Expression and specificity of FcIgG receptor sites on neoplastic lymphocytes.

We investigated the ability of an FcIgG receptor marker to discriminate between subtypes of malignant lymphoproliferative diseases that differed in their clinical presentations. A quantitative radioimmuno assay was established that enabled us to evaluate average receptor densities on a population basis. Surface receptors were first saturated with IgG complexes. The number of membrane associated IgG molecules was subsequently determined with 125I-staphylococcal protein A. Results obtained with this assay on a battery of malignant lymphocytes suggested that the range of receptor densities of malignant B and T cells might overlap each other but would correlate with the degree of tumor cell differentiation and the clinical stage of the underlying disease. This behavior limits the use of this marker in the characterization of the derivation of malignant lymphocytes; these findings, however, may be useful in the prognostic classification of lymphomas of known origin.

The retrovirus particles in human myeloma cells RPMI8226: morphological, biochemical, immunological and infective transmission studies.

The retrovirus designated RPMI8226V (isolated from human myeloma cells RPMI8226) has been characterized with respect to its morphological, biochemical and immunological properties as well as its propagation in various animal and human cells. The myeloma cells RPMI8226 produce intracytoplasmatic A-type particles and extracellular particles. The extracellular particles have been classified as immature particles with translucent core center, typical mammalian C-type virus particles and C-type particles with intermediate membrane. However, the budded particles in secondarily infected human neoplastic cells contained complete doughnut-shaped nucleoids. This type of budding is rather characteristic for B-type particles. The 3H-uridine labeled RPMI8226 viral particles have a buoyant density 1.17 g/ml in sucrose gradient containing high molecular weight RNA and the distribution of viral structural proteins in SDS-PAGE is characteristic for oncornaviruses. The internal structural proteins according to MW are ranged from 13 000 to 30 000 daltons. The virus contains a magnesium-dependent reverse transcriptase. The cellular homogenate and viral concentrate from RPMI8226 cultures do not react with antibodies against ALSV, MuLV, FeLV, RD114, MP-MV and SiSLV. The only reaction was scored with anti BLV antibodies. However, anti BLV serum inhibiting the reverse transcriptase activity of BLV to 60% does not cross-react with the reverse transcriptase of RPMI8226V. In contrast to BLV concentrates, neither XC nor KC cells show syncytia formation by RPMI8226V. The RPMI8226V replication is restricted to human tumor and normal human glia-like cells. The possible origin of the virus is discussed.

Surface glycoprotein patterns of normal and malignant human lymphoid cells. I. T cells T blasts and leukemic T cell lines.

Exposed surface glycoproteins of resting and in vitro activated human T lymphocytes and leukemic T-cell lines were labelled by the galactose oxidase-tritiated sodium borohydride method. The labelled glycoproteins were separated by polyacrylamide slab gel electrophoresis and visualized by autoradiography. The basic glycoprotein patterns of the T lymphocytes and T blasts were found also in the leukemic T cells. The glycoprotein pattern of the T-cell lines was easily distinguishable from that of other hematopoietic cell lines. The findings suggest that: (1) surface glycoprotein analysis might be useful for the identification of cell lines and for the differential diagnosis of hematopoietic malignancies; and (2) cells of cultured T lines may be used for the identification and preparation of T lymphoid differentiation antigens and perhaps also tumor-associated surface molecules.

Tumorigenicity of human hematopoietic cell lines in athymic nude mice.

Human hematopoietic cell lines, which had been classified on the basis of studies on clonality, and morphological, chromosomal and functional parameters as lymphoblastoid cell lines (LCL) of presumed non-neoplastic origin, and lymphoma, myeloma and leukemia lines of proven malignant origin, were tested for tumorigenic potential on subcutaneous transplantation to nude mice and for capacity to grow in semi-solid medium in vitro. Recently established LCL failed to grow both in nude mice and in agarose. In contrast, some of the LCL which had developed secondary chromosomal alterations during continuous cultivation for periods exceeding several years were tumorigenic and/or had the capacity to form colonies in agarose. Most lymphoma lines formed colonies in agarose and tumors in the mice. One of the two myeloma lines formed subcutaneous tumor which, however, showed no progressive growth. The other myeloma line failed to grow. Both myeloma lines, however, formed colonies in agarose. The myeloid leukemia line was tumorigenic while two of the three tested lymphocytic leukemia lines failed to grow in the mice. All leukemia lines formed colonies in agarose. We conclude from this study that: (1) Of the two types of Epstein-Barr virus containing cell lines [LCL and Burkitt lymphoma (BL) lines], only BL lines were shown to form tumors when inoculated subcutaneously in nude mice and had the capacity to grow in agarose in vitro. This shows that EBV transformation per se does not necessarily render lymphocytes tumorigenic in nude mice. The capacity to form colonies in agarose is not acquired either. (2) Changes of the karyotype and several phenotypic characteristics which occur in the originally diploid LCL during prolonged cultivation in vitro may be accompanied by the acquisition of the potential to grow subcutaneously in nude mice and in agarose in vitro. (3) The inconsistency with regard to the capacity of come of the neoplastic cell lines to grow in nude mice or in agarose seems to underline that neither of the two tests is a reliable criterion for malignancy of human lymphoma, leukemia and myeloma cell lines.

Cytochemical profile of human haematopoietic biopsy cells and derived cell lines.

