Wear of artificial denture teeth by use of toothbrushes. Part 1: Abrasive wear of anterior teeth.

High-strength denture teeth (HS teeth) were developed in order to improve the hardness and wear resistance of conventional plastic denture teeth (PL teeth), while retaining their feature of easy occlusal adjustment. The objective of this study was to evaluate the abrasive wear resistance of HS teeth. We conducted wear tests and measured surface roughness using six types of anterior artificial teeth, i.e., three types of HS teeth and three types of PL teeth, the latter serving as the control. The results of the toothbrush abrasion test revealed that the HS teeth had about 5 times greater wear resistance than the PL teeth. It was also found that the type of artificial teeth and the number of abrasive wear-testing strokes had a significant (P less than 0.05) influence on the surface roughness of artificial teeth.

Studies on the castability of Co-Cr alloy for cast plates. Part 2: Effect of form of sprue attachment to the wax pattern.

Co-Cr alloy is used more frequently than Ni-Cr alloy as a non-precious alloy for cast plates in Japan. However, since the melting point of Co-Cr alloy is very high, about 1300 degrees C, and since it oxidizes easily, a vacuum-pressure casting machine capable of melting this alloy in a reducing atmosphere has recently been developed. Using this vacuum-pressure casting machine, the authors studied the effects on the castability of Co-Cr alloy due to the form of sprue attachment to the wax pattern. The results clarified that in the vacuum-pressure casting method, the form of sprue attachment to the wax pattern has a significant effect (p less than 0.01) on the castability of Co-Cr alloy.

A region-matched hydrophobic interaction between melittin and dimyristoylphosphatidylcholine in a ternary mixture of phosphatidylcholines.

Interaction of melittin with phosphatidylcholine molecules in pure vesicles, binary mixtures and a ternary mixture of dimyristoylphosphatidylcholine IDMPC), dipalmitoylphosphatidylcholine (DPPC) and distearoylphosphatidylcholine (DSPC) was investigated by differential scanning calorimetry. Melittin binds preferentially with DMPC, and results in segregation of DMPC in binary mixtures of DMPC/DPPC and DMPC/DSPC and in a ternary mixture of DMPC/DPPC/DSPC. The results indicate that the hydrophobic part of peptide interacts preferentially with the phospholipid which has the same size of hydrophobic region or fatty acyl chains.

Porcine luteinizing hormone isoform(s): relationship between their molecular structures, and renotropic versus gonadotropic activities.

We recently demonstrated the renotropic activity in ovine LH isoform(s). In this study we purified porcine (p) LH isoforms and analyzed their structures and bioactivities. Purified pLH preparation (G-100 fraction 3) was dissociated into alpha- and beta-subunits, followed by isolation with reverse phase HPLC. Six components were isolated and analyzed for their amino acid compositions and amino-terminal amino acid sequences. alpha-Subunits were found to have heterogeneous N-terminal sequences, which started at Phe-1, Gly-4, Phe-6, or Thr-7. This N-terminal heterogeneity and the presence of the initial 6 amino acids has not been reported previously to the best of our knowledge. Moreover, the first 10 residues, Phe-Pro-Asp-Gly-Glu-Phe-Thr-Met-Gln-Gly, were identical with those of the ovine LH alpha subunit. At least 3 different beta-subunits were identified as heterogeneous in their carboxyl-terminal amino acids. The pLH preparation (G-100-fraction 3) was then chromatographed by an isoelectrofocusing (IEF) column. Four IEF fractions were obtained. Their amino acid structures appeared to be identical, as judged by the identical elution profiles on reverse phase HPLC, but their carbohydrate compositions were slightly different, especially in N-acetylgalactosamine in alpha-subunits. Thus, fractionation on IEF depended on the heterogeneity in carbohydrate structures. Each IEF fraction had different potencies in terms of its gonadotropic activity (in vitro) and renotropic activity (in vivo and in vitro). The discrepancy was observed not only between gonadotropic activity and renotropic activity, but also between in vivo and in vitro renotropic activity. This study identified the structural heterogeneity of pLH isoforms and demonstrated their biological heterogeneity. We concluded that the carbohydrate structure of the pLH isoform is important for expressing its biological heterogeneity (gonadotropic and renotropic activity.

Sparsomycin analogs. VI. Synthesis and antitumor activity of octylsparsomycin analogs.

