Retroviral transduction of a mutant methylguanine DNA methyltransferase gene into human CD34 cells confers resistance to O6-benzylguanine plus 1,3-bis(2-chloroethyl)-1-nitrosourea.

Human CD34 cells express low levels of the DNA repair protein O6-alkylguanine-DNA alkyltransferase (AGT) and are sensitive to 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU). Gene transfer of the AGT gene, methylguanine DNA methyltransferase (MGMT), results in only modest BCNU resistance. Recently, an AGT inhibitor, O6-benzylguanine (BG), entered clinical trials. In preclinical studies, BG potentiated the cytotoxic effect of BCNU in tumors but increased toxicity to normal CD34 cells. We transferred a mutant MGMT containing a glycine-to-alanine mutation at position 156, resulting in marked resistance to BG, into Chinese hamster cells; the K562 cell line and human CD34 cells used the retroviral backbone MFG. In each instance, cells expressed increased AGT and were much more resistant to the combination of BG and BCNU than the parental cells or cells transduced with wild-type MGMT. Furthermore, the transduction efficiency in human CD34 cells was in excess of 70%, and the proportion of CD34 transduced cells resistant to the combination was > 30%. Thus, retroviral-mediated transduction of a mutant MGMT into CD34 cells appears to be an effective way to induce selective resistance to a drug combination designed to overcome a significant resistance mechanism to nitrosoureas in tumors.

Identification and characterization of various differentiative growth plate chondrocytes from porcine by countercurrent centrifugal elutriation.

Countercurrent centrifugal elutriation was used to separate growth plate chondrocytes from porcine basing on their differences in sizes and densities. Eighteen fractions of cells with different sizes and densities were obtained. The mean cellular volumes increased progressively in each of successive fractions, and that increase was associated with specific phenotypic changes, such as biochemical differences in DNA synthesis, proteoglycan synthesis, and activities of alkaline phosphatase. Three distinct chondrocyte subpopulations with their unique characteristics were identified among the elutriated fractions. The resting chondrocytes were found to be small in size and quiescent. The hypertrophic chondrocytes were found to be large in size and metabolically active both in alkaline phosphatase and in proteoglycan productions. The proliferative chondrocytes exhibited a high DNA synthesis rate, and their sizes were found to be between those of the resting and hypertrophic chondrocytes.

Sensitivity-enhanced echo-planar MRI at 1.5T using a 5 x 5 mesh dome resonator.

In this work a 5 x 5 mesh dome resonator that has been optimized for functional brain imaging is presented. The resonator was reduced in length and diameter compared with previous versions to reduce sample losses, thus enhancing the signal-to-noise ratio of the acquired data. In addition, a 5 x 5 mesh design was employed, which offered improved axial homogeneity over an earlier 3 x 3 mesh version. The new resonator exhibited high sensitivity and good homogeneity over the brain volume, permitting analysis of functional activation over large areas of the cerebral cortex. In a direct comparison with a standard clinical head-imaging resonator, the high sensitivity of the 5 x 5 mesh dome resonator resulted in greater statistical confidence in functional activation.

Characterization of presystemic elimination of trichloroethylene and its nonlinear kinetics in rats.

