Effects of protease inhibitors on vasopressin transport across rat alveolar epithelial cell monolayers.

The transepithelial transport of arginine vasopressin (AVP) across cultured rat alveolar epithelial cell monolayers was studied. At 0.1 nM donor [125I]AVP, the radiolabel flux measured in the apical-to-basolateral (AB) direction was about 10 times greater than that in the reverse (BA) direction. HPLC analyses of the basolateral receiver fluid collected at the end of these flux measurements showed that about 97% of the total [125I]label represented subspecies of AVP, whereas the apical receiver fluid contained largely intact AVP (approximately 85% of total [125I]label). Both donor fluids contained virtually no degradation products of AVP (> 99%). In the presence of an excess 0.1 mM unlabeled AVP in the apical donor fluid, the Papp for radiolabeled AVP in the AB direction was decreased by approximately 68%, while the fraction of intact AVP in the basolateral receiver fluid was increased six-fold as compared to that observed at 0.1 nM [125I]AVP alone. Under this condition, the flux of intact AVP was approximately the same in both directions. When the concentration of apical camostat mesylate, an aminopeptidase inhibitor, was varied from 0 to 2 mM, the radiolabeled flux in the AB direction (with 0.1 nM [125I]AVP in the donor fluid) was significantly decreased in a dose-dependent manner, yielding commensurably elevated concentrations of intact AVP in the basolateral receiver fluid. In contrast, leupeptin (0.5 mM), a serine protease inhibitor, was without effect. These data, taken together, suggest that apically-presented AVP undergoes proteolysis (most likely by peptidases localized at apical cell membranes of alveolar epithelium.(ABSTRACT TRUNCATED AT 250 WORDS)

Increased production of eicosanoids, TXA2, PGI2 and LTC4 in experimental spinal cord injuries.

Arachidonate metabolites have many kinds of bioactivities. Thromboxane A2 (TXA2) stimulates platelet aggregation and vasoconstriction, whereas prostaglandin I2 (PGI2) antagonises its activities. Thromboxane B2 (TXB2) and 6-keto-prostaglandin F1 alpha (6-keto-PGF1 alpha) are determined in biological materials. Production of TXB2, 6-keto-PGF1 alpha and leukotriene C4 (LTC4), which have potent vascular permeability, was measured by radioimmunoassay in experimental spinal cord injured animals. TXB2 level in the rat spinal cord reached a peak concentration of 133.6 +/- 3.8 pmol/g cord, and 6-keto-PGF1 alpha increased to 26.2 +/- 11.7 pmol/g cord 5 minutes after the injury. There was good correlation between TXB2 production and vascular damage as monitored by fluorescein uptake. When OKY-046 ((E)-3-[4-(1-imidazolylmethyl) phenyl]-2-propenoic acid), which selectively inhibits TXA2 synthetase activity, was administered 10 minutes before injury, the increase in TXB2 production was inhibited by more than 80%, but the degree of vascular damage was reduced by only 40%. In the guinea pig spinal cord, LTC4 levels reached a peak concentration of 2.2 +/- 0.4 pmol/g cord 10 minutes after compression, while that of TXB2 reached 146.8 +/- 6.2 pmol/g cord. The increased production of TXB2 was correlated with the degree of compression injury while that of LTC4 production did not. These findings suggest that vasoactive eicosanoids, TXA2, PGI2 and LTC4, play important roles in secondary damage following spinal cord injury, although their roles may be different among species of animals.

Crocidolite asbestos increased 8-hydroxydeoxyguanosine levels in cellular DNA of a human promyelocytic leukemia cell line, HL60.

Crocidolite, one of the most carcinogenic asbestos fibers, induces the release of reactive oxygen species (ROS) from neutrophils and macrophages. Using HPLC combined with electrochemical detection, we determined that 8-hydroxydeoxyguanosine (8OHdG), a molecule typical of mutagenic oxidative DNA damage, was induced in the cellular DNA of a human promyelocytic leukemia cell line, HL60, incubated with crocidolite. Crocidolite increased 8OHdG in the cellular DNA of phorbol myristate acetate (PMA)-differentiated HL60, which phagocytosed crocidolite. PMA-differentiated HL60 released ROS spontaneously, as determined by ESR with 5,5-dimethylpyrrolone-N-oxide as a spin trap. However, the release of ROS from the cell line did not increase after the addition of crocidolite. The addition of superoxide dismutase at a sufficient concentration to scavenge ROS released from the cell did not inhibit the 8OHdG increase induced by crocidolite. Cytochalasin B, which inhibited phagocytosis, did not inhibit the release of ROS. However, it inhibited the crocidolite-induced 8OHdG increase by 48.3%. Contrary to PMA-differentiated HL60, undifferentiated HL60 neither phagocytosed crocidolite nor showed a crocidolite-induced increase in 8OHdG formation. The 8OHdG increase induced by crocidolite was not correlated with ROS release, but with the internalization of crocidolite, suggesting that the increase was not due to an increase in ROS release from the cell but was due to the conversion of relatively inert ROS to highly reactive ROS, such as hydroxyl radicals, by crocidolite that was internalized and close to DNA.

