Histopathological studies on experimental cryptococcosis in nude mice.

Cryptococcosis in nude (nu/nu) mice was examined histopathologically. In addition, effects of carrageenan and lymph node cell transfer against Cryptococcus infection were investigated. As controls, heterozygous littermates (nu/+mice) and mice of strain ddy (ddy mice) were employed. Each mouse was inoculated intravenously with 10(5) yeast cells of Cryptococcus neoformans suspended in 0.2 ml phosphate-buffered saline. Two mice out of each group were sacrificed at appropriate intervals up to 25 days after inoculation and histopathological sections were prepared from them. They were stained with H & E and by PAS method. Histopathological characteristic in the brain was cyst formation with no cellular response. The brain was more severely in the nu/nu mice than in either the nu/+ ddy mice. In the liver, there was a major difference in histopathological findings between the nu/nu and either of the other groups of mouse. In the nu/nu mice, cyst formation with no cellular response was induced, and on the contrary granuloma formation in the nu/+ and ddy mice. However, the granuloma formation was inhibited in the livers of the nu/+ and ddy mice by administration of carrageenan, and induced in the nu/nu mice be cell transfer. In the spleen and lymph nodes, lesions were severer in nu/nu mice than in either the nu/+ or ddy mice. These results suggested that the fungus' invasiveness of mice was strongly influenced by T-cell dependent mechanism.

Potentiating mechanism of bradykinin action on smooth muscle by sulfhydryl compounds.

(4R)-3-[(2S)-3-Mercapto-2-methylpropanoyl]-4-thiazolidinecarboxylic acid (YS-980), a potent angiotensin I (AI) converting enzyme inhibitor, inhibited the contractile response of isolated guinea-pig ileum to AI but not to AII, while it potentiated the response to bradykinin. Other sulfhydryl compuonds also produced inhibition of AI action, and the effects were closely correlated with their potentiating activities against bradykinin action. These results suggest that it is mainly inhibition of the enzyme in the tissue which participates in the mechanism of potentiation of kinin action by YS-980.

Real time observation of left atrial myxoma with high speed B mode echocardiography.

Four patients with left atrial myxoma were studied using high speed B mode echocardiography with a mechanical sector scanning system. In all four patients, a mass of echoes was recognized in the left atrial chamber during systole. Its lower portion protruded into the left ventricle through the mitral orifice in diastole. The degree of protrusion of the tumor echo varied. An analysis of three cross sections, long axis of left ventricle, sagittal and horizontal axes, showed that the tumor moved anterosuperiorly to posteroinferiorly in the left-sided heart chambers. Its movement was clearly observed in real time. The echo pattern from the tumor had an appearance of mottling, not layering. The myxomas were successfully removed from the left atrial chambers by surgical intervention in all cases.

Clinical study on the right-sided Austin Flint murmur using intracardiac phonocardiography.

Right heart catheterization was carried out on 14 patients with pulmonic regurgitation using intracardiac phonocardiography. All the patients showed pulmonic regurgitant murmur in the right ventricular outflow tract. In addition, seven out of the 14 patients showed mid-diastolic and presystolic murmurs maximally in the outflow tract of the right ventricle. Furthermore, inspiration increased the loudness of these diastolic murmurs in four patients. These findings were compatible with those of right-sided Austin Flint murmur due to functional tricuspid stenosis in pulmonic incompetence. Ten out of the 14 patients had pulmonary hypertension and all the subjects with a rightsided Austin Flint murmur showed elevated pulmonary arterial pressure. Thus, pulmonic regurgitation with pulmonary hypertension is thought to be closely related to the right-sided Austin Flint murmur and the turbulence resulting from antegrade flow across a closing tricuspid valve may be responsible for the genesis of the murmur.

Ocular manifestations of the 1976 rubella epidemic in Japan.

Of 79 cases of rubella during the 1976--1977 rubella epidemic in Japan, 56 (70.9%) had mild conjunctivitis. Six (7.6%) showed epithelial keratitis, but the epidemically estimated incidence of keratitis was about 2% (two cases with rubella keratitis per 75 cases of rubella). No evidence of disease of the anterior chamber or the fundus was observed. Characteristic features of keratitis in juvenile rubella were fine punctate epithelial opacities mainly disseminated in the central area of the cornea. The punctate lesions were about 0.1 mm in diameter on an average, but varied in number from a few to about 100. In many cases, keratitis developed about one week after skin rash and disappeared within one week with no sequelae.

