Alterations in glycosaminoglycan composition of methylcholanthrene-induced sarcoma at various stages of the tumour growth.

The methylcholanthrene-induced sarcoma contains several types of glycosaminoglycans, hyaluronic acid being the major component. Furthermore it contains all sulphated glycosaminoglycans present in the skin: chondroitin-4-sulphate, chondroitin-6-sulphate, keratan sulphate, dermatan sulphate, heparan sulphate and heparin. It was found that the amount of all glycosaminoglycans distinctly increased during tumour growth. At the same time the amount of collagen significantly decreased. It is suggested that some of the GAGs participate in the creation of a storage depot for biologically active molecules (growth factors, enzymes) which are thereby stabilized and protected. Hydrolytic degradation of some GAGs may result in the release of some cytokines which may stimulate or inhibit the tumour growth. The changes in the quantities of various glycosaminoglycans during the tumour growth may be responsible for a dual-phase growth of this tumour.

Chronic advanced pancreatitis--diagnosis, biochemical and ultrastructural changes and surgical procedures.

On the basis of 162 cases of chronic advanced pancreatitis seen at our Department during the last 15 years the diagnosis, indications and surgical procedures are described in this paper. Quantitative and qualitative changes of pancreatic collagen in chronic inflammation were also analysed. Results of these investigations were compared with histological and ultrastructural pictures of pancreatic tissue. Amongst 162 patients, 142 (88%) were operated upon. 71 underwent pancreatectomy or pancreatoduodenectomy, 14 anastomotic procedures and rest of them various operation of the bile ducts. Overall, 18 (13%) patients experienced significant complications after resectional procedures and 6 (4%) died as well after anastomotic procedures.

Collagen, elastin and glycosaminoglycans in aortic aneurysms.

The walls of human abdominal aortas and atherosclerosis-induced aneurysms contain similar amounts of collagen. The quantitative ratio between collagens of various types of this protein does not differ significantly either, whereas solubility of the collagen in aneurysmal wall and its susceptibility to the action of EDTA are distinctly decreased. In contrast with collagen, the amount of elastin in aneurysms is significantly lower. Total amount of glycosaminoglycans slightly decreased as compared with that of normal tissue, but the ratio of particular compounds varies. The percentage of chondroitin sulphate is increased and that of heparan sulphate significantly decreased. The significance of these changes in pathogenesis of aneurysms is discussed.

Rat fibrosarcoma as a source of stable co-polymer of type I and type III collagens.

During conventional procedures for the extraction of collagen from rat fibrosarcoma it was observed that a significant fraction of the acid-soluble collagen remained insoluble when subsequently fractionated at neutral pH. Analysis of this collagenous component, using amino acid analysis, cyanogen bromide peptide mapping, proteinase susceptibility and the action of denaturing and reducing agents, revealed different properties when compared to other collagen species. The results indicate that the tumour collagen fraction is probably a stable complex of type I and III collagens. This native collagen heteropolymer represented approximately 18% of the total collagen from rat fibrosarcoma and as such provides a unique natural source of this collagen complex for further analytical studies.

Solcoseryl prevents early postradiation changes in the lungs.

The aim of the study was to test the influence of Solcoseryl on early postradiation changes in the lungs of rats. The chests of the rats were irradiated with Co-60, fractional dose 250 cGy/DT, total dose 2500 cGy/DT. Solcoseryl was given intraperitoneally in quantities of 0,1ml daily, during 60 days of the experiment. Morphological examinations of the lungs revealed that Solcoseryl decreased significantly the effects of early postradiation damage of the respiratory organ in rats.

Liver collagen of rats submitted to chronic intoxication with acetaldehyde.

