Localized prevention of postsurgical adhesion formation and reformation with oxidized regenerated cellulose.

We evaluated oxidized regenerated cellulose (ORC) in experimental rabbit and dog models to assess its potential for reducing or preventing adhesion formation and reformation. Evaluation was performed in the following anatomical sites: uterine horn trauma; small bowel resection and anastomosis; parietal peritoneum excision; and adhesion reformation in the uterine horn. Ten to 14 days after ORC treatment, adhesion formation was macroscopically and microscopically evaluated. Results demonstrate that ORC produced graded reduction in adhesion formation and significantly prevented adhesion reformation.

Fungi in bathwater and sludge of bathroom drainpipes. 1. Frequent isolation of Exophiala species.

Samples of bathwater from 14 homes and 22 public bathhouses and sludge in drainpipes from 19 household bathrooms were plated out onto potato dextrose agar supplemented with chloramphenicol. Several media were used to study colony morphology of the isolates and the thermotolerance and alkaline tolerance of each isolate were examined. Eleven sludge samples produced 12 isolates of Exophiala jeanselmei, 2 of E. dermatitidis and 1 of E. moniliae. Five household bathwater samples produced 2 isolates of E. jeanselmei, 4 of E. dermatitidis and 1 of E. alcalophila. One isolate of E. jeanselmei, 2 of E. dermatitidis, 3 of E. moniliae and 2 of unidentified Exophiala species were recovered from 6 samples of the bathwater dissolving 'Chinese medicine' in the bathtubs of public bathhouses. One isolate of E. jeanselmei was recovered from the 15 samples of bathwater from public bathhouses. Bathwater and sludge in bathroom drainpipes may be an important habitat of Exophiala species.

Copurification and separation of beta-galactosidase and sialidase from porcine testis.

A soluble sialidase was copurified apparently as an enzyme complex with acid beta-galactosidase from porcine testis. The sialidase exhibited its maximum activity at acidic pH. It was efficiently active towards 4-methylumbelliferyl-alpha-D-N-acetyl-neuraminic acid and sialyllactose, relatively inactive towards glycoproteins, and had little activity towards glycolipids. The complex could be separated by sucrose gradient centrifugation or isoelectric focusing. The separated enzymes had molecular weights about 600,000 for beta-galactosidase and more than about 1,000,000 for sialidase by Sepharose 4B gel filtration. SDS-polyacrylamide gel electrophoresis of the beta-galactosidase showed three protein bands with molecular weights of 63,000, 31,000 and 20,000.

Effect of multiporous microspheres derived from chitin and partially deacetylated chitin on the activation of mouse peritoneal macrophages.

Multiporous microspheres were prepared from 80% deacetylated chitin (DAC-80) and chitin, and their effects on the activation of murine peritoneal macrophages in vivo and on the production of monokines such as colony-stimulating factor (CSF) and interleukin 1 (IL-1) were examined. Multiporous DAC-80 microspheres of mean diameter 2.5 microns [MS-DAC-80(2.5)] enhanced the cytolytic activity of peritoneal macrophages and the production of CSF in vitro by macrophages, spleen cells and bone marrow cells, and in vivo. MS-DAC-80(2.5) also stimulated the production of IL-1 by both resident and thioglycolate-induced peritoneal macrophages. Multiporous chitin microspheres [MS-chitin(2.5)] showed no effect on the activation of peritoneal macrophages in vivo and on the production of IL-1 in vitro, but slightly enhanced the production of CSF in serum in vivo.

Stimulation of non-specific host resistance against Sendai virus and Escherichia coli infections by chitin derivatives in mice.

The efficacy of chitin derivatives on non-specific host resistance to Sendai virus and Escherichia coli infections was studied in mice. Seventy percent deacetylated chitin (DAC-70) and N-trimethylated DAC-70 [DAC-70(Me)3] showed protective activity against Sendai virus infection; however, carboxymethyl-chitin (CM-chitin) did not. DAC-70 also showed protective activity against E. coli infection.

Phytosphingosine is a characteristic component of the glycolipids in the vertebrate intestine.