[Effect of 1 alpha-hydroxycholecalciferol on cellular calcium metabolism of kidney cortex (author's transl)].

Rats were provided a normal laboratory diet or a low Ca.vitamin D-deficient diet. After the administration of 1 alpha-hydroxycholecalciferol, mitochondria, microsomes and slices were prepared from kidney cortex of both control and treated rats. When 1 alpha-hydroxycholecalciferol was given to normal and low Ca.vitamin D-deficient rats, Ca accumulation in mitochondria was stimulated during 30 minutes and the high calcium content was maintained at the subsequent incubation. There was a significant decrease of mitochondrial Ca2+-ATPase and Mg2+-ATPase activities with low Ca.vitamin D depletion, but both enzyme activities were restored by 1 alpha-hydroxycholecalciferol treatment of the depleted rats. Ca2+-ATPase and Mg2+-ATPase activities of microsomes were not altered with the administration of 1 alha-hydroxycholecalciferol. In contrast to results of mitochondrial Ca transport, changes in Ca influx and efflux of slices were not significant in response to the treatment of low Ca.vitamin D-deficient rats with 1 alpha-hydroxycholecalciferol. The results of the present study suggest that 1 alpha-hydroxycholecalciferol plays a role in the process of Ca accumulation and ATP hydrolysis of mitochondria in kidney cortex.

Purification and properties of angiotensin I-converting enzyme in human lung and its role on the metabolism of vasoactive peptides in pulmonary circulation.

Purification of angiotensin I-converting enzyme from human lung and characteristics of the enzyme was studied. Experimental pneumonitis was produced in rabbits and the change of the activity of angiotensin I-converting enzyme was studied in purpose to clarify the role of this enzyme in the metabolism of vasoactive peptides in the lung. Purification was performed using trypsin treatment, acid treatment, DE52-cellulose column chromatography, hydroxyapatite chromatography and Sephadex G-200 gel filtration. The enzyme after final step showed a single band on disc gel electrophoresis. Experimental pneumonitis was produced by injection of Complete Freund's adjuvant (acute pneumonitis) and of N-nitroso-N-methylurethane (chronic pneumonitis). In acute experiment, angiotensin I-converting enzyme activity in pulmonary tissue and in plasma was significantly decreased. In perfusion experiment, conversion of angiotensin I to angiotensin II and inactivation of bradykinin were also significantly decreased. In case of decreased activity of angiotensin I-converting enzyme in the lung, less angiotensin II will be released into systemic circulation and bradykinin will pass through the pulmonary circulation into systemic circulation, thus this may result in the decrease of systemic blood pressure.

Angiotensin I-converting enzyme in human urine.

It was demonstrated that angiotensin I-converting enzyme was excreted in human urine. The mean activity of the enzyme in normal urine was found to be 0.38 +/- 0.04 (S.E.M.) units/day (n = 18) and the enzymic activity correlated well with the concentration of the excreted sodium (r = 0.76, p less than 0.005). Urinary angiotensin I-converting enzyme was partially purified. Three different molecular weights of enzyme (greater than 400 000, 290 000 and 140 000) were demonstrated by Sephadex G-200 gel filtration. The enzymic properties of these three enzymes were identical with those of angiotensin I-converting enzyme from human lung with regard to inhibitory effects (bradykinin potentiator c and Arg-Pro-Pro), Cl- dependency, pH optimum and KM value.

Potentiative effects of sulfhydryl compounds on carrageenin-induced oedema in rats and relationship to their potencies as inhibitors of angiostin-coverting enzyme in vivo.

Carrageenin-induced oedema in rats was potentiated by oral administration of (4R)-3-[(2S)-3-mercapto-2-methylpropanoyl]-4-thiazolidinecarboxylic acid (SA291) and related sulfhydryl compounds, and the effect was closely correlated with their potencies as inhibitors of angiotensin-converting enzyme in vivo.
