Relationship between visual field defect and multifocal electroretinogram.

We investigated the effect of artificial parafoveal scotomata on the multifocal electroretinogram (M-ERG). M-ERGs were recorded from normal subjects using a monitor with several different sizes of black paper attached. A lower response density area around the 10 to 15 degree parafoveal region was not recognized for scotomata up to 3 degrees but was observed in scotomata above 5 degrees (visual angle) in the field topography of M-ERG. The shape of the scotomata was not circular but somewhat oval. The results from two cases of parafoveal retinal degeneration accorded well with this basic study.

Cell motility and thermodynamic fluctuations tailoring quantum mechanics for biology.

Cell motility underlying muscle contraction is an instance of thermodynamics tailoring quantum mechanics for biology. Thermodynamics is intrinsically multi-agential in admitting energy consumers in the form of energy-deficient thermodynamic fluctuations. The onset of sliding movement of an actin filament on myosin molecules in the presence of ATP molecules to be hydrolyzed demonstrates that thermodynamic fluctuations transform their nature so as to accommodate themselves to energy transduction subject to the first law of thermodynamics. The transition from transversal to longitudinal fluctuations of an actin filament with the increase of ATP concentration coincides with the change in the nature of energy consumers acting upon thermal energy in the light of the first law, eventually embodying a uniform sliding movement of an actin filament.

The internalist enterprise on constructing cell motility in a bottom-up manner.

Quantum coherence in the biological realm is constructed internally in a bottom-up manner. In particular, an actin filament sliding on myosin molecules in the presence of ATP to be hydrolyzed as a functional unit of muscle contraction exhibits magnetization as a marker of quantum coherence. The uniqueness of quantum coherence in biology is found in precipitating synchronous time in interaction from the interacting energy quanta, each of which has carried with itself synchronous time unique to the quantum in isolation. It exhibits a marked contrast to quantum coherence met in low temperature physics, in the latter of which no transformation of the nature of synchronous time is entertained.

Postrevascularization recovery of fatty acid utilization in ischemic myocardium: a randomized clinical trial of potassium channel opener.

BACKGROUND: Abnormal fatty acid metabolism persists in hibernating myocardium, even after reperfusion. This study was designed to determine whether the K+ channel opener, nicorandil, improves fatty acid utilization after percutaneous transluminal coronary angioplasty (PTCA). METHODS: Patients undergoing elective PTCA were randomly assigned to treatment (group N, n = 26) or control groups (group C, n = 22). Group N received intracoronary and intravenous nicorandil during PTCA. Myocardial fatty acid use and perfusion were quantitatively evaluated by means of iodine-123-beta-methyl-p-iodophenyl-pentadecanoic acid single photon emission computed tomography (I-123 BMIPP SPECT) and thallium-201 (Tl-201) imaging before PTCA, 72 hours after PTCA, and 3 months after PTCA. Left ventricular function was also evaluated by means of contrast ventriculography before and 3 to 6 months after PTCA. RESULTS: The 1-123 BMIPP defect score in group N significantly decreased, from 28%+/-13% to 20%+/-20% after PTCA and to 18%+/-17% 3 months later. In contrast, the I-123 BMIPP defect score in group C increased from 28%+/-20% to 36%+/-15% (P<.05 versus group N) after PTCA, then returned to 28%+/-17% (P<.05 versus group N) 3 months after PTCA. Recovery of left ventricular function paralleled the recovery of I-123 BMIPP uptake. CONCLUSIONS: Nicorandil improves the recovery of myocardial fatty acid utilization and cardiac function after PTCA. K(ATP) channel activation may have a protective effect during coronary artery occlusion and improve subsequent recovery.

ALDH2 polymorphism and biological monitoring of toluene.

In this study, we evaluated the effects of the genetic polymorphism of ALDH2 (low Km aldehyde dehydrogenase) on toluene metabolism and determined biological exposure indices (BEIs) of the genotypes of ALDH2. The study subjects included 45 toluene workers and 122 nonexposed students. The genotype of ALDH2 was classified into the homozygous genotype of normal ALDH2 (NN), the homozygous genotype of an inactive one (DD), and the heterozygous genotype of normal and inactive ones (ND) by means of artificial-RFLP. The personal exposure levels to toluene were monitored, using diffusion type samplers and urinary hippuric acid (HA) and creatine concentrations were determined. The urinary HA levels of the three genotypes of ALDH2 of nonexposed students did not differ. In the toluene workers, positive correlations between the personal exposure to toluene and urinary HA levels were observed in the NN, ND, and DD groups. The urinary HA level of the DD group was significantly lower than that of the NN or ND group. Regression lines were used to estimate the BEIs of the NN, ND, and DD groups, which were approximately 2.9 g/g creatinine, 3.3 g/g creatinine, and 1.9 g/g creatinine, respectively.

Anti-metastatic and anti-invasive effects of polymeric Arg-Gly-Asp (RGD) peptide, poly(RGD), and its analogues.

