Internalist stance and the physics of information.

Information is by itself intrinsically diachronic, whereas synchronic information conceived by referring to an invariant source matrix of information is no more than a theoretical artifact constructed in the limit of ignoring the historicity. Information is the basic attribute of local dynamics of internal measurement, in which any dynamics is taken to proceed through detection internal to interacting material participants of whatever kind. Information dynamics of a local character is semantic by virtue of constructing the global principle of conservation of energy from the inside. Material embodiment of information actualizes in the form of energy consumers acting toward the outside from their inside. Global dynamics such as mechanics is simply a phenomenon emerging from information dynamics.

Inhibition of platelet adhesion to collagen by monoclonal anti-CD36 antibodies.

Monoclonal anti CD36 antibodies capable of inhibiting platelet adhesion to collagen have not previously been identified. We have now prepared two groups of monoclonal antibodies. One group was prepared using, as immunogen, highly purified (99+%) CD36 prepared by a denaturing procedure. These antibodies (Mo series) reacted strongly with CD36 on protein blots but did not immunoprecipitate native CD36 from platelet lysates nor inhibit platelet adhesion to collagen. The second group of monoclonal antibodies (131 series) was prepared using CD36 purified to >95% by a non-denaturing procedure. These antibodies reacted with control platelets, but not Nak(a)-negative platelets which lack CD36, as measured by flow cytometry and immunoprecipitation. Three monoclonal antibodies of this latter group (131.4, 131.5 and 131.7) inhibited platelet adhesion to collagen in static systems under Mg2+ -independent conditions but had lit tle effect in the presence of Mg2+. 131.4 and 131.7 also inhibited adhesion to collagen using citrated whole blood in a parallel plate flow chamber at physiological shear rates (800s-1), whereas 131.5 was without effect. These are the first anti-CD36 monoclonal antibodies shown to be capable of inhibiting platelet adhesion to collagen and provide further evidence that CD36 plays a role in platelet-collagen interaction.

The treatment of hydrofluoric acid burns.

Hydrofluoric acid is a colourless or almost colourless, fuming liquid having a highly caustic and corrosive effect on organic tissue. Many kinds of hydrofluoric acid in concentrations are used in Industrial processes and laboratory uses because of its useful chemical properties. Hydrofluoric acid causes severe and painful burns to the skin and eyes. In this paper, the immediate symptoms, the typical treatments in the literature, first aid and therapy of hydrofluoric acid burns are reviewed.

Involvement of sigma-receptors in the increase in contraction of mouse vas deferens induced by exogenous ATP.

The effects of sigma-receptor ligands on the twitch contraction elicited by the exogenous application of adenosine 5'-triphosphate (ATP) in the unstimulated mouse vas deferens were studied. (-)-Pentazocine, 1,3-di(2-tolyl)guanidine(DTG) and two pairs of optical isomers of 3-(3-hydroxyphenyl)-N-(1-propyl)piperidine(3-PPP) and N-allylnormetazocine (SKF-10,047) potentiated the exogenous application of ATP-induced twitch-type contraction in a concentration-dependent manner, while (+)-pentazocine did not affect it. The order of potentiating ability was: (+)-3-PPP > (-)pentazocine > (-)-SKF-10,047> DTG > (-)-3-PPP > (+)-SKF-10,047. On the other hand, haloperidol and rimcazole, putataive sigma-receptor antagonists, suppressed this twitch contraction. In addition, these antagonists significantly blocked the (+)-3-PPP- and (-)-pentazocine-induced potentiation at concentrations which did not affect contractions per se. These findings indicate that the exogenous application of ATP-induced twitch contraction in the mouse vas deferens is regulated by sigma-receptors. In addition, the present ranking order suggests that the sigma-receptor potentiating the ATP-induced twitch contraction at post-junctional sites may differ from the sigma 1- and/or sigma 2-receptor subtypes.

Evaluation of 6.5-mm BENT incision to reduce postoperative astigmatism.

We examined surgically induced astigmatism following 6.5-mm incisions between 9 and 12 o'clock (BENT incision) in 50 cases with cataracts who underwent phacoemulsification-aspiration and 6-mm intraocular lens implantation. The surgically induced astigmatism (n = 50) shifted to against-the-rule astigmatism (ATR; -0.03 +/- 0.95 dpt) 1 day after the operation and then tended to shift to with-the-rule astigmatism (WTR). The results of a group with preexisting ATR (> 2 dpt; n = 4) were similar to the results of the aforementioned group, whereas the results of a group (n = 7) with preexisting WTR (> 2 dpt) were different. Astigmatism in this group shifted to ATR of about 1.0 dpt. This study suggests that the BENT incision procedure can reduce both preexisting WTR > 2.0 dpt and preexisting ATR > 2 dpt.

