Partial inhibitory seizures: a report on two cases.

This paper described two epileptic children with the clinical features of tonic-clonic seizures, brief atonic episodes, drop attacks and myoclonic jerks. Patient 2 also displayed absences and behavioral disturbance with episodes of minor status epilepticus. When the patients held both arms outstretched, polygraphic records revealed the spike-and-wave (sp-w) discharges in association with the sudden dropping of an outstretched arm. The drops were due to a loss of tone recorded in deltoid muscles, and coincided with the slow wave components of bilateral sp-w complexes, predominantly on the contralateral side. In the present paper, the clinical and electrophysiologic features of this particular type of seizure are presented in detail. A cortical origin of these episodes is suggested.

Quantitative immunoferritin localization of [Na+,K+]ATPase on canine hepatocyte cell surface.

Distribution of [Na+,K+]ATPase on the cell surface of canine hepatocytes was investigated quantitatively by incubating prefixed and dissociated liver cells with ferritin antibody conjugates against canine kidney holo[Na+,K+]ATPase. We found that [Na+,K+]-ATPase exists bilaterally both on the bile canalicular and sinusoid-lateral surfaces. The particle density on the bile canalicular surface was much higher (approximately 2.5 times) than that on the sinusoid-lateral surface. In the latter region, the enzyme was detected almost equally both on the sinusoidal and lateral surfaces. On all the surfaces, the distribution of the enzyme was homogeneous and no clustering of the enzyme was detected. Total number of the enzyme on the sinusoid-lateral surface was, however, approximately three times higher than that on the bile canalicular region, because the sinusoid-lateral surface represents approximately 87% of the total cell surface of a hepatocyte. We suggest that the [Na+, K+]ATPase on the bile canalicular surface is responsible for the bile acid-independent bile flow and the other transport processes on the bile canalicular cell surface, while that on the sinusoid-lateral surface is responsible not only for the active transport of Na+ but also for the secondary active transport of various substances in this region.

Estradiol-17 beta and estradiol-17-glucosiduronate before and at delivery.

The maternal peripheral serum level of estradiol-17 beta (E2), which reaches a peak a few weeks before delivery, may play a role in initiating delivery. In this study, the steroidal activity regulated by both the production and conjugation was estimated by measuring estradiol-17-glucosiduronate (E2-17-G), E2, estriol-16-glucosiduronate (E3-16-G), and estriol (E3) before and at delivery. At delivery, levels of E2 and E2-17-G were higher in the maternal peripheral vein (MP) than in the umbilical vein (UV) and umbilical artery (UA), but those of E3 and E3-16-G were higher in UV and UA than in MP. These results suggested that the production and conjugation of E2 were mainly regulated on the maternal side, but those of E3 were regulated mainly in the feto-placental unit. The ratios of E2/E2-17-G in MP were or became high a few weeks before delivery but decreased remarkably as pregnancy progressed to delivery. The activity of E2 seems to be regulated at least in part by conjugation in late pregnancy and at delivery.

Isolation of the alpha and beta subunits of canine (Na+,K+)ATPase by using SDS-PAGE and lectin-Sepharose.

(Na+,K+)ATPase from dog kidney was solubilized and denatured by SDS treatment, then applied to a Con A- and WGA-Sepharose column. While the alpha subunit of the ATPase had no affinity for either of the lectin-Sepharoses, the beta subunit specifically bound to WGA-Sepharose and was eluted with N-acetylglucosamine. This property was utilized for the isolation of the alpha and beta subunits by using lectin-Sepharoses and SDS-polyacrylamide gel electrophoresis. The amino acid composition of the alpha subunit thus isolated was in reasonable agreement with the data reported by Kyte (Kyte, J. (1972) J. Biol. Chem. 247, 7642-7649). The amino acid and carbohydrate compositions of the beta subunit were, however, different from his data. The beta subunit contained little histidine (0.1 mol/100 mol amino acid) and a very large amount of carbohydrates (33%). The antibody raised against alpha or beta subunit reacted specifically with the corresponding subunit and with protease-fragmented alpha subunit and neuraminidase-treated beta subunit, respectively, but no cross-reactivity was observed between the two subunits. These results indicate that our alpha and beta subunits were highly purified.

l-Serine Production by a Mutant of Sarcina albida Defective in l-Serine Degradation.

For improved l-serine production, an l-serine dehydratase-defective mutant of Sarcina albida IAM 1012 was obtained. In the mutant, the activities of the enzymes responsible for l-serine production were as high as those in the parent strain, and, at a low glycine concentration, the mutant accumulated l-serine more efficiently than the parent. Under optimum conditions, 21 mg of l-serine per ml accumulated from 100 mg of glycine per ml. l-Serine was isolated from a reaction mixture as l-serine m-xylene-4-sulfonate, and free amino acid was obtained in high yields by use of an ion-exchange resin. Residual glycine was recovered at a yield of 61%.

[Dynamics of estriol-16-glucosiduronate (E3-16-G) in early pregnancy -- its prognostic value in complicated pregnancy (author's transl)].

Urinary estriol-16-glucosiduronate (E3-16-G) in early pregnancy was measured by a direct radioimmunoassay which was highly specific for the compound and did not require hydrolysis or chromatography. The level of E3-16-G in normal pregnant women increased as pregnancy progressed and was over the level in non-pregnant women after 6 weeks gestation. The levels were lower in complicated pregnancies, namely threatened abortion, intra uterine fetal death, ectopic pregnancy, anencephalus and so on. Decreases of urinary E3-16-G in threatened abortion were more rapid than those of urinary hCG, which was used as a useful prognostic indicator in those cases. These results suggested the possibility that the fetus played some role in the production of E3-16-G in early pregnancy. In order to clarify the role of the fetus in the formation of E3-16-G, it was measured in the maternal peripheral vein, uterine artery and vein, and in the fetal tissue at hysterectomy in complicated pregnancies (three cases). These results also suggested the possibility that E3-16-G was produced by the fetus. In the in vitro experiments where 14C-E3 was incubated with various tissues in early pregnancy, it was demonstrated that 14C-E3 was conjugated to be E3-16-G by homogenate of the fetal tissue in 8 weeks gestation, and by the liver and by the kidney in 13 weeks gestation but not by chorionic tissue. These results indicated that the conjugation process of E3, though perhaps not entirely, proceeds in the fetus and the urinary determination of E3-16-G might give us good information about the fetus in pregnant women.
