Antimetastatic activity of cell-wall skeleton of Propionibacterium acnes C7 (P. acnes-CWS) in C57BL/6 mice varied depending on the injection route. The kinetics of antimetastatic effect against Lewis lung carcinoma (3LL) revealed that the sooner intratumoral (it) injection of P. acnes-CWS was carried out, the better the result. However, intravenous (iv) injection of P. acnes-CWS produced the best result if P. acnes-CWS was injected at about the time when metastases began to develop in the lungs. Pretreatment with intrafootpad (ifp) injection of P. acnes-CWS inhibited primary tumor growth and subsequent pulmonary metastases especially when given 7 days before tumor inoculation into the same footpad, but tended to enhance artificial pulmonary metastases when given 7 days before iv injection of tumor cells. In contrast, pretreatment with iv injection of P. acnes-CWS enhanced spontaneous pulmonary metastases especially when given 7 days before ifp inoculation of tumor cells, but inhibited artificial pulmonary metastases when given one day or 7 days before iv injection of tumor cells. These results suggest the difficulty of treatment of tumor metastases with immunological adjuvants. In T-cell-deprived mice, it injection of P. acnes-CWS showed no antimetastatic effect against 3LL, but iv injection was still effective. This indicates that T-cells are required for the antimetastatic effect of it injection of P. acnes-CWS, but iv injection of P. acnes-CWS is able to inhibit pulmonary metastases in T-cell-deprived mice.

Changes in molecular forms of hepatic cytosolic glutathione S-transferases (GST) during rat chemical hepatocarcinogenesis were investigated. GST activities toward 1-chloro-2,4-dinitrobenzene and 1,2-dichloro-4-nitrobenzene increased with the increased area of gamma-glutamyltranspeptidase-positive foci and hyperplastic nodules induced by diethylnitrosamine followed by 2-acetylaminofluorene plus hepatectomy. Among GSTs with high activities toward 1,2-dichloro-4-nitrobenzene, which were separated by carboxymethyl Sephadex column chromatography, the activity of GST-A ( YbYb ) markedly increased with increased activity towards 1,2-dichloro-4-nitrobenzene in livers bearing foci and nodules and in isolated nodules and hepatomas, while activities of GST-C ( YbYb ') and -D (Yb'Yb') changed little. It was demonstrated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and two-dimensional gel electrophoresis that Yb as well as Ya , a subunit of ligandin ( YaYa ) and GST-B ( YaYc ), increased in livers bearing foci and nodules, while Yc as well as Yb' changed little. A new placental GST form (GST-P), which has a subunit molecular weight of 21,500 or 26,000, according to the marker proteins used, and neutral pls of 6.8 and 6.3, is immunologically different from any form of basic GSTs and is very low in normal liver; also, it was markedly induced in livers bearing foci and nodules and in well-differentiated hepatomas but not by short-term administration of drugs such as 2-acetylaminofluorene, in contrast to GST-A and ligandin. These results indicate that GST-A and more especially GST-P could be new preneoplastic marker enzymes for chemical hepatocarcinogenesis.
Benign nonepithelial tumor of the urinary bladder is very rare. We experienced a case of a 33-year-old woman who complained of total urinary retention. Vaginal examination revealed a hen's egg sized retrovesical tumor. IVP revealed a filling defect on the cystogram. Cystoscopy revealed protrusion of left side of the bladder neck and intact mucosa. Transvaginal needle biopsy of the tumor was done, and pathohistological diagnosis of the tumor was done, and pathohistological diagnosis of the specimen was leiomyoma. The tumor was intramural type and was resected. It was 5.5 X 5 X 5 cm in size, weighed 80 g, and pathohistological diagnosis was leiomyoma of the urinary bladder. 57 cases of leiomyoma of the urinary bladder including this case have been reported in Japan. No special method of diagnosing leiomyoma of the urinary bladder exists, but in some cases, needle biopsy is very effective.
The temporal modulation transfer function (temporal MTF) was measured in normal controls and in patients with retinal diseases, optic neuritis and amblyopia. The temporal MTF of controls showed "band-pass" filter properties, having a peak sensitivity at about 15 Hz at 1,000 trolands of mean retinal illuminance. As the retinal illuminance was reduced, the sensitivity decreased, first at high temporal frequencies, then the peak sensitivity decreased. Finally at low temporal frequencies the sensitivity decreased and the peak shifted toward the lower frequency. In retinal diseases, abnormal temporal MTF was detected, and the curves were very similar to those of normal controls when retinal illuminance was reduced. These patterns were not specific to any retinal diseases, and the severity of the condition seemed to determine the pattern. In optic neuritis, an attenuated sensitivity was detected in all frequency ranges; however, the degree of attenuation was more severe than that found in retinal diseases even when the visual acuity was similar. At the convalescent stage, the low frequency sensitivity was restored first, and high frequency loss was observed even when the visual acuity was recovered to 1.0. Two amblyopic patients showed an attenuated sensitivity in the high and intermediate frequency ranges, and in one case sensitivity was attenuated in all frequencies. Three other patients showed normal sensitivity. By occlusion of the nonamblyopic eye, recovery of the sensitivity was observed in one patient whose visual acuity improved.

