[Effect of propranolol on lipid metabolism in rats].

The effects of propranolol on lipid metabolism were studied in rats. Male Sprague Dawley rats at 5 weeks of age were used. Rats were given 5-10 mg/kg/day of propranolol subcutaneously or 10 mg/kg/day orally for 8 weeks. Serum non-esterified fatty acids and triglyceride were lowered in the oral treatment with propranolol but not changed in the subcutaneous one. Total serum cholesterol tended to decrease at the early weeks of treatment and increased at later weeks of treatment as compared with the control group. alpha-Lipoprotein was higher and beta-lipoprotein was lower in propranolol-treated groups than that in the control group. Serum lecithin cholesterol acyl transferase activity and high density lipoprotein were higher in propranolol-treated groups at 1st and 8th week of treatment. Total cholesterol in the liver decreased at the 8th week of treatment, but triglyceride in the liver did not change. These results suggest that propranolol inhibits the hormone sensitive lipase activity through adenyl cyclase at the early weeks of treatment, but this effect is reduced by homeostasis at the later weeks of treatment. There were some differences in serum lipids and enzyme activities between the oral and subcutaneous treatments. This would be related to the first pass effect of propranolol.

Antibodies to Epstein-Barr virus-specific antigens in patients with hairy-cell leukemia.

Antibody titers to Epstein-Barr virus (EBV)-specific antigens of 19 patients with hairy-cell leukemia (HCL) were markedly elevated. All patients showed high titers of IgG anti-viral capsid antigen (VCA) reactivity equal to or greater than 320 (reciprocal titer). The anti-VCA reciprocal geometric mean titer (GTM) was 1106 in contrast to a GMT of 80 for healthy controls (p less than 0.001). No IgM anti-VCA antibody was detected, but three patients had IgA anti-VCA antibodies. Fourteen patients had elevated anti-early antigen (EA) titers (GMT = 177) which were indicative of active infections with EBV. Seven of the 14 patients demonstrated the diffuse component antibody of the early antigen (EA) complex at a GMT of 98. Anti-Epstein-Barr virus nuclear antigen (EBNA) was detected in sera of 16 patients (GMT = 64, controls GMT = 79). It is noteworthy that three patients with anti-VCA titers lacked anti-EBNA titers. These anti-body responses suggest that immunodeficiency secondary to HCL allowed reactivation of the virus.

Diagnostic procedures for assessment of disease extent in prostatic carcinoma.

Staging of carcinoma of the prostate was performed on patients using Watanabe's method of transrectal ultrasonotomography (N = 37), pedal lymphography (N = 10), and bone scanning with technetium phosphate (N = 35). Of 14 patients found to have tumor confined within the prostate following routine examination, 4 were elevated to a higher disease stage after sonographic evaluation. Thirty-two percent of 25 patients with normal bone X-rays had abnormal bone scans. Four cases with positive node metastases on lymphogram had already presented with skeletal involvement. The values of these techniques in the assessment of disease extent in prostatic carcinoma are compared and briefly discussed.

Arteriovenous malformation of the ileum with chronic intestinal bleeding.

A 47 year old man had a ten year history of intestinal bleeding due to arteriovenous malformation of the distal ileum. Selective angiography is the most reliable technique for the diagnosis of the lesion and a correct diagnosis was not made until th selective superior mesenteric arteriography was done after repetitive laparotomy and intestinal resection. Transcatheter embolization was transiently successful. This case seemed to belong to type 2 according to Moore's classification and was probably of congenital origin.

Essential Role of T cells in the postexposure prophylaxis of rabies in mice.

Athymic nude mice injected intramuscularly with a street stain of rabies virus were not protected against rabies by postexposure administration of beta-propiolactone-inactivated rabies vaccine. In contrast, their normal littermates were completely protected from death by the same vaccination regimens. Nude mice did not produce IgG antibody as a result of the vaccine during the test period of 15 days, whereas normal littermates produced IgG antibody from day 5 after vaccination. However, passive immunization with antirabies hyperimmune mouse ascites showed that antibody was completely ineffective in protecting either nude mice or their normal littermates against rabies when given later than 2 days after infection. No significant difference in the induction of circulating interferon by the vaccination was noted in these mice. Passive transfer of immune spleen cells to nude mice immediately after infection resulted in 30 to 37.5% protection of the mice. Passively transferred spleen cells did not produce detectable amounts of neutralizing antibody in the recipient mice except on day 2 after the transfer, when a low level of antibody was detected. These observations demonstrate the essential role of T cells in the postexposure prophylaxis of rabies in mice. The mechanisms of the failure of postexposure vaccination in nude mice are discussed.

