Mechanism of inactivation of single sodium channels after modification by chloramine-T, sea anemone toxin and scorpion toxin.

Single sodium-channel currents were measured in neuroblastoma cells after inhibition of inactivation by chloramine-T (CHL-T), sea anemone toxin II (ATX-II) and scorpion toxin (SCT). The decaying phase of the averaged single-channel currents recorded with 90-msec pulses in cell-attached patches was clearly slower than that of the ummodified channels, suggesting inhibition of macroscopic inactivation. Each substance caused repetitive openings and a moderate increase in the channel open time. At Vm = RP + 20 mV and T = 12 degrees C, the mean channel open times were 1.4, 1.6 and 1.8 msec for CHL-T, ATX-II and SCT, respectively, as opposed to 1.07 msec for native channels. Open-time histograms could be best fitted by the sum of two exponentials. The time constants of the fits were similar for histograms constructed from single openings and from openings during bursts. This suggests that the population of channels is homogeneous and that in bursts the same open conformations of channels occur as in single openings. Mean burst durations for bursts consisting of more than one opening at Vm = RP + 20 mV were 4.9, 5.8 and 6.1 msec for CHL-T, ATX-II and SCT, respectively. Burst open-time histograms constructed from two or three openings were fitted by the gamma function. The different time constants of the fits obtained for ATX-II and SCT suggested multiple open conformations of channels for openings of bursts. However, significantly different open-time histograms constructed from the first, second and third openings of bursts could not be obtained systematically. A positive correlation was found for the dwell time of the first and the second, as well as for the second and the third opening of bursts with each substance, but a negative one for the dwell time of an opening and the neighboring closing of bursts with ATX-II. The results suggest a model with multiple open and inactivated states. In this model the inactivated states are weakly absorbing.

Longitudinal examinations in the course of dietotherapy of mentally retarded obese in-patients.

Mentally retarded obese in-patients were fed by low-energy diet (4.2-4.6 MJ) for 9 months. During this period, an average of 13 +/- 4.5 kg loss of body mass occurred in men and 16 +/- 2.7 kg in women. Anthropometric measurements were performed before starting the dietotherapy and in the ninth month. Changes of body fat could be followed well when calculated according to BMI. Less reliable results were obtained with skinfold thickness measurements, presumably due to body deformities. Results of clinical laboratory tests, which were carried out before starting the dietotherapy and in the fourth and seventh months, suggested that a low-energy-containing diet with balanced nutrient content and adequate protein intake did not impair protein metabolism, favourably affected serum triglyceride and cholesterol levels, but resulted in an unfavourable decrease in the HDL-cholesterol content.

Diffusion of HIV-1 virus into non-habitual categories at risk in European countries.

The ECP Working Group on AIDS has evaluated the available data on seropositivity to HIV-1 supplied by research groups in 12 Eastern and Western European countries. The period covered is 1985 and 1986. A significantly elevated incidence of seropositives was observed in both females and males in heterosexual contact with members of high risk groups. In contrast heterosexuals with no such contact had an incidence below 1%. For male homosexuals from Italy, Denmark and Switzerland the trend was no detectable increase in prevalence from 1985 to 1986. Hungary and Poland now have a few per cent seropositive male homosexuals, but no seropositives were found in a group of Polish drug abusers.

Comparative studies of single Na-channels in different neuroblastoma cell lines.

Single sodium channel currents were compared in two neuroblastoma (N1E 115, N18) and in two neuroblastoma x glioma (108CC5, 108CC15) hybrid clonal cell lines. Similar single channel parameters were reached from every cell line. The single channel conductance was 11 pS for N1E 115 and N18 cells, and was 10 pS for both hybrid cells at 7-8 degrees C. Except for N18 cells, the amplitude histograms could be best fitted by the sum of two Gaussian functions. The mean channel open time was 1.5-2.0 ms for N1E 115 and the hybrid cells and it was shorter than 1 ms for N18 cells. Open time histograms from N18 cells could be fitted by a single exponential at most potentials, but the sum of two exponentials resulted in a best fit for N1E 115 and the hybrid cells. Differentiation of N1E 115 cells elicited by dibutyryl cAMP instead of dimethylsulfoxide caused only a slight increase in the single channel conductance and open time. The results indicate a certain uniformity of the sodium channels in these cell line.

