A case of immunoblastic lymphadenopathy (IBL)-like T-cell lymphoma with unusual clinical manifestations.

A 67-year-old male consulted an otolaryngologist with a complaint of dysphagia due to a large polypoid mass in the hypopharynx. A biopsy of the mass revealed immunoblastic lymphadenopathy (IBL)-like T-cell lymphoma of helper/inducer subset occurring in lymphoid tissues of the hypopharynx. Although general lymphadenopathy was not seen, further biopsy of an inguinal lymph node showed small foci of lymphoma cells, suggesting the generalized distribution of this malignancy. To our knowledge, this is the first report of this malignancy mimicking primary pharyngeal sarcoma.

Effects of human recombinant interleukin 5 and 3 on the differentiation of cord blood-derived eosinophils and basophils.

When mononuclear cells from umbilical-cord blood were cultured for 3 weeks, low concentrations of interleukin 3 supported the preferential growth of basophils, with eosinophils comprising a smaller proportion. These basophils contained 0.15-0.3 microgram histamine per 10(6) cells, and released histamine by the IgE-dependent and -independent stimuli. Interleukin 5 increased the number and proportion of eosinophils in a dose-dependent manner without affecting the proliferation of other cell types in the interleukin 3-supplemented cultures. These cultured eosinophils could be activated by platelet-activating factor.

[Effects of various factors involved in bone metabolism on 45Ca2+ incorporation and alkaline phosphatase activity of ROS 17/2.8 cells].

The effects of interleukin 1, transforming growth factor-beta (coupling factors), prostaglandin E1, and prostaglandin E2 on incorporation of 45Ca2+ and on alkaline phosphatase activity were studied using cultured ROS 17/2.8 cells, one of cell lines derived from rat osteosarcoma. We found that all these factors stimulate both the incorporation of 45Ca2+ and alkaline phosphatase activity of these cells. On the other hand, one of the bone resorption hormones, parathyroid hormone (PTH), suppressed the proliferation of cells and decreased the alkaline phosphatase activity at considerably low concentrations (1 X 10(-12)-1 X 10(-11) M). However, the hormone stimulated the incorporation of 45Ca2+ by these cells in a dose-dependent manner; the maximum stimulation on day 3 was observed at 1 X 10(-7) M and it was approximately 3 times the control value. The data suggest therefore, that the osteoblasts incorporated calcium ions and transported them while bone resorption was occurring. Thus the ROS 17/2.8 cell line appears to be an advantageous experimental system for the study of calcium metabolism of osteoblasts in vitro.

[Reconsideration of organ weight in Japanese (1). Heart].

The heart weight measured at medicolegal autopsies in these days are usually heavier than the data of the mean value of Japanese in textbooks and papers which were surveyed around the Second World War. As it was supposed that some improvement of the nutrition and physique since the end of the war should make the heart weight heavier, the value on 1,399 cases of medicolegal autopsy was measured and analyzed. The results revealed that the heart weight in the late of teens reaches the old value of grown-ups and the mean values of men (360 g) and women (300 g) earned by the present study are far more than those described in the former reports. It was presumed that periodic researches for the standard heart weight are indispensable in the consequence of the present study.

A case of suicide by drowning with hesitation marks on the back.

A case of a 78-year-old woman suspected of committing suicide is reported; an analysis of the evidence suggesting that she threw herself into the river after stabbing and cutting her neck, left forearm, abdomen and back. Hesitation marks on the back are relatively rare in cases of suicide. We discuss the manner of death and assume the mode in which her injuries were inflicted.

The activated eosinophil in allergy and asthma.

Our studies indicate that human eosinophils can be activated in terms of up-regulation of surface receptors, increased membrane lipid generation, and enhanced cytotoxicity by a variety of pharmacological mediators, through IgE-, IgG-, and complement-dependent pathways. PAF is an impressive activator of eosinophil function and is the most potent chemotactic mediator for eosinophils so far described. It is not selective in that neutrophils are also attracted by PAF. In our hands, IL-1, IL-2, granulocyte/macrophage-colony stimulating factor, tumor necrosis factor, interferon-gamma, and IL-5 tested over a wide concentration range gave very weak chemotaxis for eosinophils in vitro. Nevertheless, in vivo infiltration of eosinophils into the site of allergic tissue reactions is closely associated with the local accumulation of activated (CD4+, IL-2 rec+) T cells. We suggest, therefore, that the selective eosinophil infiltration seen in allergic inflammation may result from the initial recruitment and activation of granulocytes by lipid mediators and the subsequent selective activation of eosinophils by cytokines.

The effect on the cornea of alpha cyclodextrin vehicle for cyclosporin eye drops.

We tested four different combinations of alpha-CD and CYA: 0.075% and 80 mg/mL alpha-CD, 0.025% CYA and 40 mg/mL alpha-CD, 0.009% CYA and 20 mg/mL alpha-CD, and 0.003% CYA and 10 mg/mL alpha-CD. We found that 0.025% CYA (alpha-CD[40 mg/mL]) resulted in the least corneal toxicity and penetrated in the cornea 5 to 10 times more than did lipophilic vehicle with CYA.

