Induction of cytosolic Ca2+ elevation mediated by Mas-7 occurs through membrane pore formation.

Mas-7, a mastoparan derivative, induces elevation of intracellular free Ca2+ concentration ([Ca2+]i) along two independent pathways. The minor contribution occurs via phospholipase C activation and is negatively regulated by treatment with phorbol 12-myristate 13-acetate, a protein kinase C activator. The major contribution involves plasma membrane pores allowing not only Ca2+, Mn2+, and Na+ to enter but also the uptake of ethidium bromide (314 Da) and lucifer yellow (457 Da), but not fura-2 (831 Da), Evans blue (961 Da), and fluorescein-conjugate phalloidin (1,175 Da). Mas-7-induced current, as measured in planar lipid bilayers, reveals that Mas-7-induced pores have two slope conductances, 290 and 94 pS, and that the pores are nonselective for cations. The results also indicate that Mas-7 can produce pores by direct interaction with the plasma membrane without the involvement of membrane proteins and cytosolic factors. Besides in human neuroblastoma cells, similar Mas-7 effects were also observed in other cell lines such as HL-60, 1321N1 human astrocytoma, and bovine chromaffin cells. The data suggest that the Mas-7-induced [Ca2+]i elevation is the combined result of Ca2+ release from stores via phosphoinositide turnover and prolonged Ca2+ influx through membrane pores.

Synergistic activation of adenylyl cyclase is dependent upon phospholipase C-mediated processes in human neuroblastoma SK-N-BE(2)C cells.

1-[6-[17 beta-3-Methoxyestra-1,3.5(10)-trien-17-yl]amino]hexyl]-1 H-pyrrole-2,5-dione (U-73122), an inhibitor of processes involved in the activation of phospholipase C, was used to assess the role of phospholipase C activation in the synergistic elevation of cAMP induced by carbachol and prostaglandin E2 in human neuroblastoma (SK-N-BE(2)C cells. Pre-treatment of the cells with U-73122 resulted in inhibition of carbachol-induced intracellular Ca2+ ([Ca2+]i) rise and inositol 1,4,5-trisphosphate (InsP3) generation, with maximal and half maximal inhibition (IC50) occurring at approximately 15 microM and 3.2 microM, respectively. U-731222 also inhibited the synergistic enhancement of cAMP accumulation induced by carbachol and prostaglandin E2 in a concentration-dependent manner with maximum and IC50 at 12 +/- 4 microM and 3.4 +/- 0.3 microM, respectively. However, U-73122 did significantly inhibit prostaglandin E2-induced production. While 1,2-bis(o-aminophenoxy)ethane-N,N,N,'N'-tetraacetic acid (BAPTA/AM) treatment decreased the synergistic cAMP accumulation by 28%m addition U-73122 further decreased it down to complete inhibition. Furthermore, GTP gamma S- and A1F4(-)-induced InsP3 generation in digitonin-mediated permeabilized cells was also inhibited by U-73122 treatment. Pre-treatment of the cells with neomycin, another blocker of the phospholipase C pathway, also resulted in inhibition of the carbachol-induced [Ca2+]i rise, InsP3 generation, and the enhancing effect on cAMP accumulation, to a comparable extent. But, Ca2+ chelation by BAPTA/AM in addition to neomycin treatment further decreased the cAMP accumulation. These results suggest that the increase in cytosolic Ca2+ and the coupling process between muscarinic receptor-like G-protein and phospholipase C are important for the synergistic activation of adenylyl cyclase in SK-N-BE(2)C cells.

The high-latitude cloud MBM 7. I. H I and CO observations.

