Analysis of the transformation of human lymphocytes by Epstein-Barr virus. I. Sequential occurrence from the virus-determined nuclear antigen synthesis, to blastogenesis, to DNA synthesis.

A B-cell population of human cord blood lymphocytes was exposed to the B95-8 strain of Epstein-Barr virus (EBV), and simultaneous observations of immunofluorescence, cellular morphology and autoradiography were carried out in each individual cell. It was evident that EBV-determined nuclear antigen (EBNA) synthesis occurred prior to blastogenic response and DNA synthesis. EBNA-positive cells could be observed as early as 12 h after infection and reached a maximum of 17% at 24 h, followed by a plateau for a subsequent 12 h. The positive cells were seen exclusively as morphologically normal lymphocytes until 18 h; at 24 h, blastogenesis became evident, without cell division. DNA synthesis was detected at 36 h in EBNA-positive blast cells, after which these cells increased rapidly. EBNA synthesis was similarly evident in the presence of cytosine arabinoside, but was significantly inhibited by a short-term exposure to cycloheximide immediately after infection. These findings suggest that the early events in EBV-induced transformation of human lymphocytes occur sequentially from EBNA synthesis, to blastogenesis, to DNA synthesis and that the crucial step of such transformation is probably involved in protein synthesis occurring in the very early stage of EBV infection.

Impedance cardiography for the assessment of cardiac function during exercise.

1. Impedance cardiography proved valuable to estimate the stroke volume and other cardiac parameters noninvasively, when the electrical resistivity of the blood was corrected with hematocrit. 2. Cardiac response to graded exercise on a bicycle ergometer up to the maximal load in healthy young and active middle-aged subjects was divided into four phases from the changes in cardiac parameters such as stroke volume, cardiac output, ejection index and height of the atrial wave. 3. The ischemic patients did not tolerate well the exercise after the second phase or drift phase of exercise. 4. Physical training enhanced the cardiac function in work through increase in cardiac output and work load.

A rapid and precise method for the determination of vitamin D3 in rat skin by high-performance liquid chromatography.

In order to develop the investigations into photobiogenesis of vitamin D3, a rapid and precise method for the determination of the vitamin in rat skin was established by using high-performance liquid chromatography (HPLC). The proposed method included saponification of small pieces of rat skin, extraction of the unsaponifiable matter and application to HPLC using "Zorbax SIL" (straight-phase) as an adsorbent and 0.5% isopropanol in n-hexane as a mobile phase. The applicable lower limit of the method was 2ng of vitamin D3/cm2 of subcutaneous tissue-removed skin and it was possible to assay a concentration higher than 2 ng/cm2. The proposed method was applied to determine the content of vitamin D3 in rat skin obtained from in vivo and in vitro irradiation experiments. In the in vitro experiment, the yield of vitamin D3 increased in proportion to the irradiation time. On the other hand, the yield in the in vivo experiment showed a proportional increase similar to the in vitro experiment until 60 min irradiation, while a nearly constant value was obtained by irradiation for longer than 60 min. When the rat skin obtained from the in vitro experiment was irradiated with monochromatic UV rays in the range s60-350 nm, the most effective wavelength for the formation of vitamin D3 was confirmed to be 303 nm, which differs from the result obtained from the experiment in a test tube (295 nm). Moreover, the yield of vitamin D3 by irradiation with UV rays below 288 nm was extremely low, which again differed from the results of a test tube experiment. These differences were thought to be due to the filter effect of the malpighian layer in the epidermis of rat skin.

Assay of 25-hydroxyvitamin D3 in human plasma by high-performance liquid chromatography.

A method using preparative and analytical high-performance liquid chromatography (HPLC) is proposed for the assay of 25-hydroxyvitamin D3 (25-OH-D3) in human plasma. A constant volume (0.2--2.0 ml) of a plasma sample was saponified. The unsaponifiable matter was first applied to preparative HPLC using a column of the straight-phase type (Zorbax SIL) in order to separate a 25-OH-D3 fraction from lipophilic concomitants giving ultraviolet-absorbing noise. Then, the separated 25-OH-D3 fraction was applied to analytical HPLC using a column of the reversed-phase type (Zorbax ODS) in order to measure the content of 25-OH-D3 from the peak height. This is a revised method from Jones (1978): Clin. Chem., 24, 287--298). The results showed that the clean-up procedure by the first preparative HPLC was successfully performed because the peak corresponding to 25-OH-D3 on the chromatogram of the second analytical HPLC was not disturbed by any other interfering peaks. Moreover, recovery through the whole procedure was satisfacotry (about 100%) and the procedures of saponification and isolation of the unsaponifiable matter diminished the overload to the columns. These are the revised points of Jones' method. When two determinations were performed on 12 samples of plasma taken from normal adults in October, the values were 22.6 +/- 4.8 and 21.0 +/- 3.6 (mean +/- SD) ng/ml, respectively.

