Prognostic significance of the blast self-renewal capacity in patients with acute myeloid leukemia.

BACKGROUND: The proliferation of leukemic cells in acute myeloid leukemia (AML) is supported by a small subpopulation of leukemic blast progenitors, which can be detected in methylcellulose and suspension cultures. It is important to determine the association between the biologic properties of leukemic blast progenitors and the clinical prognosis of patients with AML. METHODS: Ninety-five patients with AML and two patients with chronic myelocytic leukemia in blast crisis were studied. T-cell depleted mononuclear cells obtained from the peripheral blood cells were cultured in both methylcellulose and suspension cultures. In methylcellulose culture, primary blast colony formation (PE1) and secondary blast colony formation (PE2) were enumerated. The recovery of clonogenic cells was determined in suspension culture. The association between PE1, PE2, or clonogenic cell recovery in suspension and the remission induction outcome or survival duration of the patients were examined. RESULTS: PE1 was not associated with the remission induction outcome. PE2 and clonogenic cells recovered in suspension were significantly associated with the remission induction. Furthermore, the survival duration of the patients who achieved complete remission was associated significantly with the number of clonogenic cells recovered in suspension. CONCLUSIONS: PE1 reflects the terminal divisions of leukemic blast progenitors, whereas PE2 and the recovery of clonogenic cells in suspension culture are considered to reflect the self-renewal of leukemic blast progenitors. The results suggest that the self-renewal capacity of leukemic blast progenitors is predictive of not only remission induction outcome but also survival duration of patients with AML.

Type III collagen is a major component of interodontoblastic fibers of the developing mouse molar root.

BACKGROUND: Recently, collagenous interodontoblastic fibers (IOF) were reported in some particular developmental stages and/or locations of the tooth. However, it remained unclear whether these fibers were identical to so-called von Korff fibers. METHODS: To clarify this issue, we examined the developing mouse molar by three-dimensional reconstruction of 8 confocal images within a 6 micron-thick section using laser scanning confocal microscopy, and confirmed our findings using immunoelectron microscopy. RESULTS: In the root pulp during circumpulpal dentin formation, the IOF stained weakly for type I collagen, but stained strongly for type III collagen by a double-staining technique. It could be clearly seen that many immunoreactive fibers ran spirally among the odontoblasts and entered the predentin. This distribution pattern of IOF was similar to that of the classical von Korff fibers. Furthermore, the existence of anti-type III reactive collagen fibrils between odontoblasts was confirmed, whereas IOF were not observed in the coronal pulp during circumpulpal dentin formation. CONCLUSIONS: This study presents for the first time, immunohistochemical observations which demonstrate the presence of IOF at least during root circumpulpal dentin formation and which reveal that type III collagen is a major component of IOF.

Enhancement of differentiation induction of mouse myelomonocytic leukemic cells by extracellular ATP.

We examined the effect of ATP on the terminal differentiation of mouse myelomonocytic leukemic M1 cells to macrophages. Although ATP alone did not induce M1 cell differentiation, addition of ATP with the differentiation inducer, interleukin 6 (IL-6), enhanced the induction of differentiation by IL-6 about two-fold. Comparison among several adenine nucleotides revealed that the order of effectiveness on differentiation enhancement was ATP > ADP > AMP > or = adenosine. Using reactive blue 2, a P2 receptor antagonist, we confirmed that the effect of ATP on the stimulation of differentiation was mediated through the P2 purinergic receptor. Examination of cytosolic [Ca2+] elevation by ATP and comparison of potency of differentiation enhancement among several ATP analogs demonstrated that the effect of differentiation enhancement was mediated through P2y purinergic receptors expressed on M1 cell surface. Within 3 h of exposure, ATP alone slightly increased the expression of differentiation-related immediate early response genes, c-myc and JunB, and ATP also enhanced the IL-6-induced expression of these genes. Induction of JunB expression by ATP analogs correlated with their potency of differentiation enhancement, which suggested that induction of JunB by ATP is one of signaling pathways involved in the exertion of its differentiation-enhancing effect.

