Effect of isoprenaline on tissue elastin content and serum elastolytic activity in normal and hydrazinophthalazine-treated rats.

Cardiomyopathy was produced by isoprenaline injections to healthy and hydrazinophthalazine-treated rats. The elastin content in the heart muscle and the aortic wall as well as the level of serum elastolytic activity were determined. An increase of elastin content in both tissues was found, and its progress was concomitant with the development of isoprenaline injury. It was also found that the elastolytic activity increased on the second day after the last isoprenaline injection, and on the seventh day of the experiment it began to decrease. Changes in hydrazinophthalazine-treated rats after dosing them with isoprenaline had a similar tendency but they were significantly lower than those found in healthy animals receiving isoprenaline only.

Effect of some selected anti-inflammatory steroids on elastin content in liver of rats treated with carbon tetrachloride.

Hepatic fibrosis was induced in rats by poisoning them with carbon tetrachloride. The influence of anti-inflammatory steroids (hydrocortisone, prednisone, dexamathasone) upon elastin content in liver was investigated in healthy and CCl4 intoxicated rats. An increase of liver elastin content was observed in animals with CCl4-induced hepatic fibrosis Opposite results were obtained in normal rats after treating them with glucocorticosteroids. A decrease of elastin content after steroid treatment was also found in rats with injured liver, as compared to animals receiving CCl4 only. Presumable mechanisms of the results are discussed.

Leukocyte collagenolytic activity in normal and hydrazinophthalazine-treated rats during isoprenaline-induced cardiomyopathy.

Cardiomyopathy was produced by isoprenaline injections to healthy and hydrazinophthalazine-treated rats. The activity of leukocyte collagenase and collagenolytic cathepsin was determined on the 2nd, 7th, 19th and 21st day after the last isoprenaline injection. At the same time morphological studies of the heart muscles were carried out. It was found that alterations in enzyme activity were different in healthy and hydrazinophthalazine-treated rats. A negative synergism of isoprenaline as regards the hydrazinophthalazine action was observed in biochemical and morphological studies.

Effect of anti-inflammatory steroids on the activity of selected lysosomal hydrolases in serum of rats with experimental liver fibrosis.

Liver fibrosis was produced in rats by subcutaneous injections of carbon tetrachloride. The effect of anti-inflammatory steroids (hydrocortisone, prednisone, dexamethasone) on the activity of serum lysosomal hydrolases (beta-N-acetylglucosaminidase, beta-glucosidase and alpha-mannosidase) was investigated in healthy and CCl4 poisoned rats. The marked increase of enzymatic activity in blood serum was observed in animals with CCl4-induced liver fibrosis. Quite opposite results were found in normal rats treated with glucocorticosteroids. In rats which were given both carbon tetrachloride and corticosteroids the obtained values of serum enzymatic activity did not have a uniform character as compared to controls or CCl4 intoxicated rats. Presumable mechanisms of the results obtained are discussed.

Studies on collagenolytic cathepsin activity in rats with experimental collagen-like syndrome.

Collagenolytic cathepsin activity was determined in the heart muscle, liver, kidney spleen, skin samples and leukocytes of normal and hydrazinophthalazine treated rats Isolated collagen susceptibility to the crude liver enzyme action was estimated as well The products of enzymatic degradation were analyzed by polyacrylamide gel electrophoresis. An increase of enzyme activity in all studied organs and leukocytes was observed. It has also been found that the crude liver enzyme is able to degrade the polymeric collagen gamma-chains.

Elastin and collagen metabolism in the arterial wall of rats fed an atherogenic diet.

Experimental atherosclerosis in rats was produced by feeding them atherogenic diet for ten months. Elastin and collagen content of the arterial wall as well as some aspects of the metabolism of these proteins were studied. A decrease of elastin content was accompanied by its enhanced susceptibility of enzymatic hydrolysis in vitro. An increase of soluble collagen fractions in tissue and a simultaneously enhanced level of collagen catabolites in serum and urine were found. The present work deals with a disturbed elastin and collagen metabolism in experimental atherosclerosis and shows simultaneous participation of these compounds in pathogenesis of this disease.

[Experimental evaluation of toxicity of selected mixtures used in metallurgy].