The high-latitude cloud (HLC) MBM 7 has been observed in the 21 cm H I line and the 12CO(1-0) and 13CO(1-0) lines with similar spatial resolutions. The data reveal a total mass approximately 30 M solar for MBM 7 and a complex morphology. The cloud consists of a cold dense core of 5 M solar surrounded by atomic and molecular gas with about 25 M solar, which is embedded in hotter and more diffuse H I gas. We derive a total column density N(H I + 2H2) of 1 x 10(21) cm-2 toward the center and 1 x 10(20) cm-3 toward the envelope of MBM 7. The CO line indicates the existence of dense cores [n(H2) > or = 2000 cm-3] of size (FWHM) approximately 0.5 pc. The morphology suggests shock compression from the southwest direction, which can form molecular cores along the direction perpendicular to the H I distribution. The H I cloud extends to the northeast, and the velocity gradient appears to be about 2.8 km s-1 pc-1 in this direction, which indicates a systematic outward motion which will disrupt the cloud in approximately 10(6) yr. The observed large line widths of approximately 2 km s-1 for CO suggest that turbulent motions exist in the cloud, and hydrodynamical turbulence may dominate the line broadening. Considering the energy and pressure of MBM 7, the dense cores appear not to be bound by gravity, and the whole cloud including the dense cores seem to be expanding. The distance to HLCs suggest that they belong to the galactic plane, since the scale height of the cloud is < or approximately equal to 100 pc. Compared to the more familiar dense dark clouds, HLCs may differ only in their small mass and low density, with their proximity reducing the filling factor and enhancing the contrast of the core and envelope structure.

Transcriptional enhancement of tyrosine hydroxylase by prostaglandin E2 in SK-N-BE(2) C cells.

Transcriptional regulation of the rat tyrosine hydroxylase (TH) gene by prostaglandin E2 (PGE2) was investigated in human neuroblastoma SK-N-BE(2)C cells. Prostaglandins increased intracellular cAMP in the presence of 3-isobutyl-1-methylxanthine (IBMX), a cAMP phosphodiesterase inhibitor. Among the prostaglandins tested for their cAMP raising property PGE2 was the most effective. The results suggest that the cells express adenylyl cyclase-linked prostanoid receptors that have a higher affinity for PGE2 than for any other naturally occurring prostaglandin. The treatment of cells with PGE2 increased the TH gene expression approximately 2-fold, even though the cAMP accumulation induced by PGE2 alone was almost negligible. Simultaneous treatment with PGE2 and IBMX enhanced the gene expression concomitantly with a marked accumulation of cAMP. Transient transfection assays with 5' upstream serially deleted constructs of the rat TH gene promoter region fused to the chloramphenicol acetyltransferase (CAT) gene revealed that a cAMP response element (CRE) located at -45 to -38 from the start of the TH gene was essential for the enhancement of TH gene expression by PGE2. Site-directed mutagenesis and specific deletion within the sequence of the CRE motif abolished the transcriptional enhancement by PGE2. In addition, a protein kinase A (PKA) inhibitor, H89, specifically blocked the PGE2 effect on TH gene expression. Northern blot analysis revealed that the increase in TH gene transcription with PGE2 is associated with an elevated TH mRNA level. Gel retardation and competition assays confirmed that the binding of nuclear factors to the CRE site was sequence specific and was augmented by PGE2. Our data indicate that PGE2 enhances transcription of the TH gene mediated by the CRE motif through the activation of PKA. They also suggest that the signal flow from the adenylyl cyclase-linked prostanoid receptor to the nucleus is efficient although cAMP accumulation is not prominent.

Change of glutathione S-transferases in the skin by ultraviolet B irradiation.

Glutathione S-transferases (GSTs) may play an important role in protecting skin from ultraviolet radiation (UVR). However, the study on the response of GST to UVR is limited at present. We have examined the effects of a single exposure to ultraviolet B (UVB) radiation on GST in cultured human keratinocytes and the epidermis of SKH/hr-1 hairless mice. We have also investigated the changes of skin GST by chronic irradiation of UVB on the hairless mice. Significant decreases in GST activities in vitro and in vivo were observed at 24 h after 30 and 50 mJ/cm2 UVB irradiation. Chronic UVB exposure also caused decrease in GST activities of the skin tissue. However, any changes in mRNA expression or protein amount of GST have not been observed by Northern blot analysis and Western blot analysis after 30 mJ/cm2 UVB irradiation in cultured human keratinocytes, which suggests that mRNA expression and protein amount of GST are not affected by UVB. These results suggest that UVB irradiation results in inhibitory effect on GST activity in the skin.

