Peritoneal serous papillary carcinoma: radiological appearance.

BACKGROUND: The radiological appearance of peritoneal serous papillary carcinoma (PSPC) is described. METHODS: Three cases of PSPC were analyzed retrospectively with regard to the radiological appearance and histopathological features. RESULTS: All three patients were women, aged 44-71 years. Massive ascites and a greater omentum tumor were observed on computed tomography in all patients. Double-contrast enema performed in one patient showed irregularity on the upper aspect of the transverse colon. Radiological examinations excluded primary tumors in both gastrointestinal and genital organs in all patients. Histological diagnosis was made from the surgical specimen in two patients and from an autopsy specimen in one patient. All patients had a large omental tumor involving the transverse colon, but the ovaries were not involved or only minimally involved on the surface. Serum CA125 was markedly elevated, and immunohistochemical staining for CA125 was positive within the tumor cell cytoplasm in all three patients. CONCLUSION: PSPC cannot be diagnosed from radiological findings alone because of its similarity to metastatic peritoneal carcinomatosis and peritoneal mesothelioma. Marked elevation of serum CA125 may help with PSPC diagnosis. Response to treatment is promising, and exploratory laparotomy is thus justified when a patient shows characteristic radiological findings and high CA125 level.

Antitumor effect of palmitoleic acid on Ehrlich ascites tumor.

Palmitoleic acid (PLOA) markedly prolonged the survival time of mice bearing Ehrlich ascites tumor at doses of 37.5-150 mg/kg/day X 10, but the antitumor activity of oleic acid (OA) was weaker than that of PLOA. The total lipid and phospholipid contents in the tumor cells treated with PLOA were significantly decreased. In addition, the fatty acid pattern of phosphatidyl choline, cholesterol esters and triglycerides from the PLOA-treated Ehrlich tumor cells differed markedly from that of the corresponding lipids from the control tumor cells.

Antitumor effect of dicyclohexylammonium sulfate, a potent inhibitor of spermidine synthase against P388 leukemia.

The antitumor effect of dicyclohexylammonium sulfate (DCHA), a potent inhibitor of spermidine synthase, was tested on BDF1 mice inoculated i.p. with P388 leukemia (1 X 10(6) cells/mouse). DCHA prolonged the survival time of mice bearing P388 leukemia at the doses of 10-100 mg/kg administered daily for 6 days. The spleen weight increased by 30% at 7 days after tumor inoculation. DCHA treatment had no effect on the tumor-induced increase in splenic weight. The spermidine concentration of the ascites tumor cells and spleens of mice bearing the tumor was lowered by the treatment, while spermine concentration hardly changed. The depletion of spermidine in the ascites tumor cells and spleens might be a cause of the suppression of tumor growth.

In vitro isolation and characterisation of a bovine Neospora species in Japan.

Eleven aborted bovine fetuses and five calves suspected as having neosporosis were necropsied and tissues from these animals were inoculated into bovine cardiopulmonary aortic endothelial cells and monkey kidney cells and maintained at 37 degrees C with 5 per cent CO2. Neospora tachyzoites were observed in one cell 49 days after inoculation. The isolated parasite (JPA1) was morphologically identical to the previously reported bovine Neospora species (BPA1) and confirmed by its strong antigenic reactivity with bovine control antisera to Neospora species and its lack of reactivity with Toxoplasma gondii and Sarcocystis cruzi antisera. This is the first bovine Neospora species isolate in Asia and further studies with this isolate are now expected.

The usefulness and limits of magnetic resonance imaging in the differential diagnosis of pelvic tumors.

Three cases of benign pelvic tumors are presented (2 leiomyomas and 1 fibroma). All three tumors were suspected of being malignant neoplasms because they were visualized as heterogeneous high signal intensity on T2-weighted images, and thus they were difficult to diagnose preoperatively. One of the leiomyomas was located in the retroperitoneum and had been misdiagnosed as an ovarian tumor. All three tumors exhibited secondary myxoid changes, these changes may have been responsible for the high signal intensity on the T2-weighted MR images. Since benign tumors sometimes mimic malignant tumors on MR images, exploratory laparotomy is essential to make a definitive diagnosis.

Primary peritoneal clear cell carcinoma: excellent results from paclitaxel and carboplatin combination chemotherapy.

While papillary serous adenocarcinomas are histocytologically common in primary peritoneal carcinomas, clear cell carcinomas are rare. We report a new regimen for the treatment of recurrent clear cell carcinomas of the peritoneum. A 45-year-old woman was referred to our hospital and underwent optimal debulking surgery. Thirty-two months after the operation, lymph node swelling and elevation of serum CA19-9 were detected and recurrence was diagnosed. Paclitaxel (175 mg/m(2)) in a 3-hour and carboplatin (300 mg/m(2)) in a 1-hour infusion were repeated at three-week intervals. After completion of four courses, abdominal CT and serum CA19-9 were undertaken and results compared. Lymph node swelling was significantly decreased and the serum CA19-9 level was decreased to within a normal range. Paclitaxel and carboplatin combination chemotherapy may be effective in preventing the recurrence of peritoneal carcinomas.

