Percutaneous absorption of chlorquinaldol (Sterosan).

The percutaneous absorption of chlorquinaldol from three different preparations for skin treatment (Sterosan-hydrocortison ointment and cream, Sterosan ointment) was evaluated by measuring the cumulative urinary excretion of chlorquinaldol and was compared to that found after an equivalent oral dose. In the blood the concentrations were below the detection limit of the analytical procedure, i.e., smaller than 0.02 microgram/ml. The study was carried out in 3 healthy volunteers using a four-period change-over design. The topical preparations were applied under occlusive dressings. Following epicutaneous application of the three topicals in quantities containing 30 mg chlorquinaldol each, the mean urinary excretion of the drug amounted to 11.1 +/- 6.6, 19.5 +/- 3.5 and 13.0 +/- 2.5% of the applied dose. When the same dose of chlorquinaldol was administered orally, 67.6 +/- 9.5% of the dose was excreted in the urine. Taking the urinary elimination as the minimal amount of drug absorbed, the extent of percutaneous absorption from the three dermatological preparations varied between 4.2 and 23.5% of the applied dose. There was no difference in the pattern of urinary excretion products among the three topicals and the oral formulation. The bulk of chlorquinaldol (mean 98.0 +/- 0.1%) was excreted as sulfate (mean 1.3 +/- 0.3%) or unchanged drug (mean 0.8 +/- 0.1%).

Percutaneous absorption of clioquinol (Vioform).

The percutaneous absorption of clioquinol from three different preparations for skin treatment (Vioform cream, Locacorten-Vioform cream and Vioform-Hydrocortisone cream) was evaluated. After topical dosages corresponding to 30 mg clioquinol, concentrations in the blood were below the detection limit of the analytical procedure, i.e., smaller than 0.02 micrograms/ml; therefore the percutaneous absorption was evaluated by measuring cumulative urinary excretion of clioquinol and was compared to that found after an equivalent oral dose. The study was carried out in 4 healthy volunteers. The topical preparations were applied under occlusive dressings. Following epicutaneous application of the three topicals in quantities containing 30 mg clioquinol each, the urinary excretion of the drug was between 1.2 and 3.6% of the applied dose. When the same dose of clioquinol was administered orally to two volunteers, 52.4 and 92.9% of the dose was excreted in the urine. Taking the urinary elimination as the minimal amount of drug absorbed, the extent of percutaneous absorption from the three dermatological preparations amounted to 1.2-3.6% of the applied dose. There was no difference in the pattern of urinary excretion products among the three topicals and the oral formulation. The bulk of clioquinol was excreted as glucuronide (mean: 96 +/- 3%) and only a small fraction was excreted as sulfate (mean: 3.8 +/- 3%). A small amount of free clioquinol (1.1%) was measured in 1 subject only after the oral dose.

[Membrana interossea antebrachii--a common ligament of the radius-ulna joint. II: Submacroscopic and microscopic structure].

The membrana interossea natebrachii was investigated submacroscopically and microscopically in human cadaveric specimens. Stereomicroscopical and histological investigations under polarized and non-polarized light were performed. Due to the textological structure and due to its osseous insertion the membrana interossea antebrachii represents in its middle and proximal part all morphological features of an interosseous ligament with uniform direction of its fibers. In this middle and proximal part the membrana interossea has to be regarded as a strong ligament coordinating both radioulnar joints which act by the cardan shaft of the forearm as a functional unit.

Purine nucleoside phosphorylase from human erythrocytes: physiocochemical properties of the crystalline enzyme.