The activity of ornithine decarboxylase, the rate-limiting enzyme in polyamine synthesis, increases as granulosa cells proliferate in developing follicles. Both luteinizing hormone and prostaglandins stimulate ornithine decarboxylase activity. Here, we sought to determine the relative contributions of both trophic stimuli to ornithine decarboxylase activity in the preovulatory rabbit ovary. Baseline ovarian ornithine decarboxylase activity, determined by measuring the release of CO2 from (1-14C)-ornithine, was 13.4 +/- 1.27 (mean +/- SD) pmol of carbon dioxide per hour per milligram of protein. Treatment with ibuprofen, a prostaglandin synthetase inhibitor, led to a significant (p less than 0.05) decrease in the baseline ovarian ornithine decarboxylase activity (4.7 +/- 0.29 pmol of carbon dioxide per hour per milligram of protein). Administration of human chorionic gonadotropin (hCG) (50 IU/kg intramuscularly) to adult rabbits (2.5 to 3.5 kg) elicited a 1,200% elevation of ovarian ornithine decarboxylase activity 5 hours after injection; there was a return to baseline by 11 hours after injection. Stimulation with human chorionic gonadotropin led to ovulation in 22.2%, 25%, and 60% of rabbits at 7, 9, and 11 hours after treatment, respectively. Single-dose ibuprofen treatment (5 mg/kg intramuscularly), 7 hours after human chorionic gonadotropin administration inhibited ovulation and elevated ovarian ornithine decarboxylase activity. These results indicate that ibuprofen effectively inhibits ovulation in hCG-stimulated rabbit ovaries in the presence of a significant (p less than 0.001) elevation in ovarian ornithine decarboxylase activity. Thus, different intracellular mechanisms are involved in the prostaglandin modulation of basal and hCG-stimulated cells during the course of preovulatory follicle maturation.
We did this experiment to clarify the mechanism of granuloma formation and the killing functions of granuloma in nude mice against Blastomyces dermatitidis and Paracoccidioides brasiliensis infections. B. dermatitidis A-295 and P. brasiliensis B-1183 were the cultures used. Congenitally athymic nude (nu/nu) mice and their heterozygous (nu/+) littermates of BALB/c background were the test animals. From culture A-295, 0.1% and 1% cell suspensions (wet weight) were prepared and from culture B-1183 0.2% and 2% cells suspensions were prepared. Ten nu/+ and 10 nu/nu mice were allotted to each of four cell suspensions. For experimental blastomycosis each mouse was inoculated intravenously with 0.2 ml of the cell suspension of A-295 and for experimental paracoccidioidomycosis, with 0.15 ml of the cell suspension of B-1183. Two mice from each of the four groups were killed at 5, 8, 12, 18 and 25 days after inoculation, and histopathologic sections, stained with H&E or by PAS, were prepared from various internal organs. In the nu/nu mice inoculated with B. dermatitidis A-295 granuloma was formed in the brain tissue after the 12th day. However, mononuclear cells, which formed the granuloma, did not kill the fungal cells, and the fungal cells continued to multiply in the granuloma. On the other hand, in the heart, kidney and fat tissue, their histopathological findings after the 18th day were clumps of fungal cells with slight cell reactions. In these organs the exertion of cell-mediated immunity was necessary for granuloma formation against the fungal infection. In the nu/nu mice infected with P. brasiliensis B-1183, granuloma appeared in the brain and kidney after the 18th day and fungal cells continued to multiply within the granuloma as well as in those inoculated with culture A-295. These results show that the exertion of cell-mediated immunity plays an important role in the mouse's defense mechanisms against these fungal infections. We assume that the defense mechanisms of immunocompetent mice against B. dermatitidis or P. brasiliensis infection consist chiefly of two steps: in the first step phagocytosis by PMNs occurs and in the second step cell-mediated immunity enters into play.
A 12-year-old boy with T-cell ALL was found to have occult papillary thyroid carcinoma at autopsy. This patient was treated with chemotherapy but no radiotherapy was utilized. Family history was not contributory. Because of short latent period (14 months) and no history of radiotherapy, an intrinsic factor might have played a major role in developing this second malignancy. Currently 11 solid tumors have been reported as second malignant neoplasms after ALL in childhood. Four (including this case) of 11 were thyroid carcinoma. Two of them did not receive any radiotherapy. Special interrelation between ALL and thyroid carcinoma may be considered. And this interrelation should be taken into account in following the patients with ALL in the future.
