The amino acid sequence of a newly isolated pentapeptide, neo-kyotorphin from bovine brain was synthetically verified to be Thr-Ser-Lys-Tyr-Arg corresponding to the C-terminal portion of hemoglobin alpha-chain. The synthetic neo-kyotorphin showed the dose-dependent analgesia in mice which was approximately equal to that of Leu-enkephalin.
A novel analgesic peptide, isolated from the bovine brain, was found to contain the Tyr-Arg (kyotorphin) unit at the C-terminal portion. This peptide, in the methanol-soluble fraction, was isolated by gel filtration and cation exchange chromatography. This peptide was termed "neo-kyotorphin" and the amino acid sequence is Thr-Ser-Lys-Tyr-Arg. The synthetically prepared neo-kyotorphin proved to have a dose-dependent analgesic effect in mice. The median analgesic dose, ED50 of neo-kyotorphin was 195 nmol/mouse (intracisternal injection), with the tail pinch test, and the potency was approximately equal that of Leu-enkephalin.

We conducted a randomized double blind comparison of cefamandole (CMD) and cefmetazole (CMZ) in the treatment of 193 patients with complicated urinary tract infections. The patients received 1 gram of CMD or CMZ twice a day intravenously by drip infusion over 1 hour for 5 days. Pretreatment urinary leukocyte counts and urinary bacterial counts were at least 5 cells/hpf and 10(4) bacteria/ml, respectively. Each patient was randomly allocated either to CMD or CMZ group. There were 93 patients in CMD group and 100 patients in CMZ group. Clinical efficacy was evaluated based on the effect of treatment on bacteriuria and pyuria according to the criteria set by the UTI Committee, Japan. The response to CMD treatment was excellent in 18 cases (19.4%), moderate in 38 cases (40.9%) and poor in 37 cases (39.8%) with an overall effectiveness of 60.2%, whereas the response to CMZ was excellent in 19 cases (19.0%), moderate in 40 cases (40.0%) and poor in 41 cases (41.0%) with an overall effectiveness of 59.0%. No statistical significant difference was found between 2 treatment groups. Comparison of the bacteriological response between 2 groups showed that the eradication rate for strains of Gram-positive cocci were significantly higher in those patients treated with CMD. Gram-negative rods were eradicated from 68.4% of cases treated with CMD, and 78.0% of those with CMZ, but the difference was not significant. Adverse reactions were observed in 3 patients receiving CMD 1 case each of diarrhea, eruption and epigastric pain. Abnormality in laboratory tests was found in 6 patients in each treatment group. The results indicate that CMD is effective, safe and useful in the treatment of patients with complicated urinary tract infections, and its efficacy, safety and usefulness are comparable with those of CMZ.

Anaerobic power output was measured by the staircase climb test in 14 obese, 16 lean, and 21 ordinary men aged from 18--22 years. Fat storage rate (%fat) was estimated by densitometry. The obese group ranked highest with an average power output of 1,012 W. This value was significantly higher than those of the other two groups, 890 W for lean subjects and 855 W for ordinary subjects. The power output per kilogram of lean body mass of the obese group was the highest also. However, the vertical velocity was the lowest although the difference among the three average values was not statistically significant. To investigate the effect of excess fat, eight non-obese subjects engaged in an added-weight experiment. The value obtained was almost the same as for the obese group. The added weights made the vertical velocity decrease but the power output increase. Consequently, it was obvious that the excess fat of an obese man played a role only as an inert mass in the power output measurement. A significantly higher power output of the obese group might be due to more excess fat, and obesity itself was an advantage in the staircase climb test.
1. The present experiments were designed to further characterize the specific [3H]substance P (SP) binding to membrane fractions from rabbit brain. 2. The specific binding was resistant to treatment with proteolytic enzymes whereas phospholipase A(0.7 unit/ml), C(0.025 unit/ml) and D(6.2 unit/ml) reduced the specific binding without decreasing total binding. Neuraminidase (0.01 unit/ml) decreased total binding as well as the specific binding. 3. High concentration of Triton X-100 reduced the specific binding whereas deoxycholate, at 0.05%, reduced both total and the specific binding. 4. Na+ (50--200 mM) reduced the specific binding, depending on the concentrations employed, whereas K+ decreased the specific binding at 10 and 100 mM. The binding was reduced considerably by 100 mmol/l of CaCl2. 5. From these results it is suggested that the specific [3H]SP binding sites in membrane fractions from rabbit brain could be phospholipids.
