The immunization of C3H mice with irradiated syngeneic MM48 tumor cells induced specific tumor-neutralizing cells (TNC). The TNC activity was mediated by the L3T4+, Ly-2- T cell population, and the generation of TNC coincided with the appearance of delayed-type hypersensitivity response against MM48 antigen. The administration of an auto-I-Ak reactive T helper cell clone MS202 to normal C3H mice resulted in the facilitation of growth of MM48 tumor due to the induction of T suppressor (Ts) cells. Splenic T cells from animals given this T cell clone inhibited the TNC activity of immunized mice resulting in the escape of MM48 from the TNC effect. The surface phenotype of the Ts cells was L3T4+, Ly-2-. The Ts cells induced by the clone MS202 were totally antigen-nonspecific, and were able to suppress both the effector and inductive processes of TNC. The results suggest the presence of a physiological MHC-restricted T cell circuit that regulates immune responses against the growing tumors.
We cultured the cervical lymph node lymphocytes of a patient suffering from cutaneous T-cell lymphoma. His anti-ATLV antibody was positive by indirect immunofluorescent method (IF). ATLV was detected on these cultured cells by IF. Type C particles were observed in the cultured cells by electron microscopy. These particles were measured to be 60 to 120 nm in diameter with electron dense core, and were considered as ATLV. This case showed a possibility of detecting ATLV by culture of lymph node lymphocytes from such a patient.
To investigate a non-RI test which is equivalent to the reverse transcriptase inhibiting antibody test, a reverse transcriptase inhibiting antibody was compared to the absolute number of CD-4 or CD-8 cells or CD-4/-8 ratio and also to photodensitometric analysis for western blotting. There is no correlation of the reverse transcriptase with any test for cell numbers and their ratio. In photodensitometry, relative units of anti-p65 and anti-p51 were compared with reverse transcriptase inhibiting antibody. The reverse transcriptase inhibiting antibody showed a higher correlation to the relative unit of p65 antibodies than that of p51 antibodies. The photodensitometric analysis of western blotting for a serum test may be a possible method to find a prognostic marker in HIV-1 infection.
A cross-sectional study of 128 individuals infected with human immunodeficiency virus type 1 (HIV-1) was conducted to determine the correlation of reverse transcriptase-inhibiting (RTI) antibody to clinical disease. Thirty-two individuals were studied in each of four clinical groups: asymptomatic individuals, those with persistent generalized lymphadenopathy, those with acquired immune deficiency syndrome (AIDS)-related complex, and those with AIDS. Our study showed that 78% of asymptomatic individuals, 53% of those with persistent generalized lymphadenopathy, 50% of those with AIDS-related complex, and only 25% of those with AIDS have RTI antibody. Concurrent measurement of measles antibody level was used as an indicator of the immune status of these individuals. Measles antibody did not decline in persons with clinical disease, but asymptomatic individuals had lower antibody titers, possibly due to hypergammaglobulinemia associated with advanced HIV infection. These results indicate that more HIV-infected asymptomatic individuals than symptomatic individuals have RTI antibody. This suggests either that the RTI antibody level decreases with the progression of disease in HIV infection or that symptomatic individuals do not produce RTI antibody. The presence or absence of RTI antibody can thus be used as a marker of advanced disease.
A cross sectional study of risk factors in respiratory diseases was carried out in August 1986, in Beijing, China. Inhabitants greater than or equal to 40 years old were selected at random from a rural area, a residential area and an industrial area, using a two stage sampling method. The analysis presented here is based on the sample population of adults who (1) were prepared to be interviewed, using the British Medical Research Council's questionnaire translated into Chinese (n = 3423) and (2) had lung function measurements at the same time (n = 3373). Obstructive lung disease was defined as forced expiratory volume in 1s (FEV1) less than 68% of forced vital capacity (FVC). Seven variables were considered as potential risk factors or confounding factors: area of residence, sex, age, cigarette smoking, history of respiratory disease, socio-economic status and familial component. A modified binary variable regression method developed by Feldstein was used for the adjustment of rate ratios. The adjusted prevalence of obstructive lung disease was highest in the rural area and lowest in the residential area(s). An increase in age, cigarette smoking, low socio-economic status and positive history of respiratory diseases were associated with significantly higher rates of impaired pulmonary function. The other measured factors did not appear to be related to impaired pulmonary function.
As a surgical therapy for intravaginal testicular torsion, we developed a testicular fixation method considering the testis in intravaginal testicular torsion in comparison with the normal one. With our method, the lower part of the testis is fixed in a ring form around the testicular long axis to show the wide extravaginal area similar to that of a normal testis. We believe that this method is simple, logical and effective to fix the testis in intravaginal testicular torsion.