This paper deals mainly with the conidium ontogenesis and phylogenesis of black yeasts such as E. jeanselmei, E. gougerotii, E. dermatitidis and E. spinifera. The conidium ontogenesis of E. jeanselmei, E. gougerotii and E. dermatitidis was almost the same. One to five annellated tips were observed through a scanning microscope at the apices of conidiogenous cells, which were bottle- or jar-shaped. Annellations on the tips looked like fringes and the conidiogenous cells of these three species were annellides. Annellated projections occurred on hyphae and annelloconidia were also produced from them. Occasionally, secondary annellides occurred from primary ones. They looked like moniliform hyphae. Daughter conidia sometimes budded directly from mother cells. The shapes and sizes of the conidia of these species were very similar to each other. The conidium ontogenesis of E. spinifera was annellidic as well. However, a single annellated tip usually occurred on an annellide. The annellated tips of the fungus were long and more than 20 annellations were observed on their walls. The conidiogenesis of the four species of Exophiala is only annellidic. There were no differences in the biological examinations except KNO3 assimilation among these four species. The growth of E. jeanselmei and E. gougerotii was poor at 37 degrees C. The GC contents of E. jeanselmei 1171, E. gougerotii B-1800, E. dermatitidis MM-7 and E. spinifera DU-3342 were 54.6, 54.6, 56.6 and 59.7%, respectively.

The syndrome of inappropriate secretion of antidiuretic hormone is a rare but well-recognized neurotoxic side effect of vincristine therapy. The first neuropathological report of a case is presented. A 6-month-old boy with skin leukemia developed inappropriate secretion of antidiuretic hormone caused by vincristine. Postmortem examination revealed axonal spheroids in the ansa lenticularis and the area surrounded by the substantia innominata, amygdala and supraoptic nucleus. The lesion was confined to that area and the neurosecretory neurons were intact with well preserved neurophysin. The pathological findings suggest that these fibers play a role in the development of inappropriate secretion of antidiuretic hormone caused by vincristine therapy.
Oxidized regenerated cellulose (ORC) is widely used to cover the denuded mesothelium and to promote hemostasis. We evaluated ORC in a rabbit model to assess the potential for adhesion reduction after uterine trauma and intestinal anastomosis. After surgery, a second laparotomy was done on postoperative day 7 for macroscopic evaluation of adhesion formation. The traumatized uterine horns wrapped in ORC had more numerous and higher grade adhesions compared with the non-surgical left, intact horns. However, when compared to the contralateral uterine horn which underwent trauma without ORC, a significant decrease in the grade of adhesion formation was noted. Rabbits undergoing intestinal anastomosis had the most pronounced degree of adhesion formation. Typically, the area of healing was encapsulated in a fibrin clot with adhesions attached to adjacent viscera. When ORC remained between the lesion and adjacent organs during mesothelial regeneration, adhesion formation decreased. However, the soft, gelatinous mass of ORC was also an effective nidus for clot formation. Where post- and intra-operative ooze produced a fibrin clot in the ORC, dense adhesions formed.
Proteins central to normal wound repair, including collagen and proteoglycans, were extracted during postoperative mesothelial regeneration, then the quantitation was correlated to macroscopic observations of normal peritoneal reepithelialization and/or postoperative adhesion formation. Sixty-three New Zealand white female rabbits of reproductive age were prospectively assigned to either Group A, untreated control; Group B, which received intramuscular injections of ibuprofen, 70 mg/kg per injection (immediately and 6 hr after surgery); or Group C, which received 5 intramuscular injections of ibuprofen (4 hr before surgery, and immediately, 6, 12, and 18 hr after surgery). The right uterine horn underwent one of three standardized surgical traumas: (1) abrasion of the peritoneal surface with a scalpel until punctate bleeding developed, (2) ischemia of the uterine horn by removal of the collateral blood supply (devascularization), (3) crushing of the uterine horn by cross clamping for 3 min with a Kelley hemostat. Thereafter, 10 microCi of C-14-labeled glucosamine and 10 microCi of C-14-labeled proline were injected into the marginal ear vein of each rabbit. All rabbits underwent a laparotomy on the fifth postoperative day for evaluation of adhesion formation and tissue biopsy for protein extraction. No reduction in adhesion formation was found using a 2-dose postoperative treatment regimen. However, using a 70 mg/kg X 5-doses regimen in the immediate perioperative interval, a significant reduction in both adhesion formation and severe adhesion formation (both P less than 0.025) were found following standardized surgical injury. The extent of adhesion formation was correlated with the extractable glycosaminoglycan and collagen concentrations. As determined by recovered glucosamine and proline, a positive correlation was apparent between the severity of adhesion grade and formation of new glycosaminoglycans or collagens. Thus, ibuprofen appears to inhibit adhesion formation through suppression of fibroproliferative inflammation.