Indoor pollution from heating.

The concentrations of indoor pollutants generated from types of heaters were measured in a model room of 20m2 in area and 45m3 in capacity. We used six different heaters: three kerosene heaters of different types, town and propane gas heaters, and an electric heater. Three ventilation conditions were introduced into each experiment: non-ventilation, fan-on ventilation with closed door and fan-off ventilation with half-opened door. The results obtained by heating under non-ventilation condition were as follows: The concentrations of NO2 and CO2 were comparatively high and the values obtained from all the heaters except the electric heater exceeded the 1-hr Environmental Quality Standards, Japan (EQS NO2: 0.04-0.06 ppm) and the Building Sanitation Management Standards, Japan (BSMS CO2: 1,000 ppm), respectively. The CO concentration emitted from reflection kerosene and town gas heaters slightly exceeded the BSMS (10 ppm). The concentrations of suspended particulate matter and polynuclear aromatic hydrocarbons showed an increasing tendency during the use of kerosene-fueled heaters. Under two ventilating conditions, NOx concentration decreased to less than a third in comparison with non-ventilating condition.

Enhancement of acetylcholine release by SA4503, a novel sigma 1 receptor agonist, in the rat brain.

We found that 1-(3,4-dimethoxyphenethyl)-4-(3-phenylpropyl) piperazine dihydrochloride (SA4503), a potent and selective sigma 1 receptor agonist, significantly enhanced the cerebral acetylcholine (ACh) release in the rat using in vivo brain microdialysis technique. Interestingly, the significant enhancement of ACh release elicited by SA4503 was observed in the rat frontal cortex and hippocampus, although the striatal ACh release was unchanged. This cortical ACh release was fully reversed by haloperidol, a prototype sigma receptor antagonist, or by N, N-dipropyl-2-(4-methoxy-3-(2-phenylethoxy)phenyl)ethylamine monohydrochloride, a putative sigma 1 receptor antagonist. In addition, this enhanced ACh release by SA4503 was inhibited by tetrodotoxin, a Na+ channel blocker. However, tetrahydroaminoacridine, an acetylcholinesterase inhibitor, significantly increased the extracellular ACh level in the rat frontal cortex and weakly increased the hippocampal level. This compound also showed the significant increase of extracellular ACh level in the rat striatum. Moreover, tetrahydroaminoacridine markedly produced cholinomimetic side-effects, such as hypothermia, tremor, miosis and lacrimation. However, SA4503 did not produce these cholinomimetic side-effects. These findings suggest that SA4503 enhances the ACh release that is mediated through a novel mechanism, namely sigma 1 receptor subtype. Furthermore, SA4503 has regional differences in the enhancement of cerebral ACh release, and did not produce cholinomimetic side-effects. These profiles are different from tetrahydroaminoacridine.

[Neuropharmacological effects of sigma receptor ligands: anxiolytic, anti-amnesic and neuroprotective effects].

There is evidence for the existence of two classes of sigma binding sites, termed "site 1" and "site 2", that are distinct from opioid and PCP receptors. Sigma receptor ligands may be useful in the treatment of schizophrenia, since they improve not only positive but also negative symptoms with little extrapyramidal side effects in animal models. In addition, recent experiments have demonstrated that sigma receptor ligands attenuate the motor suppression and colonic motor disturbances seen under mentally stressful situations, stimulate the central cholinergic function thereby ameliorating impairment of learning and memory, and protect cerebral neurons against cerebral ischemic insult. The present review describes the neuropharmacological effects of sigma receptor ligands, especially anxiolytic (anti-stress) effects, ameliorating effects on impairment of learning and memory, and neuroprotective effects.

[Cross sectional study of the relationship between bone density to diet and life style using ultrasound bone densitometry].

The relationship between bone density to diet and life style was investigated in pre- and postmenopausal women in Kyoto Prefecture in 1994 by a cross-sectional study. Bone densities of 453 women aged 30-86 years were measured by ultrasound bone densitometry. History of pregnancy and delivery, menstruation, medical history, bone and arthral symptoms, life style, food intake frequency, current and past intake of dairy products, and physical activity were examined by self-administered questionnaire. Analysis of covariance and multiple-regression analysis were performed to determine the relation between bone density and life style adjusted for age and obesity index among 151 premenopausal women (PRE), 244 postmenopausal but not sedentary (under 65 years of age) women (POST), and 58 sedentary (older than 65 years of age) women (SED). The results were as follows; 1) A marked age-related decline in bone density was observed at 45-55 years of age. The correlation coefficient between age and bone density was significant at -0.65 (p < 0.01). 2) Obesity index and bone density were positively correlated in each group. 3) Among the PRE group women, there was no relation between life style and bone density. Those who experienced bone fractures tended toward low bone density. Among the POST group, time since menopause, exercise, and current milk intake were significantly correlated with bone density. In the SED group, women with arthralgia showed significantly lower densities. 4) From multiple-regression analysis, age, obesity index, and milk intake during childhood were shown to be related to bone density in each group.