We observed the effects of a new thyrotropin-releasing hormone derivative, YM-14673 (N alpha-[[(S)-4-oxo-2-azetidinyl]carbonyl]-L-histidyl-L-prolinamide dihydrate), on behavioral changes in rats for 3 weeks after focal cerebral ischemia. Under halothane anesthesia, the left middle cerebral artery was occluded via a transretro-orbital approach. YM-14673 was administered just after the operation and once a day for 3 weeks. Neurologic deficits, including hemiplegia and abnormal posture, and disturbance of passive avoidance learning were present in solvent-treated control rats for the entire 3 weeks. YM-14673 at 0.1 or 0.3 mg/kg i.p. or 1 mg/kg p.o. significantly accelerated the recovery of neurologic deficits and ameliorated cognitive disturbance compared with the solvent-treated controls although the drug at 0.1 and 0.3 mg/kg i.p. did not influence the size of the ischemic infarct. YM-14673 mitigated the behavioral disturbances in this model of chronic focal cerebral ischemia. We also discuss the suitability of this model for the evaluation of drugs.
We observed the effects of a new thyrotropin-releasing hormone analogue, YM-14673 (N alpha-[[(S)-4-oxo-2-azetidinyl]carbonyl]-L-histidyl-L-prolinamide dihydrate) on behavioral changes in 26 rats subjected to middle cerebral artery occlusion. The administration of 0.1 mg/kg i.p. YM-14673 was started 1 week after occlusion and was repeated daily for 2 weeks. YM-14673 significantly accelerated the recovery of neurologic deficits and ameliorated the disturbance of passive avoidance learning. Thus, YM-14673 improved behavioral response in a model of chronic focal cerebral ischemia. The availability of a chronic middle cerebral artery occlusion model for the evaluation of drugs is also discussed.
We investigated the activity and the content of copper and zinc-containing superoxide dismutase (SOD) and superoxide anion (O2-) production in polymorphonuclear leukocytes (PMN) from healthy term newborn infants, very low birth wt infants, and healthy adults. SOD activity in PMN was measured with nitroblue tetrazolium reduction assay on PAGE, and the SOD content in PMN was determined with an ELISA using a monoclonal antibody against human copper and zinc-containing SOD. The activity and the content of SOD in the term neonatal PMN and VLBW-infants' PMN were significantly lower than those in the adults' PMN (term newborn infants, 6.6 +/- 0.6 U/mg protein and 170.4 +/- 16.3 ng/mg protein, n = 10; VLBW infants, 6.8 +/- 0.9 and 173.0 +/- 16.2, n = 6; adults, 10.3 +/- 0.6 and 241.9 +/- 13.3, n = 10; values were expressed as mean +/- SEM). Both the phorbol myristate acetate- and the N-formyl-methionyl-leucyl-phenylalanine-induced O2- production rates of VLBW-infants' PMN were significantly higher than those of the term neonatal PMN. The phorbol myristate acetate-induced O2- production rate of the term neonatal PMN was significantly lower than that of the adults' PMN. The phorbol myristate acetate-induced H2O2 production rate of the term neonatal PMN was significantly lower than that of the adults' PMN. The conversion rate from O2- to H2O2 of the neonatal PMN was similar to that of the adults' PMN.(ABSTRACT TRUNCATED AT 250 WORDS)

Morphometric assessment including epithelial indices which express morphological features of the epithelium, mitotic index, mean nuclear area, mean form factor of the nucleus and cellular infiltration in the stroma was performed in 14 cases with oral non-dysplastic epithelium and 66 cases with dysplastic epithelium. The results from morphometry showed a close relationship to the histological severity of dysplasia determined by the histological criteria of Bánóczy. Immunohistochemical localization of carcinoembryonic antigen (CEA), epithelial membrane antigen (EMA) and epidermal growth factor (EGF) were investigated in 14 cases of non-dysplastic epithelium, 66 cases of dysplastic epithelium and 16 cases of squamous cell carcinoma, and compared with the morphometric results. Positive rates of CEA and EMA reaction in epithelial dysplasia increased with the advance of the dysplastic grade. Those of EGF reaction decreased with the advance of the dysplastic grade. It is suggested that morphometry and immunohistochemistry are useful in confirmation of the histological severity of oral epithelial dysplasia.
