Interaction between human lymphoblastoid interferon and chemotherapeutic agents in vitro.

The combined effect of human lymphoblastoid interferon (HLBI) and anticancer agents on the growth of MOLT-4 was studied in vitro. The interferon showed a strikingly synergistic interaction in combination with aclarubicin, cytosine arabinoside or prednisolone. It was moderately synergistic in combination with adriamycin or 5-fluorouracil and tended to show additive effects with daunorubicin or vincristine. In vitro studies of combination chemotherapy with interferon and anticancer agents should yield valuable information as to the best combination for man.

An adult case of chronic myelogenous leukemia with myeloblastic involvement of the central nervous system.

A 31-year-old female with chronic myelogenous leukemia, who developed myeloblastic involvement of the central nervous system during acute myeloblastic transformation of the disease, was treated with methotrexate intrathecally. The therapy produced prompt clinical response and complete reversal of abnormal cerebrospinal fluid findings. However, the patient expired 10 months following the acute blastic crisis.

A case of chronic neutrophilic leukemia with paraproteinemia (IgG Type lambda and IgA Type K).

An unusual case of chronic neutrophilic leukemia associated with paraproteinemia (IgG Type lambda and IgA Type K) was followed for 3 years. Clinical and hematological findings obtained during the entire course fulfil the previously reported criteria for the diagnosis of chronic neutrophilic leukemia. A marked infiltration of mature neutrophils in the spleen and liver was confirmed by postmortem examination.

[PS (peplomycin-silicone) bullet catheter therapy for bladder tumor: combination of hyperthermic treatment and cepharanthine].

A peplomycin-silicone (PS) bullet containing 130 mg peplomycin, which was steadily released over a long period, was prepared. The PS bullet was then affixed to the top of a 3-way balloon catheter, to form a so called "PS bullet catheter". About 80% of the peplomycin was gradually released over 2 weeks from the PS bullet catheter. Using this device hyperthermic treatment using cepharanthine (biscoclaurin ++ alkaloid) in the irrigant was performed in 15 cases of superficial bladder tumor. Five daily hyperthermic treatments were carried out and all patients retained the PS bullet catheter in the urinary bladder for 2 weeks. Marked tumor regression was noted in 10 of the 15 cases. In addition, partial tumor regression occurred in 4 of the remaining 5 cases. None of these cases had irreversible side effects. This therapy had a good effect on superficial bladder tumor and it is considered that the PS bullet catheter would be useful for cancer therapy.

Isolation and structures of the third major type of carbohydrate units in polysialoglycoproteins from rainbow trout eggs.

The structures of the third major type of oligosialyloligosaccharides obtained by alkali-borohydride treatment of polysialoglycoproteins of the unfertilized eggs of rainbow trout have been determined to be: alpha-L-fucosyl-(1----3)-beta-N-acetyl-D-galactosaminyl-(1----3)-beta-D- galactosyl-(1----4)-beta-D-galactosyl-(1----3)-[[(----8)-alpha-N- glycolylneuraminyl-(2----)]n-(----6)]-N-acetyl-D-galactosaminitol with n = 1 to 4. The proportion of the fucose-containing units in 3 major carbohydrate units present in fish egg polysialoglycoproteins was found to be highly species-specific and only 6% in the eggs of rainbow trout.

[Therapeutic results of 43 cases of adult acute lymphocytic leukemia].

Between July, 1976 and June, 1984, 43 adults with acute lymphocytic leukemia were treated with a V(V') P [vincristine (vindesine), prednisolone] followed by DV(V')MP [daunomycin, vincristine (vindesine), 6-mercaptopurine, prednisolone] and V(V')AP [vincristine (vindesine), 1-asparaginase, prednisolone] regimen. They were all previously untreated and aged 15 and over. Complete remission (CR) was attained in 41.9% of cases by V(V')P alone, the CR rate being increased up to 62.8% by the sequential administration of DV(V')MP, and to 74.4% by the further administration of V(V')AP. The median duration of CR was 5.6 months; it was 10.9 months for patients with the maintenance therapy and 1.3 months for patients without it. For patients who achieved CR, the median survival time (MST) was 21.3 months compared to 2.7 months for those who failed to achieve CR. As for the maintenance therapy, MST was 22.3 months for patients who received it, and 9.5 months for patients who did not. Factors associated with significantly lower rate of CR were: lymphadenopathy and hepatomegaly and/or splenomegaly. The CR rate was somewhat lower for patients aged over 20 and with hyperleukocytosis (above 40,000/cmm). Shorter remissions tended to be associated with ages over 20 and with hepatomegaly and/or splenomegaly. Patients who obtained CR by the sequential administration of the V(V')AP regimen showed somewhat shorter CR duration compared with to those who obtained CR by V(V')P alone and by the sequential administration of DV(V')MP. Survival was significantly shorter for patients who failed to achieve CR, with B-ALL and with hyperleukocytosis. Shorter survival was also observed among patients with ages above 60 compared to those with ages below 20.

Voice onset in pathologic speakers.

The time interval between the release and beginning of vocal cord vibration in word-initial /p/ and /b/ was measured. The data from subjects with laryngeal pathology are described. /b/ showed larger values compared with the data from normals. It is suggested that such a test of vocal initiation could provide a useful measure for the evaluation of laryngeal dysfunction.

Adult T-cell leukemia. Chromosome analysis of 15 cases.