Five sparsomycin analogs (9-13) were prepared and examined for their ability to inhibit deoxyribonucleic acid (DNA) synthesis in L5178Y lymphoma cells. All of the compounds showed significant activity in the DNA synthesis assay. The compounds having Rc configuration exhibited almost the same activities independently of the configuration at the sulfoxide sulfur atom. Among the Sc isomers, the Rs configuration was advantageous for the appearance of activity.

[A questionnaire about the dental treatment for visiting patients at welfare center for the aged people].

A questionnaire about the dental treatment for visiting patients at Welfare center for the aged people. Nowadays, 11 percent of the population is over 65 in Japan. According to the Minister of Health and Welfare's Population Projection for Japan, it will grow to 20% at the beginning of the 21st century. Problems concerning old people will become some of Japan's biggest challenges. In that situations, the dental treatments for visiting aged patients will become more important. Then, authors sent out a questionnaire about it on aged people who have no need of anyone's assistance in their home lives. And we examined concrete countermeasures and problems about the dental treatments for visiting patients. The findings were as follows: 1. Even the aged people who have no need of anyone's assistance in their home lives, they hope much carrying out the dental treatments for visiting them. 2. The dental treatments for visiting aged patients have some problems. But it will become one of the most important way to supply their demand for dental treatments, in spite of their latent requirements, a lot of old people give up to undergo dental treatments.

Establishment and characterization of factor-dependent macrophage cell lines.

Three macrophage cell lines from bone marrow cells of C3H/HeN mice were isolated by successive transfer of the cells in culture with L-cell-conditioned medium (LCM) or WEHI-3 cell-conditioned medium (WEHI-3CM). These cell lines, which express Fc receptors, are involved in Fc-mediated phagocytosis and possess nonspecific esterase activity. Two (BDM-1 and BDM-2) of three cell lines show dependency for growth on either macrophage colony-stimulating factor (M-CSF) (CSF-1) or granulocyte-macrophage colony-stimulating factor (GM-CSF) and do not respond to interleukin 3 (IL-3). The third clone (BDM-3) proliferates in response to IL-3 as well as to GM-CSF and weakly responds to M-CSF and to interleukin 4 (IL-4). GM-CSF, in combination with the suboptimal concentration of M-CSF, acted synergistically on the proliferation of BDM-1 cells. The tumor-promoting phorbol diester, 12-o-tetradecanoyl-phorbol-13-acetate (TPA) also acted synergistically with the three CSFs (IL-3, GM-CSF, and M-CSF) to stimulate the proliferation of BDM-1 cells. The synergistic effect was observed when cells were pretreated with TPA and subsequently stimulated with IL-3. The calcium ionophore A23187 enhanced the proliferation of BDM-1 cells costimulated with TPA and IL-3. These factor-dependent macrophage cell lines should be useful for studying signal transduction mechanisms in the regulation of cell growth.

Ca2+-induced lateral phase separation in ternary mixtures of phosphatidic acid, phosphatidylcholine, and phosphatidylethanolamine inferred by calorimetry.

Phase transition characteristics of ternary mixtures of dipalmitoylphosphatidic acid, dipalmitoylphosphatidylcholine, and phosphatidylethanolamine (dilauroyl-, dimyristoyl-, or dipalmitoyl-phosphatidylethanolamine) were examined by differential scanning calorimetry at various concentrations of calcium ions. In the absence of calcium ion, these ternary mixtures showed a broad phase transition, which suggested a high miscibility of these components. Addition of a low concentration of calcium ions showed a tendency to induce separation of the transition into a major one and a small one. As the concentration of calcium ions increased, the separation became more distinct and the transition enthalpy of the major transition decreased. At a Ca2+/dipalmitoylphosphatidic acid ratio (mol/mol) of 1.5, the major transition became similar to the transition of dipalmitoylphosphatidylcholine and the phosphatidylethanolamine binary mixture. On the other hand, in a binary mixture dipalmitoylphosphatidic acid and dipalmitoylphosphatidylcholine, the Ca2+-induced phase separation was distinct even at the lowest concentration of calcium ions used in the present experiment. The results indicate that a high concentration of calcium ion is required for inducing complete phase separation of the transition event in the ternary mixture because of its high miscibility. It is suggested that the phase separation revealed by spin-labeled phospholipid in ternary mixtures at a low Ca2+ concentration might be a phase separation in a local domain.

Influence of manganese deficiency and toxicity on isoprenoid syntheses.