1,1,2-Trichloroethylene (TCE) is a volatile organic chemical which contaminates drinking water and food supplies and is primarily of concern because of the risk of cancer it may pose. The objectives of the present study were to evaluate the efficiency and dose dependency of presystemic elimination of TCE in rats. Cannulas were surgically implanted into male Sprague-Dawley rats (330-380 g) 24 hr before TCE dosing. TCE (0.17, 0.33, 0.71, 2, 8, 16, and 64 mg/kg) in a 5% aqueous Alkamuls emulsion was administered over 30 sec into the carotid artery, jugular vein (JV), hepatic portal vein, or the stomach. Serial arterial blood samples of 1-500 microliters were collected for up to 12 hr from the unanesthetized animals and analyzed for TCE content by headspace gas chromatography. Pharmacokinetic analyses indicated that TCE was eliminated through dose-dependent nonlinear processes. A three-compartment model with Michaelis-Menten and first-order elimination was derived to fit simultaneously the TCE blood data following JV administration. Total presystemic elimination of TCE was inversely related to dose, ranging from approximately 60 to < 1%. A dose-dependent decrease in hepatic extraction was primarily responsible for the reduction in total first-pass elimination at high doses, whereas pulmonary extraction (i.e., 5-8%) was relatively constant over the dosage range. When metabolic saturation was minimal or absent, hepatic presystemic elimination of TCE accounted for approximately 45-55% of the administered dose. These findings indicate that a substantial proportion of trace amounts of VOCs ingested in environmental media may not enter the systemic circulation nor reach extrahepatic target organs.

The pattern of organochlorines in mussels Mytilus edulis L. from the south west Baltic Sea.

The concentration of organochlorines (PCB, DDT and its metabolites, HCH isomers and HCB) were determined in mussel, Mytilus edulis, from the south west Baltic Sea. The harbor Kiel Innenförde showed the highest contamination with PCB (487 microg/kg dry weight), the Gelting bathing beach area the lowest contamination (38 microg/kg). The highest contamination with DDT was measured in Lübeck bay, which may be explained by the long term application of DDT in the former German Democratic Republic. Penta- and hexa-chlorinated PCB were selectively accumulated in Mytilus edulis whereas highly lipophilic PCB with more than seven chlorine atoms were found in negligible concentrations. The accumulation of these compounds in mussels is determined by equilibrium partitioning with ambient water.

Information integration in cross-modal pattern recognition: an argument for acquired modularity.

Three experiments addressed the issue of whether modularized operations (Fodor, 1983) in pattern recognition could be acquired. In Experiment 1, blind and sighted individuals were compared in the speech-tactile domain of pattern recognition. The results were inconsistent with the assumptions of the fuzzy logical model of perception (FLMP) (Oden and Massaro, 1978) and with the assumptions of modularized pattern recognition processes, but were consistent with the assumptions of 'penetrability' into non-modularized operations. Experiments 2 and 3 compared sighted individuals across speech-visual and speech-tactile domains, revealing fundamentally different patterns of performance that we argue demonstrate modularized and non-modularized FLMP operations in pattern recognition, respectively.

Protective effect of nitric oxide in an endotoxin-induced septic shock.

BACKGROUND: Calcium ion (Ca++)-independent nitric oxide (NO) synthase activity in animals was markedly induced by treatment with endotoxin, but NO levels in various tissues removed from endotoxin-treated animals have not been reported. The role of NO during an endotoxin-induced septic shock remains controversial. METHODS: ICR mice, randomly divided into one of six treatment groups, received intraperitoneal injections as follows: phosphate-buffered saline; Escherichia coli LPS (LPS); N(omega)-nitro-L-arginine (L-NNA); N(omega)-nitro-D-arginine (D-NNA); LPS plus L-NNA; and LPS plus D-NNA. The mice were either monitored for mortality or killed for nitrite/nitrate assays and histologic analysis. RESULTS: NO levels in many tissues were markedly increased by injection of LPS, and administration of L-NNA increased mortality rates of LPS-treated mice, in association with an increase in tissue damage in the lung, liver, and kidney. CONCLUSIONS: The endogenous NO generated during LPS-mediated septic shock could be protective.

Accuracy and efficiency of X-ray requests initiated by triage nurses in an accident and emergency department.