Trialwise tracking method for measuring drug-affected sensory threshold changes in animals.

Rats and rhesus monkeys were trained under a multiple schedule, the components of which were random ratio schedules for food presentation and for shock presentation. The discriminative stimulus for the shock presentation component was a pure tone for the rats and a light for the rhesus monkeys. In the test session under the extinction condition for the shock presentation component, the intensity of the discriminative stimulus was successively either decreased by fixed units when the conditioned suppression was observed or increased when the conditioned suppression was not observed. The levels finally oscillated within a narrow range around the threshold. The auditory thresholds of rats were increased by intramuscular administration of quinidine at 20 mg/kg and also by repeated intramuscular administration of kanamycin at 250 and 500 mg/kg/day. In rhesus monkeys, visual thresholds were raised by application of pilocarpine at 0.02-0.16 mg/kg to the eyes and also by subcutaneous administration of LSD-25 at 4-8 micrograms/kg in one monkey and at 20-30 micrograms/kg in another. The method used for tracking the animals' sensory thresholds was sensitive enough to test the selective effect of the drugs and was also a relatively easy way to obtain a stable behavioral baseline for experimental purposes.

Horseradish peroxidase-labelled cells in cervical sympathetic and trigeminal nervous systems after injection of HRP into tooth pulp in monkeys and tree shrews (a preliminary report).

The neurons that project to the tooth pulp were examined by the HRP-retrograde transport technique, using 4 monkeys (3 Japanese monkeys and a crab-eating monkey) and 6 tree shrews. The present investigation showed that a large number of sympathetic cervical ganglion neurons, small and medium in size, project to the molar teeth from the superior, middle and inferior ganglia and a small number of Gasserian gangilon neurons (small in size) to the molar teeth ipsilaterally.

The simultaneous observations of immunofluorescence and cellular morphology in viral infections: a new procedure.

A simple method for comparative observations of immunofluorescence and cellular morphology has been developed in the investigation of Epstein-Barr virus (EBV) infection and oncogenesis. When cells were stained for immunofluorescence of EBV-determined nuclear antigen, early antigens, or the viral capsid antigen, followed by treatment with hematoxylin, viral marker expression and cellular morphology could be simultaneously observed in individual cells by simply changing ultraviolet illumination to visible light. In our double-staining method, hematoxylin was not counteractive to specific immunofluorescence, and pretreatment of cells with antibodies did not affect hematoxylin staining. This method may possibly be more widely applied in virology and other fields.

Malignant fibrous histiocytoma of the temporomandiblar joint. Report of a case.

A case of malignant fibrous histiocytoma of the temporomandibular joint in a 21-year-old Japanese female was presented. The patient died of the tumor with local aggressive growth but without any organ metastasis, despite the intensive treatment including radical operation of the tumor, irradiation, and chemotherapy. This case suggested that radical curative operation was the only method of the treatment for this tumor. The clinicopathological features of the tumor are discussed, making reference to the malignant fibrous histiocytoma.

Impedance cardiography for estimating cardiac output during submaximal and maximal work.

Impedance cardiography was used to estimate cardiac output in 10 men during rest and within 5 s after exercise on a bicycle ergometer, including work up to and including maximal aerobic capacity. An indwelling venous catheter permitted simultaneous sampling of venous blood for observing changes in hematocrit associated with each exercise level. Cardiac output, calculated from a standard equation which assumes a constant value of 150 omega.cm for the electrical resistivity of blood, was compared with corresponding calculations in which blood resistivity was individually determined as a function of hematocrit. It is concluded that many of the discrepancies in the literature related to values for cardiac output obtained during exercise by the impedance method may be inherent in calculations that do not consider the changing electrical resistivity of the blood with a changing hematocrit.

Chromogenic substrates for horseshoe crab clotting enzyme. Its application for the assay of bacterial endotoxins.

An endotoxin-activated hemocyte lysate from the horseshoe crab (Tachypleus and Limulus) was found to hydrolyze Bz-Ile-Glu-(gamma-OR)-Gly-Arg-p-nitroanilide (PNA), Bz-Val-Gly-Arg-PNA, Boc-Val-Leu-Gly-Arg-PNA, and Boc-Leu-Gly-Arg-PNA, all of which have the COOH-terminal Gly-Arg sequence. This amidase activity was due to a clottting enzyme contained in the lysate. Furthermore, the amidase activity increased by increasing the concentration of bacterial endotoxin (E. coli, 0111-B4) added to the lysate. Therefore, the measurement of the endotoxin-induced amidase activity made it possible to determine the concentration of the endotoxin.