Isolation of dendritic cells in the rat liver lymph.

Lymphadenectomy of the regional lymph nodes of the rat liver resulted in the direct influx of peripheral hepatic lymph into the thoracic duct after regeneration of lymphatic vessels. Thus, we could obtain the dendritic cells in the hepatic lymph by cannulating the thoracic duct. By the double immunostaining, dendritic cells in cytosmears could be easily determined as non-B, non-T, and MHC class II-positive cells. The yield of dendritic cells after enrichment by the metrizamide density gradient was about 5 x 10(5)/first 16 hr collection/rat, with viability of more than 95% and purity of more than 70%. About 80% of dendritic cells were positive for OX62, which recognized the rat dendritic cell subpopulation. They showed strong stimulating activity in the primary allogeneic mixed leukocyte reaction. The method presented here should be applicable to studies of the roles of liver dendritic cells, especially in transplantation immunity.

Involvement of sigma 1 receptor in (+)-N-allylnormetazocine-stimulated hippocampal cholinergic functions in rats.

The effects of the stereoisomers of N-allylnormetazocine (SKF-10,047) on the hippocampal cholinergic functions were compared in rats. A putative sigma 1 receptor agonist, (+)-SKF-10,047, elicited an increase of hippocampal extracellular acetylcholine level and anti-amnesic effect against scopolamine-induced memory dysfunctions in rats. These phenomena were not produced by (-)-SKF-10,047, and were reversed by haloperidol, a putative sigma 1 receptor antagonist. Such stereoselectivity and antagonism imply an involvement of sigma 1 receptors in these (+)-SKF-10,047-stimulated hippocampal cholinergic functions.

Decorating actin filaments with troponin T-I complexes and acceleration of their sliding movement on myosin molecules.

Actin filaments, when partially decorated with troponin T-I complexes, can slide faster on myosin heads than those with no decoration. Purified troponin T-I complexes bind to actins, and inhibit the actin activated myosin adenosine 5'-triphosphatase activity completely when the molar ratio of troponin T-I complex to actin is increased to 1 to 1. Those actin filaments decorated with troponin T-I complexes up to 20 to 50% of their molar ratio exhibit enhancement of the velocity of sliding on myosins up to 20% compared to those without such decoration. As the molar ratio of decoration further increases, the sliding velocity decreases. These results are consistent with the observation that even if some of actin monomers do not participate in the ATPase activity directly, they can interact with myosin heads and take part in the sliding movement.

Notch signaling.

The Notch/Lin-12/Glp-1 receptor family mediates the specification of numerous cell fates during development in Drosophila and Caenorhabditis elegans. Studies on the expression, mutant phenotypes, and developmental consequences of unregulated receptor activation have implicated these proteins in a general mechanism of local cell signaling, which includes interactions between equivalent cells and between different cell types. Genetic approaches in flies and worms have identified putative components of the signaling cascade, including a conserved family of extracellular ligands and two cellular factors that may associate with the Notch Intracellular domain. One factor, the Drosophila Suppressor of Hairless protein, is a DNA-binding protein, which suggests that Notch signaling may involve relatively direct signal transmission from the cell surface to the nucleus. Several vertebrate Notch receptors have also been discovered recently and play important roles in normal development and tumorigenesis.

Consumer power as the major evolutionary force.

Each trophic level in evolving ecosystems can behave as either a supplier toward the downstream of trophic energy flow or a consumer toward the upstream, or an arbitrary mixture of the two. We show that each trophic level behaves almost completely as a consumer in evolutionarily sustainable ecosystems. The present prevalence of consumers in evolving ecosystems derives from a model trophic dynamics minimizing the artificial constraints to be applied arbitrarily.

Characterization of hepatic allograft infiltrates in rats pretreated with donor-specific blood transfusion (DST).

