C-19393 S2 and H2, new carbapenem antibiotics. IV. Inhibitory activity against beta-lactamases.

New carbapenem antibiotics, C-19393 S2 and H2, have been found to be potent and broad-spectrum inhibitors of beta-lactamases. Among 11 types of beta-lactamases tested, those from Escherichia coli (plasmid-bearing), Klebsiella pneumoniae, Proteus vulgaris, Serratia marcescens and Bacteroides fragilis were especially sensitive. They also inhibited cephalosporinases insensitive to clavulanic acid. The inhibition by C-19393 S2 and H2 was of progressive type, except for the inhibition of E. coli enzyme (plasmid-mediated type I) by C-19393 H2. The inhibition of E. coli beta-lactamase by C-19393 S2 was irreversible, while that by C-19393 H2 was reversible.

C-19393 S2 and H2, new carbapenem antibiotics. I. Taxonomy of the producing strain, fermentation and antibacterial properties.

C-19393 S2 and H2 are new carbapenem antibiotics produced by a streptomycete. The producing strain was taxonomically studied and named Streptomyces griseus subsp. cryophilus. Cobaltous compounds were necessary for production of the antibiotics. C-19393 S2 and H2 showed a broad spectrum of antibacterial activities with C-19393 H2 being 8 approximately 120 times more active than C-19393 S2. They also exhibited beta-lactamase-inhibiting activities and acted synergistically with ampicillin and cefotiam against clinical isolates resistant to beta-lactam antibiotics.

Japanese psychoanalysis and the Ajase complex (Kosawa).

In 1932, Kosawa visited Freud at 19 Berggasse, and submitted to him a paper entitled: 'Two kinds of guilt feelings, the Ajase complex. In his paper, Kosawa expounded his theory of the Ajase complex, rooted in Buddhist thought. He discussed the psychological peculiarity of the Japanese as evidenced by the acceptance of oral dependency toward the mother, the repression of the resentment at the mother and 'guilt feeling over being forgiven' and a need to make restitution for harboring this evil intent. This type of guilt feeling is quite different from the guilt feeling as a fear of punishment which Freud embodied in his theory of the Oedipus complex.

Therapeutic efficacy of cefozopran in a murine model of haematogenous pneumococcal meningitis.

Antimicrobial regimens for the treatment of pneumococcal meningitis are not established. We have produced a murine model of haematogenous pneumococcal meningitis and have examined its usefulness for determining the required dosage and term of antimicrobial agents. Streptococcus pneumoniae serotype 6 was injected intraperitoneally (inoculum: about 1 x 10(4) CFU) into mice. Although half of the mice died within 2 days, the surviving mice showed positive bacterial cultures, increase of the protein level, decrease of the glucose level and infiltration of polymorphonuclear leucocytes into cerebrospinal fluids (CSF). When cefozopran was administered subcutaneously twice a day for 1-3 days starting 2 days after infection, dose- and duration-dependent effects were observed and all mice treated with 20 mg/kg of cefozopran for 3 days survived. The penetration rate of cefozopran from blood to CSF in infected mice was 44.7%, which was 6 times higher than that obtained in uninfected mice. This model may be useful for investigating the pathogenesis of haematogenous pneumococcal meningitis and its therapy.