Cyclic AMP-independent inhibition of voltage-sensitive calcium channels by forskolin in PC12 cells.

Forskolin has been used to stimulate adenylyl cyclase. However, we found that forskolin inhibited voltage-sensitive Ca2+ channels (VSCCs) in a cyclic AMP (cAMP)-independent manner in PC12 cells. Ca2+ influx induced by membrane depolarization with 70 mM K+ was inhibited when cells were preincubated with 10 microM forskolin. Almost maximum inhibitory effect on Ca2+ influx without any significant increase in cellular cAMP level was observed in PC12 cells exposed to forskolin for 1 min. In addition, the forskolin effect on Ca2+ influx was not affected by the presence of 2',5'-dideoxyadenosine, an inhibitor of adenylyl cyclase that reduces dramatically forskolin-induced cAMP production. 1,9-Dideoxyforskolin, an inactive analogue of forskolin, also inhibited approximately 80% of Ca2+ influx induced by 70 mM K+ without any increase in cAMP. The data suggest that forskolin and its analogue inhibit VSCCs in PC12 cells and that the inhibition is independent of cAMP generation.

AP1-mediated transcriptional enhancement of the rat tyrosine hydroxylase gene by muscarinic stimulation.

We investigated transcriptional regulation of the rat tyrosine hydroxylase (TH) gene by muscarinic stimulation in human neuroblastoma SK-N-BE(2)M17 cells. Carbachol treatment increased the levels of intracellular Ca2+ and inositol 1,4,5-trisphosphate (IP3) and enhanced transcription of the TH gene. The muscarinic receptor antagonist atropine completely abolished the carbachol effect on TH gene expression. When cells were loaded with 50 microM 1,2-bis(2-aminophenoxy) ethane-N,N,N',N'-tetraacetic acid/acetoxymethyl ester (BAPTA/AM) to chelate intracellular Ca2+, carbachol still raised intracellular IP3 level and enhanced TH gene expression. Transient transfection analysis of the 5' upstream region of TH gene revealed that the AP1 cis-acting element at -205 to -199 bp was responsible for carbachol stimulation. But carbachol did not enhance TH gene expression in protein kinase C (PKC)-activated or down-regulated cells that had been induced by 5-min or 24-h exposure to phorbol 12-myristate 13-acetate (PMA), respectively. Thus, Ca(2+)-independent PKC may play a role in carbachol-induced TH gene expression. We demonstrated by gel retardation and competition assays that a DNA sequence containing the wild-type AP1 site formed the specific DNA-protein complex. However, treatment with carbachol or PMA did not change the amount of the specific DNA-protein complex. Our results indicate that stimulation of phospholipase C-linked muscarinic receptors leads to elevated TH gene expression via AP1-mediated enhancement in a PKC-dependent pathway.

Characterization of Na+ influx mediated by ATP(4-)-activated P2 purinoceptors in PC12 cells.

1. Micromolar levels of extracellular ATP increased cytosolic Na+ concentration ([Na+]i) as well as cytosolic Ca2+ concentration ([Ca2+]i) in PC12 cells. 2. Pretreatment of cells with tetrodotoxin, benzamil or thapsigargin did not alter the ATP-induced Na+ influx. 3. Increased extracellular Mg2+ concentration decreased the ATP effect. Furthermore, when the extracellular ATP pool was treated to contain corresponding calculated concentrations of ATP4-, the increase in [Na+]i stayed linked to the ATP4- concentration rather than to the total ATP concentrations in the stimulants. 4. Extracellular ATP does not create nonselective pores as shown by the fact that ethidium bromide does not enter the cells upon ATP stimulation. 5. Among the tested nucleotides, only adenosine 5'-O-(3-thiotriphosphate), 2-methylthio ATP and 2-chloro ATP also caused Na+ influx. 6. Reactive Blue 2 specifically decreased the ATP effect in a concentration-dependent manner. 7. The results suggest that extracellular ATP triggers Na+ influx through a P2 purinoceptor which is activated by ATP4- in PC12 cells.