[Studies of specific IgE and IgG antibodies in allergic children. Changes in specific IgE and IgG antibodies to egg white, milk, soybean and mite from children with atopic dermatitis].

Egg, milk, soybean, mite (DF)-specific IgG and IgE antibodies were measured in 276 atopic dermatitis children in whom dietary restrictions had never been done. We recommend cleaning house and bed clothes to decrease number of mites and elimination diet according to clinical symptoms and laboratory findings. After elimination periods (approximately 140 days) the same specific antibodies were examined again. In the patients who improved clinically by elimination of foods, the specific IgG antibodies to the foods decreased significantly. The other patients did not improved clinically suggesting that inadequate elimination diet were done and cleaning house and bed clothes was incomplete and/or other types of allergic reactions might exist.

Large olfactory responses of the carp after complete removal of olfactory cilia.

To study the role of olfactory cilia on olfactory reception, the carp olfactory cilia were removed by modified "ethanol-calcium shock" and the bulbar responses were recorded before and after deciliation. Large olfactory responses to various amino acids were observed after complete deciliation. The relation between magnitude of olfactory response and alanine concentration before and after deciliation was essentially unchanged. The present results suggests that the olfactory cilia may not be necessary for receptor neuron function in the carp.

Temperature dependence of amiloride-sensitive and -insensitive components of rat taste nerve response to NaCl.

The temperature dependence of the rat taste response to NaCl was examined by measuring the chorda tympani activities in response to NaCl solution of varying temperatures before and after amiloride treatment. The response to NaCl was composed of 3 components: component I which was not suppressed by amiloride and components II and III which were suppressed by amiloride. Components I, II and III show maximal responses around 30, 30 and 10 degrees C, respectively. The response to NaCl at 10 degrees C was composed mostly of amiloride-sensitive component (component III). Magnitude of the amiloride-insensitive component (I) was greatly affected by variation of anion species. The amiloride-sensitive component II also showed anion influence but the influence was different from that for component I. Another amiloride-sensitive component (III) showed practically no anion influence. Threshold concentrations of the components were 0.1 mM for component I, 3 mM for component II and 0.3 mM for component III. Equilibrium potentials of Na+ at the threshold concentrations across the apical membrane of taste cells were calculated to be -120.2 mV for component I, -31.4 mV for component II and -85.4 mV for components III. Thus the equilibrium potentials of Na+ at the thresholds for components I and III were more negative than the resting potentials reported, which suggested that an increase in permeabilities of Na+ through receptor sites (or channels) for these components will lead to hyperpolarization, not depolarization of taste cells.

Liposomes as model for taste cells: receptor sites for bitter substances including N-C=S substances and mechanism of membrane potential changes.

Various bitter substances were found to depolarize liposomes. The results obtained are as follows: (1) Changes in the membrane potential of azolectin liposomes in response to various bitter substances were monitored by measuring changes in the fluorescence intensity of 3,3'-dipropylthiocarbocyanine iodide [diS-C3(5)]. All the bitter substances examined increased the fluorescence intensity of the liposome-dye suspension, which indicates that the substances depolarize the liposomes. There existed a good correlation between the minimum concentrations of the bitter substances to depolarize the liposomes and the taste thresholds in humans. (2) The effects of changed lipid composition of liposomes on the responses to various bitter substances vary greatly among bitter substances, suggesting that the receptor sites for bitter substances are multiple. The responses to N-C=S substances and sucrose octaacetate especially greatly depended on the lipid composition; these compounds depolarized only liposomes having certain lipid composition, while no or hyperpolarizing responses to these compounds were observed in other liposomes examined. This suggested that the difference in "taster" and "nontaster" for these substances can be explained in terms of difference in the lipid composition of taste receptor membranes. (3) It was confirmed that the membrane potential of the planar lipid bilayer is changed in response to bitter substances. The membrane potential changes in the planar lipid bilayer as well as in liposomes in response to the bitter substances occurred under the condition that there is no ion gradient across the membranes. These results suggested that the membrane potential changes in response to bitter substances stem from the phase boundary potential changes induced by adsorption of the substances on the hydrophobic region of the membranes.

Antimycobacterial activities in vitro and in vivo and pharmacokinetics of dihydromycoplanecin A.

