Variation in response of xenografts of colo-rectal carcinoma to chemotherapy.

Ten xenograft lines of human colonic and rectal carcinomas have been established in immune-suppressed mice. Mice bearing tumours in the 2nd to 6th passage were treated with maximum tolerated doses of 8 chemotherapeutic agents and tumour growth delay was estimated in terms of the number of volume doubling times gained by the treatment. The average response corresponded to a delay of only about 0.5 doubling times, but some tumour lines showed a good response to some agents. Twenty-three out of about 700 treated tumours failed to regrow. Statistical analysis showed no consistent difference in sensitivity among the tumour lines, but melphalan, 5-fluorouracil and hexamethylmelamine stood out as the most effective agents. A study of two-drug combinations showed that their order of administration had little effect on response. Only 4 of the patients who donated the xenografts were treated with chemotherapy, but among these there was some evidence that response in the xenografts correlated with response in the patient.

[Studies on non-metal materials in oral surgery].

Two non-metallic implant materials are introduced and the indications for their use are described. A maxillo-facial indication exists in the reconstruction of the temporo-mandibular joint and the orbital floor. We find dental-surgical indications for the Aluminium-Oxide ceramics as well as for implants in the replacement of single tooth gaps and unilateral as well as bilateral shortened dental rows. The indication in the toothless jaw is the most unfavourable and should be restricted to replacement of both canine teeth.

Amino acid sequence of the basic trypsin inhibitor from bovine splenic capsule. A new fluorescent reagent for protein sequence analysis.

The amino acid sequences of tryptic peptides from the performic acid-oxidized trypsin inhibitor were determined. The degradation was performed with a new reagent, 3-isothiocyanato-4-methoxy-4'-nitrostilbene, and compared with the dansyl-Edman technique. The amino acid sequence of the trypsin inhibitor from bovine splenic capsule was found to be the same as that of the basic trypsin inhibitor from bovine pancreas, established by Kress & Laskowski (J. Biol. Chem. 1967, 242, 4925-4928).

Amino acid sequence of human muscle glyceraldehyde-3-phosphate dehydrogenase. Isolation and amino acid sequences of tryptic peptides.

1. The amino acid compostion, N- and C-terminal amino acid sequences, and the subunit molecular weight of glyceraldehyde phosphate dehydrogenase from human muscle, were determined. The obtained results and the maps of tryptic peptides suggest that the enzyme is composed of four identical or very similar polypeptide chains. 2. From the tryptic digest of performic acid-oxidized enzyme, 32 peptides were isolated. The amino acid sequence analysis showed a high degree of homology with the corresponding tryptic peptides of the dehydrogenase from pig muscle, with 9 replacements and probably two additional amino acids in the examined sequences of the human muscle enzyme.

Surgical complications of implanting pacemakers.

An analysis of the complications of implanting pacemakers at St George's Hospital between 1967 and 1973 highlights the difficulties of implanting foreign bodies. During this 7 years 1543 pacemakers were implanted in 779 adults. In 91 patients there were 118 implants with surgical complications, most of them due to a pressure necrosis or local infection; 3 septicaemias occurred. The various complications are classified and related to technique of implantation. Methods of management of the complications are presented. As an Appendix the operative techniques of first and subsequent implantations are outlined.