Genetic evidence for two sequentially occupied K+ binding sites in the Kdp transport ATPase.

Substrate binding sites in Kdp, a P-type ATPase of Escherichia coli, were identified by the isolation and characterization of mutants with reduced affinity for K+, its cation substrate. Most of the mutants have an altered KdpA subunit, a hydrophobic subunit not found in other P-type ATPases. Topological analysis of KdpA and the locations of the residues changed in the mutants suggest that KdpA has 10 membrane-spanning segments and forms two separate and distinct sites where K+ is bound. One site is formed by three periplasmic loops of the protein and is inferred to be the site of initial binding. The other site is cytoplasmic. We believe K+ moves from the periplasmic site through the membrane to the cytoplasmic site where it becomes "occluded," i.e. inexchangeable with K+ outside the membrane. Membrane-spanning parts of KdpA probably form the path for transmembrane movement of K+. The kinetics of cation transport in the mutants indicate that each of the two binding sites contributes to the observed Km for cations as well as to the marked discrimination between K+ and Rb+ characteristic of wild-type Kdp. Energy coupling in Kdp, mediated by the KdpB subunit, is performed by a different subunit from the one that mediates transport.

Cytosolic calcium is essential in the basal expression of tyrosine hydroxylase gene.

To test the role of cytosolic calcium in the basal expression of a neuronal gene, promoter activity of the rat tyrosine hydroxylase (TH) gene was monitored upon reduced resting level of intracellular calcium. TH promoter activity was decreased by cell-permeable calcium chelator, BAPTA/AM, in SK-N-BE(2)C human neuroblastoma cells. The cAMP response element (CRE) was mapped to the calcium responsible element by mutational and deletional analysis of the 5' upstream promoter region. Gel shift assay showed 2 CRE-specific DNA-protein complexes. The quantities of specific complexes were markedly decreased in BAPTA/AM-treated cells. These data suggest that resting level of intracellular calcium has a critical role in the basal expression of TH gene through the regulation of the binding of nuclear proteins to the CRE motif in the promoter.

Signal flows from two phospholipase C-linked receptors are independent in PC12 cells.

Bradykinin (BK) receptor and P2-purinergic receptor are known to be coupled to phospholipase C (PLC) in PC12 cells. To study the interaction between these two PLC-linked receptors, the presence of both receptors on individual cells was demonstrated by sequential Ca2+ spikes caused by BK and ATP in a single fura-2-loaded cell. BK- and ATP-induced catecholamine (CA) secretions were desensitized within 5 min. However, in the sequential experiment, the BK-induced homologous desensitization of CA secretion did not block the ATP-induced secretion, and vice versa. Each agonist-induced an increase in inositol 1,4,5-trisphosphate (IP3) production and intracellular free Ca2+ concentration also led to homologous desensitization. However, there was no heterologous desensitization between the two agonists. When the cells were treated with both BK and ATP simultaneously, the amounts of CA secretion, IP3 production, internal Ca2+ mobilization, and Ca2+ influx were all additive. We also found that both IP3-induced Ca2+ release from intracellular Ca2+ stores and Ca2+ influx from extracellular space were able to release [3H]norepinephrine, and the secretion induced by both agonists was exactly additive in the absence or presence of extracellular Ca2+. The data suggest that the CA secretions caused by BK or ATP may have separate secretory pathways even though they activate identical second messenger pathways.

Stimulation of adenylyl cyclase mediated by phospholipase C-linked M3 muscarinic receptor in human neuroblastoma SK-N-BE (2) C cells.

