Human infection with the pancreas fluke, Eurytrema pancreaticum.

A 70-year-old Japanese woman with gastric cancer in Fukuoka Prefecture, Japan, was found at autopsy to have about 15 adult flukes, Eurytrema pancreaticum, in the dilated pancreatic ducts. The number of eosinophils in the blood was within normal limits. The two worms which were least damaged when removed from the gland were used for stained specimens, and another two were macerated to collect the eggs. The parasites (10 X 5, 11 X 7 mm in size) were broad, flattened, oval to fusiform. The suckers were large, the oral (2.1 X 2.0, 2.0 X 1.9 mm) being larger than the ventral (1.6 X 1.6, 1.4 X 1.5 mm). The eggs (47.06 +/- 3.88 X 30.35 +/- 2.74 microns) were embryonated in the uterus. These findings led to the identification of E. pancreaticum. This is the second documented case of eurytremiasis in man.

Characterization of tyrosine hydroxylase from bovine adrenal medulla.

Tyrosine hydroxylase purified to apparent homogeneity from the soluble fraction of bovine adrenal medulla had an apparent Mr of about 280,000 by Bio-Gel A-1.5m chromatography, and gave a single band with a Mr of 60,000 by sodium dodesyl sulfate polyacrylamide gel electrophoresis. The enzyme is considered to be composed of four identical subunits. Isoelectric point of purified enzyme was pH 6.0. The amino acid composition of the enzyme was characterized by fairly high contents of glutamic acid and alanine residues. The N-terminal amino acid was determined to be glutamic acid.

The role of Kupffer's cells in disseminated intravascular coagulation. A morphologic study in thrombin-infused rabbits.

We studied ultrastructural changes of Kupffer's cells in rabbits during experimental disseminated intravascular coagulation (DIC) induced by thrombin infusion. We observed three steps of fibrin clearance by Kupffer's cells: adsorption on the cell coat, invagination of the plasma membrane, and formation of bristle-coated vesicles adjoining the invaginated fibrin. There was no direct evidence of phagocytosis of fibrin by Kupffer's cells. Pinocytosis revealed by bristle-coated vesicles seemed to be one of the microthrombiclearing mechanisms of Kupffer's cells. Dissemination of microthrombi to systemic organs seemed to depend on the speed of fibrin formation and the clearing ability of Kupffer's cells. This hypothesis was supported by the finding that pretreatment with latex particles increased the severity of DIC.

Distribution patterns of microthrombi in disseminated intravascular coagulation.

Distribution of microthrombi in 37 autopsy cases of untreated disseminated intravascular coagulation (DIC) was studied histologically. According to the organs predominantly involved, four patterns of distribution were distinguished: hepatic, hepatorenal, renal, and undetermined. The results suggested that these patterns indicated the course of DIC and the function of Kupffer's cells in the liver, whose role in the course of DIC was evaluated.

Highly sensitive assay for phenylethanolamine N-methyltransferase activity in rat brain by high-performance liquid chromatography with electrochemical detection.

This paper describes a new and highly sensitive assay for phenylethanolamine N-methyltransferase (PNMT) activity with noradrenaline as substrate in various rat brain regions by high-performance liquid chromatography with electrochemical detection. Commercially available noradrenaline contained about 0.27% of contaminating adrenaline, which was removed to reduce the blank value. Enzymatically formed adrenaline was adsorbed on an aluminum oxide column, eluted with 0.5 M hydrochloric acid, separated by high-performance reversed-phase paired-ion chromatography and measured with electrochemical detection. 3,4-Dihydroxybenzylamine was added to the incubation mixture as an internal standard after the reaction. This assay was very sensitive and 0.5 pmol of adrenaline formed enzymatically could be detected. This assay method was applied to measure PNMT activity in various rat brain regions. The highest activity was observed in the hypothalamus, pons plus medulla oblongata, septum, lower brain stem, and cerebral cortex; the lowest activity was in the striatum, hippocampus, cerebellum, and limbic brain.

Transrectal longitudinal ultrasonotomography of the prostate by electronic linear scanning.

Transrectal longitudinal ultrasonotomography of the prostate with the newly developed electronic linear scanning method was done on 100 cases and the characteristic echo patterns for each prostatic disease were obtained. With this method we can observe real-time longitudinal tomograms of the prostate, seminal vesicles and bladder. Real-time observations make it easier to examine the relationship between the prostate and its surrounding organs, especially the bladder neck and the prostatic urethra, at the time of urination.

An electronmicroscopical study on peroxidase activity of rabbit monocytes, resident and exudate macrophages.

Changes of peroxidase (PO) activity in blood monocytes, untreated or stimulated peritoneal macrophages and subcutaneous exudate macrophages of rabbits were investigated with the glucose-oxidase method in the culture experiments of these cells after their in vitro or in vivo adherence to the surface of foreign bodies. 1) PO activity was demonstrated in the nuclear envelope (NE) and rough endoplasmic reticulum (rER) in 75% of the untreated peritoneal macrophages. 2) Blood monocytes, most of the stimulated peritoneal macrophages, and subcutaneous exudate macrophages showed no PO positivity. 3) In the in vitro culture of blood monocytes and stimulated peritoneal macrophages, there appeared from 2 hours cells showing PO positivity in NE and rER. 4) Although blood monocytes, stimulated peritoneal macrophages, and subcutaneous exudate macrophages after their in vivo surface adherence showed no PO positivity, it appeared in these cells when further cultured in vitro. From the fact that such a positive change of PO activity in these cultured cells is not only a transient phenomenon but also in vitro cultural one, we cannot subscribe to the view that blood monocyte is the precursor of resident macrophages.

Simultaneous fluorometry of urinary dopamine, norepinephrine, and epinephrine compared with liquid chromatography with electrochemical detection.

A simple fluorometric method is described for simultaneous assay of urinary norepinephrine, epinephrine, and dopamine. The catecholamines are separated by boric acid gel chromatography and subsequently determined by use of a modified trihydroxyindole reaction. By the separation with boric acid gel, dopa can be eliminated, which interferes with dopamine measurement; consequently, the reaction is specific for catecholamines. This simple, rapid method was applied to urine specimens from normal adults. Values for urinary dopamine, norepinephrine, and epinephrine agreed with those obtained by "high-performance" liquid chromatography with electrochemical detection.
