gp130 and the IL-6 family of cytokines: signaling mechanisms and thrombopoietic activities.

gp130 is a common signal transducing component of the functional receptor complexes for the interleukin (IL)-6 family of cytokines, ie, IL-6, IL-11, leukemia inhibitory factor (LIF), oncostatin M, ciliary neurotrophic factor, and cardiotrophin-1. These cytokines exhibit pleiotropic biological activities in, for instance, immune, hematopoietic, and neural systems, and function in a redundant manner owing to the shared usage of gp130. Thrombopoiesis is regulated by a variety of polypeptide factors including some from the IL-6 family. A polypeptide that has the most potent thrombopoietic activity among the thus far identified molecules is thrombopoietin (TPO). TPO exerts its biological functions through its cognate receptor, c-MPL, that interestingly is classified into the hematopoietic cytokine receptor family in which gp130 also belongs. Although each member of the IL-6 family as well as TPO signals through the distinct receptor complex, the underlying signaling mechanism is similar in each case: the first event after cytokine stimulation is dimerization of a respective set of receptor components, and activation of receptor-associated tyrosine kinases in the janus kinase (JAK) family then follows. Activation of JAK kinases is the pivotal step to initiate downstream cytoplasmic signaling cascades involving signal transducer and activator of transcription transcription factors and mitogen activated protein kinase. In this article, we focus on gp130-mediated cytokine signal transduction by comparing that mediated by c-MPL.

[Peripheral eosinophil counts relates the improvement of the skin lesions of atopic dermatitis patients more sensitive than serum eosinophil cationic protein levels].

We described the severity of skin lesions and detected peripheral eosinophil counts (Eos) and serum eosinophil cationic protein levels (s-ECP) in 81 patients with atopic dermatitis (male 33, female 48, age 20.7 +/- 8.8) at the first consultation. Eos were 583.3 +/- 560.2/mm3 and s-ECP were 18.0 +/- 21.7 micrograms/l (normal 6.88 +/- 3.46). After approximately 30 days treatment, the improvement of dermatitis were estimated and those lavatory findings were rechecked. Eos were 446.7 +/- 367.6/mm3 and s-ECP were 13.2 +/- 11.7 micrograms/l. At 5 percent level, positive correlations were observed between Eos and s-ECP (R = 0.75), Eos and the severity of dermatitis (R = 0.52) and s-ECP and the severity of dermatitis (R = 0.51) at the first consultation. We found a positive correlation between Eos and the improvement of dermatitis (R = 0.40) but no correlation between s-ECP and the improvement of dermatitis (R = 0.10). Eos related the improvement of the skin lesions of atopic dermatitis more sensitive than s-ECP.

[A resected case of a bronchiolo-alveolar cell carcinoma of the lung accompanying pneumonia-like shadow on chest roentgenogram].

A 67-year-old female was admitted to our hospital because of pneumonia-like shadow on chest roentgenogram with persistent cough and sputum of 4 months duration. Diagnosis as lung cancer was delayed more than 4 months. She showed fever and inflammatory reactions. Antibiotics were effective to inflammatory reactions, but not effective to pneumonia-like shadow. Transbronchial lung biopsy was useful for the diagnosis. Right lower lobectomy was performed. In this case, tumor extents were limited within one lobe. Tumor cells did not invade blood and lymphatic vessels, and extrathoracic metastases were not detected. The prognosis of bronchiolo-alveolar cell carcinoma was determined by intra-pulmonary tumor extent. Based on a comparison with the outcome of unresected cases, bronchiolo-alveolar cell carcinoma limited within one lobe should be surgically resected.

Effect of dentin treatment with citric acid/ferric chloride solutions on glass ionomer bond strength.