The in vitro activity of dihydromycoplanecin A (DHMP A), a new cyclic peptide antibiotic, was compared with those of antimycobacterial drugs such as streptomycin, isoniazid (INH), rifampin, and ofloxacin against several clinically isolated species of mycobacteria, including Mycobacterium tuberculosis, M. intracellulare, and M. kansasii. DHMP A demonstrated stronger activities than other drugs against all species of mycobacteria tested at concentrations of less than 0.0125 to 25 microgram/ml. A marked synergism between DHMP A and INH was demonstrated by the checkerboard technique against M. tuberculosis, M. intracellulare, and M. smegmatis, and the synergistic effect was observed by treatment of the culture of M. smegmatis with DHMP A for at least 3 h prior to treatment with INH. It was also shown that both absorption and excretion of INH in mice were faster than those of DHMP A. On the basis of these results, combination therapy with DHMP A and INH was successfully carried out in experimental tuberculosis in mice infected with M. bovis Ravenel. After a single intravenous administration of 10 mg of DHMP A per kg, its half-life in serum in mice was about 0.5 h and in dogs it was 5.5 h. A single oral administration to dogs of 12.5 mg/kg gave a peak of 5.0 micrograms/ml at 3 h. In these experiments, urinary recoveries within 48 h were 21.0% in mice and 25.2% in dogs. The tissue distribution level of DHMP A in mice after oral administration was in the order of liver greater than kidney greater than serum greater than spleen = lung. The 50% lethal doses of DHMP A for mice were more than 6,000 mg/kg orally and 1,840 mg/kg intraperitoneally.

Liposomes as a model for olfactory cells: changes in membrane potential in response to various odorants.

Various odorants were found to depolarize azolectin liposomes. The results obtained are as follows. (1) Changes in the membrane potential of azolectin liposomes in response to various odorants were monitored by measuring changes in the fluorescence intensity of 3,3'-dipropylthiocarbocyanine iodide [disS-C3(5)]. Ten odorants examined increased the fluorescence intensity of the liposome-dye suspensions in a dose-dependent manner, which indicates that odorants depolarize the liposomes. Concentrations of odorants that depolarized the liposomes greatly varied among the odorants. There existed a good correlation between the minimum concentrations of odorants to depolarize the liposomes and the thresholds of respective odorants in the frog or porcine olfactory responses. (2) Addition of sphingomyelin (SM) to azolectin led to a large enhancement of depolarizations by nonanol, citral, and n-amyl acetate. The results indicate that lipid composition of liposomes is one of the factors that control the sensitivity to odorants. (3) Odorants changed the membrane fluidity of the liposomes, which was monitored by changes in the fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene (DPH). The membrane fluidity was changed in concentration ranges of odorants similar to those where the membrane potential changes occurred, which suggests that changes in the membrane fluidity are related to generation of the membrane potential changes. (4) Changes in the membrane potential in response to odorants were electrically measured with the planar lipid bilayer made of an azolectin-SM (2:1 w/w) mixture. It was shown that odorants (nonanol, citral, and n-amyl acetate) depolarized the planar lipid bilayer.(ABSTRACT TRUNCATED AT 250 WORDS)

Effects of changed lipid composition on responses of liposomes to various odorants: possible mechanism of odor discrimination.

In a previous paper [Nomura, T., & Kurihara, K. (1987) Biochemistry (preceding paper in this issue)], we showed that azolectin liposomes are depolarized by various odorants and there is a good correlation between the responses in the liposomes and the frog or porcine olfactory responses. In this study, we examined effects of changed lipid composition on responses of liposomes to various odorants. The membrane potential changes in response to odorants were monitored with the fluorescent dye 3,3'-dipropylthiocarbocyanine iodide [diS-C3(5)]. Egg phosphatidylcholine (PC) liposomes showed depolarizing responses to nine odorants among ten odorants tested. The magnitudes of depolarization by alcohols were similar to those in azolectin liposomes, but those by other odorants were much less than those in azolectin liposomes. Addition of sphingomyelin (SM) to PC led to an increase in the magnitude of depolarization by most odorants. Addition of phosphatidylethanolamine (PE) to PC (PE/PC = 0.25) led to depolarizing responses to four odorants among six odorants tested, and a further increase in PE content (PE/PC = 0.54) led to depolarizing responses only to two odorants. Addition of SM to the lipids of this composition of PC and PE [SM/(PC + PE) = 0.22] led to depolarizing responses to four odorants again. Liposomes made of a mixture of SM, PE, and PC exhibited depolarizing responses to four odorants tested, and addition of cholesterol to the lipids [cholesterol/(PC + PE + SM) = 0.05 and 0.11] led to depolarizing responses only to two and one odorant, respectively. Thus, changes in lipid composition of liposomes led to great changes in specificity of the responses to odorants.(ABSTRACT TRUNCATED AT 250 WORDS)

ADP-ribosylation of specific membrane proteins in pheochromocytoma and primary-cultured brain cells by botulinum neurotoxins type C and D.

Type C1 and D toxins produced by Clostridium botulinum caused ADP-ribosylation of a protein of 24 kDa in membrane preparations of rat clonal pheochromocytoma cells (PC12) and of proteins of 25 and 26 kDa in neuron-rich culture of fetal rat brain cells. The ADP-ribosylation reaction was dependent on the presence of MgCl2, GTP and GTP gamma S. The results obtained suggested that the ADP-ribosylation reaction is responsible for the development of the biological activity of the botulinum neurotoxins and that the target of this reaction may be novel GTP-binding proteins localized on cell membranes.
