Synthesis and secretion of alpha 1-microglobulin by human lymphocytes.

alpha 1-Microglobulin (alpha 1-m) was purified by column chromatography from a supernatant fluid of cultured T and B lymphocytes stimulated with mitogens. This protein had a molecular weight of 33,000 as determined by sodium dodecyl sulphate (SDS) polyacrylamide gel electrophoresis, migrated in the alpha 1-region on immunoelectrophoresis and proved immunologically identical to alpha 1-m which had been purified from the urine of patients with renal tubular disorders. Using indirect immunofluorescence, alpha 1-m was detected on the surface of both T and B lymphocytes, displaying the same intensity on each cell type. These findings indicate that alpha 1-m is actively produced and secreted by T and B lymphocytes.

Effect of papaverine on Ca2+ uptake by a mitochondrial fraction isolated from rat uterine smooth muscle.

A mitochondrial fraction was isolated from rat uterine smooth muscle. The effect of papaverine (3 X 10(-6), 10(-5), 3 X 10(-5), 10(-4) M) on Ca2+ accumulation by the fraction was studied. The effect of papaverine was changed by using different substrates for Ca2+ accumulation. Papaverine inhibited the Ca2+ accumulation supported by glutamate plus ATP concentration-dependently. The Ca2+ accumulation supported by ATP alone was also inhibited. On the other hand, the Ca2+ accumulation supported by succinate and ATP was increased by papaverine. The effect was not concentration-dependent; papaverine (3 X 10(-5) M) was the most effective concentration for the effect with a shorter incubation (2 min).

Immobilization of plasminogen activator, urokinase, on nylon.

Urokinase (UK), a fibrinolytic enzyme activator purified from human material was immobilized on nylon using different procedures. One was a modified method of immobilization of antigen or antibody initially carried out by Edelman and others in 1971 (Procedure I). The other was our newly devised method (Procedure II) (Sugitachi et al. 1976). Major specificities of the immobilized UK are as follows: 1. The UK revealed properties of a plasminogen activator and the optimum pH of the immobilized UK was between 7.2 and 7.4, these values being in good parallel with that of soluble UK. The immobilized UK maintained a stable fibrinolytic activity after long-term preservation and heat-treatment. 2. As the fibrinolytic activity of immobilized UK was found to be inhibited by the antiplasmin in human plasma, an antiplasmin inhibitor was immobilized on the nylon together with the UK. The antiplasmin activity was to some extent prevented using this procedure. 3. Nylon tubes immobilized with UK and antiplasmin inhibitor were used for thrombotic coagulation studies carried out according to the method of Chandler. Thrombus formation time (TFT) of UK-immobilized tubes was 30 min, while that of the non-treated tubes was no longer than 10 min.

Radioimmunoassay of an early plasmin degradation product of human fibrinogen, "fragment A", and its clinical application.

Upon the plasmin digestion of human fibrinogen, an early cleavage product, which has been designated as fragment A, was isolated, and to study the action of plasmin in the circulation, radioimmunoassay for fragment A was carried out. This assay used rabbit immune serum obtained by injection of fragment A mixed with complete Freund's adjuvant, and fragment A was labeled with 125I using the Chloramin-T method. In 20 normal healthy donors its serum level was 3.57 +/- 1.62 microgram/ml (mean +/- SD), and it was increased significantly in certain diseases, such as acute leukemias, cardiovascular disorders, malignancies, renal failure, systemic lupus erythematosus and sepsis.

Strain differences in aspirin-induced gastric ulceration in rats.

We found that there are strain differences in aspirin-induced ulceration in pylorus-ligated rats; the ulcer indices varied, from high to low, in the following order: Donryu greater than Sprague-Dawley greater than Wistar. Several experiments including analysis of gastric contents or ionic flux, determination of serum aspirin esterase activity, absorption of aspirin from the stomach, prothrombin time and hexosamine content in gastric mucosa and juice were performed to elucidate the origin of the differences. A significantly higher acid output in Donryu rats, and higher hexosamine content in the gastric mucosa of Wistar rats were noted. However, it appears unlikely that these factors only contribute to the marked strain difference in aspirin-induced ulcers. The possible different sensitivity of gastric mucosal cell itself to aspirin must be considered.

Antithrombogenicity of immobilized urokinase--clinical application.

A plasminogen activator, urokinase (UK) was immobilized on four different polymer surfaces: nylon, silicone rubber, poly (vinyl chloride) and a polyester, using our newly devised technique. Each material proved to have plasminogen activating activity. In vitro the thrombus formation time (TFT) of the UK-immobilized nylon tubes was over 45 minutes and the TFT of UK-immobilized silicone rubber tubes varied. When applied clinically, the UK-immobilized nylon tubes showed excellent antithrombogenicity.

A simple and practical method for isolation of an early plasmin degradation product of human fibrinogen.

One of the earliest plasmin degradation products of human fibrinogen, so-called fragment A, was isolated by a simple method. This peptide has a molecular weight of approximately 22,500, migrating electrophoretically at beta-area, and its amino acid composition shows a very high content of glycine, serine, threonine and proline, and a markedly low content of hydrophobic amino acids. This fragment does not react against anti-fibrinogen; however, the anti-serum of this fragment reacts strongly with fibrinogen.

Effects of cimetidine and atropine sulfate on gastric secretion and healing of gastric and duodenal ulcers in rats.

Cimetidine, a new histamine H2-receptor antagonist (50 or 100 mg/kg) and atropine sulfate (15 mg/kg) given intraduodenally, markedly inhibited gastric secretion in pylorus-ligated rats. Cimetidine (100 or 200 mg/kg/day) given for 10 or 12 consecutive days orally in two divided doses, significantly promoted the healing rate of both gastric and duodenal ulcers induced in rats. Atropine (30 mg/kg/day) also significantly accelerated the healing of duodenal ulcers but failed to affect gastric ulcers.