Adenosine induced delay of expression of AIDS virus, HIV, in H9T cells.

Adenosine reduced the expression of CD4 antigen both in HIV infected and uninfected H9 T cells. The continuous presence of adenosine (1 mumol/ml) resulted in a remarkable delay of HIV expression in the infected cells. A relationship is suggested in the effects of HIV and adenosine on the human T cells.

Prevalence of HIV-antibodies in patients with haemophilia in Hungary.

Incidence of HIV antibodies have been studied in 617 patients with congenital bleeding disorders. Screening was performed with the Organon ELISA test, repeatedly positive samples were tested with four different confirmatory assays. HIV antibodies were found in 3/356 patients with haemophilia A, 21/114 patients with haemophilia B, 3/123 patients with von Willebrand disease and in 1/24 patients with other types of congenital coagulation disorders. All the 28 seropositive patients were treated with imported coagulation factor, mostly with factor IX concentrates. No patient treated exclusively with nationally produced factor preparations between 1981 and 1985 was found to be anti-HIV positive.

Subconductance states of single sodium channels modified by chloramine-T and sea anemone toxin in neuroblastoma cells.

Single channel currents of chloramine-T (Chl-T) and sea anemone toxin (ATX-II) modified sodium channels were studied in neuroblastoma cells. With both substances similar subconductance states have been observed. The conductances of the sublevels were multiples of the "unit" step which was about one-forth of the most frequently occurring main conductance. Thus, the current levels observed were one fourth, half and five-fourths of the main current size. Both substances caused a slower decay of the averaged current compared to the current of the native channels. The main single-channel conductance was 15.2 pS (T = 16 degrees C) for the Chl-T and 10.8 pS (T = 12 degrees C) for the ATX-II modified channels. The channel open time was doubled by ATX-II, but was not increased significantly by Chl-T. The existence of the subconductance states suggests that the native channels may also have multiple open conformations.

Evidence for multiple open states of sodium channels in neuroblastoma cells.

Open times of voltage-gated sodium channels in neuroblastoma cells were measured during repolarization (following a short depolarizing conditioning pulse) and during moderate depolarization. Conditional and unconditional channel open-time histograms were best fitted by the sum of two exponentials. (The conditional open time was measured from the end of the conditioning pulse until an open channel shuts provided it was open at t = 0). Time constants of both histograms depended on the post-pulse and were shifted to more positive potentials with increasing conditioning pulse potential. This shift could be explained by assuming more than two time constants in the histograms, which could not be separated. Channel open-time histograms from single-pulse experiments showed a maximum at t greater than 0. These histograms could be best fitted by an exponential function with three time constants. One term of this function included the difference of two exponentials resulting in a maximum at t greater than 0. Open-time histograms showed a definite time dependence. At 2 to 6.5 msec after the beginning of the depolarization the best fit could be obtained by the difference of two exponentials. To these components another term had to be added at 0 to 2 msec. Between 6.5 and 14.0 msec the sum of two exponentials, and after 14.0 msec a single exponential resulted in a good fit. The results support the hypothesis that sodium channels in neuroblastoma cells may have multiple open states. Two of these states are irreversibly coupled.

Ultrastructure and immunoelectron microscopy of human T-cell leukaemia virus type I-producing lymphoid and non-lymphoid human tumour cells.

The ultrastructure and distribution of some viral and cellular antigens was examined in lymphoid C91/PL and fibroblastic HOS/PL cells infected with human T-cell leukaemia virus type I. Tubuloreticular structures in the endoplasmic reticulum, characteristic of virus-infected or interferon-treated cells, were found in both cell types. Virions were observed particularly in the HOS/PL cells in budding and mature forms and in coated pits, coated vesicles and lysosomes, indicative of receptor-mediated endocytosis. Using immunogold indirect labelling, viral antigens, recognized by sera of infected patients and by monoclonal antibody, were detected in patches on the cell surface and in material loosely attached to the envelope of some virions. Beta 2-microglobulin was associated with virion envelopes from C91/PL and HOS/PL cells, but major histocompatibility complex class 1 (HLA) antigens were not associated with virions even when produced by HLA-positive HOS/PL cells.

Morphologic and immunoelectronmicroscopic identification of human T-cell lymphotropic virus type III (HTLV-III).