PURPOSE: To investigate the effect of dentin treatment with mixed solutions of citric acid and ferric chloride on the tensile bond strength of resin-modified glass ionomers. MATERIALS AND METHODS: 120 freshly extracted bovine incisors were ground to expose dentin surface. Vitremer and Fuji II LC were bonded to dentin surfaces that were (1) rinsed with distilled water and dried with oil-free compressed air, (2) treated with Super Bond Dentin Activator for 30 seconds, (3) treated with Bondwell LC Conditioner for 20 seconds, (4) treated with Scotchbond Multi-purpose Etchant for 15 seconds, (5) treated with Dentin Conditioner for 20 seconds, or (6) treated with K-etchant for 40 seconds following manufacturers' recommendations. Specimens were stored in water at 37 degrees C for 24 hours. The tensile bond strength was measured at a crosshead speed of 0.5 mm/minute. The data were statistically analyzed by a one-way ANOVA and Dunnett's Post-hoc Procedure (P < 0.05). RESULTS: Super Bond Dentin Activator and Bondwell LC Conditioner increased the tensile bond strength of Vitremer and Fuji II LC. Scotchbond Multi-purpose Etchant, Dentin Conditioner, and K-etchant did not improve the tensile bond strength of Vitremer and Fuji II LC.

[A case of intrathoracic lipoma arising from the chest wall resected by video-assisted thoracoscopic surgery].

We present a case of lipoma arising from the chest wall spreading into the thoracic cavity. Although asymptomatic, a 65-year-old female was pointed out an abnormal shadow on the chest X-ray film taken at the mass survey. The tumor, measuring 3.4 x 3.0 x 2.0 cm in diameter, was surgically removed under thoracoscopic visualization through a small thoracotomy incision of lt. 1st intercostal space, and the diagnosis of lipoma was confirmed postoperatively by histopathologic examination.

Synovial immunohistochemical changes after high tibial osteotomy for osteoarthritis of the knee. Two-year prospective follow-up.

The synovium of the osteoarthritic knee was investigated for pathological changes after high tibial osteotomy (HTO) in a prospective two-year follow-up study. The series included 11 osteoarthritic knees in 9 patients with a mean age of 57 +/- 8 years (range: 47 to 70 years). Synovial specimens were obtained from the medial part of the knee just above the medial meniscus at the time of HTO, and taken from a different medial portion from the initial biopsy at the time of removal of the internal fixation (mean interval: 21 +/- 4 months). Lining cell hyperplasia, villous hypertrophy, cell infiltration, and vascular proliferation, observed in the first biopsy specimens, were less discernible in the samples from the second biopsy, which were obtained at the time of the removal of the fixation device. Immunohistochemical staining of the specimens revealed that cartilage fragments positive for type 2 collagen were surrounded by proliferating lining cells at the time of osteotomy. These lining cells were predominantly stained with anti-CD68 antibody for macrophage lineage cells. The mean numbers of type 2 collagen-positive fragments and CD68-positive cells in the lining layer at HTO were 9.1 +/- 6.2 (3 to 26) and 30.8 +/- 11.0 (17 to 55) per high power field (x400), respectively, while they were 1.1 +/- 1.2 (0 to 4) (p < 0.005) and 14.8 +/- 4.7 (7 to 21) (p < 0.01), respectively, at the time of removal. The number of these cartilage fragments decreased markedly after correction of the varus deformity by HTO. In conclusion, the excessive pressure on the medial compartment of the osteoarthritic knee with a varus deformity can cause wearing of the articular cartilage and fragmentation. The cartilage fragments might stimulate macrophage lineage cells in the lining layer and might cause synovitis in osteoarthritis. After correction of the varus deformity by HTO, the synovitis subsided because there were fewer cartilage fragments and less wear.

Defective mammopoiesis, but normal hematopoiesis, in mice with a targeted disruption of the prolactin gene.