It was found that chronic intoxication of rats with acetaldehyde results in a distinct, progressive increase of 5-3H-proline incorporation into collagen synthesized by liver. At the same time biosynthesis of other proline-containing (noncollagenous) proteins does not change significantly. On the other hand the collagen content in the rat liver did not increase in the early stage of acetaldehyde administration, but increased when acetaldehyde feeding was continued for 6 months. About 40% increase of total collagen content was found in livers of the intoxicated animals. All the investigated collagen types (I, III, IV and V) grew in the same degree. No changes in proportional relationships between collagens of different types were found.

Acetaldehyde↗

Chronic intoxication with acetaldehyde stimulates collagen biosynthesis in rat liver.

It was found that chronic intoxication of rats with acetaldehyde results in a distinct, progressive increase of 5(3)H-proline incorporation into collagen synthesized by liver. At the same time, biosynthesis of other proline-containing (noncollagenous) proteins does not change significantly. The effects are similar to those induced by chronic intoxication of rats with ethanol. Since acetaldehyde is an intermediary metabolite formed during ethanol oxidation in liver, it may be concluded that acetaldehyde is a factor responsible for alcohol-induced liver fibrosis.

Effect of lathyrogen on collagen of methylcholanthrene-induced sarcoma of rat.

Since the lathyrogen beta-aminopropionitrile is known to affect the fibrogenic and physical properties of collagen polymers, we have examined its effects on collagenous components of methylcholanthrene-induced fibrosarcoma in rat. Lathyrogen treatment reduced total collagen content of tumours from approximately 17 to 12 mg collagen/g tissue. It also proportionately increased the solubility of specific collagen fractions, the summation of all extractable solubilized collagens reflecting 37 and 67% of total collagen content for control and lathyric tumours, respectively. Although lathyrogen had no significant effect on the growth and overall size of fibrosarcoma, histological studies confirmed that changes had occurred to appearance and distribution of collagenous components of extracellular matrix.

Triacylglycerol and glycogen contents in the human gastric mucosa: effect of histamine H2, muscarinic and gastrin receptors blockade.

Triacylglycerol and glycogen contents in the gastric mucosa were studied in 20 patients with duodenal ulcer before and after two-week treatment with cimetidine, pirenzepine or proglumide. Determinations of both metabolic substrates were performed in mucosal slices taken from gastric corpus and antrum during endoscopic examination: the results were expressed per mg of protein. Concentrations of triacylglycerol were significantly elevated after treatment with any of the blocking agents: in the gastric corpus by 88% after cimetidine application, 69% after pirenzepine and 102% after proglumide; in the antrum by 76, 59 and 132%, respectively. No significant changes in the mucosal contents of glycogen were observed. It is concluded that an increase in the mucosal concentration of triacylglycerol can be connected with an inhibition of the gastric acid secretion following treatment with receptor-blocking drugs.

Heterogeneity of collagen isolated from methylcholanthrene-induced sarcoma.

Rat fibrosarcoma induced by subcutaneous injection of methylcholanthrene was found to contain at least three different types of collagen. Two of them were identified as type I and type III collagens, the third (fraction B) seems to be specific for this tumour. The ratio of type I to type III collagen is lower in fibrosarcoma than in normal rat skin. The number of hydroxyproline residues in alpha 1 (I), alpha 2 (I) and alpha 1 (III) chains of tumour collagen appears to be higher than in the corresponding chains of rat skin collagen. Fraction B is composed of three identical alpha chains connected with disulphide bonds. It contains a relatively low amount of glycine: 234 molecules per 1000 residues. The amount of hydroxyproline and cysteine is similar to that found in the type III collagen.

[Leucocyte alkaline phosphatase in normal and pathological pregnancy (author's transl)].

The activities of leucocyte alkaline phosphatase were determined in 511 patients with normal and pathological pregnancy. Mean values were compared and the enzyme followed up, and the conclusion was drawn that leucocyte alkaline phosphatase was no safe indicator of foetal condition. No direct relationship were found to exist between leucocyte alkaline phosphatase, total oestrogens, HSAP, HLAP, HPL, and oxytocinase.