We have investigated the anti-metastatic and anti-invasive activities of polypeptide analogues based on the Arg-Gly-Asp (RGD) adhesive signal in fibronectin, poly(RGD), poly(RGDS)[Arg-Gly-Asp-Ser] and poly(RGDT)[Arg-Gly-Asp-Thr]. These polypeptides containing repetitive RGD sequences were able to inhibit experimental and spontaneous lung metastases of B16-BL6 cells more effectively than the corresponding monomer peptides. In the spontaneous metastasis model, multiple i.v. administrations of these polymeric peptides before or after surgical excision of the primary tumor resulted in a significant reduction of lung tumor colonies. However, there was no significant difference in ability to inhibit spontaneous lung metastasis among poly(RGD), poly(RGDS) and poly(RGDT), although the carboxy-terminal amino acid residue (i.e., Xaa in -RGDXaa-) has been shown to play an important role in the expression of cell adhesive character. The treatment with poly(RGD) substantially prolonged the survival time for mice injected s.c. with B16-BL6 melanoma as compared with the untreated control. We also found that the polypeptides were potently able to inhibit the invasion and migration of tumor cells in vitro. Since these polypeptide analogues showed no antigenicity in the host and had no toxic effect on tumor cells in vitro, they may be potentially useful in the prevention of cancer metastasis.

Familial gastric cancer in the Japanese population is frequently located at the cardiac region.

The clinical features of familial gastric cancer are still unknown. To approach this question, we investigated the clinicopathological characteristics of 16 cases of familial gastric cancer. In this study the criteria used to define familial gastric cancer was the existence of three or more family members with gastric cancer in at least two successive generations. The clinicopathological characteristics of cases who fulfilled this criteria were studied. This study contained 16 familial gastric cancer probands. Seven cases (44%) of gastric cancer had developed at the cardiac region of the stomach. This frequency was significantly higher than for gastric cancer in the general population in Japan (15.4%, p < 0.01). Undifferentiated types were dominant in familial gastric cancer (69%, p < 0.05). Furthermore, the frequency of disseminated peritoneal (40%) and liver metastases (20%) in familial gastric cancer was also significantly higher than for gastric cancer in the general population in Japan (10.9%, p < 0.01, and 4.4%, p < 0.05, respectively). Familial gastric cancers were frequently located at the cardiac region and appeared to be more aggressive than sporadic gastric cancers. The unique characteristics of familial gastric cancer suggest a genetic background in their etiology.

Retinoic acids reduce matrilysin (matrix metalloproteinase 7) and inhibit tumor cell invasion in human colon cancer.

All-trans retinoic acid (ATRA), 9-cis retinoic acid and 13-cis retinoic acid are naturally occurring retinoids used in the prevention and therapy of various preneoplastic and neoplastic diseases. It was previously reported that matrilysin, one of the matrix metalloproteinases (MMP-7), plays a critical role in the invasion and metastasis of gastrointestinal cancers. Moreover, it has been shown that ATRA downregulates matrilysin expression and prevents in vitro invasion by colon cancer cells. In this study, three retinoids were used, both in Matrigel invasion assays and in subcutaneous xenografts in mice, to evaluate the effects of retinoids on invasion by colon cancer cell lines (CHC-Y1, DLD-1, HT-29, BM314, CaR-1 and WiDr). All three retinoic acids tested reduced matrilysin expression and suppressed the invasiveness of colon cancer cell lines in vitro. Retinoic acids also reduced tumor invasion in mice without influencing tumor growth. Matrilysin expression in these tumors was clearly reduced. These data support the use of retinoic acids as useful reagents to manage patients with colorectal carcinoma.

Comparative study of retinal nerve fiber layer loss in normal-tension glaucoma and chronic open-angle glaucoma.

PURPOSE: To clarify whether loss of the retinal nerve fiber layer in normal-tension glaucoma (NTG) differs from that in chronic open-angle glaucoma (COAG). METHODS: Using a scanning laser polarimeter, retinal nerve fiber layer thickness (RNFLT) was measured in 21 patients with NTG, 21 patients with COAG and 21 normal controls. The three groups were matched for age, and the NTG and COAG groups were matched for visual field loss. RESULTS: The mean RNFLT was significantly less in the NTG than in the control group and less in the COAG than in the controls and NTG group. Although the correlations between the parameters of RNFLT and homotopic visual field measurements were significant in COAG, none of the correlations were significant in NTG. CONCLUSIONS: The present results suggest that loss of the retinal nerve fiber layer in NTG differs from that in COAG.

Cardiohemodynamic effect of a novel calcium antagonist, SD-3211, in the dog.

The cardiohemodynamic effects of SD-3211, a calcium antagonist possessing a unique structure, were studied in anesthetized open-chest dogs and in conscious dogs. SD-3211 (10-300 micrograms/kg, i.v.) increased coronary and vertebral artery blood flow dose-dependently while lowering blood pressure, indicating a vasodilatation of these arteries. SD-3211 caused a significant increase in heart rate at 10-100 micrograms/kg, but a significant decrease at 300 micrograms/kg. Left ventricular dp dtmax was dose-dependently decreased at the dose range examined, and the change was significant at 300 micrograms/kg. When compared with the cardiohemodynamic effects of diltiazem (10 300 micrograms/kg, i.v.) and nicardipine (1-30 micrograms/kg, i.v.), the selectivity of SD-3211 with regard to vasodilatation as compared to cardiac depression, manifested by a reduction in heart rate and myocardial contractility, was greater than that of diltiazem, but less than that of nicardipine. Furthermore, a comparative study of the effects of orally administered SD-3211 and diltiazem on blood pressure and atrioventricular conduction in conscious, normotensive dogs, demonstrated that SD-3211 had a more potent and long-lasting hypotensive effect, but a much weaker effect on atrioventricular conduction prolongation than diltiazem. Thus, these in vivo cardiohemodynamic studies show that SD-3211 possesses a tissue-selectivity for vasculature, respectively cardiac tissues, which is intermediate between diltiazem and nicardipine.
