The occurrence of phthalic acid esters in various samples of commercially available sodium chloride injections (Indian Pharmacopoeia).

Conclusive evidence for the presence of di(2-ethylhexyl) phthalate (DEHP) in 15 out of 17 commercially available sodium chloride injection (Indian Pharmacopoeia) samples was obtained and concentration levels as high as 11.0 mg/500 ml of saline were detected. The presence of such contaminants in i.v. fluids, avoidable by the institution of appropriate quality control measures before manufacturing and marketing, is a matter of serious concern. Stipulated specifications in different pharmacopoeias are meant only to ensure sterility and pyrogenicity, and it is advisable that cognisance be taken of the presence of toxic contaminants e.g. di(2-ethylhexyl) phthalate.

Studies on the distribution of beta- and gamma-isomers of hexachlorocyclohexane in rat tissues.

A study of the comparative distribution pattern of beta- and gamma-isomers of hexachlorocyclohexane (HCH) in albino rat tissues as a function of duration of treatment at two dose levels revealed different accumulation patterns for the two isomers in tissues. In general, the accumulation pattern was found to be in the order: fat greater than kidney greater than lungs greater than liver greater than muscle greater than heart greater than spleen greater than brain greater than blood for beta-HCH and fat greater than brain greater than kidney greater than muscle greater than lungs greater than heart greater than spleen greater than liver greater than blood for gamma-HCH. The tissue accumulation of beta-isomer was always greater than that of gamma-isomer in any tissue except in brain. Brain beta-HCH residue levels were relatively lower than in many other tissues except blood, whereas brain gamma-HCH concentration was only next to fat. A higher dose or a longer period of treatment generally produced a more marked elevation of tissue residue levels in beta-HCH fed rats, but not in gamma-HCH treated animals.

Cerebral venous and arterial thrombosis in pregnancy and puerperium. A study of 135 patients.

Cerebral venous thrombosis occurring in puerperium is about 10 to 12 times more frequent in India than in Western countries. A clinical study of 135 patients with cerebrovascular accidents in early puerperium is reported. Cortical vein thrombosis is common and arterial thrombosis rare (6/135). The illness usually occurs within the first two weeks, after normal delivery at full term, in a multiparous woman, with multi-focal seizures, stupor or coma, regressing focal signs or at times as pseudotumour cerebri. Deep leg vein thrombosis and pelvic sepsis are rare. Mortality has been high reaching 28 to 33 percent in both Indian and Western countries. With the use of anticoagulants in some selected patients and earlier; energetic, supportive and symptomatic measures in others the mortality could be reduced to less than 20 percent. The quality of survival is good since those who survive have only minimal physical disability.

Substrate specificity and other properties of DOPA decarboxylase from guinea pig kidneys.

DOPA decarboxylase (aromatic-l-amino-acid carboxy-lyase, EC 4.1.1.28) from guinea pig kidneys has been purified to a specific activity of 9370 or 330-fold. Efficient purification was possible by employing apolar interaction chromatography. The purified enzyme gives a single component on polyacrylamide gel electrophoresis and the absorption spectrum of the enzyme reveals two forms of binding of pyridoxal 5-phosphate. The pure enzyme decarboxylates l-DOPA, 5-hydroxytryptophan, o-tyrosine and m-tyrosine but it is inactive towards phenylalanine, tyrosine, tryptophan, histidine and 3-methoxy-phenylalanine. The enzyme behaves as an undissociated enzyme but only towards 5-hydroxytryptophan. It behaves as an enzyme from which the coenzyme is partially dissociated when it attacks l-DOPA, o-tyrosine and m-tyrosine.

Effect of cholesterol feeding and estrogen treatment on synthesis of fatty acids in liver.

The effect of cholesterol feeding and estrogen administration on synthesis of fatty acids in liver mitochondria, microsomes and cytoplasm of male rabbits has been investigated. The synthesis was measured by the incorporation of [1(-14)C] acetyl CoA or [2(-14)C]malonyl CoA into long chain fatty acids under optimal conditions. It was found that atherogenesis markedly decreased the fatty acid synthesis in cytoplasm. The mitochondrial fatty acid synthesis was not affected by the disease. There was a small but measurable decrease in the synthesis of fatty acids in microsomes. Estrogen had no effect on the synthesis of fatty acids in mitochondria or microsomes. But if effectively counteracted, after a short lag period, the decreased synthesis of cytoplasmic fatty acids observed in atherosclerosis. It is possible that liver fatty acid synthetase is one of the enzyme systems through which estrogens exert their atherosclerosis-retarding effect. The decreased cytoplasmic fatty acid synthesis observed in atherosclerosis might account for the low levels of saturated fatty acids reported in liver and plasma lipids of atherosclerotic animals.

A giant schwannoma of cauda equina.

A case of giant schwannoma occurring in a woman 23 years of age with deformity of the foot developing at the age of five years is reported. The gross radiological changes of increased interpedicular distance and erosion of the posterior surface of the vertebra suggested the massive size of the tumour (6'' x 21/2'' x 11/2''). Successful surgical excision of such a large size schwannoma has not been reported previously.

Protein Breakdown and Formation of Protease in Attached and Detached Cotyledons of Phaseolus vulgaris L.

In contrast to earlier reported results of similar experiments in peas, in which almost no increase in protease activity occurred in incubated detached cotyledons, we report here an increase in protease activity in both attached and detached bean cotyledons. Detached bean cotyledons showed continually increasing protease activity up to the 12th day, while that in attached cotyledons declined after 6 days. The free amino acid level in detached cotyledons reached a maximum at the 11th day; protease formation leveled off after 50% of the original seed protein was digested. These data suggest that high free amino acid levels may inhibit protease formation.The activity of partially purified protease in aqueous extracts was enhanced by 10 mm 2-mercaptoethanol or cysteine, indicating a sulfhydryl requirement for activation. Protease formation in detached cotyledons was inhibited 30% by 10 mug/ml cycloheximide and 50% by 100 mum abscisic acid. In contrast, alpha-amylase formation was inhibited 90% by 10 mug/ml cycloheximide and 95% by 20 mum abscisic acid. The cycloheximide data suggest that only a part of the protease, but all of the alpha-amylase, is synthesized de novo; the similar pattern of inhibition by abscisic acid emphasizes the concept that protease may exist in two forms.