Evaluation of 8-hydroxydeoxyguanosine, a typical oxidative DNA damage, in human leukocytes.

8-Hydroxydeoxyguanosine (8-OHdG) is a typical form of oxidative DNA damage which causes mutation in vitro and in vivo. We investigated potential factors confounding 8-OHdG determination and, based on the results, then determined the 8-OHdG levels in human peripheral blood leukocytes. 8-OHdG was detected electrochemically after extraction of DNA from the cells without the use of phenol by a DNA extractor under helium. In the preliminary experiments, the mononuclear leukocytes (MN) in blood samples obtained from 19 laboratory workers and students were separated from the polymorphonuclear leukocytes (PMN) with Mono-Poly resolving medium. The 8-OHdG in the MN (1.157 +/- 0.414 molecules per 10(5) deoxyguanosine) did not differ significantly from that in PMN (1.131 +/- 0.418). The effect of red blood cells (RBC) on 8-OHdG formation during DNA extraction was then examined by adding RBC to the human lymphoblastoid cell line FA72. Addition of RBC at ratios of up to 4 RBC per FA72 cell did not increase 8-OHdG levels, while addition at a RBC/FA72 cell ratio of 20 increased the 8-OHdG level 1.43-fold over that without RBC. The potential effect of histidine, a scavenger of both hydroxyl radicals and singlet oxygen, on reduction of artificial 8-OHdG formation during DNA extraction was examined during DNA extraction in the human promyelocytic leukemia cell line HL60. Addition of His decreased the 8-OHdG level dose-dependently (30% reduction at 30 mM His concentration). Based on these results, we determined the 8-OHdG levels in human leukocyte samples obtained from 79 healthy male factory workers aged 24-59 years. The leukocyte fraction containing both MN and PMN was separated from RBC with Mono-Poly resolving medium and DNA was extracted from the leukocytes in the presence of 30 mM His. The mean 8-OHdG level in these samples was 1.072 +/- 0.230. To evaluate the reliability of the assay, FA72 was used as a standard sample in all assay determinations and the 8-OHdG levels of both the leukocyte samples and the FA72 sample(s) were measured in each determination. The inter- and intra-assay coefficients of variation (CV) were calculated to be 14.4% (n = 14) and 3.9-13.5% (n = 3-5 per assay) respectively. The 8-OHdG level was measured twice in 19 leukocyte samples; the value at the first determination was not correlated with that at the second determination. The range of 8-OHdG levels in the samples was relatively small compared with the CV of the assay.(ABSTRACT TRUNCATED AT 400 WORDS)

A new approach to the evaluation of broad-spectrum sunscreens against ultraviolet and visible light-induced delayed tanning.

Protection against UV and visible light-induced delayed tanning (DT) and immediate tanning (IT) of two broad-spectrum sunscreens with the same sun protection factor was investigated. Protection factors (PFs) against DT (24 h and 7 days) of the sunscreens differed significantly, but PFs against IT did not. This study indicates that using PF against 24 h DT is more appropriate for an evaluation of broad-spectrum sunscreens against UVA and visible light than IT.

Linkage between severe atopy and chromosome 11q13 in Japanese families.

Atopy, characterised by allergic asthma and rhinitis, is due to increased IgE responses to common aeroallergens. An Oxford group has described maternal inheritance of atopy, where there is significant linkage between IgE responsiveness and a VNTR marker D11S97 and a CA microsatellite within a candidate gene, the high affinity IgE receptor beta subunit (Fc epsilon RI beta), on chromosome 11q. Attempts at independent replication have produced conflicting results. We therefore recruited 270 atopic asthmatic probands in a Japanese community population for genetic linkage analysis. Four families, each with more than 15 meioses and a clear phenotype for atopy, were selected for genetic analysis. Atopy was defined as presence of all of raised total IgE, positive RAST and skin tests to three or more aeroallergens; non-atopy, as absence of all these criteria. Linkage analysis showed a maximum two-point lod score of 9.35 for D11S97 and Fc epsilon RI beta under the assumption of unequal rates of maternal and paternal recombination. Two families showed close genetic linkage with Fc epsilon RI beta with a pattern of maternal inheritance. These results from a Japanese population provide further evidence for genetic linkage between severe atopy and chromosome 11q13 and the likelihood of genomic imprinting at the locus.

Reduction of aluminium concentration in albumin products.

The aluminium concentration in albumin products has been reduced to < or = 100 ppb by an anion exchanger treatment. Furthermore, we have found that this low aluminium concentration in a albumin products has been maintained for extensive periods of time, when they are stored in de-alkalized soft glass containers.

