In cancer therapy, the authors have attempted to decrease the side effects. Since 1975 they have used immuno-thermo-chemotherapy(so-called ITC therapy). At this time the ITC therapy is used in combination with induced-hypertension-chemotherapy (IHC therapy). This new approach has been applied to patients with terminal or advanced malignancy, which have been unsuccessfully treated by conventional ITC therapy. This report compares the clinical effects of conventional ITC therapy those of new modality in the same patient. The clinical results are 6 cases of I-A and one case of I-B according to Karnofsky's criteria. This New modality is more effective than the conventional ITC therapy in all 7 patients.

The replication origin of the E. coli K-12 chromosome has been isolated as autonomously replicating molecules(oriC plasmid), and the DNA region essential for replicating function(oriC) has been localized to a sequence of 232-245 base-pairs(bp) by deletion analysis. In this report, the functional role of oriC was analysed by using an in vitro replication system and various OriC+ and OriC- plasmids previously constructed. The results obtained were summarized as follows: (1) The oriC sequence contained information enough to direct bidirectional replication. (2) The actual DNA replication began at a region near, but outside, oriC and progressed bidirectionally. (3) Initiation of DNA synthesis at the specific region required the dnaA-complementing fraction from cells harboring a dnaA-carrying plasmid.
The nucleotide sequences of the termini of mature DNA from T3 phage were determined. A directly repeated sequence of 230 base pairs, corresponding to the terminal redundancy of T3 DNA, was found in the left and right end of the genome. The sequence of the terminal regions of T3 DNA was compared with that of T7 DNA. There are discrete sequences with significant homology, specifically in the regions around the two ends of the terminally redundant sequences. 5'-CCTAAAG and its variants appear frequently in both ends of the genome. T3 DNA fragments carrying the terminally redundant sequences were isolated from concatemeric DNA and analyzed by restriction enzymes.
Binding of 8-fluoro-8-demethylriboflavin to riboflavin binding protein was studied 19F-NMR spectroscopically. The pH dependence of 19F chemical shift of this fluoro derivative both free and bound to the protein revealed a shift of pKa associated with N(3)-H to alkali when bound to the protein. 19F chemical shift of the fluoro derivative bound to the protein is in a higher magnetic field than that of the free derivative in the neutral to acidic pH region. These results are interpreted in terms of the environmental changes at the 8- and 3-positions of isoalloxazine, indicating the applicability of this derivative for probing flavin-protein interaction at these positions.
Compressed tablets were prepared from theophylline and hydroxypropylcellulose. Effects of the viscosity grades of the polymer, the mixing ratios of two polymers with different viscosity grades, and the polymer contents in the tablets on release patterns of theophylline were examined in vitro. Release rate was decreased with increasing viscosity designation and polymer contents in the tablets. In salivary level profiles of theophylline following oral administration of sustained-release tablets to five human volunteers, a low but sustained level was noted indicating sustained release of the drug from the tablets in vivo.
A special cell type was identified in the mucosal epithelium of the large intestine of the tadpole of the bullfrog, Rana catesbeiana. It is a slender, columnar cell, with a dark, basally situated nucleus. By electron microscopy the cell displays prominent bundles of filaments emerging from each microvillus and extending deep into the cytoplasm without ending in the terminal web. It has longer and more crowded microvilli than the absorptive cell. The specialized cell is also characterized by the presence of many apical vesicles and numerous subapical dense bodies. These cytological features suggest that it may be a brush cell (Rhodin and Dalhamn 1956). These cells displayed endogenous peroxidase activity in smooth and rough endoplasmic reticulum, in the well-developed Golgi apparatus and in apical vesicles. Furthermore, peroxidase reaction product was frequently observed on their luminal surface membrane. These findings suggest that the brush cell in the large intestine of the bullfrog tadpole may be a secretory cell.
In order to clarify the pathogenesis of carbon disulfide retinopathy, fluorescein fundus angiograms were analyzed for 143 workers exposed to CS2 and 40 unexposed workers (reference) from viscose rayon factories. Retinal abnormalities, characterized by saccular microaneurysms, ellipsoidal microaneurysms and loop formation of small vessels, and atrophic/degenerative changes of pigmentary epithelium, were found in workers exposed to CS2. Saccular microaneurysms were found in 45.5% of the CS2 workers and 7.5% of the reference workers (P less than 0.05). Ellipsoidal microaneurysms or loop formation were found in 14.0% of the CS2 workers and 5.0% of the reference workers. Atrophic/degenerative changes of the pigmentary epithelium were found in 32.9% of the CS2 workers and 15.0% of the reference workers (P less than 0.05). Significant correlations were found between the prevalence of these retinal abnormalities and the CS2 exposure index.
