Studies of the sensitivity and reproducibility of commercial kits to detect antibodies to the human immunodeficiency virus.

Nine serial three-fold dilutions (1:1 to 1:6561) were prepared from 18 sera obtained from hemophiliacs confirmed to have antibodies to the human immunodeficiency virus. The dilutions were tested with five different commercial enzyme immunoassay kits and twelve sera were retested 5 to 7 months later by different lots of three kits. The dilution that gave an absorbance (OD) equal to the cut-off OD was considered as the titer of antibody. Sensitivities of the kits were compared by statistical and regression analysis; the same approach was used for studying reproducibility from the results of retesting. The highest titers were found with the Wellcome kit, the lowest with Organon and Pasteur kits, whereas intermediate values were found with the Sorin/Biomedica and Electronucleonics kits. With the Organon and Wellcome kits, excellent reproducibility was observed on later retesting; however, a significant change in titers was seen on retesting the Sorin kit.

Multicentric Castleman disease and systemic lupus erythematosus phenotype in a boy with Klinefelter syndrome: long-term disease stabilization with interferon therapy.

An 11-year-old boy with Klinefelter syndrome had Castleman disease (CD) of plasma cell type develop. Nonregulated antibody production mimicked systemic lupus erythematosus (SLE). Hepatitis C virus (HCV) infection caused significant disease worsening. The patient was treated with a daily dosage of 2 million units/m2 of IFN-alpha. Dramatic clinical improvement and decreasing autoimmune phenomenon were observed. HCV RNA were cleared. Hypergammaglobulinemia did not change. The boy has been living for 8 years with his disease. Plasma cell type CD can mimic collagenosis. Disease worsening is caused by HCV, though it can be reversed with IFN-alpha. Klinefelter syndrome may be a genetic susceptibility factor for CD in some cases.

TGF beta 1 induces caspase-dependent but death-receptor independent apoptosis in lymphoid cells.

Transforming growth factor beta 1 (TGF beta 1) is an antiproliferative and proapoptotic cytokine for normal B-cells, however many B-cell lymphomas have lost their response to TGF beta 1. The aim of this study was to identify the sequence of events in apoptosis induced by TGF beta 1 in an EBV negative, human B-cell lymphoma line (HT58). The proportion of apoptotic cells increased gradually (up to 72 hr) at an optimal dose range of 0.5-1.0 ng/ml. The induced cell death required the action of downstream caspases. Caspase activation was accompanied by an increase in the permeability of mitochondrial membranes, but there was no change in the expression of certain members of Bcl-2 family (Bcl-2, Bax, Bcl-XL). Similarly, none of the death receptors or ligands were involved in apoptosis induction. Further study will include the participation of TGF beta 1 target genes in the pore formation of mitochondrial membranes and/or the elimination of a putative survival signal.

[Experimental 3-month study of safety of textured soybeans Becsei].

The authors conducted a three-month safety test of a textured soy (TS) product manufactured by the Agricultural Plant, Boly. Young male and female rats received semisynthetic diets containing 8, 18 and 38% of TS protein or casein supplemented with methionine, as the only source of protein. The effect of the diet containing 18% of TS protein on the growth, on certain parameters of protein and fat metabolism and on the zinc and copper status corresponded to that produced by 18% of casein supplemented with methionine. When the content of TS protein was lower or higher than the optimal (18%), the development of rats fell behind the control ones receiving adequate amount of casein. High doses of TS protein produced a favourable effect on lipid metabolism in the liver, but its effect on zinc metabolism was unfavourable. No signs of toxic action of the TS product were recorded during the three-month safety test of TS.

[Microbial study of the surface of malignant tumors of the oral cavity].

Malodour and other infection sequelae may increase the morbidity, compromise the oral well-being of the patient suffering from maxillo-facial neoplasia. The purpose of our study was to evaluate the differences of the oral bacterial flora attached to the tumor and the contra lateral sound surfaces. Swabs were obtained and samples were microbiologically cultured. It was concluded, that carcinoma surface biofilm harbors increased levels of both aerobs and anaerobs.