Relationship between mean arterial pressure and muscle cell pH during forearm ischaemia after sustained handgrip.

Recent evidence suggests that there is a close correlation between the physiological responses to muscle chemoreflex and the decrease in intracellular pH during ischaemia after handgrip. This study evaluated whether the relationship is linear or has an apparent threshold. We measured muscle cellular pH through phosphorous nuclear magnetic resonance spectroscopy (31P-NMR), mean arterial blood pressure (MAP) and heart rate (HR) during ischaemia after sustained handgrip exercise at 50% of maximum voluntary contraction (MVC). Contraction was sustained for 15, 30, 45 and 60 s, followed by 2 min of circulatory arrest, respectively. Muscular pH during the ischaemia decreased linearly with increasing contraction time, from the base-line level of 7.11 +/- 0.03 units (U) to 6.98 +/- 0.03, 6.90 +/- 0.04, 6.72 +/- 0.06 and 6.54 +/- 0.06 U after 15-, 30, 45-, and 60-s contractions, respectively. The MAP was 86 +/- 2 mmHg at rest and did not change during the ischaemia after 15- and 30-s contractions. However, it significantly increased to 95 +/- 2 and 107 +/- 2 mmHg, after 45- and 60-s contractions, respectively. These data indicate that the relationship between MAP and pH is not a single linear relationship, showing one breaking point around the pH of 6.90 units. It suggests that the muscle chemoreflex has a clear threshold around 6.90 units of muscle pH, and below this pH, MAP increased linearly with decreasing muscle cellular pH.

Enhancement of parasympathetic cardiac activity during activation of muscle metaboreflex in humans.

We measured the changes in heart rate (HR) variability estimated from the standard deviation of the R-R intervals to evaluate cardiac parasympathetic tone noninvasively before and during activation of muscle metaboreflex induced by postexercise muscle ischemia. Eight healthy male subjects performed sustained handgrip at 50% maximal voluntary contraction followed by forearm occlusion. Mean arterial pressure, cardiac stroke volume, and ratio of cardiac preejection period to left ventricular ejection time (PEP/LVET) were also measured. During the 2-min occlusion after 60 s of handgrip with voluntary respiration, HR variability and mean arterial pressure were significantly increased from baseline (54.4 +/- 6.1 to 80.1 +/- 12.8 ms and 81 +/- 1 to 99 +/- 3 mmHg, respectively) and PEP/LVET was decreased from resting level of 0.404 +/- 0.022 to 0.363 +/- 0.036. During occlusion and recovery, HR did not change from baseline level in any experiment. There was no influence of occlusion itself or of cessation of exercise per se on any parameters. Although overall enhanced HR variability was seen, probably due to lower breathing frequency and larger tidal volume, similar results were also obtained from an experiment with controlled respiration, showing that the increase in HR variability was not due to the changes in tidal volume or breathing frequency during occlusion. In conclusion, the HR variability is increased during activation of the muscle metaboreflex induced by postexercise muscle ischemia in humans. This finding shows that the parasympathetic cardiac tone is enhanced during activation of the muscle metaboreflex in humans and balances enhanced cardiac sympathetic activity to result in an unchanged HR.(ABSTRACT TRUNCATED AT 250 WORDS)

[Evaluation of genotoxicity by DNA damages].

A great number of genotoxins are known to be present in the environment. These genotoxins induce many kinds of DNA damage, and may cause changes in genetic information and cancer. Therefore, evaluation of such DNA damage is important to keep genetic information stable and to prevent cancer. We reviewed here methods used to assay the damage and the importance of this DNA damage in mutation. DNA damage is categorized into two groups, strand breaks and base modifications. To assay DNA strand breaks, the alkaline elution method, pulsed-field gel electrophoresis and single-cell gel assay are being used. The alkaline elution method determines both single- and double-strand breaks sensitively and quantitatively. Pulsed-field gel electrophoresis preferentially determines double-strand breaks, and the results of the method appeal to the eye as an electrophoretogram. The single-cell gel assay could determine both single- and double-strand breaks even in a single cell, and could evaluate susceptibility to the damage in individual cells. To assay base modifications, methods to detect differences in the physicochemical properties of the damage (physicochemical methods), immunoassays and the 32P-postlabeling method are being used. Physicochemical methods are suitable for chemically minor and abundant modifications such as those in 8-hydroxyguanine, using high-performance liquid chromatography or gas chromatography/mass spectroscopy. Immunoassays, by the use of specific antibodies against DNA damage, are highly sensitive to DNA damage such as changes in O6-methylguanine and thymine glycol, and simple once the assay systems have been established.(ABSTRACT TRUNCATED AT 250 WORDS)

[A study of preventive medicine in relation to mental health among middle-management employees (Part 1)--Relationship between lifestyles and working-life satisfaction].