Induction of superoxide anion production from monocytes an neutrophils by activated platelets through the P-selectin-sialyl Lewis X interaction.

Activated platelets expressing P-selectin on their surface are known to adhere to monocytes and neutrophils. We examined the possibility that the leukocytes were activated by their adhesion to activated platelets and demonstrated that P-selectin-dependent platelet adhesion to neutrophils and monocytes induced production of extracellular superoxide anion (O2-) by these leukocytes. Leukocyte membrane glycoproteins containing Ser/Thr-linked carbohydrate chains were responsible for the signal reception leading to the leukocyte activation. Cytokines were shown to influence these processes. For example, treatments of neutrophils with interleukin-8 (IL-8) or granulocyte colony-stimulating factor (G-CSF) potentiated the P-selectin-induced O2- production. Furthermore, interleukin-1 (IL-1) and interferon-gamma (IFN-gamma) induced surface expression of P-selectin on platelets in the presence of a low concentration of thrombin and consequently enhanced their adhesion capacity to leukocytes. These results indicated that the adhesion of activated platelets to the leukocytes through the interaction between P-selectin and its carbohydrate ligand, sialyl Lewis X (LeX), was a crucial step for the activation of leukocyte function and supported the notion that activated platelets were actively involved in the inflammatory processes.

Two collagen-binding proteins, osteonectin and HSP47, are coordinately induced in transformed keratinocytes by heat and other stresses.

pSE48 was one of six clones selected by differential colony hybridization as a cDNA coding for mRNA expressed in parietal endoderm-like F9 cells and not in primitive endoderm-like F9 cells. It was sequenced and identified as a segment of mouse osteonectin (SPARC) cDNA. We found osteonectin to be heat-inducible in some cells. Expression and secretion of osteonectin were then investigated using mouse (Pam 212) and human (HSC-1) keratinocyte cell lines. Both the mRNA levels and the secretion of osteonectin increased concurrently when Pam and HSC-1 cells cultured in low calcium medium were exposed to various stresses including heat shock and treatment with sodium arsenite or L-azetidine-2-carboxylic acid. Another collagen-binding stress protein, HSP47, was also found to be expressed, synthesized, and stress-inducible in the keratinocyte cell line. The degree of HSP47 induction by various stresses was not so prominent as that of HSP70 but greater than that of osteonectin. The time courses of osteonectin and HSP47 induction by heat shock were similar to each other and distinct from HSP70; they were slower and more persistent than HSP70. We identified a heat shock element-like sequence in the promoter region of the mouse and bovine osteonectin genes. This sequence might participate in the stress induction of osteonectin. Thus, osteonectin and HSP47 share another common feature, stress-inducibility, as well as collagen-binding capacity and inducibility through differentiation, although they are quite distinct in their amino acid sequence and distribution.

Quantitative distribution of rat brain monoamine oxidase A by [14C]clorgyline autoradiography.

The distribution of functionally active monoamine oxidase type A (MAO-A) was investigated by in vivo quantitative autoradiography using [14C]clorgyline in normal, conscious rat brain. [14C]clorgyline was synthesized by the methylation reaction of N-desmethylclorgyline using [14C]methyliodide. Sixty minutes after [14C]clorgyline administration (1.58 MBq/animal i.v.), the brains were removed and prepared for autoradiography by washing the brain sections with 5% trichloroacetic acid solution to remove the nonbinding free tracer. The amount of MAO-A was calculated from the regional acid-insoluble tissue radioactivity and the specific activity of the tracer. The highest amount of MAO-A (5.84 nmol/g tissue) was found in the locus coeruleus. The interpeduncular nucleus, habenular nucleus, fasciculus retroflexus, and solitary tract nucleus possessed over 1.6 nmol/g tissue of MAO-A. Among 23 regions of interest, the lowest amount of MAO-A (0.37 nmol/g tissue) was found in the globus pallidus. The findings of this study suggest that the pattern of MAO-A parallels both in neuroanatomical distribution and in density that of norepinephrine and serotonin innervation. The MAO-A concentration was, however, relatively low in the dopamine-related areas. This corresponded to the previous results obtained by histochemical analysis. In addition, among the white matter structures, a high amount of MAO-A was found specifically in the fasciculus retroflexus.