A study was carried out from 1 March 1995 to 31 March 1995 in the Accident & Emergency department, Pamela Youde Nethersole Eastern Hospital, Hong Kong. The aim was to assess the feasibility of requesting X-rays by triage nurses based on a pre-set protocol prior to patients being seen by a doctor. Judgement of the attending physician was used as the gold standard by which to judge the need for radiographs requested by the triage nurse. In addition, the senior medical officer or consultant would judge whether the triage nurse had applied the protocol correctly. A total of 934 patients were included in the study and 883 (94.54%) patients had X-rays requested by the triage nurse. Only 5.44% of the radiographs requested by nurses were considered to haven been unnecessary by the doctor. There was no statistically significant difference in agreement for the child and adult groups. In 917 (98%) of cases, nurses had adhered to the protocol. A saving of an average of 18.59 minutes of total service time was obtained during the study. Requesting of X-ray by triage nurse was feasible

Characterization of monoclonal antibodies to the 26-kDa glutathione S-transferase of Schistosoma japonicum.

Six monoclonal antibodies (MAbs) were raised in mice against the 26-kDa glutathione S-transferase (GST) of the parasite Schistosoma japonicum. These MAbs were originally selected for their specific binding to the recombinant GST (r-GST) generated in E. coli by an enzyme-linked immunosorbent assay. A further study demonstrated that all these MAbs bound to plate-coated GST affinity-purified from the parasite Schistosoma japonicum. However, in Western blotting analysis only a single monoclonal antibody (MAb Y3D7) yielded positive binding. The binding of MAb Y3D7 on Western blotting was further characterized; specific binding was found on other GST fusion proteins and on the authentic 26-kDa GST but not the 28-kDa GST in the total soluble worm proteins from Schistosoma japonicum. Using protein-A-mediated immunoprecipitation, MAbs Y3D7 and Y5D5 precipitated r-GST while in parallel experiments the remaining MAbs did not generate r-GST precipitation. In an alternative co-precipitation experiment, r-GST was first bound to glutathione (GSH) Sepharose beads and subsequently tested for interaction with the MAbs. In this manner, all MAbs except MAb Y5D5 were co-precipitated with the complexes. Thus, these select MAbs readily reacted with GST although their binding characteristics were different. Because GST has been widely used in the generation of fusion proteins for various purposes and is a potential vaccine candidate in controlling schistosomiasis, these MAbs should prove valuable for their application to molecular biology and parasitology.

Factors affecting the pullout strength of cancellous bone screws.

Screws placed into cancellous bone in orthopedic surgical applications, such as fixation of fractures of the femoral neck or the lumbar spine, can be subjected to high loads. Screw pullout is a possibility, especially if low density osteoporotic bone is encountered. The overall goal of this study was to determine how screw thread geometry, tapping, and cannulation affect the holding power of screws in cancellous bone and determine whether current designs achieve maximum purchase strength. Twelve types of commercially available cannulated and noncannulated cancellous bone screws were tested for pullout strength in rigid unicellular polyurethane foams of apparent densities and shear strengths within the range reported for human cancellous bone. The experimentally derived pullout strength was compared to a predicted shear failure force of the internal threads formed in the polyurethane foam. Screws embedded in porous materials pullout by shearing the internal threads in the porous material. Experimental pullout force was highly correlated to the predicted shear failure force (slope = 1.05, R2 = 0.947) demonstrating that it is controlled by the major diameter of the screw, the length of engagement of the thread, the shear strength of the material into which the screw is embedded, and a thread shape factor (TSF) which accounts for screw thread depth and pitch. The average TSF for cannulated screws was 17 percent lower than that of noncannulated cancellous screws, and the pullout force was correspondingly less. Increasing the TSF, a result of decreasing thread pitch or increasing thread depth, increases screw purchase strength in porous materials. Tapping was found to reduce pullout force by an average of 8 percent compared with nontapped holes (p = 0.0001). Tapping in porous materials decreases screw pullout strength because the removal of material by the tap enlarges hole volume by an average of 27 percent, in effect decreasing the depth and shear area of the internal threads in the porous material.

Tyrosine phosphorylation and activation of pp60c-src and pp125FAK in bradykinin-stimulated fibroblasts.