A single intravenous injection of 1 ml freshly heparinized donor blood given 7 days prior to transplantation prolonged significantly the survival of subsequent hepatic allografts in fully allogeneic ACI(RT1a)-to-LEW(RT1(1)) rats. The cellular identity of allograft infiltrates was investigated at various times after transplantation using OX8 (CD8) and W3/25 (CD4) monoclonal antibodies. The number of CD8+ cells increased rapidly and reached a peak on Day 3 after transplantation of the untreated allografts. Similarly, the number of CD8+ cells in the allografts from DST-treated rats was maximum on Day 3 and decreased gradually thereafter. The maximum number of CD4+ cells was found on Day 3 in untreated allografts. In contrast, no significant infiltration of CD4+ cells occurred during the first 7 days after transplantation in DST-treated allografts. Thereafter, the number of CD4+ cells increased rapidly and reached a peak on Day 14. CD4+ cells remained persistently elevated in hepatic allografts of rats pretreated with DST, but did not become functionally competent or initiate rejection. These findings suggest that persistent infiltration by CD4+ cells is a characteristic feature of long-surviving hepatic allografts in rats pretreated with DST.

Evaluation of G-to-A substitution in the apolipoprotein A-I gene promoter as a determinant of high-density lipoprotein cholesterol level in subjects with and without cholesteryl ester transfer protein deficiency.

The effect of a polymorphism, guanine (G) to adenine (A) substitution in the promoter of apolipoprotein A-I gene at a position 78 bp upstream of the transcription initiation site, on the serum high-density lipoprotein (HDL)-cholesterol level was studied in 168 Japanese subjects with HDL-cholesterol levels ranging from 26 to 171 mg/dl. Considering the significant effect of cholesteryl ester transfer protein (CETP) on the HDL-cholesterol level and the common occurrence of its deficiency, we performed statistical analyses separately for two groups: one without CETP deficiency (n = 126) and the other with CETP deficiency (n = 42). In the group without CETP deficiency, in which the numbers of G/G, G/A, and A/A genotypes were 92 (73.0%), 28 (22.2%), and 6 (4.8%), respectively, the frequency of the A allele in the subjects with HDL-cholesterol levels of > or = 70 mg/dl did not differ from subjects with HDL-cholesterol levels of < or = 69 mg/dl, irrespective of gender: 0.154 and 0.145 in males, and 0.182 and 0.174 in females, respectively, for the > or = 70 mg/dl and < or = 69 mg/dl groups. Additionally, the HDL-cholesterol levels for the subjects with the G/G genotype did not differ from those for the subjects with the A allele: 64 +/- 22, 58 +/- 14, 77 +/- 14 and 62 +/- 16 mg/dl, respectively, for the G/G, G/A, A/A, and G/A + A/A in males, and 72 +/- 18, 74 +/- 24, 63 +/- 4, and 73 +/- 23 mg/dl in females. For the group with CETP deficiency, in which the numbers of G/G and G/A + A/A genotypes were 25 (59.5%) and 17 (40.5%), the HDL-cholesterol levels also did not differ: 98 +/- 24 mg/dl and 99 +/- 30 mg/dl, respectively, for the G/G and G/A + A/A genotypes. Thus, there is no evidence that the polymorphism has any effect on serum HDL-cholesterol levels regardless of CETP status. We conclude that the G-to-A substitution in the promoter of apolipoprotein A-I gene does not significantly alter serum HDL-cholesterol level.

Acute effects of sigma ligands on the extracellular DOPAC level in rat frontal cortex and striatum.

Acute administration of (+)-N-allylnormetazocine ((+)-SKF-10,047) and (+/-)-pentazocine, was found to increase the extracellular level of 3,4-dihydroxyphenylacetic acid (DOPAC), a major dopamine (DA) metabolite, in the rat frontal cortex. By contrast, these benzomorphan sigma ligands did not change the extracellular DOPAC level in the rat striatum. On the other hand, 1,3-di(2-tolyl)guanidine (DTG) increased the extracellular DOPAC level in the frontal cortex, while it decreased that level in the striatum. Another non-benzomorphan sigma ligand, (+)-3-(3-hydroxyphenyl)-N-(1-propyl)piperidine ((+)-3-PPP) decreased the extracellular DOPAC level in both frontal cortex and striatum. Moreover, the increase of the extracellular DOPAC level elicited by (+)-SKF-10,047 was significantly inhibited by rimcazole, a putative sigma antagonist, while the DTG-induced increment was not reversed by rimcazole. These findings indicated that the effects of sigma ligands on the mesocortical DA neurons differed from those on the nigrostriatal DA neurons. In addition, the effects of benzomorphan sigma ligands on the central DA neurons were different from those of non-benzomorphan sigma ligands.