Prolactin (PRL) has been implicated in numerous physiological and developmental processes. The mouse PRL gene was disrupted by homologous recombination. The mutation caused infertility in female mice, but did not prevent female mice from manifesting spontaneous maternal behaviors. PRL-deficient males were fertile and produced offspring with normal Mendelian gender and genotype ratios when they were mated with heterozygous females. Mammary glands of mutant female mice developed a normal ductal tree, but the ducts failed to develop lobular decorations, which is a characteristic of the normal virgin adult mammary gland. The potential effect of PRL gene disruption on antigen-independent primary hematopoiesis was assessed. The results of this analysis indicated that myelopoiesis and primary lymphopoiesis were unaltered in the mutant mice. Consistent with these observations in PRL mutant mice, PRL failed to correct the bone marrow B cell deficiency of Snell dwarf mice. These results argue that PRL does not play any indispensable role in primary lymphocyte development and homeostasis, or in myeloid differentiation. The PRL-/- mouse model provides a new research tool with which to resolve a variety of questions regarding the involvement of both endocrine and paracrine sources of PRL in reproduction, lactogenesis, tumorigenesis and immunoregulation.

Cerebrospinal fluid apo E and apo A-I concentrations in early- and late-onset Alzheimer's disease.

We compared cerebrospinal fluid (CSF) apolipoprotein (apo) A-I and apo E concentrations in early- and late-onset Alzheimer's disease (EOAD (n = 11), LOAD (n = 15), respectively) with those in control subjects (n = 23). CSF apo A-I levels in both EOAD and LOAD were consistent with control subjects. However, CSF apo E levels were significantly lower in EOAD group (mean +/- SD; 2.65 +/- 1.69 mg/l, P < 0.05) and higher in LOAD group (5.90 +/- 1.94 mg/l, P < 0.01) than those in control group (4.16 +/- 1.69 mg/l). In addition, the epsilon4 allele frequency was not different between EOAD and LOAD groups. Although the reason for the difference in CSF apo E concentrations between two groups is unknown, CSF apo E concentration seems to be associated with the pathogenesis of EOAD and LOAD. The rate of apo E production and/or catabolism in the brain may be different between them.

High-performance liquid chromatography of methamphetamine and its related compounds in human urine following derivatization with fluorescein isothiocyanate.

A high-performance liquid chromatographic method with fluorescence detection for the determination of methamphetamine and its related compounds is reported. Methamphetamine, amphetamine, norephedrine, p-hydroxymethamphetamine and 1-phenylethylamine as an internal standard were extracted from human urine, derivatized with fluorescein-4-isothiocyanate, and then separated on a reversed-phase column within 36 min. The fluorescence intensity of the effluent was monitored at excitation and emission wavelengths of 496 and 518 nm, respectively. Calibration curves were confirmed to be linear up to at least 100 pmol on the column with a correlation coefficient (r) of 0.994-0.999 for the target compounds. The detection limits (S/N=3) were 55-105 fmol per 20-microl injection. The method was successfully applied to urine samples taken from methamphetamine addicts.

Secretion of authentic 20-kDa human growth hormone (20K hGH) in Escherichia coli and properties of the purified product.

Using Bacillus amyloliquefaciens neutral protease gene (npr), we have constructed a secretion system of 20-kDA human growth hormone (20K hGH) in E. coli. The secretion-signal region from npr was modified inserting a fragment coding a 2Lys-5Leu cluster. In this system we found that co-expression of glutathione reductase remarkably increased accumulation level of 20K hGH in periplasm and confirmed that secreted 20K hGH was correctly processed. The recombinant 20K hGH was highly purified and subjected to analyses of physicochemical properties and biological activities which are still unclear and controversial due to difficulty in preparing the sample with authentic structure. The secreted recombinant product had authentic disulfide linkages and showed molecular weight of 20,270.5 +/- 3.7 (theoretical value, 20,269.9). The results suggest that the recombinant 20K hGH is a full agonist on rat growth promotion and lipolysis stimulation in isolated rat adipose tissues. In particular, the lipolysis-stimulating activity of 20K hGH was distinct as compared with that of 22K hGH under physiological concentration. Cell proliferation activity via prolactin-receptor in Nb-2 lymphoma was obviously low as compared with that of 22K hGH.