Sphingoids in the intestinal lipids of an agnatha, a chondrichthyes, two osteichthyes, three amphibia, three reptiles and two avian species were analyzed by reversed phase high performance liquid chromatography. The glycolipid fraction of all the samples studied contained 4-D-hydroxysphinganine as the major component together with sphingosine and sphinganine. While the trihydroxy base was not found in their sphingomyelin fraction. The trihydroxy base was considered to be a characteristic component of the intestinal glycolipids for the vertebrates in general. Its concentration in the intestinal tissue had little correlation with the food habitat of the animals.

A case of laparoscopically detected cholangiocarcinoma of a few millimeters in size, with portal fibrosis.

A 54-year-old male with no subjective complaints was admitted to our hospital with fatty liver and low echoic lesion of 3 X 4 cm, thereafter revealed to be a hemangioma. A white lesion of few millimeters in size was detected in the left lobe of the liver at laparoscopic examination, and histologically diagnosed in an aimed biopsy as cholangiocarcinoma. Another 5 lesions (three white spots, a white longitudinal protrusion and a white lesion with a lacy pattern) suggestive of malignancy were also selectively biopsied, but all proved histologically negative. Microwave coagulation was applied to all biopsied sites as anti-cancer therapy.

Evaluation of the effect of metoprolol on energy metabolism in the ischaemic myocardium in relation to regional myocardial blood flow.

To evaluate the effect of beta blockade on energy metabolism in ischaemic myocardium metoprolol tartrate in doses of 0.2 or 0.5 mg.kg-1 or saline was injected into 24 anaesthetised dogs 20 min after coronary ligation. Regional myocardial blood flow was measured by the hydrogen gas clearance method, and the heart rate and aortic pressure were recorded. After 60 min coronary occlusion myocardial biopsy specimens were removed from five areas where myocardial blood flow had been recorded to determine their content of adenosine-5'-triphosphate (ATP). Respiratory function and acyl-coenzyme A contents of mitochondria isolated from ischaemic and non-ischaemic myocardium were also measured. In the control group the ATP content in severely ischaemic myocardium where myocardial blood flow was less than 20 ml.min-1.100 g-1 was decreased to 27.5% of that in the non-ischaemic area, whereas the administration of metoprolol 0.5 mg.kg-1 maintained ATP content at 46.3%. In ischaemic myocardium metoprolol inhibited the accumulation of long chain acyl-coenzyme A in mitochondria and significantly preserved their respiratory function. In contrast, metoprolol did not affect myocardial blood flow in myocardium where flow was less than 60 ml.min-1.100 g-1. Heart rate was decreased by metoprolol in proportion to the dose, whereas mean aortic pressure was not changed. These results suggest that beta blockade had beneficial effects on the energy metabolism of ischaemic myocardium and that such effects might be due not only to a reduction in cardiac work but also to a reduction in the vicious circle of fatty acid metabolism in ischaemic myocardium.

A Lewis X hapten is the major glycolipid of Japanese quail intestine.

A fucolipid isolated from Japanese quail intestine was identified as beta-galactosyl-1,4-(alpha-fucosyl-1,3-) beta-N-acetylglucosaminyl-1,3-beta-galactosyl-1,4-beta-glucosyl-1,1-cera mide, a glycolipid which exhibits X-hapten activity. Analysis of the tissue at various embryonic stages showed that the fucolipid is characteristically present at later stages of organogenesis.

Effects of an angiotensin-converting enzyme (ACE) inhibitor, SA446, on tissue ACE activity in normotensive, spontaneously hypertensive, and renal hypertensive rats.