This study examined the cross-sectional relationship between health practices and working-life satisfaction, which we used as a subjective index of Quality of Life (QOL), based on data obtained from a survey of 3,928 middle-management employees (1,026 department chiefs and 2,902 section chiefs) in 110 major companies in 1990. The results are summarized as follows. (1) The section chiefs had more poor health practices than the department chiefs. The section chiefs appeared to have significantly more poor habits in terms of cigarette smoking, eating breakfast, nutritional balance, working hours, snacking, salt consumption, obesity, enjoying hobbies and physical status than the department chiefs. Surprisingly, 66% of department chiefs and 77% of section chiefs worked more than 10 hours per day. (2) Both the Health Practice Index (HPI) and working-life satisfaction tended to be higher as their ages were higher. (3) The department chiefs who had a poor nutritional balance, did not maintain an adequate stress level, had poor eating habits, poor sleeping habits and physical inactivity appeared to have lower working-life satisfaction. (4) The working-life satisfaction of the management staff was significantly affected by health practices, occupational stress, physical health status and position after controlling simultaneously for the other potential confounders. From these results, it may be concluded that the behavioral lifestyle change of the middle-management employee is an important factor to promote mental health as evaluated by working-life satisfaction.

Quantitative assessment of stressors and stress reaction: a review.

To survey methods for assessing stress and to examine the relationship of stress to health and the factors of lifestyle, we reviewed the literature on stressors and stress reactions. Firstly, we reviewed the representative methods for assessing stressors. Secondly, self-report questionnaires based on a theoretical model of occupational stressors were surveyed and factors in work stress were listed. Then, we reviewed stress reactions including physiological, psychological and behavioral reactions. Finally, we examined the relationship of perceived stress measured by a single question to mental health status determined by the General Health Questionnaire (GHQ-28), lifestyles and personality based on the Egogram in Transactional Analysis. It is suggested that we need to assess subjective aspects as well as using objective indices such as data from physical examinations, to evaluate the level of stress and to promote mental health.

Benign intracerebellar cyst without epithelial lining.

A case of benign intracerebellar cyst is reported in a 61-year-old woman who presented progressive neurological deficits. MRI and operative findings could not reveal any evidence of neoplastic growth. The biopsied cyst wall revealed normal cerebellar tissue. The simple cyst of the cerebellum is rare, thus we discuss the clinical features and etiology of this rare entity.

Neuroblastomas of the central nervous system--clinicopathological features of a cerebellar neuroblastoma in comparison with a metastatic cerebral neuroblastoma.

Two patients with neuroblastoma of the central nervous system are described. The first, a 10-month-old male infant with symptoms of vomiting and weakness of the right upper extremity, was found on magnetic resonance imaging (MRI) to have enhanced mass lesion in the left hemisphere, vermis of the cerebellum and part of brain stem. The second, a 3-year-old boy who had swelling of the right side of the head, was found on computed tomography (CT) to have enhanced mass lesion of the frontal hemisphere and frontal bone. Histological examination of the surgical specimen of both cases demonstrated neuronal differentiation. The clinicopathological features of cerebellar neuroblastoma in comparison with metastatic neuroblastoma of the frontal lobe were discussed.

Genotoxic potentials of lifestyles assessed by urinary mutagenicity.

The relationships between lifestyles and urinary mutagenicity were investigated by using blue rayon extraction from 33 healthy male workers' urine. Subjects were classified into three groups, as "good", "moderate", and "poor" according to their responses on a questionnaire regarding eight health practices (cigarette smoking, alcohol consumption, eating breakfast, hours of sleep, hours of work, physical exercise, caring about nutritional balance, mental stress). The better lifestyle groups exhibited the lower mutagenicity. Subjects in a "good" group showed significantly lower urinary mutagenicity than those both in a "moderate" (p < 0.05) and a "poor" (p < 0.05) groups at fraction number 1 to 3 that were given after ingesting fried beef. These tendencies also found at fraction number 8 to 9 that were given after smoking, although not significant. The lifestyles were significantly associated with the urinary mutagenicity, and the results suggested that not only particular lifestyle factor but also some combinations with smoking significantly enhanced with the urinary mutagenicity.

[A case of bronchogenic cyst at the anterior chest wall].

Bronchogenic cyst in the subcutaneous region is rare and only 5 cases including this case have been reported in Japan. A 2-year-old boy was admitted to our hospital because of congenital mass at the anterior chest wall. Ultrasonography showed a 29 x 9 mm sized well-defined tumor with homogeneous low echogenicity in the subcutaneous tissue. Extirpation of the tumor was performed. Histologically, ciliated columnar epithelia with the surrounding smooth muscle and seromucous glands were observed. Diagnosis of bronchogenic cyst was made. Postoperative course was uneventful and no tumor has recurred.
