[A case of colonic carcinoma associated with intestinal tuberculosis, and an analysis of 26 cases reported in Japan].

A case of tuberculosis and adenocarcinoma co-existing in the ileocecal region in a 61-yr-old woman is described. The Japanese literature is reviewed and several points discussed regarding the epidemiological and morphological features in a colon carcinoma associated with tuberculosis. (1) Females predominate by a ratio of 17:9 in such cases. (2) The tumor has been found in the right side of the colon in 17 out of 26 cases. (3) The most characteristic histological finding is that of the cancer, which shows a well differentiated adenocarcinoma with a tendency to produce a mucin. These unique findings are consistent with those in our case. It is of interest that these unique findings may support the possibility that the cancer originated from a tuberculous lesion.

Temporal patterns of protein phosphorylation after angiotensin II, A23187 and/or 12-O-tetradecanoylphorbol 13-acetate in adrenal glomerulosa cells.

The temporal patterns of protein phosphorylation in the adrenal glomerulosa cell were analysed by two-dimensional electrophoresis after stimulation with 10 nM-angiotensin II or various agents [10 nM-12-O-tetradecanoylphorbol 13-acetate (TPA), 50 nM-A23187, 1 microM-nitrendipine], administered singly or in combination. These patterns were compared with the temporal patterns of aldosterone secretion induced by the same agonists and antagonists. After 1 and 30 min of stimulation with angiotensin II, different patterns of protein phosphorylation were observed. A comparison of these patterns reveals that: the phosphorylation of only one protein was persistently enhanced during the continuous incubation with angiotensin II; the phosphorylation of five proteins was transiently enhanced (at 1 min but not 30 min); and the phosphorylation of three proteins did not occur at 1 min but was seen at 30 min. Addition of the phorbol ester TPA alone, which at 30 min is without effect in enhancing aldosterone production, has no effect on protein phosphorylation. The combined addition of TPA and the Ca2+ ionophore, A23187, which, like angiotensin II, evokes a sustained increase in aldosterone production, reproduced the temporal patterns of protein phosphorylation seen after angiotensin II action. Manipulations (A23187 alone, angiotensin II plus nitrendipine) which evoke only a transient rise in aldosterone production rate induce a transient rise in cellular protein phosphorylation. The 1 min patterns of phosphorylation seen after A23187 or combined angiotensin II and nitrendipine (a Ca2+ channel antagonist) are similar to those observed after 1 min of angiotensin II stimulation. These results suggest that, when angiotensin II acts, the initial cellular response is mediated by a different mechanism than that responsible for the sustained response.

Short term memory in the calcium messenger system. Evidence for a sustained activation of protein kinase C in adrenal glomerulosa cells.

If adrenal glomerulosa cells are treated with angiotensin II for a period of 20-30 min, their subsequent response to either a rechallenge with the same concentration of angiotensin II or treatment with BAY K 8644, a calcium channel agonist, differs from the responses of control cells. Perifusion of control cells with 10 nM-angiotensin II leads to an increase in aldosterone secretory rate from 44 +/- 7 to 166 +/- 9 pg/min per 10(6) cells, but perifusion of cells pretreated for a 20 min period with angiotensin II leads to an increase in secretory rate from 51 +/- 9 to 209 +/- 18 pg/min per 10(6) cells. Likewise, treatment of control cells with 10 nM-BAY K 8644 leads to no significant increase in aldosterone secretory rate, but treatment of previously exposed cells to angiotensin II leads to an increase in rate from 51 +/- 9 to 130 +/- 11 pg/min per 10(6) cells. This memory effect is time-dependent in two ways: cells must be exposed to angiotensin II for 20 min or more before it is apparent; the longer the time between removal of angiotensin II and the rechallenge, the less effect these agents have on aldosterone secretory rate. When cells are exposed to angiotensin II for 20 min and then treated with [Sar1,Ala8]angiotensin II, a competitive antagonist of angiotensin II action, the aldosterone secretory rate falls to basal with a half time of 5-7 min. If BAY K 8644 is added simultaneously with [Sar1,Ala8]angiotensin II, the secretory rate falls with a halftime of 35-60 min. BAY K 8644 increases Ca2+ influx rate to the same extent in the presence or absence of [Sar1,Ala8]angiotensin II, and does not alter the effect of either angiotensin II or [Sar1,Ala8]angiotensin II on the production of inositol tris-, bis-, or mono-phosphate. In cells treated with 10 nM-angiotensin II for either 20, 30 or 45 min, the extent of phosphorylation of four cellular proteins is increased. If cells treated for 20 min with angiotensin II are then treated with [Sar1,Ala8]angiotensin II, and examined 15 min later (35 min), there is no longer an increase in the extent of phosphorylation of any of the four proteins. If such cells are then treated with 10 nM-BAY K 8644 and re-examined 5 min later (40 min), all four patients show an increase in the extent of phosphorylation.(ABSTRACT TRUNCATED AT 400 WORDS)

Mesocestoides lineatus: trypsin induced development to adult mediated by Ca2+ and protein kinase C.