Twenty-three human haematopoietic cell lines, normal and mitogen stimulated peripheral blood lymphocytes and tumour material from fresh leukaemias, myelomas and lymphomas were investigated with a panel of cytochemical reactions. Normal and mitogen stimulated lymphocytes, non-neoplastic lymphoblastoid cell lines (LCL), lymphoma lines with B-lymphocyte characteristics, chronic lymphocytic leukaemia and fresh lymphocytic lymphomas reacted weakly or negatively with all stains. T-lymphocyte acute leukaemia lines were PAS and alpha-naphtyl acetate esterase positive. Myeloma lines and fresh myelomas were strongly beta-glucoronidase positive. A histiocytic lymphoma cell line was strongly esterase positive with naphtol AS-D acetate esterase inhibited by NaF. The three fresh histiocytic lymphomas, however, reacted as the lymphocytic lymphomas suggesting a lymphoid origin. A myeloid leukaemia line was strongly positive for acid phsophatase. No major disagreement was noted between the reactivity of established neoplastic lines and the corresponding fresh biopsy cells indicating an unaltered qualitative expression of enzyme production after prolonged in vitro culture.

Quantitation of beta2-microglobulin and HLA on the surface of human cells. I. T and B lymphocytes and lymphoblasts.

Relative amounts of beta2-microglobulin (beta2m) and of HLA specificities were analysed on the surface of resting unfractionated peripheral human lymphocytes, enriched B and T cells, and on in-vivo- and in-vitro-stimulated lymphoblasts. Single-cell cytofluorometry and a very sensitive radioimmunoassay were used to determine as closely as possible the absolute amounts of membrane-bound beta2m/HLA antigens. B and T "resting" and "stimulated" lymphoid cells express very similar numbers of beta2m and HLA antigenic determinants, respectively, per unit of surface area when compared with each group, although beta2m was found to exist in two- to three-fold excess of HLA.

Quantitation of beta2-microglobulin and HLA on the surface of human cells. II. In vitro cell lines and their hybrids.

Beta2-microglobulin (beta2m) participates as an integral part in molecules of the major histocompatibility complex (MHC) type. Absence of beta2m makes the residual heavy MHC chain largely inactive as antigen. Striking reductions in the density per unit surface area of beta2m in seven out of nine malignant lymphoid tumour lines in comparison with normal lymphocytes or "immortalized" Epstein-Barr-virus-transformed lymphoblastoid lines were found is this study. This would seemingly represent a specific reduction in the ability of the malignant cells to express actively produced beta2m, since their HLA antigenic determinants were not reduced to the same extent and no indications were obtained suggesting that free beta2m could transfer from one cell to another. However, that beta2m is important in conveying serological specificities of MHC type to cells was shown by fusion of beta2m-negative and beta2m-positive cells, yielding hybrid cells with synergistically increased numbers of detectable, HLA-related determinants. Whether the reduction of beta2m on malignant versus nonmalignant lymphoid cells bears any relevance as to emergence of the malignant clones and resistance to possible anti-tumour reactions would now be an issue for study.

Distortion of non-invasive cardiac pulse curves. A capillary-damped pick-up and a calibration unit forapex cardiograms and other pulse curves.

A calibration unit and a capillary-damped pick-up, of funnel type, intended for apex cardiograms and other non-invasive cardiac pulse curves are presented. The physical properties of a recording system, suitable for non-invasive cardiac diagnostic techniques, are reviewed. In 4 identical recording units consisting of a funnel with and without capillary damping, connected via air transmission to a transducer, the overamplification in the underdamped systems varied between 2-4 and 4-3, and in the capillary-damped systems between 1-1 and 1-2. The capillary damping gave an approximately linear relation between phase shift and frequency in the high frequency range with a calculated delay of about 4ms. The low frequency time constant in the systems varied between 1-9 and 4-6 s, depending on the amplifier setting used, which for sine curves of frequency 0-5 and 2 Hz means an apparent prematurity for their maxima and minima varying between 53 and 1 ms, this being larger with lower frequency time constants and lower frequency curves.

Lysozyme synthesis by established human and murine histiocytic lymphoma cell lines.

A human cell line established in culture from a histiocytic lymphoma patient synthesizes and secretes the monocyte-granulocyte specific enzyme lysozyme. 18 other human cell lines with characteristics of T-lymphocyte, B-lymphocyte, Burkitt's lymphoma, non-Burkitt's lymphoma, myeloma, and bone marrow epithelial cells were not associated with lysozyme. Among murine cell lines, lysozyme was produced by (a) three histiocytic lymphoma or macrophage lines, which mediate antibody-dependent phagocytosis and cytolysis; (b) myelomonocytic leukemia line which also secretes myeloid colony-stimulating factor; and (c) a spontaneous lymphoma and an Abelson leukemia virus-induced lymphoma. Lysozyme-negative lines include another Abelson lymphoma, myelomas, T lymphomas, and mastocytoma.

Establishment and characterization of a human histiocytic lymphoma cell line (U-937).

A human hematopoietic cell line (U-937) with exceptional characteristics was derived from a patient with generalized histiocytic lymphoma. The morphology of the cell line was identical to that of the tumor cells in the pleural effusion from which the line was derived. Since Epstein-Barr virus (EBV) carrying diploid lymphoblastoid cell lines unrelated to the tumor population often become established in vitro from non-Burkitt lymphoma explants, several parameters were studied to discriminate the U-937 from such lines: morphology in vitro, growth characteristics, cytochemistry, surface receptor pattern, Ig production, lysozyme production, beta2-microglobulin production, presence of EBV genome and karyotype. In all these respects U-937 differed from prototype lymphoblastoid cell lines. The histiocytic origin of the cell line was shown by its capacity for lysozyme production and the strong esterase activity (naphtol AS-D acetate esterase inhibited by NaF) of the cells. It is therefore concluded that the U-937 is a neoplastic, histiocytic cell line.