8-Fluoro-8-demethylriboflavin was shown to be a potential active-site-directed reagent for flavoproteins. It reacted with the nucleophiles of N-acetylcysteine (-SH), N-acetyltyrosine (-OH), alpha-N-acetyllysine, and glycine (epsilon- and alpha-NH2, respectively) under fairly mild conditions, and the reaction products were identified. The reactivity of the fluoroflavin was higher than that of 8-chloro-8-demethylriboflavin, which reacted only with the cysteine derivative among the amino acid derivatives used, and whose pseudo first order rate constant at 23 degrees C was 1/23 of that of the fluoroflavin. The reactivity of the fluoroflavin was also estimated by 13C and 19F NMR spectroscopy. The results showed that this compound is more reactive than the chloroflavin.
To clarify the pathological difference of retinal changes between carbon disulfide (CS2) and diabetic retinopathies, fluorescein fundus angiograms of 92 workers with CS2 retinopathy (Group I) were compared with those of 37 cases with diabetic retinopathy (Group II) and 40 workers who had not been exposed to CS2 (Group III). The results were: 1) Ellipsoid or loop-shaped microaneurysms were observed in 25% of Group I, and in 5% of Groups II and III. 2) Fluorescein leakage from vessels was observed in Group II, but not in Groups I and III. 3) An avascular area of the retina was observed in Groups I and II. The avascular area in CS2 retinopathy was small and local and showed no tendency of enlarging. 4) Atrophic/degenerative changes of the pigmentary epithelium were observed in 40% of Group I, 11% of Group II and 15% of Group III.
In order to clarify the relationship between exposure to hard metal dust containing cobalt and bronchopulmonary diseases, cross-sectional medical and occupational hygienic investigations were conducted in a hard metal tool manufacturing factory. As a part of the environmental investigation, general air concentrations and personal exposure to both total dust and cobalt dust were measured. The highest total dust and cobalt concentrations in general atmosphere and personal samples were found in a powder handling room among all departments. The highest personal exposure concentration of cobalt was 1.29 mg/m3 (arithmetic mean value) in the dry grinder using diamond wheels. Powder handlers were exposed to cobalt 0.56 mg/m3, press operators to 0.66 mg/m3, shapers to 0.19 mg/m3, wet grinders to 0.07 mg/m3, and sintering workers to 0.03 mg/m3. In Japan TLV for cobalt has not yet been established. Dust exposure measurement should be carried out in all hard metal industries. TLV for cobalt is discussed by means of literature.
Using 3 kinds of medium, Mueller-Hinton (M-H) agar, M-H agar added with defibrinated horse blood and lysed horse blood, susceptibilities of Campylobacter were tested quantitatively to 12 antibacterial agents. The result showed no remarkable difference between them. MIC was examined with the cell concentration of McFarland unit 0.5 and 100-times dilution, and there was no significant difference between them except for 2 strains, on which erythromycin showed MIC of 6.25 micrograms/ml at McFarland unit 0.5 and 0.39 micrograms/ml at 100-times dilution. Eighty-two clinical isolates of C. jejuni and 6 of C. coli were tested for their susceptibility to 12 antibacterial agents. These strains were most susceptible to gentamicin and amikacin and less susceptible to benzylpenicillin, carbenicillin and cephaloridine with the MIC of 25 micrograms/ml. There were 2 peaks of susceptibility distribution to minocycline (0.2 and 50 micrograms/ml). In tests for beta-lactamase of C. jejuni and C. coli, 3 strains obtained from 1 patient were positive.

The replication origin region of the Escherichia coli K-12 chromosome has been cloned, and a region of 245 base-pairs has been shown to contain all the information for autonomous replication (defined ori). In order to obtain further information on the sequence organization in the defined ori stretch, various types of mutation were introduced by in vitro techniques at a restriction site (AvaII site) which locates near the middle of ori. When the correlation between these mutations and replicating function was examined, different effects were obtained with the types of mutation: the replicating function was completely destroyed by either insertion or deletion of short sequences, but not by base-substitutions. Based on these observations and on the fact that multi-gene products are involved in the initiation of replication, we assume that two categories of sequences are present within the ori stretch, one specifying interaction with initiation factors (recognition sequences) and the other spacing the recognition sequences in appropriate distances (spacer sequences), and that the AvaII site is located in the spacer region.
1. After the frog tongue was adapted for 10 sec to various salts and sugars, the initial phasic component of gustatory neural responses to almost all of quinine hydrochloride (Q-HCl), quinine sulfate (Q-H2SO4). Brucine, caffeine and picric acid was suppressed. 2. Following 10 sec adaptation to acetic acid, the phasic responses to Q-HCl and Q-H2SO4 were unchanged, those to brucine and caffeine were enhanced, and that to picric acid was depressed slightly. 3. The response to any one of Q-HCl, Q-H2SO4. brucine and caffeine was suppressed after adaptation to the other three, while those to picric acid and nicotine were unchanged or enhanced after adaptation to another bitter solution.
