-inflammatory drugs (author's transl)].

Carrageenin was injected into the knee joint cavity of rats, and influence of anti-inflammatory drugs on prostaglandin (PG) release in vitro from inflamed synovial membrane was studied. Release of PGE and F from inflamed synovial membrane was increased. Aspirin suppressed PGE and F release from non-inflamed and inflamed synovial membrane in vivo and in vitro, but suppression of PGF release was more intense. Hydrocortisone suppressed release of PGE and F from inflamed synovial membrane in vivo, and did not influence PG release in vitro. Release of PGE and F fron non-inflamed synovial membrane was not influenced by hydrocortisone in vivo and in vitro. PGE content in inflamed synovial membrane was increased markedly, and PGE and F content in inflamed and non-inflamed synovial membrane was decreased markedly by aspirin in vivo. Content of PGE and F in non-inflamed and inflamed synovial membrane showed a tendency to decrease with hydrocortisone administration in vivo, but such was not significant.

Effects of cimetidine on healing of chronic gastric and duodenal ulcers in dogs.

The effects of cimetidine on healing of gastric and duodenal ulcers induced in mongrel dogs were studied. Gastric ulcers were produced by subserosal injection of acetic acid solution and duodenal ulcers by topical application of acetic acid on the serosal surface of the duodenum. Oral treatment with cimetidine, 450 mg/dog/day in three divided doses for 14 days, produced a remarkable acceleration of healing of duodenal ulcers but exerted little influence on gastric ulcers.

Surgical treatment of the aortic aneurysm.

For 93 cases of thoracic and 118 cases of abdominal aneurysms, the over-all operative mortality rate was 24.7 per cent and 9.3 per cent, respectively. Although the over-all operative mortality rate for 37 patients with aneurysms of the ascending aorta or aortic arch had been 40.5 per cent, recent advances in surgical technique led to a higher survival rate so that since 1975 no death occurred among 14 consecutive surgery cases. Cardiopulmonary bypass with or without selective perfusion of the carotid arteries or temporary external bypass procedures were employed in these 14 cases. The over-all operative mortality rate for 56 patients with aneurysms of the descending thoracic aorta was 14.3 per cent. Temporary external bypass procedures were employed in 49 cases. The operative mortality rate for 99 patients with unruptured abdominal aneurysms was five per cent, and that for 19 patients with ruptured aneurysms was 31.5 per cent.

Elementary steps in the acto-H-meromyosin ATPase reaction to arterial smooth muscle.

Transient and steady state kinetics were studied in the interactions of ATP with acto-H-meromyosin reconstituted from bovine arterial heavy-meromyosin (HMM) and rabbit skeletal muscle F-actin. The results showed that the rate of dissociation of the hybrid acto-HMM induced by ATP was slower than the rate of the fluorescence enhancement of HMM, and that the rate of the P1 burst of HMM was unaffected by addition of skeletal muscle F-actin. The ATPase [EC 3.6.1.3] activity of arterial HMM was activated only slightly even with addition of high concentrations of skeletal muscle F-actin. Furthermore, the rates of dissociation of the hybrid acto-HMM induced by ATP and reassociation of dissociated arterial HMM with skeletal muscle F-actin after decomposition of ATP were much lower than those of skeletal muscle acto-HMM.

Strain differences in aspirin-induced gastric ulceration in rats.

We found that there are strain differences in aspirin-induced ulceration in pylorus-ligated rats; the ulcer indices varied, from high to low, in the following order: Donryu greater than Sprague-Dawley greater than Wistar. Several experiments including analysis of gastric contents or ionic flux, determination of serum aspirin esterase activity, absorption of aspirin from the stomach, prothrombin time and hexosamine content in gastric mucosa and juice were performed to elucidate the origin of the differences. A significantly higher acid output in Donryu rats, and higher hexosamine content in the gastric mucosa of Wistar rats were noted. However, it appears unlikely that these factors only contribute to the marked strain difference in aspirin-induced ulcers. The possible different sensitivity of gastric mucosal cell itself to aspirin must be considered.