The AIDS associated HTLV-III virus infected H9 cells were extensively studied using light, scanning and transmission electronmicroscopy. It was demonstrated that the morphological features of HTLV-III are different from the C-type particles and are similar to those of lentiviridae. For immunological identification high titer pre-AIDS patient sera served as the anti-HTLV-III envelope antibody source. The immunoelectronmicroscopic method was able to identify the viral envelope antigen on the surface of infected cells and in certain areas of the viral envelope. This is the first application of immunoelectronmicroscopy for the identification of the HTLV-III virus.

Studies on the effect of iron overload on rat cortex synaptosomal membranes.

Iron as ferrous ammonium sulfate was injected into the cerebral spinal fluid of rats. After three consecutive days of injection of 4 mumol of iron, the total iron content of brain cortex synaptosomes from the iron-treated animals was 2-fold higher than that from control animals receiving the saline vehicle only. Spin label studies of the synaptosomal membranes demonstrated that the lipid region of the membranes became more rigid and, in addition, the mobility of labeled SH groups of membrane proteins decreased after the iron treatment. The cholesterol content was significantly higher in iron-treated animals as compared to controls. Centrophenoxine pretreatment (100 mg/kg body weight daily for 6 weeks) diminished the iron effects. Synaptosomal membrane alterations observed after iron treatment were similar to changes observed previously during aging. This lends support to the notion that free-radical induced damage occurs in brain membranes with increasing age.

Interaction between sodium channels in mouse neuroblastoma cells.

Single sodium channels in mouse neuroblastoma cells (N1E 115) were studied in cell-attached patches. During a series of consecutive responses to depolarizing pulses, records with and without channel opening were seen to form clusters rather than appearing randomly. The probability of finding open channels on a record seemed to increase with increasing number of channel openings. The open times of channels became shorter with increasing closed time interval measured between consecutive channel openings. Overlapping openings showed a voltage-dependent open time, in contrast to single openings which had voltage-independent open time. On the basis of these observations interaction between neighbouring sodium channels is suggested.

Immunological alterations in anti-HTLV-III negative haemophiliacs and homosexual men in Hungary.

Hungary can be considered as a low risk area for AIDS since no patient with full-blown AIDS or AIDS-related complex has been found in the country. A complex clinical and immunological (T cell subsets, DNCB sensitization test, circulating immune complexes, acid-labile alpha interferon) investigation was performed between November, 1983 and June, 1984 in order to study whether alterations found in symptom-free homosexuals and haemophiliacs in the risk areas for AIDS can be observed in Hungary as well. 38 patients with mild haemophilia, 35 patients with severe haemophilia and 40 homosexual men were investigated in parallel to 37 heterosexual blood donors as controls. Anti-HTLV-III antibodies were measured later in the stored serum aliquots from the same subjects by the indirect membrane immunofluorescence assay. Although specific anti-HTLV-III antibodies were not detected in the haemophiliacs or homosexuals, immunological alterations characteristic for the members of AIDS risk groups in the high risk areas (decrease in the percentage of OKT4 cells and/or decrease of the OKT4/8 ratio) were found in one-third of the homosexuals and haemophiliacs tested. In addition, a significant part of these subjects did not develop delayed type hypersensitivity skin reaction on DNCB rechallenge. These findings indicate that an immunodeficiency independent of HTLV-III infection can be present in two major AIDS risk groups, in homosexual men and haemophiliacs.

The shedding of viable cells into the local lymph by tumours growing in the gut of rats.

Suspensions of syngeneic sarcoma cells were injected into the Peyer's patches of rats from which the mesenteric nodes had been removed. By later cannulating the thoracic duct of such rats it was possible to collect peripheral intestinal lymph that had come directly from the tumour bearing area without being filtered through a regional node. The number of viable tumour cells in the lymph coming from the tumours was monitored by culturing the whole lymph cells in a limiting dilution assay. The tumours grew to a diameter of approximately 1 cm in 25 days and during this time tumour cells were present in the lymph at a ratio of approximately 1 tumour cell per 10(5) lymph cells. In euthymic rats this number declined as the immune response developed. In athymic rats the number increased by approximately 10 fold during the experiments. It was concluded that the shedding of viable cells parallels the linear, not the volumetric dimensions of the tumour.