Isolation and partial characterization of crystal matrix protein as a potent inhibitor of calcium oxalate crystal aggregation: evidence of activation peptide of human prothrombin.

In order to clarify the characteristics of crystal matrix protein (CMP), which exhibits a remarkable affinity for calcium oxalate crystals and may be important in stone pathogenesis, we have isolated CMP from macromolecular matrix substances of newly-formed calcium oxalate crystals. Purification of CMP consisted of calcium oxalate crystal formation, dissolution of crystals, electrodialysis, anion exchange chromatography and high-performance liquid chromatography. CMP showed the protein band of 31 kDa in sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The N-terminal amino acid sequence of CMP was identical to that of human prothrombin. Both anti-CMP polyclonal antibody and anti-human prothrombin antibody cross-reacted well with human prothrombin and CMP in Western blotting. Its amino acid composition and its molecular weight of 31 kDa strongly suggest that CMP is the activation peptide of human prothrombin.

Structural similarity and diversity of sulfotransferases.

In the present study, four new forms of aryl sulfotransferase cDNAs have been isolated and their structures determined. A compilation of primary structures of 16 different sulfotransferases, including enzymes metabolizing endogenous chemicals and xenobiotics, showed a considerable extent of similarity among bacterial, plant and mammalian species, and indicates that these enzymes constitute a supergene family. Aryl sulfotransferase and estrogen sulfotransferase are shown to belong to a single gene family (ST1) which consists of at least four subfamilies, whereas, based on the sequence similarity, hydroxysteroid sulfotransferases constitute a distinct family (ST2). Little or no clear similarity was observed between the primary structures of enzymes N-sulfating aminosugars and those sulfating hydrophobic chemicals such as phenols, alcohols or amines, indicating that both types of enzymes diverged early in their evolutionary history. Two regions in the C-terminal parts are, however, conserved among all enzymes examined, which suggests a possibly essential role of these sites for the binding of a PAPS cofactor or for sulfate transfer.

Localization of a low Mr GTP-binding protein, rap1 protein, in plasma membranes and secretory granule membranes of rat parotid gland.

Subcellular fractions were prepared from rat parotid gland by sequential centrifugation, Percoll gradient centrifugation and divalent-cation precipitation, and the localization of a low Mr GTP-binding protein, rap1 protein (rap1p) was analyzed by immunoblotting using a specific antibody. rap1p was found to be located in apical and basolateral plasma membranes, and secretory granule membranes in rat parotid gland. On the other hand, the beta gamma subunits of heterotrimeric Gt protein was localized in plasma membranes but not in granule membranes.

Predictive factors for the histologic nature of residual tumor mass after chemotherapy in patients with advanced testicular cancer.

OBJECTIVES: To further improve the accuracy of the prediction of histologic findings in the residual mass after chemotherapy in patients with metastatic germ cell tumors, various predictive factors were investigated. METHODS: A total of 23 patients with advanced testicular cancer underwent combination chemotherapy based on cisplatin following surgical removal of residual tumor mass in 20 patients and observation in 3. RESULTS: Complete fibrosis and/or necrosis was found in 6 (30%) operative specimens, teratoma in 3 (15%), and viable malignant tumor in 11 (55%). Three patients had no evidence of disease for a mean follow-up of 45 months. Five of the 6 patients and all of the 11 patients' results could be predicted by a combination of 4 factors, including absence of teratomatous element in the primary tumor, greater than 85% tumor reduction rate after chemotherapy, normal alpha-fetoprotein and beta-subunit of human chorionic gonadotropin levels after chemotherapy, and actual half-life of alpha-fetoprotein less than 7 days. Among these factors the tumor reduction rate and the actual half-life of alpha-fetoprotein were the most important predictions statistically on multivariate analysis. With the use of these criteria, the sensitivity rate, specificity rate, and overall predictive value of diagnosis for existence of viable malignant tumors were 83.3%, 100%, and 94.1%, respectively. CONCLUSIONS: By determining these factors it seems possible to avoid surgical removal of the postchemotherapy residual mass in approximately 30% of patients with advanced metastatic testicular cancer.