Muscarinic receptor in human neuroblastoma SK-N-BE(2)C cells was identified and characterized. Treatment of the cells with carbachol evoked the generation of inositol 1,4,5-trisphosphate (IP3) with a peak level reached at 1 min after stimulation. Carbachol increased intracellular Ca2+ ([Ca2+]i) with an EC50 value of 35 microM. In addition, carbachol produced a 1.3-3-fold increase in the cyclic AMP (cAMP) level compared with untreated control and elevated synergistically the cAMP level in the treatment with prostaglandin E2 (PGE2). The M3 antagonist p-fluorohexahydrosiladifenidol (IC50 = 0.5-0.8 microM) inhibited the increases in [Ca2+]i, IP3, and cAMP more effectively than the M1 antagonist pirenzepine (IC50 = 5-9 microM) and the M2 antagonist methoctramine (IC50 = 20-30 microM). The involvements of [Ca2+]i elevation and protein kinase C activation induced by phospholipase C activation were tested in the carbachol-induced cAMP production. The calcium chelator BAPTA/AM (75 microM) inhibited significantly the synergistic effects of carbachol and PGE2 on the production of cAMP, whereas the Ca2+ ionophore ionomycin (1 microM) clearly enhanced PGE2-induced cAMP production. However, phorbol 12-myristate 13-acetate did not enhance PGE2-stimulated cAMP production. These data suggest that phospholipase C-linked M3 receptors are present and that stimulation of the receptors activates adenylyl cyclase, at least in part, by the Ca(2+)-dependent system in the neuronal cells.

Inhibition of bradykinin-induced cytosolic Ca2+ elevation by muscarinic stimulation without attenuation of inositol 1,4,5-trisphosphate production in human neuroblastoma SK-N-BE(2)C cells.

Cross talk between two phospholipase C (PLC)-linked receptor signalings was investigated in SK-N-BE(2)C human neuroblastoma cells. Sequential stimulation with two agonists at 5-min intervals was performed to examine the interaction between muscarinic and bradykinin (BK) receptors. Pretreatment of cells with a maximal effective concentration (5 microM) of BK did not affect the subsequent carbachol (CCh)-induced [Ca2+]i rise, but CCh (1 mM) pretreatment completely abolished the BK-induced [Ca2+]i rise without inhibition of BK-induced inositol 1,4,5-trisphosphate (IP3) generation. Thapsigargin (1 microM) pretreatment abolished the subsequent BK- and CCh-induced [Ca2+]i rise, though it did not affect agonist-induced IP3 generation. However, the addition of atropine at plateau phases of CCh-induced [Ca2+]i rise and IP3 production caused a rapid decline to the basal levels and then restored the [Ca2+]i rise by BK. Treatment of cells with both CCh and BK at the same time showed additive effects in IP3 production. However, the [Ca2+]i rise induced by both agonists in the presence or absence of extracellular Ca2+ was the same as the responses triggered by CCh alone. The results suggest that each receptor or receptor-linked PLC activity is not influenced by pretreatment with the other agonist but IP3-sensitive Ca2+ stores are shared by signal pathways from both receptors.

Inhibition by ethaverine of catecholamine secretion through blocking L-type Ca2+ channels in PC12 cells.

Ethaverine, a derivative of papaverine, is used as a vasodilator and antispasmodic drug. We have investigated the effects of ethaverine on the secretion of [3H]norepinephrine from PC12 cells, of neuroendocrine origin. Treatment with ethaverine reduced catecholamine secretion in a concentration-dependent manner. The maximal inhibitory effect (90%) was achieved with 10 microM ethaverine, and the IC50 for secretion was approximately 2 microM. Ethaverine pretreatment for 1 min prior to stimulation by 70 mM K+ also decreased the level of intracellular Ca2+ in a concentration-dependent manner, as measured by fura-2 fluorescence. The IC50 for the inhibition of the increase in intracellular Ca2+ was approximately 2 microM. Nifedipine, a dihydropyridine L-type Ca2+ channel blocker, did not enhance the inhibitory effect of ethaverine on the 70 mM K(+)-induced increase in [Ca2+]i or catecholamine secretion. In contrast, the addition of the N-type voltage-sensitive Ca2+ channel antagonist omega-conotoxin with ethaverine resulted in further reductions in the increase in [Ca2+]i and catecholamine release. Maximally effective concentrations of ethaverine and nifedipine showed the same inhibitory effect on the 70 mM K(+)-evoked responses. However, ethaverine pretreatment did not inhibit the bradykinin-induced secretion and [Ca2+]i rise, which are known to be produced through the receptor-operated Ca2+ channels. We conclude that ethaverine reduces catecholamine secretion by blocking the L-type voltage-sensitive Ca2+ channel.