Complete nucleotide sequence of the chloroplast genome from the green alga Chlorella vulgaris: the existence of genes possibly involved in chloroplast division.

The complete nucleotide sequence of the chloroplast genome (150,613 bp) from the unicellular green alga Chlorella vulgaris C-27 has been determined. The genome contains no large inverted repeat and has one copy of rRNA gene cluster consisting of 16S, 23S, and 5S rRNA genes. It contains 31 tRNA genes, of which the tRNALeu(GAG) gene has not been found in land plant chloroplast DNAs analyzed so far. Sixty-nine protein genes and eight ORFs conserved with those found in land plant chloroplasts have also been found. The most striking is the existence of two adjacent genes homologous to bacterial genes involved in cell division, minD and minE, which are arranged in the same order in Escherichia coli. This finding suggests that the mechanism of chloroplast division is similar to bacterial division. Other than minD and minE homologues, genes encoding ribosomal proteins L5, L12, L19, and S9 (rpl5, rpl12, rpl19, and rps9); a chlorophyll biosynthesis Mg chelating subunit (chlI); and elongation factor EF-Tu (tufA), which have not been reported from land plant chloroplast DNAs, are present in this genome. However, many of the new chloroplast genes recently found in red and brown algae have not been found in C. vulgaris. Furthermore, this algal species possesses two long ORFs related to ycf1 and ycf2 that are exclusively found in land plants. These observations suggest that C. vulgaris is closer to land plants than to red and brown algae.

Structures of genes encoding phospholipase A2 inhibitors from the serum of Trimeresurus flavoviridis snake.

Inhibitors (PLIs) against snake venom gland phospholipases A2 (PLA2s) have been found in their sera. A cDNA encoding a PLI from Trimeresurus flavoviridis (Tf, habu snake, Crotalinae) serum, cPLI-A, was isolated from the Tf liver cDNA library and sequenced. Northern blot analysis with cPLI-A showed that PLIs are expressed only in liver. Genes for PLIs, gPLI-A and gPLI-B, were isolated from the Tf genomic DNA library and their nucleotide (nt) sequences were determined. The genes consisted of four exons and three introns, and exon 4 encoded the carbohydrate recognition domain (CRD)-like motif. Comparison of the nt sequences between gPLI-A and gPLI-B showed that these genes are highly homologous, including introns, except that exon 3 is rich in nonsynonymous nt substitutions which are almost four times as frequent as synonymous nt substitutions. This evolutionary feature of PLI genes is different from that of venom gland PLA2 isozyme genes in which nonsynonymous nt substitutions are spread over the entire mature protein-coding region.

Interstitial deletion of 6q21-q23 associated with split hand.

We report on a 7-month-old boy with interstitial deletion of 6q21-q23 and split-hand defect. He died at 7 months. This is the fifth patient with distal limb anomaly associated with a rearrangement of 6q21 region, and supports previous suggestions that there may be candidate gene(s) for distal limb development in the 6q21 region.

Effects of concanavalin A on chondrocyte hypertrophy and matrix calcification.

Resting chondrocytes do not usually undergo differentiation to the hypertrophic stage and calcification. However, incubating these cells with concanavalin A resulted in 10-100-fold increases in alkaline phosphatase activity, binding of 1,25(OH)2-vitamin D3, type X collagen synthesis, 45Ca incorporation into insoluble material, and calcium content. On the other hand, other lectins tested (including wheat germ agglutinin, lentil lectin, pea lectin, phytohemagglutinin-L, and phytohemagglutinin-E) marginally affected alkaline phosphatase activity, although they activate lymphocytes. Methylmannoside reversed the effect of concanavalin A on alkaline phosphatase within 48 h. Concanavalin A did not increase alkaline phosphatase activity in articular chondrocyte cultures. In resting chondrocyte cultures, succinyl concanavalin A was as potent as concanavalin A in increasing alkaline phosphatase activity, the incorporation of [35S]sulfate, D-[3H]glucosamine, and [3H]serine into proteoglycans, and the incorporation of [3H]serine into protein, although concanavalin A, but not succinyl concanavalin A, induced a rapid change in the shape of the cells from flat to spherical. These findings suggest that concanavalin A induces a switch from the resting, to the growth-plate stage, and that this action of concanavalin A is not secondary to changes in the cytoskeleton. Chondrocytes exposed to concanavalin A may be useful as a novel model of endochondral bone formation.