Structure of the N-terminal SH3 domain of GRB2 complexed with a peptide from the guanine nucleotide releasing factor Sos.

Src-homology 3 (SH3) domains mediate signal transduction by binding to proline-rich motifs in target proteins. We have determined the high-resolution NMR structure of the complex between the amino-terminal SH3 domain of GRB2 and a ten amino acid peptide derived from the guanine nucleotide releasing factor Sos. The NMR data show that the peptide adopts the conformation of a left-handed polyproline type II helix and interacts with three major sites on the SH3 domain. The orientation of the bound peptide is opposite to that of proline-rich peptides bound to the SH3 domains of Abl, Fyn and p85.

Infratentorial infarction: correlation of MR findings with neurological and angiographical features.

Using magnetic resonance imaging, we reviewed 141 infratentorial infarcts in 81 consecutive cases: 65 infarcts were seen in the paramedian pons, while 18 in the posterior inferior cerebellar artery (PICA) territory and 17 in the watershed area between PICA and superior cerebellar artery. No comparable sign or symptom was observed for 44 (31.2%) infarcts, whereas 66 (46.8%) infarcts appeared symptomatic and 31 (22.0%) infarcts were regarded as equivocal mainly due to the coexisting supratentorial lesions or non-localizing symptoms. The frequent coexistence of basal ganglionic small infarcts in those with infratentorial small (< 15 mm) infarcts implicated their common pathogenetic background. The fact that atrial fibrillation was seen in 33.3% of those with large (> 15 mm) infarcts whereas it was seen in only 6.5% of those with small infarcts may suggest a cardiogenic embolism as a possible cause of infratentorial large infarcts. Major artery occlusive lesion was seen in 15 of 22 cases with cerebellar infarction, whereas no occlusive lesion was seen in the majority of cases with pontine small infarcts. With MR imaging, infratentorial infarcts were detected more frequently than in the previous studies based on X-ray CT, and they can be considered as a benign condition.

Monoclonal antibody specifically reacting against 73-kilodalton heat shock cognate protein: possible expression on mammalian cell surface.

The heat shock proteins (hsp) are regarded as being immunogenic to the animal hosts. Although certain hsp are suggested to be expressed on the cell surface, further evidence for the cell surface expression of these proteins has been required. In this article we report the development of a MAb NT22. This antibody reacted with ATP-binding proteins (which contain a large amount of 70-kDa hsp family) of HeLa cells, and with purified bovine 70-kDa hsp. It did not react with the E. coli lysate, but clearly reacted with the recombinant rat hsc73. However, NT22 failed to react with hsp72. Furthermore, stress treatment of cells also indicated that considerable amounts of NT22-defined antigen translocated into the nucleus from the cell cytoplasm. These results suggest that NT22 is a novel MAb that reacts specifically to the mammalian hsc73. Moreover, this antibody could detect the constitutive and stress-induced cell surface expression of its relevant antigen. It is expressed preferentially on EBV-transformed B cell and certain epithelial cancer cell lines. However, resting B cells did not express this antigen on the cell surface. These data indicate that hsc73 could be expressed on the cell surface of certain cells, and suggest that hsc73 may interact with the host immune system.

Primary structure and molecular basis of polymorphic appearance of an acetyltransferase (AT-II)* in hamsters.