Inhibition of lung metastasis by a calmodulin antagonist, N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7), in mice bearing Lewis lung carcinoma.

The antimetastatic effect of calmodulin antagonist, N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7) was examined in an experimental model of lung metastasis induced by Lewis lung carcinomas (3LL) in C57BL/6crSlc mice. 3LL cells were implanted into the footpad of the mice. Ten days later, the implanted primary tumors were surgically removed. Injection of W-7 i.v. after removal of the implanted primary tumor caused the greatest inhibition of development of lung metastases. The number of peritoneal macrophages, total cells and macrophages in the lung increased in mice treated with W-7. Binding of the third component of complement (C3) cleavage products (C3b) to the C3 receptor on peritoneal macrophages after i.v. injection of W-7 were enhanced, as shown by the fluorescent antibody technique. Lung metastases were inhibited by i.v. injection of peritoneal macrophages activated with W-7. A possible mechanism of inhibition of lung metastases by treatment with W-7 could be interpreted as due to C3 activation and macrophage activation. These findings raise the possibility that W-7 may have clinical value in the prevention of cancer metastasis.

Antitumor effect of a new organic selenium compound, 6-phenyl-7 (6H)-isoselenazolo [4,3-d] pyrimidone (ISP), on the growth of P388 mouse leukemia.

The organic selenium compound, ISP [6-phenyl-7(6H)-isoselenazolo [4,3-d] pyrimidone] markedly inhibited the growth of P388 mouse leukemia at dose of 100 micrograms/mouse per day x 10 with no sign of toxicity. The antitumor activity of 4,5-dihydro-4-methyl-6-oxo-5-phenyl-6H-pyrazolo [4,5-c] isoselenazole (PIS) was weaker than that of ISP. The total lipid and phospholipid contents in the leukemic cells treated with ISP were significantly decreased. The fatty acid pattern of cholesterol esters, phosphatidyl choline and phosphatidyl ethanolamine from the ISP-treated P388 leukemic cells differed markedly from that of the corresponding lipids from the control leukemic cells. In addition, the synthesis of DNA or RNA was depressed in the ISP-treated leukemic cells. The present results indicate that ISP may open new perspectives in cancer chemotherapy.

Resistance of chicks against reinfection with Leucocytozoon caulleryi.

Chickens were inoculated with five to 40 sporozoites of Leucocytozoon caulleryi at 1 to 9 days of age. Mortality, parasitemia, appearance of serum-soluble antigen, and antibodies were examined. Chickens that survived primary infection were reinoculated with 3 x 10(3) to 1 x 10(4) sporozoites and examined for resistance against reinfection by the same criteria used in primary infection. After primary infection, up to 80% of chickens died of leucocytozoonosis. Surviving chickens showed parasitemia from 13 days to 23 or 24 days after inoculation. Serum-soluble antigen and antibodies were detected in most chickens, except for some chickens that were infected at 1 to 3 days of age. After secondary infection, only a few chickens that received primary infection at 1 to 7 days of age showed a small number of parasites in their peripheral blood. Most of the other chickens were resistant to reinfection. Resistance to reinfection was recognized in some chickens that had not showed antibody production before secondary infection.

Antitumor effects of a new polysaccharide-protein complex (ATOM) prepared from Agaricus blazei (Iwade strain 101) "Himematsutake" and its mechanisms in tumor-bearing mice.

The antitumor activity of the i.p. or p.o. administration of polysaccharide-protein complex, ATOM (antitumor organic substance Mie) prepared from cultured mycelia of Agaricus blazei (Iwade strain 101) "Himematsutake" examined against four kinds of established mouse tumors. ATOM was highly effective at the doses of 10 and 20 mg/kg/day x 10 on subcutaneously implanted Sarcoma 180 in mice, and was also active against Ehrlich ascites carcinoma, Shionogi carcinoma 42 and Meth A fibrosarcoma at doses of 50 and 100 mg/kg/day x 10. ATOM has no direct cytotoxic action on tumor cells in vitro. Thus the tumor growth-inhibitory effect of ATOM is apparently due to immunological host-mediated mechanisms. The number of peritoneal macrophages, the phagocytosis of polystyrene latex beads and the proportion of the third component of complement (C3)-positive fluorescent cells were increased in the mice treated with ATOM. These results suggest that the macrophage activa-tion and alterations of the C3 are necessary for the induction of an antitumor effect of ATOM.