A Halophilic vibrio isolated from a case of chronic cholecystitis.

We found a halophilic vibrio in B bile from a 75-year-old female patient with chronic cholecystitis, and examined its biochemical characteristics. The organisms are gram-negative short rods or comma shaped, with some ring forms. They have a single polar flagellum, but not capsule. The strains can grow in peptone water with 1.0 to 4.0% NaC1, but not with no NaC1 or 6.0% NaC1. The characteristics of the organisms are positive dextrose fermentation, catalase, oxidase, and ornithine decarboxylase, and negative lysine decarboxylase, arginine dihydrolase, and absence of gas from glucose. They are sensitive to 2,4-diamine-6,7-diisopropyl pteridine (0/129). These characteristics indicate that the isolated strain should be a halophilic vibrio. however, no growth on Salmonella-Shigella (SS), SS with added sucrose and bromcresol purple (SS-SB), MacConkey's or thiosulfate citrate bile salts (TCBS) agar plates was demonstrated. Nitrate reduction, Simmons' citrate agar, indole, omicron-nitrophenol-beta-d-galactopyranoside (ONPG), motility and esculin hydrolysis were positive. Urease, gelatinase, Voges-Proskauer, phenylalanine deaminase and malonate reactions were negative. Acid was produced from amygdalin, arabinose, cellobiose, fructose, galactose, glucose, lactose, maltose, mannitol, salicin, starch, sucrose, trehalose, and xylose, but not from adonitol, dulcitol, inositol, mannose, melibiose, raffinose, rhamnose and sorbitol. From these characteristics the isolate is considered to be not identical with V. parahaemolyticus, V. Cholerae, V. Vulnificus or other vibrios. It can be presumed that this isolate represents another species of halophilic vibrio.

[Effect of oxythiamine and pyrithiamine on thiamine levels in the blood, liver and urine of rats (author's transl)].

Thiamine (T) deficient state and various functional and morphological changes in rat liver were induced, after several days, with T antagonists, oxythiamine (OT) or pyrithiamine (PT), and a thiamine deficient diet (TDD). The relationship between the extent of liver damage and T levels in blood, liver and urine were studied in OT treated rats fed a TDD (OTD group). PT treated rats fed a TDD (PTD group), OT treated rats (OT group), PT treated rats (PT group) and rats fed a TDD (TDD group). The T levels in blood and liver of OT group, PT group, OTD group, PTD group and TDD group were 89% and 91%, 76% and 88%, 25% and 14%, 33% and 16%, 24% and 15% compared with the data in the control group, respectively. Effects of OT or PT on T levels were not remarkable in some cases, and such may be due to the duration time of T antagonists, which was considered to be 3-15 hours in PT treated rats, and 0-18 hours in PT treated rats, as assessed by urinary T measurements. Treatment of T antagonist twice daily is recommended to obtain a definite T deficient state. Our results suggest that the extent of liver damage is related to the T levels in tissue.

Cellular immune defects to Epstein-Barr virus-determined antigens in young males.

Three males with the X-linked lymphoproliferative syndrome (XLP) with hypo- or agammaglobulinemia following Epstein-Barr virus (EBV) infection and two males with the chronic mononucleosis syndrome were investigated for immune responses to EBV-determined antigens. Males with XLP showed profound cellular immune defects. Markedly diminished responses of natural killer cell and interferon-activated killer cell activities and impaired leukocyte migration inhibition responses to phytohemagglutinin were determined in patients with XLP. The two patients with chronic mononucleosis showed less severe defects. All patients showed partial or complete impairment of their EBV-specific immune responses as measured by leukocyte migration inhibition. EBV-specific antibodies were markedly diminished against EBV-associated nuclear antigen, early antigen, and viral capsid antigen in males with XLP. In contrast, patients with chronic mononucleosis had elevated antibodies to most EBV-specific antigens. Individuals with life-threatening EBV-induced lymphoproliferative disorders may exhibit multiple defective immune mechanisms against the virus.