Chromosome studies was conducted on 15 patients with adult T-cell leukemia. Cells with chromosomal abnormality were seen in 14 of the 15 patients. The modal chromosome number was near diploid range in all the patients. The most common abnormality was 14q+ marker chromosome and partial deletion of the long arm of chromosome 6, i.e., 6q-, which were seen in eight and seven cases, respectively. Donor chromosomes involved in the 14q+ marker chromosome varies, i.e., Yq, #5p, #5q, #9q, #10q or #12q, except for two patients whose donor chromosome origins were unable to determine. The break point in 14q+ marker chromosome was band at q32. The 6q- chromosome was due to a deletion in one patient and interstitial deletion in six patients. A 14q- chromosome having break point at q24 was found in one patient and duplication of Yq chromosome in two patients. In addition, four patients showed a 5q- chromosome or a 9q- chromosome which was due to a translocation or deletion. The significance of these chromosome abnormalities was discussed.

Adult T-cell leukemia occurring in mother and son.

We report two cases of adult T-cell leukemia in which the disease developed in a mother, aged 62 years, and her son, aged 41 years, less than four months apart. Both mother and son showed abnormal karyotypes and high titers of adult T-cell leukemia-associated antibody.

Superoxide anion (O2-) production by neutrophils in refractory anemia with excess of blasts.

The O2- production by neutrophils was examined in 4 cases of refractory anemia with excess of blasts (RAEB) in order to evaluate the possible causes of enhanced susceptibility to infection and to gain some informations on the differentiation of neutrophils in this hematological disorder. In three of the four RAEB cases there was little O2- production by neutrophils, in addition to there being morphological anomalies of the neutrophils such as a Pelger-Hüet-like anomaly, granular deficiency and binucleated cells. These results suggest that the impairment of O2- production by neutrophils in RAEB is one of the possible causes of susceptibility to infection and also suggest that the differentiation of neutrophils in this hematological disorder is faulty. The estimation of O2- production by neutrophils may be a useful diagnostic method for preleukemia.

[Clinical evaluation of piperacillin in the urological field].

The sensitivity of 249 etiological bacterial strains causing inflammation isolated from patients with infections of the urogenital tract who were under treatment on an in-patient basis in our Department was examined by the Disc (3 concentration) assay method. Piperacillin (PIPC) was employed for prophylactic treatment in 18 patients against postoperative infections and for therapeutic treatment in 3 patients against complicated infections of urogenital tracts. The results of these studies are reported along with some discussion on the utility of PIPC in the urological field. The frequency of detection of bacteria isolated from the patients on their admission into our hospital was in the decreasing order of Streptococcus Sp., Proteus Sp. and E. coli Pseudomonas aeruginosa and Klebsiella Sp. also were noted in some of the patients. Pseudomonas aeruginosa, nonfermentative Gram's-negative bacilli (NF- GNB ), and Streptococcus Sp. were detected in many of the patients who had had an operation or who had been in hospital for a long time. The sensitivity of E. coli, Proteus Sp., Streptococcus Sp. and Pseudomonas aeruginosa was invariably more than 80%, while that of NF- GNB was more than 50%. PIPC was used for prophylactic treatment of patients against infections at the postoperative stage in the urogenital field. Out of 18 cases, it proved to be markedly effective in 14 cases and moderately effective in 4 cases. There was no poorly effective case. PIPC was employed for treatment in 5 cases complicated infections of urogenital tracts. There was no markedly effective case, and 2 moderately effective and 3 poorly effective cases resulted according to the UTI efficacy evaluation standards However, its bacteriological efficacy could be identified in all the treated cases. Therefore, PIPC is a valuable prophylactic antibiotic agent against infections after an operation in the urological field. It may also prove valuable in the therapeutic treatment of complicated infections in the urogenital tracts. PIPC has a relatively large margin of safety in terms of side effects as in the case of conventional penicillin group antibiotics.

[Phase II study of mitoxantrone for hematologic malignancies].

A phase II study of mitoxantrone (MIT) was performed in 21 patients with hematologic malignancies refractory to combination chemotherapy including anthracyclines. MIT was administered intravenously at doses of 8 to 13 mg/m2 on day 1 for 12 malignant lymphoma patients, and 1 to 3.3 mg/m2 on day 1 through 5 for 7 acute leukemia patients and 2 malignant lymphoma patients. Four malignant lymphoma patients (2 each of Hodgkin's disease and non-Hodgkin's lymphoma) achieved partial response lasting 19, 14, 8+, and 4 weeks, respectively. Although no definite response was obtained in acute leukemia patients, a marked cytoreduction was observed in 2 patients. Myelosuppression was a major toxicity, however, life-threatening toxicities were not observed in this study.

Effect of pentazocine and related compounds on the lipid composition of Ehrlich ascites tumor cells.

The effect of pentazocine and its related compounds on the lipid composition of Ehrlich ascites tumor cells was examined. Ehrlich tumor cells were suspended in Hanks balanced salt solution (pH 7.4) supplemented with 2% bovine albumin (2 x 10(6) cells/ml) and incubated at 37 degrees C for 120 min with and without 10(-3) M of pentazocine, morphine or dihydrocodeine. After incubation, the tumor cell lipids were extracted with chloroform-methanol (2:1, v/v), and they were analyzed quantitatively by the dichromate reduction procedure of Amenta. The tumor cells treated with pentazocine contained lower levels of triglycerides and cholesterol esters as compared with the tumor cells incubated alone. The amounts of triglycerides and cholesterol esters in the tumor cells treated with and without pentazocine were about 82 and 23 mg/10(10) cells and 182 and 55 mg/10(10) cells, respectively (P less than 0.01), whereas there was only a slight difference in the contents of these neutral lipids between the tumor cells treated with and without (10(-3) M morphine or dihydrocodeine. In addition, the fatty acid pattern of triglycerides and cholesterol esters from the pentazocine-treated tumor cells differed markedly from that of the corresponding lipids from the tumor cells incubated alone.