The investigation is aimed at evaluation of the effects of copper-coating and bonderizing solutions and oil--emulsion used in technological processes in the metallurgic industry upon hematological and biochemical changes in rats' peripheral blood. The studies were performed on male Wistar rats. The solutions were carried onto hairy and depilated skin. Acute toxicity was evaluated in two-weeks' experiment and chronic toxicity in three--months' experiment. The copper-coating solution was found to be most hazardous, whereas the oil emulsion-the least hazardous. The copper-coating and bonderizing solutions, when chronically applied, result in anaemia and changes in the percentage picture of leucocytes, and also increased blood concentration of uric acid, creatinine and hydroxyproline. No significant changes in concentrations of the remaining biochemical indices were found. The results point to synergistic effects of oil-emulsion and copper-coating solution.

[Effects of ionizing radiation on the content of total collagen and its fractions and the activity of collagenolytic enzymes in rat tissues].

Changes in collagen fractions as well as in the activity of leucocytic collagenase and collagenolytic cathepsin of rats after 30 days of gamma-rays exposure were examined. A statistically significant decrease in total collagen content resulting from the reduction of salt--soluble (NSC) and acid--soluble (ASC) collagen fractions was found. An increased content of insoluble collagen fraction (ISC) may confirm the opinion about stimulative gamma-rays influence upon cross--links formation. An increase in collagenolytic enzymes activity may account for an elevated degradation of collagen; with a contribution of leucocytic collagenase and collagenolytic cathepsin. Perhaps the quantitative increase in insoluble collagen stimulates elevated biosynthesis of collagenolytic enzymes determining the process of protein degradation. Further investigations are needed to explain the radiobiological effects of gamma-rays on the connective tissue.

Activity of selected enzymes in arterial wall and blood serum of rats with experimental atherosclerosis.

The activity of lysosomal hydrolases (hyaluronidase and beta-glucuronidase) as well as elastolytic activity in arterial wall and blood serum of rats with experimental atherosclerosis were determined. An increase of the enzyme activity was found in arterial wall, while in blood serum the rise of lysosomal hydrolase activity was accompanied by and unchanged level of elastolytic activity, as compared to control animals. The present studies show the participation of lysosomal enzymes and elastolytic activity in the development of atherosclerotic changes.

[Disorders of lysosomal enzyme activity in rats with experimental radiation sickness].

The studies undertaken were aimed at detecting the biological effects of radioactive radiation on rat's organism, through studying the activity of lysosomal enzymes in both blood plasma and some organs. The contemporary studies suggest that lysosomes play an important role in the occurrence and course of postradiation disease. The obtained results suggest the multidirectional gamma-rays effects on lysosomal enzymes response in serum, leucocytes, liver lysosomes and in liver, kidneys, lungs, heart. Increased activity of acid phosphatase, beta-glucuronidase and beta-acetyl-glucosaminase in the tissues of irradiated animals indicates that gamma rays labiliza the lysosomal membrane. The range of changes indicates a selective nature of this phenomenon. Kidneys, lungs and liver appeared the most ray--sensitive organs. The activity of acid phosphatase was found to be most increased in blood serum and leucocytes. The activity of all the examined enzymes in liver lysosomes was decreased. Acid phosphatase exhibited the greatest activity increases. Lysosomal responses, as one of the radiobiological effects of postradiation disease are indicative of the degree of destructive or regenerative changes in the organism.

Influence of thyroid hormones and methylthiouracil upon elastin content in the liver of rats with carbon tetrachloride-induced hepatic fibrosis.

The effect of administration of thyroid extract and methylthiouracil upon the elastin content of the liver was investigated in healthy and CCl4-poisoned rats. Hyperthyroidism induced a decrease in liver elastin content and hypothyroidism led to an increase of liver elastin. Similar changes were found in rats with carbon tetrachloride-induced hepatic fibrosis. The possible mechanisms of these changes are discussed.

Influence of thyroid hormones and of methylthiouracil upon collagen content in the liver of rats with hepatic fibrosis.

Influence of the administration of thyroid extract and of methylthiouracil on the collagen content in the liver and on hydroxyproline level in serum in rats with or without carbon tetrachloride-induced fibrosis of the liver was investigated. A decrease of collagen content in the liver was found in hyperthyroid rats and an increase of the liver collagen content was observed in hypothyreosis. Similar results were found in animals with fibrosis of the liver.

Age-related changes in the content of fibrous proteins in the rat tissues.

An attempt was made to establish a relationship between the content of elastin and collagen in the rat tissues during the process of aging. The content of collagen fractions and elastin in the rat liver, lung and skin, as well as the elastolytic activity of blood serum were determined. It was found that the concentration of elastin as well as the elastolytic activity of blood serum are increasing during the maturation of rats and the total collagen content is increasing too. After the animals reached the age from twelve to twenty four months--above mentioned values began to decrease. It is concluded that the changes in the content of the two fibrous proteins of the connective tissue depend on age.