Thirty-two patients with anorectal cancer were treated with cryosurgery from April 1974 to August 1985. The purpose of the treatment was to provide palliative benefits to 27 patients. In one group (17 patients) who did have previous surgery, cryosurgery was used for the predominant symptoms of melena and/or obstruction. In the postoperative period, local bleeding was minimal, and palliation was judged significant. The survival time of this group ranged from 1 month to 18 months. In another group (10 patients) in whom recurrent cancer followed previous operative treatment, cryosurgery was used to control the local invasion and tumor bulk. All of these patients died within 17 months after recurrence was discovered. Nevertheless, treatment was judged to provide significant palliation. A small group (five patients) was treated with the intent to cure, and cryosurgical techniques were used as an adjunct to surgical excision. Four patients survived more than 8 years after the operation. Two of these patients died of illnesses other than cancer. The local destruction of an anorectal cancer by cryosurgery in these patients provided effective control of the tumor. Bleeding was controlled, and tumor bulk was reduced in size. All patients were treated without any complications caused by cryosurgery. This experience indicates that the cryosurgical technique provides therapeutic benefits for selected patients with anorectal cancer, especially those of high surgical risk.

Undertaken to ensure perfection of sterilization with ethylene oxide gas (EO gas) in routine clinical laboratory practice, an investigation was made of the relationship between sterilization procedure and sterilizing effect using the film bag delivery system and chamber system. Using a biological indicator as a monitor of sterilizing effect, the efficiency of sterilization was studied under various likely conditions and compared. The results led to the following conclusions: 1. Sterilization with EO gas by the film bag delivery system, because of this procedure being unable to be performed at a constant temperature, was shown to prove inadequate due to insufficient exposure to gas especially in winter season (when room temperature fell below 10 degrees C). 2. With the EO gas sterilization method using the film bag delivery system, which is believed to be effective within the ordinary range of humidity of the atmosphere, sterilizing effect was recognized to be affected by excess wetting of an object of sterilization that was caused by inadvertent manipulation or due to a predisposing attribute of the object, such as shape or outline. 3. All materials are sterilized upon being packaged. It is necessary therefore to make best choice of package material with respect of texture and size. Reuse influence to sterilizing effect. 4. The method of sterilization with EO gas using the chamber system has the advantage in that operating conditions are all automatically controlled. For this reason, there was not a single instance of failure to sterilize due to inappropriateness of temperature and humidity. From these results it is concluded that sterilising effect was affected by sterilization procedure and condition.(ABSTRACT TRUNCATED AT 250 WORDS)

1. The effects of leucine and its metabolites, such as alpha-ketoisocaproate and ketone bodies, on the metabolic fluxes of tryptophan were investigated in isolated rat liver cells using [benzene ring-U-14C]- or [methylene-14C]-tryptophan. 2. Tryptophan metabolized through the kynureninase flux decreased while that metabolized to acetyl CoA remained unchanged in the presence of leucine or its metabolites. Accordingly an amount of tryptophan metabolized through the quinolinate-NAD pathway, which was estimated by subtracting an amount of tryptophan metabolized via the acetyl CoA flux from that via the kynureninase flux, was decreased in the presence of leucine or its metabolites. 3. Less quinolinate accumulated during incubation with metabolites of leucine, however, the amount was still sufficient to saturate quinolinate phosphoribosyltransferase (EC 2.4.2.19). 4. Leucine and its metabolites added in vitro at 1 mM level did not inhibit quinolinate phosphoribosyltransferase activity in rat liver homogenate. 5. The results indicate that a decrease in NAD biosynthesis from tryptophan caused by metabolites of leucine in the isolated rat liver cells was neither due to insufficient supply of quinolinate nor to direct inhibitory effect on quinolinate phosphoribosyltransferase.
Acute toxicity studies of miporamicin and its trace ingredients, degradations and metabolites were conducted in mice and rats. 1. Following oral administration of miporamicin (MPM), none died among mice or rats even at the highest dosage levels. Therefore, its LD50 values were estimated to be greater than 2,500 mg/kg for mice and greater than 2,000 mg/kg for rats. The LD50 value of MPM was the highest by oral route, followed, in order, by subcutaneous route and intravenous route. There was no difference in this respect between sexes of animals studied. 2. No signs of abnormalities were observed among mice or rats following oral administration of MPM. In animals dosed with MPM by subcutaneous route, such inflammatory reactions as swelling, subcutaneous hyperemia and hemorrhage, and loss of hair incrustation at the site of injection were noted. Animals among those given MPM by intravenous injection developed postdosing depression of motor activity, respiratory depression or arrest, tremor and convulsion. 3. Deaths from administration of MPM were estimated to be due to paralysis of respiratory function inasmuch as fatally affected animals exhibited respiratory depression and cyanosis and, subsequently, respiratory arrest was followed by cardiac arrest. 4. Trace ingredients, metabolites and degradation products of MPM proved to be essentially the same as MPM in acute toxicities.