A mode of action of the inducible treatment with trypsin for the development of Mesocestoides lineatus tetrathyridium to adult was analyzed by administering various agents effective on Ca2+-dependent metabolic pathways in the cells: protein kinase C activators such as a synthetic diacylglycerol, 1-oleoyl-2-acetylglycerol, and a tumor promoting phorbol, 12-O-tetra-decanoyl-phorbol-13-acetate, enhanced the trypsin induced developmental processes. On the contrary, a calmodulin inhibitor, N-(6-aminohexyl)-5-chloro-1-naphthalene sulfonamide, cyclic adenosine 3',5'-monophosphate, and adenylate cyclase activators such as forskolin and cholera toxin, inhibited the triggering action of trypsin. Furthermore, a combined administration of Ca2+ ionophore (A23187) and the phorbol showed a similar effect with trypsin treatment, and sodium taurocholate acted as a potent enhancer like the activators of protein kinase C. These results strongly suggest that the initiation of development to adult in this cestode may be regulated synergistically by Ca2+ and protein kinase C, and that a bile acid may be involved in an activation mechanism of protein kinase C.

Suppression of in vitro human antithyroglobulin antibody secretion by private and cross-reactive anti-idiotypic antibodies.

This is a report on suppression of in vitro human antithyroglobulin antibody secretion in Epstein-Barr (EB) virus-transformed B lymphocytes by private and cross-reactive anti-idiotypic antibodies. Two polyclonal anti-idiotypic antibodies, private anti-Yo-Id and cross-reactive anti-Uc-Id antibodies, were raised in rabbits. Human IgG, lambda antithyroglobulin antibody-producing cell line (Yo3) and human IgG-producing cell line (Yo5), which did not contain the antithyroglobulin activity were established by EB virus transformation of peripheral lymphocytes of patient Yo with chronic thyroiditis. Binding of anti-Yo-Id and anti-Uc-Id antibodies to IgG F(ab')2 antithyroglobulin derived from patient Yo was significantly inhibited by human thyroglobulin and affinity-purified IgG antithyroglobulin of Yo3 cell line. Both private anti-Yo-Id and cross-reactive anti-Uc-Id antibodies suppressed the antithyroglobulin antibody secretion in Yo3 cell line. But these anti-idiotypic antibodies did not suppress the IgG secretion in Yo5 cell line. These results suggested that interactions between idiotype and anti-idiotype may play a role in the immune system of human chronic thyroiditis.

Activation of magnesium-dependent, neutral sphingomyelinase by phosphatidylserine.

The plasma membrane isolated from rat ascites hepatoma, AH 7974 cells was treated with 1% Triton X-100, which resulted in a more than 80% reduction in the phospholipid content of the plasma membrane. The delipidized plasma membrane showed only 18% of the activity of the magnesium-dependent, neutral sphingomyelinase in the untreated plasma membrane. On the addition of acidic phospholipids, especially phosphatidylserine, however, the enzyme activity in the delipidized membrane was markedly restored up to 77% of that in the untreated membrane. It was suggested that, considering the phospholipid composition of the AH 7974 plasma membrane (Koizumi, K. et al. (1977) Cell Struct. Func. 2, 145-153), phosphatidylserine may be a natural activator of neutral sphingomyelinase.

Assignment of A and B types of monoamine oxidase in NCB20 hybrid cells to those of the parental cells by peptide mapping.

The structures of [3H]pargyline-labeled, flavin-containing polypeptides of monoamine oxidase (MAO) from hybrid NCB20 cells, and their parental cells, A/J mouse brain cells and Chinese hamster brain cells, were analyzed and compared by using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and limited proteolysis and one-dimensional peptide mapping in SDS gels. After preincubation of mitochondrial preparations with deprenyl or clorgyline, the flavin-containing polypeptide of type A or type B MAO was selectively labeled with [3H]pargyline. SDS-PAGE of [3H]pargyline-labeled mitochondrial samples revealed that the polypeptide with apparent Mr of 62,000 was associated with type A activity in the three types of cells, and that the polypeptide with apparent Mr of 61,000 or 58,000 was associated with type B activity in Chinese hamster brain cells and NCB20 cells or A/J mouse brain cells, respectively. Chymotrypsin digestion of the [3H]pargyline-labeled polypeptides and the peptide mapping in SDS gels from A/J mouse and Chinese hamster brain cells produced identical map patterns between the two type A MAOs, almost the same map patterns (with the exception of one additional peptide fragment) between the two type B MAOs, and different map patterns between type A and type B MAOs. The results of identical treatments of the [3H]pargyline-labeled polypeptides of MAOs in NCB20 cells showed that type A and type B MAO in NCB20 cells were similar to type A MAO of A/J mouse and Chinese hamster brain cells and to type B MAO of Chinese hamster brain cells.(ABSTRACT TRUNCATED AT 250 WORDS)

Mechanism of cholinergic stimulation of aldosterone secretion in bovine adrenal glomerulosa cells.