Bradykinin (BK) stimulates protein tyrosine phosphorylation in human foreskin fibroblasts (K.-M. Lee, K. Toscas, and M. L. Villereal, J. Biol. Chem. 268:9945-9948, 1993). The major tyrosine phosphorylation occurs in proteins of a molecular mass of 130 and 70 kDa. In this report, we demonstrate that focal adhesion-associated tyrosine kinase, pp125FAK, is one component of the 130-kDa phosphotyrosine band. The BK-stimulated pp125FAK tyrosine phosphorylation level is well correlated with increased kinase activity, as assessed by in vitro immune complex kinase assays. We have identified paxillin, a protein that is localized in focal adhesions, as a component of the 70-kDa phosphotyrosine band. In addition to identifying the two proteins responsible for the major phosphotyrosine bands, we also report that pp60c-src is tyrosine phosphorylated and activated in response to BK, as analyzed by immunoblotting and in vitro kinase assays, respectively. These findings indicate, for the first time, that the BK receptor is coupled to the important protooncogene c-src and that the src pathway may mediate some of the events downstream from BK binding.

Pattern of emergence and the effects of temperature and light on the emergence and survival of heterophyid cercariae (Centrocestus formosanus and Haplorchis pumilio).

Production of the cercariae of Centrocestus formosanus and Haplorchis pumilio was not affected by a 10-day starvation of the snail host Melanoides tuberculata, and there was no circadian rhythm in the pattern of cercarial emergence. Daily average cercarial productions from each snail were 1,643 for C. formosanus and 689 for H. pumilio at 25 C and 500 lux illumination. The number of cercariae was moderately correlated with the size of snail host. The heaviest sheddings ever observed from 1 snail in 24 hr were 63,400 cercariae for C. formosanus and 3,470 cercariae for H. pumilio. The cercariae of C. formosanus could emerge in the dark, but the number was much less than in the light. In contrast, the cercariae of H. pumilio emerged equally well with or without light. Within the temperature range tested, the emergence of cercariae occurred at 15-35 C, but not at 10 C. Shedding of C. formosanus cercariae increased with the rise in ambient temperature, whereas the greatest shedding of H. pumilio cercariae occurred at 25 C. Life-span of the cercariae was temperature-dependent. The most favorable temperature for survival of C. formosanus was 15 C, at which some survived for 160 hr, and 20 C for H. pumilio, with the longest survival of 130 hr.

Intestinal parasites among the Southeast Asian laborers in Taiwan during 1993-1994.

BACKGROUND: A large number of foreign laborers are now working in Taiwan, the majority coming from Southeast Asia where parasitic infections are common. Since 1992, monitoring the intestinal parasitic infections of these foreigners has been carried out, particularly to prevent the introduction of some of the parasites into Taiwan. This paper reports results obtained during the year 1993-1994. METHODS: The merthiolate-iodine-formalin (MIF) concentration method was used to detect intestinal parasites; and the data were analyzed by chi 2-test. RESULTS: Among the 3,020 persons examined, 526 (17.4%) were infected and 14 kinds of parasites were identified; i.e. Ascaris lumbricoides, Trichuris trichiura, Strongyloides stercoralis, Enterobius vermicularis, hookworms, Taenia sp., Opisthorchis viverrini, Fasciolopsis buski, Schistosoma japonicum, an echinostome, Entamoeba histolytica. Entamoeba coli, Endolimax nana and Giardia lamblia. By nationality, those workers from Indonesia had the highest infection rate, at 32.3%; they were followed by the Filipinos (18.7%), the Thais (16.6%) and the Malaysians (8.0%). The three or four most prevalent parasites were T. Trichiura (21.8%), A. lumbricoides (5.6%) and hookworms (2.4%) for the Indonesians; T. Trichiura (9.1%), A. lumbricoides (3.7%), hookworms and E. histolytica (2.1% each) for the Filipinos; O. viverrini (8.4%), hookworms (3.3%) and E. histolytica (1.2%) for the Thais; T. trichiura (2.2%), A. lumbricoides and E. histolytica (1.8% each) and hookworms (1.4%) for the Malaysians. Among the 526 persons infected, 92.2% had 1 species, 7.2% had 2 species, 0.4% had 3 species and 0.2% had 4 species of parasites. CONCLUSIONS: The present result, a 17.4% infection rate, was significantly higher (p < 0.05) than the 12.3% obtained for the previous year (1992-1993). E. histolytica was found in 34 persons, 16 of whom were working as live-in housemaids. The epidemiological significance of some parasites is discussed here.