The major physicochemical properties of human erythrocytic purine nucleoside phosphorylase (PNPase) have been described. The molecular weight, estimated by ultracentrifugation, molecular sieving and sucrose density gradient centrifugation, ranged from 87 000 to 92 000. Other physical constants of erythrocytic PNPase were: sedimentation coefficent (s20, w), 5.4 S obtained by sedimentation analysis and 5.5 S by the sucrose density gradient procedure; Stokes radius, 38 A; calculated diffusion coefficient (D20, w), 5.7 X 10(-7) cm2 s-1; frictional ration, 1.29; and partial specific volume calculated from amino acid analysis, 0.73 cm3 g-1. The CD spectra of the human erythrocytic and bovine spleen PNPases were almost identical and indicated a very low alpha-helical content. Polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate indicated that the molecular weight of the PNPase subunit is 30 000 +/- 500. These results corroborate earlier reports that the native enzyme is a homologous trimer. Comparative studies with crystalline bovine spleen PNPase confirmed that it is also a trimer but is somewhat smaller than the human erythrocytic enzyme with a molecular weight of about 86 000.

Sequence comparison of human plasma alpha1-acid glycoprotein and the immunoglobulins.

The amino acid sequence of human plasma alpha1-acid glycoprotein, upon comparison with the sequences of other blood proteins, was shown to possess significant similarity with the immunoglobulins. Employing direct and corrected sequence identity, the average mutation value and two different computer comparisons for the evaluation of sequence similarity, the following two regions of this alpha-globulin, which account for approximately half of the total amino acid sequence of the protein, were found to possess sequence similarity with the immunoglobulins. a) The region from residues 77 through 125 proved to be related to the variable region of several human H and L chains, and b) the region from residues 136 through 166 was found to be related not only to the constant region of a human and a mouse L chain but also to the third and fourth constant region of a rabbit and a human H chain, respectively. These results suggest that alpha1-acid glycoprotein is probably related to the immunoglobulins and further suggest that it possibly diverged from the immunoglobulin evolutionary tree prior to the formation of the primitive L chain.

[Spontaneous hyperstimulation of the ovaries with luteal cysts and ascites during a twin pregnancy-extreme case of the syndrome of painfull early pregnancy (author's transl)].

Laparotomy was performed in a para II gravida II of 28 years in an early twin pregnancy. The left ovary was enlarged to 18 X 8 cm, the right ovary to 1i X 8 cm by luteal cysts. The abdominal cavity contained an ample amount of peritoneal fluid. The history is understood as spontaneous ovarian hyperstimulation, and as an extreme case of the relatively common syndrome of painfull early pregnancy, occasionally leading to unnecessary surgical intervention.

Isolation of a bone-resorptive factor from human cancer ascites fluid.

A protein fraction that induces the resorption of bone explants in organ culture was isolated from the ascitic fluid of patients with advanced cancer metastatic to the peritoneal cavity. Partial purification was achieved by means of gel filtration, affinity chromatography, and ion-exchange chromatography. The isolated fraction, the components of which have an apparent molecular weight of 60,000, was found to be heterogeneous by disc gel electrophoresis and to be composed primarily of proteins with relatively acidic electrophoretic properties. The specific bone-resorptive activity of this protein fraction was greatly increased over that of the unfractionated starting material, and the activity could be completely destroyed upon incubation with pronase and on heating. As determined by immunoassay and extraction procedures with various solvents, the bone-resorptive action of the isolated fraction was not attributable to the presence of parathyroid hormone, prostaglandin E2 or vitamin D-like sterols. In parallel experiments the supernatants of phytohemagglutinin-stimulated normal human peripheral leukocytes were subjected to identical chromatographic techniques, and a proten fraction with a molecular weight of 60,000, which resembled the resorptive fraction isolated from cancer ascites fluid and which contained significant bone-resorptive activity, was also partially purified.

A low molecular weight glycosaminoglycan from the human aorta.

A low mol. wt, dialyzable glycosaminoglycan was isolated from human aorta and was found to be homogeneous on 2 dimensional electrophoresis. As judged by its electrophoretic mobilities and its hydrolysis by chondroitin sulfatase ABC, it was concluded that this hitherto unknown glycosaminoglycan is an oversulfated chondroitin sulfate.