The effect of cholinergic stimulation on aldosterone secretion was examined in bovine adrenal glomerulosa cells. Both acetylcholine and carbachol stimulated aldosterone secretion in a dose-dependent manner. Acetylcholine-induced secretion was inhibited by atropine but not by hexamethonium, suggesting that cholinergic agonists act on muscarinic receptors. The mechanisms of cholinergic agonist action were compared with those of angiotensin II. Like angiotensin II, carbachol generated calcium signal in glomerulosa cells. When [3H]inositol-labeled cells were stimulated by carbachol, there was an immediate increase in [3H]inositol trisphosphate, followed by a relatively slow increase in [3H]inositol bisphosphate. Carbachol increased the cytosolic concentration of calcium transiently but not intracellular cAMP. Carbachol caused a rapid 3-fold increase in 45Ca fractional efflux ratio in 45Ca-prelabeled cells both in the presence and absence of extracellular calcium. Carbachol also increased calcium influx; however, carbachol-induced influx was smaller than that of angiotensin II. In a perifusion system, the time course of carbachol-induced aldosterone secretion was biphasic. However, when calcium influx was increased to a value similar to that in angiotensin II-treated cells by combination of carbachol and BAY K-8644, this combination induced a monophasic and sustained secretory pattern. These results indicate that muscarinic cholinergic agonists stimulate aldosterone secretion via the calcium messenger system, and the biphasic secretory response to cholinergic agonist is due to a smaller increase in calcium influx.

Determination of proscillaridin in plasma by radioimmunoassay.

The development of a sensitive and selective radioimmunoassay for determination of proscillaridin in plasma is described. Antiserum against proscillaridin was obtained from guinea pig immunized with an immunogen prepared by conjugating methylproscillaridin, 14 beta-hydroxy-3 beta-[(4-O-methyl-alpha-L-rhamnosyl)oxy]bufa-4,20,20, 22-trienolid to bovine serum albumin. Methyl N-[3-[(14 beta-hydroxybufa-4,20,22-trienolid-3 beta-yl)oxy-carbonyl] propanoyl]-L-[3',5'-125I2] diiodotyrosinate ([125I] SST) was used as a radioactive ligand. [125I] SST and antiserum were added to the recovered sample from plasma using a Bond Elut column. After overnight incubation, the antibody-bound and free [125I] SST were separated using polyethylene glycol. The mean coefficients of variation of intra and interassay at four different plasma concentrations were 4.0 and 5.0%, respectively. The assay was able to determine as little as 125 pg/ml of proscillaridin in plasma by using 1.5 ml of sample. Beagle dogs were orally administered one tablet (Talusin) containing 0.25 mg of proscillaridin. Proscillaridin was rapidly absorbed, exhibiting plasma maximum concentration of 2.06 ng/ml at 20 min. Thereafter, the plasma level declined biphasically with half-lives of 0.6 and 25.4 h.

Effects of fatty acid modification of ascites tumor cells on pulmonary metastasis in rat.

Yoshida sarcoma cells were incubated with each of 4 different saturated and 17 different unsaturated fatty acid methyl and ethyl esters in order to modify the fatty acid composition of the cell membrane, and a possible correlation between the lipid fluidity of the cell membrane and the metastatic efficiency was studied. Almost all the unsaturated fatty acids used were incorporated into the tumor cell membrane, resulting in an increase of fluidity. In contrast, the incorporation of saturated fatty acids brought no change in fluidity. Exogenous treatment of the cells with palmitoleic acid (16:1, cis) induced a remarkable increase of membrane lipid fluidity resulting in easier passage of modified cells through capillary vessels, which was detected in an in vitro perfusion test of the lung. Intravenous injection of the modified cells resulted in fewer metastatic foci in the lung as compared with that of unmodified cells. A similar effect on lung metastasis was also observed in exogenous modification of the cells by linoleic acid (18:2, cis) or linolenic acid (18:3, n-3). On the other hand, although exogenous eicosapentaenoic acid (20:5, n-3) and docosahexaenoic acid (22:6, n-3) treatments induced a slight increase in the lipid fluidity of the cells, intravenous injection of the cells produced a significant increase in their metastatic potential in the lung. Interestingly, the treatment with 20:5 and 22:6 produced increased stickiness of the cells to a glass surface and reduced cellular passage through the vessels as detected by the lung perfusion test. Thus, the present results suggested that selective modification of membrane fatty acid may be a useful method for artificial regulation of the ability of circulating tumor cells to pass through vessels.