Effects of an angiotensin-converting enzyme (ACE) inhibitor, SA446, on the renin-angiotensin system, particularly on tissue ACE activity, were studied in Wistar-Kyoto normotensive rats (WKY), spontaneously hypertensive rats (SHR), and two-kidney, one-clip renal hypertensive rats (RHR) by repeated oral administration for 7 days. SA446 (45 mg/kg/day p.o.) inhibited ACE activity in the lung, brain, kidney, heart, and whole blood throughout the administration period in WKY, but showed a slight hypotensive action and no inhibition of aorta ACE activity. On the other hand, SA446 had an apparent hypotensive action at the same dose in SHR and inhibited ACE activity significantly in the aorta as well as the kidney and whole blood during the administration period. Furthermore, enzyme activity in the aorta, kidney, heart, and whole blood was also inhibited at a hypotensive dose of SA446 (10 mg/kg/day p.o.) in RHR. The inhibition in whole blood and kidney was almost complete, and the inhibition in the aorta was greater on day 7 than on day 1. The maximum decrease of blood pressure was correlated with the maximum inhibition in aorta ACE activity, but not in brain, lung, or heart ACE activity. In addition, a good positive correlation was observed between the basal blood pressure and the basal aorta ACE activity in WKY, SHR, and RHR, although there was no correlation in the brain, lung, kidney, heart, or whole blood. These results suggest that the antihypertensive action of SA446 by repeated administration may be due to inhibition of arterial ACE activity in addition to inhibition of plasma and kidney ACE activity.(ABSTRACT TRUNCATED AT 250 WORDS)

Endometrial cysts of the ovary: MR imaging.

Magnetic resonance (MR) images of 15 cases of ovarian endometrial cyst were reviewed. With a high-field-strength (1.5-T) MR imager, T1- and T2-weighted images were obtained in axial and sagittal planes, with 5-mm section thickness. MR findings highly suggestive of an endometrial cyst included adhesions to the surrounding organs (e.g., loss of clear margin of the uterine body and tethered appearance of the rectum); a distinct low-intensity zone surrounding a cyst loculus on both T1- and T2-weighted images produced by a thick fibrous capsule; loculus contents with short T1 and long T2 values, attributed to hemorrhagic fluid; and prominent low intensity (shading) within a loculus on T2-weighted images, the mechanism of which is yet to be determined.

Ovarian cystic teratomas: MR imaging.

Magnetic resonance (MR) imaging characteristics of ovarian cystic teratomas were evaluated in 18 patients with 23 surgically proved tumors. The fat within the tumor demonstrated characteristic intensity similar to that of subcutaneous fat and a pathognomic pattern of chemical shift artifact that helped differentiate ovarian cystic teratomas from endometrial cysts. In addition to fat, MR images of ovarian cystic teratomas had several other characteristics that allowed confident diagnosis, including gravity-dependent layering or floating debris, palm tree-like protrusions or rounded nodules (dermoid plugs), and, occasionally fat-fluid levels and globular calcification. MR images were diagnostic in 20 tumors and were able to depict a tumor measuring 2 cm in diameter. MR imaging appears to be valuable in problematic cases in which other modalities are not diagnostic.

Vaginal agenesis: classification by MR imaging.

Vaginal agenesis was studied by magnetic resonance (MR) imaging lin a 15-year-old girl, a 19-year-old woman, and a 21-year-old woman. In each case MR studies demonstrated the presence or absence of a normal cervix and functioning endometrium. This information was useful in planning surgical management. MR imaging assisted in the classification of the abnormality by noninvasively demonstrating the details of pelvic anatomy-and the uterine morphologic characteristics.

Hepatocellular carcinoma: MR imaging.

Sixty patients with hepatocellular carcinoma (HCC) were studied with computed tomography (CT) and magnetic resonance (MR) imaging at 1.5 T. MR imaging was equivalent to CT in detection of HCC. MR imaging was superior to CT in demonstrating the details of tumors, especially pseudocapsules. In 58 cases, main tumors were detected with MR imaging. On spin-echo (SE) 600/25 (repetition time msec/echo time msec) sequences, tumors were hyperintense in 18 cases, isointense in ten, and hypointense in 30. On SE 2,000/60 sequences, all but two tumors had high signal intensity. Pseudocapsules, intratumoral septa, daughter nodules, and tumor thrombi, which are important characteristics of HCC, were demonstrated in 22, three, six, and six cases, respectively, on MR imaging. MR imaging is useful for characterizing the internal architecture of HCC.