Twenty-eight day old wheat (Triticum aestivum L. cv Stacy) response to varying Mn concentration (10.1-10,000 micromolar) in nutrient solution was measured. Manganese concentrations in the most recently matured leaves (blade 1) were 0.21 to 19.03 mmol Mn per kilogram dry weight, respectively. Fresh and dry weights increased to a maximum at the 5 micromolar Mn nutritional level (0.37 millimole Mn per kilogram dry weight) and were decreased at Mn above and below this concentration. Blade 1 chloroplast pigment concentrations increased up to the 20 micromolar Mn nutritional level (1.98 millimole Mn per kilogram dry weight) and decreased at higher Mn concentrations. Thylakoid Mn content was above 1 mole Mn/100 mole chloroplast at Mn nutrition levels which resulted in greatly decreased plant growth. Total phytoene biosynthesis was decreased by Mn deficiency and toxicity. In vitro ent- kaurene synthesis was greatly influenced by Mn concentration with a maximal biosynthesis at 1 micromolar Mn and decreases at Mn levels above and below this concentration. In vivo blade 1 gibberellic acid equivalent concentrations were maximal at 20 parts per million Mn nutrition solution levels (1.98 millimole Mn per kilogram dry weight) and decreased at Mn tissue concentrations above and below this value; additionally, gibberellic acid concentrations were reciprocal to extracted C(20) alcohol concentrations. Mn influence on gibberellin and chloroplast pigment biosyntheses exactly matched the measured changes in growth.

Dynamics of appearance and disappearance of the ripple structure in multilamellar liposomes of dipalmitoylphosphatidylcholine.

The physical properties of the pretransition (P beta'----L beta') of dipalmitoylphosphatidylcholine liposomes were investigated using freeze-fracture electron microscopy. The kinetics of pretransition examined in the previous paper using TEMPO spin probe (Tsuchida, K., et al. (1985) Biochim. Biophys. Acta 812, 249-254) was extensively studied by observing the ripple structures in the freeze-fractured surfaces at different time intervals. When the temperature is decreased from 38 degrees C to 30 degrees C, the ripple structure disappears in the following steps. The intervals between ripples begin to expand with the decrease of ripple density upon the temperature shift, and this process continues for several tens minutes. Then, each ripple disappears gradually and changes into a completely smooth surface at 3 h after the temperature shift. The comparison of relaxation times between the previous ESR measurement and the present experiment suggests that the fast relaxation observed in the previous study corresponds to the expansion of the intervals between ripples. On the other hand, the ripple structure of regular intervals appears rapidly in some places and then spreads over the whole area of fractured surface when the temperature is increased from 23 degrees C to 35 degrees C. The results obtained in this work and the previous ESR work strongly suggest that the formation and disappearance of ripple structure is closely related to the relaxation processes near the pretransition temperature.

Properties of colony-stimulating factors produced by macrophage cell lines and hybrid cells.

Colony-stimulating factors (CSFs) produced by two simian virus 40(SV40) transformed macrophage cell lines (BAM1 and BAM3), and three hybrids (HM3-11, HM3-12, and HM3-14) derived from fusion between BAM3 and a Chinese hamster cell line (hs222-16) were examined. HM3-11 and HM3-14 produce two molecular species of CSF, which are not found in the conditioned media from cultures of BAM1 and BAM3 or lipopolysaccharide (LPS), phorbolmyristate-acetate (PMA), and zymosan-stimulated BAM3. HM3-12, which is classified into another group in terms of CSF secretion, does not produce these two CSFs. On the basis of various criteria, one of these CSF species (peak 1-CSF) was characterized as a macrophage-colony-stimulating factor (M-CSF). The other CSF (peak 2-CSF) induced a group of bone marrow cells in granulocytes and macrophages as well as growth of a mast cell line, IC2. This CSF has an apparent molecular weight of 18,000, estimated by SDS-polyacrylamide gel electrophoresis. Unlike interleukin 3 (IL3) from WEHI-3 cells, the growth factor activity of peak 2-CSF binds to DEAE-Sephacel. Thus, peak 2-CSF is similar to a granulocyte-macrophage colony-stimulating factor (GM-CSF) rather than to IL3. The anti L cell CSF serum does not inhibit the CSF activity in Chinese hamster fibroblast conditioned medium, and the IC2 cells do not respond to Chinese hamster lung conditioned medium (CHLCM), suggesting that peak 1- and peak 2-CSF are of mouse origin.