Patient profile and survival in 270 computer tomography-staged patients with nasopharyngeal cancer treated at the Singapore General Hospital.

This is a report analysing the patient profile and survival results in 270 patients with nasopharyngeal carcinoma treated with radiation therapy between July 1987 and December 1988. The patients had a median age of 52.6 months, and a male-female ratio of 2.5:1. Ninety-four percent of patients were Chinese. The commonest presentation was that of a neck mass (found in approximately 65% of patients). Approximately two-thirds presented with disease more advanced than Ho's stage II. At a median follow-up of 52.6 months, the 5-year actuarial survival rate was 53% and the freedom from relapse rate was 57%. One hundred and seven patients died within the observation period. Of these, 46 patients had disease in the post nasal space, 41 in the neck and 60 in sites outside the head and neck.

Testicular seminoma--the results of treatment by radiotherapy in Singapore.

A retrospective analysis was done of 49 patients treated for testicular seminoma over a 15-year period between January 1980 and April 1995 at the Department of Therapeutic Radiology, Singapore General Hospital. Their mean age was 39 years and their median period of follow-up was 67 months. Using the Royal Marsden Staging classification, 35 (71.4%) were stage I, 10 (20.4%) were stage II, I (2%) was stage III, and 3 (6.1%) were stage IV. Five (10.2%) arose in undescended testes. Thirty-six patients were treated with inguinal orchidectomy and postoperative radiotherapy to the para-aortic and ipsilateral iliac nodes. Eight had para-aortic and bilateral iliac irradiation, and 2 had ipsilateral pelvic, para-aortic, mediastinal and supraclavicular lymph node irradiation. Six of the patients had, in addition, irradiation of the inguinal-scrotal area. Nine received both chemotherapy as well as radiotherapy. The median dose of radiation given was 30.67 Gy and where a boost was required, the median total tumour dose was 35.75 Gy. The actuarial relapse-free survival at 10 years was 88.9% and the actuarial overall survival at 10 years was also 88.9%. Long-term treatment toxicities were uncommon.

No change in bone-specific alkaline phosphatase activities in cultured rat osteoblastic cells under L-ascorbate and beta-glycerophosphate-induced mineralization.

Mineralization occurred both in fetal rat calvarial cells and UMR 106 osteoblastic cells when they were cultured in medium containing L-ascorbate and beta-glycerophosphate as evidenced by von Kóssa staining as well as deposition of calcium ions and inorganic phosphate in the cells. When compared with corresponding non-mineralized cell cultures, both the mineralized cultures of calvarial cells and UMR 106 cells did not exhibit any change in intracellular bone-specific alkaline phosphatase activities which were measured by wheatgerm lectin precipitation method. Our results support the hypothesis that mineralization may not exert any direct negative feedback on matrix protein synthesis in osteoblasts during bone formation.

A novel method for selective isotope labeling of bacterially expressed proteins.

A novel method for isotope labeling in selected amino acids is presented for use with the T7 RNA polymerase system. The protocol is illustrated with the DNA-binding domain from the E2 protein of bovine papillomavirus, BPV-1. On addition of rifampicin, protein expression occurs exclusively from the gene controlled by the T7 promoter. Since the bacteria are now dedicated to the production of E2 protein, labeling with specific amino acids is efficiently performed. For example, 10 mg/l of 15N-labeled phenylalanine is shown to be sufficient for incorporation of the label, without scrambling, and without the use of an auxotrophic strain.