Three hamster clones (clones 1, 2 and 3) were isolated from a genomic library constructed from a homozygote rapid acetylator using a cDNA (hamAT-101) of a monomorphic acetyltransferase (AT-I) as a probe. Clone 1 (13 kbp) was found to contain a gene corresponding to AT-I. The entire coding region was located in an exon and completely identical to that of AT-I cDNA. Clones 2 and 3 (14.5 and 15 kbp) each contained identical information to the AT-I-related protein (AT-B protein). The intronless coding region shared 83.7% of sequence similarly to the AT-I cDNA, and its length was identical to that of the AT-I cDNA. Clone 2 also included a nucleotide sequence identical to the 3'-portion of the AT-I gene, which is located 5'-upstream of the AT-B gene. Restricted fragment lengths of clone 3, which encompassed the entire coding region was expressed in COS-1 cells. The expressed protein migrated at a position identical to that of AT-II purified from a hamster liver on Western blots. AT-B-expressed protein catalysed acetyl CoA-dependent N-acetylation of 2-aminofluorene and p-aminobenzoic acid, but had marginal activities for O-acetylation of 2-N-hydroxyamino-6-methyl-6-methyldipyrido[1,2-a:3',2'-d]imidazole and N-hydroxyarylacetamide-dependent N-acetylation of 4-aminoazobenzene. These results are in good agreement with the data of AT-II purified from hamster livers, indicating that the AT-B gene encodes a polymorphic acetyltransferase (AT-II) in hamsters. Although the AT-II protein was undetectable in slow acetylators, specific mRNA, hybridizing with a selective oligonucleotide probe for the AT-II gene (AT-B), was detected in livers of both homozygous acetylators. Analysis of genomic DNA of a homozygous slow phenotype hamster indicates that AT-II DNA from the slow phenotype has a point mutation which causes premature termination at the 243th (Arg to stop codon) position of the deduced amino acid sequence. PCR-RFLP analysis further confirmed that the point mutation conferred a defective AT-II protein in slow phenotype hamsters.

Effect of long-term administration of probucol on triglyceride turnover in rats.

BACKGROUND: Our previous observation revealed that probucol, a well known cholesterol-lowering drug, can improve impaired catabolism of plasma triglyceride in streptozotocin-diabetic rats without reducing plasma glucose. The present study was conducted using Triton WR1339 and endogenously radiolabeled lipoproteins in order to examine the mechanisms whereby the drug can stimulate triglyceride removal from the circulation in rats independent of glucose metabolism. METHODS: The short- and long-term effects of probucol on triglyceride turnover were examined in rats. Male Wistar rats received a diet containing probucol (1%) for 2 weeks and 4 months. Triglyceride turnover was estimated in rats using Triton WR1339. RESULTS: After 2 weeks of probucol administration, triglyceride and cholesterol concentrations in plasma and in the very-low-density lipoprotein (VLDL) fraction were suppressed slightly but significantly in the probucol-administered rats compared with standard chow-fed control rats. However, triglyceride secretion rates were similar in the two groups. Probucol-administered rats showed markedly suppressed triglyceride concentration in plasma and the VLDL fraction compared with chow-fed control rats after 4 months of administration. However, triglyceride secretion rate was significantly increased in rats given probucol. The radioisotope experiment revealed that probucol-administered rats can remove VLDLs (lipoproteins either from probucol-administered or control rats) from their system faster than normal rats and that probucol-containing VLDL can be removed from the circulation of both groups of rats more rapidly than normal VLDL. Therefore, probucol-administered rats as well as probucol-containing VLDL (lipoprotein from probucol-administered rats) are responsible for accelerated catabolism of VLDL-triglyceride. CONCLUSION: No significant differences in the lipid composition of newly-secreted VLDL particles between control rats and those given probucol were noted after Triton injection. The precise mechanism behind increased triglyceride secretion rate in rats after long-term administration of probucol is not known. However, we concluded from the present data that long-term administration of probucol results in markedly accelerated VLDL-triglyceride turnover without affecting lipid composition of newly secreted VLDL particles in normal rats.