Characterization of a cartilage-derived 66-kDa protein (RGD-CAP/beta ig-h3) that binds to collagen.

A 66-kDa collagen fiber-associated protein (RGD-CAP) was isolated from a fiber-rich fraction of pig cartilage by ultrafiltration and collagen-affinity chromatography. Amino acid sequencing and cDNA cloning indicated that the RGD-CAP is identical or closely related to beta ig-h3 protein which is induced in human adenocarcinoma cells by transforming growth factor-beta (TGF-beta) (Skonier, J., Neubauer, M., Madisen, L., Bennett, K., Plowman, G.D., and Purchio, A.F. (1992) DNA Cell. Biol. 11, 511-522). The RGD-CAP, as well as beta ig-h3, has the RGD sequence in the C-terminal region. The native RGD-CAP bound to type I, II, and IV collagens even in the presence of 1 M NaCl. A recombinant preparation of RGD-CAP expressed in Escherichia coli cells also bound to collagen but not to gelatin. The RGD-CAP mRNA was expressed in chondrocytes throughout all stages, although the expression level was highest during the prehypertrophic stage. In addition, TGF-beta increased the RGD-CAP mRNA level in chondrocyte cultures. Since RGD-CAP transcripts were found in most tissues, this novel collagen-binding protein may play an important role in cell-collagen interactions in various tissues including developing cartilage.

[Nutritional study of elimination diets in the case of two-year-old children recovered from atopic dermatitis].

The aim of this study is to clarify the nutritional value of elimination diets (four types of Japanese style diets) which atopic dermatitis children had been on for seven days before the examination. The intake of energy was less than the recommended dietary allowances. This result suggests that the intake of energy is associated with the removing of vegetable oils. As 54.8% of the children had not had any vegetable oils, the intake of fat was half the recommended dietary allowance. Half of the children had not had eggs and milk at all, but the intake of protein of the boys was 31% more than the recommended dietary allowance and that of the girls was 42% more than the recommended dietary allowance. The intake of calcium by the boys was 365 +/- 199 mg and that by the girls was 452 +/- 67 mg. The intake of iron and vitamins was more than the recommended dietary allowance. In the case of the boys on elimination diets, the ratio of PFC was P = 17.1%, F = 11.6%, C = 70.8%, and in the case of the girls it was P = 17.1%, F = 13.9%, C = 69.0%. It has been clarified that the F ratio of the intake of the boys and the girls was smaller than the ratio of the recommended dietary allowances, and that P ratio and C ratio were larger.

Leptin receptor (OB-R) oligomerizes with itself but not with its closely related cytokine signal transducer gp130.

Leptin (OB) exerts weight-reducing effects in mice. The structure of the receptor for this factor, OB-R, is considerably similar to those of gp130, the common signal transducing receptor component for the interleukin-6 (IL-6) family of cytokines, and leukemia inhibitory factor receptor (LIFR). Since the IL-6 family of cytokines signal through gp130 homodimer or gp130/LIFR heterodimer, we have examined in this study the possible involvement of gp130 and LIFR in leptin signaling through OB-R. Leptin stimulation induces tyrosine phosphorylation of neither gp130 nor LIFR, while LIF stimulation does both. As examined by using two differently epitope-tagged OB-R molecules, the spontaneous homo-oligomerization of OB-R has been elucidated. Ba/F3 cells, which do not express gp130, are non-responsive to leptin and exhibit increased DNA synthesis in response to leptin after transfection of OB-R cDNA alone. OB-R appears to transduce the signal via its homo-oligomerization without interaction with gp130 or LIFR.
