Prediction of three-dimensional structure of Escherichia coli ribosomal RNA.

A model for the tertiary structure of 23S, 16S and 5S ribosomal RNA molecules interacting with three tRNA molecules is presented using the secondary structure models common to E. coli, Z. mays chloroplast, and mammalian mitochondria. This ribosomal RNA model is represented by phosphorus atoms which are separated by 5.9 A in the standard A-form double helix conformation. The accumulated proximity data summarized in Table 1 were used to deduce the most reasonable assembly of helices separated from each other by at least 6.2 A. Straight-line approximation for single strands was adopted to describe the maximum allowed distance between helices. The model of a ribosome binding three tRNA molecules by Nierhaus (1984), the stereochemical model of codon-anticodon interaction by Sundaralingam et al. (1975) and the ribosomal transpeptidation model, forming an alpha-helical nascent polypeptide, by Lim & Spirin (1986), were incorporated in this model. The distribution of chemically modified nucleotides, cross-linked sites, invariant and missing regions in mammalian mitochondrial rRNAs are indicated on the model.

An analysis of postsynaptic potentials of tectal neurons of the frog: correlation with impulses recorded from the terminals of retinotectal afferents.

The purpose of this study is to examine the synaptic action between terminals of retinal ganglion cell axons and tectal neurons. To accomplish this, an extracellular single unit identified as retinotectal fiber was first isolated from the superficial layer of the optic tectum and intracellular responses were recorded from a tectal neuron in the vicinity of the extracellular recording electrode. "On-off" retinal fibers and both E-E (EPSP at "on" and "off" of diffuse light) and EI-EI type (EPSP-IPSP combination at "on" and "off" of diffuse light) tectal neurons were selected for the pre- and postsynaptic pair. Postsynaptic responses to a small moving square were averaged by triggering with the isolated presynaptic impulses. The latency of the resultant EPSPs indicated that most of the E-E and EI-EI type tectal neurons were monosynaptically activated by "on-off" retinal fibers. One of the E-E type tectal neurons was identified as a large ganglionic neuron in layer 8.

The transmembrane segment of the human Na,K-ATPase beta-subunit acts as the membrane incorporation signal.

The human Na,K-ATPase beta-subunit is anchored to the membrane by a single stretch of 28 hydrophobic amino acids; the hydrophilic amino terminus faces the cytoplasm and the carboxyl terminus is exoplasmic. Glycosylation and insertion of the Na,K-ATPase beta-subunit into the endoplasmic reticulum membrane are shown to be co-translational and SRP-dependent. The hydrophilic amino terminus is not required for the membrane insertion. The membrane-anchor domain is necessary for membrane insertion, and a 16 amino acid stretch has been identified as an element sufficient for the insertion.

The third type of alpha-subunit of Na,K-ATPase.

The cDNA encoding the third type of alpha-subunit of NaK-ATPase was found in rat brain cDNA library. The deduced amino acid sequence was very similar to those of alpha- and alpha plus-subunits. The third one was suggested to be highly ouabain-sensitive from its aminoacid sequence. From Northern blot analysis the third type of the subunit was not expressed in adult rat kidney and heart.

[Adenocarcinoma of the kidney in a 20-year-old female].

A 20-year-old female, who was one of identical twins, was transferred from an another hospital for further therapy of a right renal tumor. There was no history of hematuria, flank pain. Routine laboratory studies were within the normal range. On excretory urogram, the tumor, 7 x 9 cm in diameter, was demonstrated in lower pole of the right kidney. A selective renal arteriography revealed that the central area of the tumor was rather hypovascular, but, some neovascularity was seen in the peripheral area. Pedal lymphography, chest X-ray and bone survey showed normal findings. A right nephrectomy was carried out. Grossly, the tumor in the lower pole of the kidney was well demarcated by a pseudocapsule and had massive necrotic areas. Pathological sections revealed a granular cell adenocarcinoma of the right kidney. No extension into the peripheric fat was present. The patient remains well and free of recurrent tumor one year post-operatively. We discussed the diagnosis and prognosis of renal cell carcinoma in young adults in the literature.

Congenital internal mammary artery-to-pulmonary artery fistulas: a case report.

A case of congenital internal mammary artery-to-pulmonary artery fistulas is reported. The patient was a 7-year-old boy in whom this condition was suspected from an abnormal shadow in the left upper lung field upon chest X-ray examination and continuous murmur. A definite diagnosis was made by selective angiography of the internal mammary artery. This condition is extremely rare. Our patient is the youngest among the 16 cases which have been reported so far in the English literature. Surgical treatment may be indicated because there is a risk of rupture of the fistula, endarteritis, and congestive heart failure.

Regenerating retinal fibers of the goldfish make temporary and unspecific but functional synapses before forming the final retinotopic projection.

Many investigators have examined the retinotectal projection following regeneration, but the mapping technique used in these studies mainly involved recording action potentials from presynaptic terminals. Hence it was not possible to analyse the postsynaptic phenomena underlying the target finding process of the retinal fibers. In the present study, the process of making functional synapses was examined using field potentials generated by small spots of light fixed in the visual field. The results show that regenerating retinal fibers first make functional but temporary, unspecific and diffuse synapses before reaching the target area to form a sharpened retinotopic projection. It is suggested that the formation of synapses subserves an important role in the target finding process.

Primary structures of two types of alpha-subunit of rat brain Na+,K+,-ATPase deduced from cDNA sequences.

A rat brain cDNA library was screened by using as a probe a fragment of cDNA encoding the alpha-subunit of human Na+,K+-ATPase. Two different cDNA clones were obtained and analyzed. One of them was concluded to be a cDNA encoding the alpha-subunit of the weakly ouabain-sensitive rat kidney-type Na+,K+-ATPase. The deduced amino acid sequence consists of 1,018 amino acids. The alpha-subunit of the rat kidney-type Na+,K+-ATPase shows 97% homology in amino acid sequence with the alpha-subunit of human, sheep, or pig enzyme and 87% with that of Torpedo. Based on a comparison of the amino acid sequence at the extracellular domain of the alpha-subunit between weakly ouabain-sensitive rat kidney-type enzyme and the ouabain-sensitive human, sheep, pig, or Torpedo enzyme, it was proposed that only two significant amino acid replacements are unique to the rat kidney-type alpha-subunit. Another cDNA clone obtained showed 72% homology in nucleotide sequence with the former cDNA coding the alpha-subunit of the rat kidney-type Na+,K+-ATPase and the deduced amino acid sequence exhibited 85% homology with that of the alpha-subunit of rat kidney-type Na+,K+-ATPase.

Epidemiology of cataract--clinical evaluation with retro-illumination photography.

Retro-illumination photography was utilized for epidemiological studies of cataract. Diagnosis of cataract was made by the existence of shadows on the photographs obtained. In the prevalence study, prevalence rate of cataract was 35.7% in all (30.2% in male and 45.1% female) 1,017 persons (643 males and 347 females). Prevalence rates were always higher in females than in males, and in diabetics as compared with non-diabetics, in all decades of life. The progression study in 338 persons (144 males and 194 females) revealed that maturing courses of cataract were most rapid in diabetic females in the fifth to seventh decades of life.