Activation of 2-hydroxyamino-1-methyl-6-phenylimidazo[4,5-b] pyridine by cDNA-expressed human and rat arylsulfotransferases.

Sulfation plays an obligatory role in the activation of N-hydroxy derivatives of carcinogenic arylamine (amide)s and heterocyclic amines. We found that the hepatic sulfotransferase-mediated covalent binding of 3H-labeled 2-hydroxyamino-1-methyl-6-phenylimidazo[4,5-b] pyridine (N-OH-PhIP) to calf thymus DNA was 3.3 and 12.9 times higher with human cytosol preparation than with male and female rat cytosol preparations, respectively, in the presence of 3'-phosphoadenosine 5'-phosphosulfate. To assess the activating capacities of individual phenol-sulfating sulfotransferases, five different forms, human ST1A2 and ST1A3 and rat ST1A1, ST1B1 and ST1C1, were expressed in heterologous cells. All five sulfotransferases mediated the activation of N-OH-PhIP to DNA-bound products. The extents of the binding, however, differed considerably among these forms. Human ST1A2 and ST1A3 mediated the activation of N-OH-PhIP at 5.2- and 6.2-fold higher rates than did rat ST1C1, a main N-hydroxy-2-acetylaminofluorene-activating sulfotransferase, in rat liver. Extents of the binding of N-OH-PhIP in human hepatic cytosols of different individuals were positively correlated with the contents of immunoreactive ST1A2/3. These results suggest a potential role of human liver sulfotransferases in N-OH-PhIP activation. In contrast, the low sulfotransferase-mediated activation of N-OH-PhIP in rat liver is consistent with the lack of PhIP hepatocarcinogenicity in this species.

Haemoglobin inhibits GTP-hydrolysis and GDP/GTP-exchange activities of a low M(r) GTP-binding protein, ras p21.

Haemoglobin was observed to inhibit the GDP/GTP-exchange activity of ras protein (ras p21) by measurement of [3H]GDP-dissociation activity in time- and dose-dependent manners. Haemoglobin also inhibited the [32P]GTP-hydrolysis activity of ras p21 time- and dose-dependently. These inhibitory effects of haemoglobin were lost after incubation of haemoglobin at 80 degrees C for 3 min. Globin showed limited inhibition on the [32P]GTP-hydrolysis activity of ras p21, and haemin had no effect, indicating that the ternary tetrameric structure of haemoglobin is essential for the inhibitory effects on ras p21 activities. Methaemoglobin also inhibited both [3H]GDP-dissociation and [32P]GTP-hydrolysis activities of ras p21 in a very similar manner to that by haemoglobin. The obtained results strongly suggest that haemoglobin suppresses the physiological function(s) of ras p21 in vivo inhibiting both [32P]GTP-hydrolysis and GDP/GTP-dissociation of ras p21 in erythrocytes.

Evolution of crossed cerebellar diaschisis in middle cerebral artery infarction.

To elucidate the evolution of crossed cerebellar diaschisis, cerebral oxygen metabolism was measured repeatedly by positron emission tomography (PET) in 35 consecutive patients with unilateral cerebral infarction within the territory of middle cerebral artery. The crossed cerebellar diaschisis was defined as significant when the laterality ratio of cerebral oxygen metabolism between the left and right cerebellar hemispheres exceeded the control range (mean +/- 2 standard deviations) as derived from 27 age-matched normal volunteers. Significant crossed cerebellar diaschisis was observed in 31 patients (89%) on the initial PET studies. Of these 31 patients, 23 with infarcts involving the frontal sensorimotor cortex persistently had crossed cerebellar diaschisis up to 5 years after onset, whereas the diaschisis disappeared in 8 patients with smaller infarcts mainly in the frontal or parietal lobe without recovery of oxygen metabolism in the infarcted areas. These present results suggest that crossed cerebellar diaschisis can exist persistently even in the late stage in those having a lesion involving the cortical pontine-cerebellar pathways.