1H magnetic resonance spectroscopy characterization of neuronal dysfunction in drug-naive, chronic schizophrenia.

RATIONALE AND OBJECTIVES: We investigated the proton metabolism of right prefrontal white matter in drug-naive, chronic schizophrenic patients (n = 23), compared with healthy normal control subjects (n = 10), by using localized, water-suppressed in vivo 1H magnetic resonance (MR) spectroscopy. METHODS: All 1H MR spectroscopy examinations were performed on a 1.5-T MR imaging/MR spectroscopy system by using a point-resolved spectroscopy pulse sequence for localized volumes of 2 x 2 x 2 cm3. Proton metabolite ratios relative to creatine (Cr) were obtained using a Marquart algorithm. RESULTS: Drug-naive, chronic schizophrenic patients demonstrated a decrease in the N-acetylaspartate (NAA):Cr and choline (Cho):Cr ratios and an increase of the complex of gamma-aminobutyric acid (GABA) and glutamate (Glu)-containing ratio [(GABA + Glu):Cr] as compared with normal control subjects. CONCLUSION: Results suggest that the reduction of NAA and Cho may indicate neuronal dysfunction and that the elevation of GABA and Glu may play a role in chronic schizophrenia. 1H MR spectroscopy may be a useful modality in research and in the clinical evaluation of chronic schizophrenic patients.

Reaction of the sera of the Korean children free from Hib invasive diseases against H. influenzae type B capsular polysaccharide antigen.

The purpose of our experiment is to examine the level of anti-Haemophilus influenza polysaccharide antibody titer in the Korean population. Using ELISA, the level of Hib-PS antibodies in 384 infants and children who were all free from Hib invasive diseases, was tested. And the blood of 50 mothers within 24 hours of delivery and cord blood from their respective full-term neonates was also tested. The transport of Hib-PS IgG and IgG subclasses in paired sera from mothers and neonates was also measured. The titer of Hib-PS IgG varies with age. At birth the mean optical density of cord blood was 1.028; however, it declined to 0.609 up to 6 months and further decline was noted up to 2 years to 0.488. Then the mean O.D. remained around 0.5 from 3 to 14 years of age. The mean O.D. of Hib-PS IgG in the mothers blood was 0.856. The ratio of mean O.D. of anti-Hib PS IgG antibody in the cord blood to that in the maternal blood was 1.20. The mean optical densities of IgG subclasses were: 1.18 for anti-Hib PS IgG1, 1.07 for anti-Hib PS IgG2, 1.01 for anti-Hib PS IgG3, and 1.09 for anti-Hib PS IgG4. The sera from Korean children of almost all age groups reacted to Hib-PS antigen on ELISA. Also the active transport of anti-Hib PS IgG antibody through placenta was observed. Among four IgG subclasses, only IgG1 transport had significant experimental meaning.

Esophageal duplication cyst complicated with intramural hematoma--case report.

Esophageal duplication cysts account for a very small percentage of benign esophageal tumors and are infrequently symptomatic. Esophageal duplication cysts result from aberrant alignment of the normal vacuolization process that produce the esophageal lumen in the 5th to 8th week of embryonic life. Complications most often are bleeding into or infection of cysts. Recently, we experienced a case of esophageal duplication cyst complicated with intramural huge hematoma and the cause of hematoma could not be identified. We report it with a review of literatures.
